Com o objetivo de desenvolver um modelo de hemodiálise (HD) em cães, foram estudados 18 animais, sem raça definida, machos, clinicamente sadios, com peso corporal variando entre sete e 14 kg. O acesso vascular foi obtido através de implantação do cateter de duplo lúmen em veia jugular externa. As sessões de HD, em número de cinco por animal, com até três horas de duração, foram realizadas em hemodialisadora de sistema proporcional com ultrafiltração (UF) controlada, com solução dialisante padrão e tampão bicarbonato. A UF foi ajustada para HD isovolêmica, utilizou-se perfil de sódio, e para anticoagulação heparina sódica. Os animais foram mantidos anestesiados com cloridrato de levomepromazina e propofol. Foram avaliados dados hematológicos, bioquímicos, hemogasometria, pressão arterial sistêmica e tempo de coagulação ativado. Foi observada diminuição do número global de hemácias, volume globular, hemoglobina e leucócitos. Em relação aos exames bioquímicos, houve manutenção nos níveis de sódio sérico, e quanto à hemogasometria, a manutenção da SO2. A pressão arterial sistêmica manteve-se constante. Os resultados obtidos no presente trabalho permitiram concluir que foi possível o desenvolvimento do modelo proposto e mostrou que a HD em cães é um método viável e seguro, que poderá contribuir para o tratamento clínico da insuficiência renal nesta espécie.

PURPOSE: To investigate the sedative and clinical effects of the pharmacopuncture with xylazine, compared to the conventional dose of a intramuscular injection in dogs.METHODS: Twelve dogs were randomly distributed in two groups of six animals and treated as follows: control group (X-IM): 1mg kg(-1) of xylazine given intramuscularly (IM); pharmacopuncture group (X-Yintang): 0.1mg kg(-1) of xylazine diluted to 0.5 mL of saline injected into the Yin Tang acupoint. Heart rate, cardiac rhythm (ECG), systolic arterial blood pressure (SABP), respiratory rate (RR), rectal temperature (RT), blood glucose concentration, degree of sedation and adverse effects were evaluated.RESULTS: Sedative effect was observed in both groups. The degree of sedation was greater in X-IM only at 15 min when compared with X-Yintang group. Cardiovascular established was observed in X-Yintang group, while marked reduction in the HR and increased incidence of ECG abnormalities were detected in X-IM. In both treatment groups, minimal changes were observed in relation to SABP, RR, RT and blood glucose. High incidence (66%) of vomiting was observed in X-IM, while this adverse effect was absent in X-Yintang.CONCLUSION: Pharmacopuncture with xylazine induced clinically relevant sedative effects in dogs, with the advantage of reduction of undesirable side effects associated with alpha(2)-agonists, including bradycardia, cardiac arrhythmias, and emesis.

Objective: The present study aimed to investigate the influence of methadone on cardiorespiratory parameters, electrocardiogram and clinical sedation in dogs. Further possible side effects are reported.Study designProspective experimental cross-over study.DogsEight, 1-4-year-old, various breeds of dogs of both genders weighing 9-36kg.MethodsEach dog was treated three times: methadone 0.3mgkg(-1) (M0.3), 0.5mgkg(-1) (M0.5) and 1.0mgkg(-1) (M1.0) intramuscularly. Respiratory rate, heart rate and arterial blood pressure were recorded as well as electrocardiographic evaluation of lead II. Clinical sedation in each treatment received a score (0-3) after drug administration and at 30minute intervals until scores and measurements returned to baseline values.ResultsA significant decrease in heart rate was seen with each dose of methadone and bradycardia (HR<60bpm) was noted in a few dogs at each dose. A clinically significant arrhythmia occurred in one dog at 1mgkg(-1) that required reversal with butorphanol. There was no significant difference in SAP, MAP and DAP between treatments. Some side effects such as salivation, defecation, vocalization and panting, after administration of methadone were observed. There were no differences in mean values of heart rate, P-wave and QRS complex duration and QT interval between treatments.Conclusion and clinical relevanceMethadone administration was associated with panting and a decrease in heart rate at all doses tested in this study. The cardiac rhythm should be monitored carefully in dogs when methadone is administered on its own, especially at higher doses.

PURPOSE: To measure the change in the minimum alveolar concentration of isoflurane (EtISO) associated with epidural nalbuphine and the postoperative analgesic requirements in dogs after ovariohysterectomy.METHODS: Twenty four healthy female dogs were randomly assigned to receive saline or nalbuphine at 0.3 or 0.6 mg/kg (n=8 for each group) administered via lumbosacral epidural catheter introduced cranially into the epidural canal. Changes in heart and respiratory rates and arterial blood pressure during surgery were recorded along with the corresponding EtISO. Immediately after tracheal extubation, analgesia, sedation, heart rate, respiratory rate, and arterial blood pressure were measured at predetermined intervals and every 60 min thereafter until the first rescue analgesic.RESULTS: A significant decrease in EtISO was associated with epidural nalbuphine at 0.3 mg/kg (26.3%) and 0.6 mg/kg (38.4%) but not with saline in ovariohysterectomized dogs. In the postoperative period, VAS and Colorado analgesic scores were lower for the dogs that received the higher nalbuphine dose, which only required supplemental analgesia 10 h following its administration, compared with dogs that received the lower dose.CONCLUSION: Epidural nalbuphine significantly reduces the intra-operative isoflurane requirement and provides prolonged postoperative analgesia after ovariohysterectomy in dogs.

Maternal undernutrition affects the foetal development, promoting renal alterations and adult hypertension. The present study investigates, in adult male rats, the effect of food restriction in utero on arterial blood pressure changes (AP), and its possible association with the number of nephrons, renal function and angiotensin II (AT1R/AT2R), glucocorticoid (GR) and mineralocorticoid (MCR) receptors expression. The daily food supply to pregnant rats was measured and one group (n=5) received normal quantity of food (NF) while the other group received 50% of that (FR50) (n=5). The AP was measured weekly. At 16 weeks of life, fractionator’s method was used to estimate glomeruli number in histological slices. The renal function was estimate by creatinine and lithium clearances. Blood and urine samples were collected to biochemical determination of creatinine, sodium, potassium and lithium. At 90th and 23rd days of life, kidneys were also processed to AT1R, AT2R, GR and MCR immunolocalization and for western blotting analysis. FR50 offspring shows a significant reduction in BW (FR50: 5.67 ± 0.16 vs. 6.84 ± 0.13g in NF, P<0.001) and increased AP from 6th to 12nd week (6thwk FR50: 149.1 ± 3.4 vs. 125.1 ± 3.2mmHg in NF, P<0.001and, 12ndwk FR50: 164.4 ± 4.9 vs. 144.0 ± 3.3 mmHg in NF, P=0.02). Expression of AT1R and AT2R were significantly decreased in FR50 (AT1, 59080 ± 2709 vs. 77000 ± 3591 in NF, P=0.05; AT2, 27500 ± 95.50 vs. 67870 ± 1509 in NF, P=0.001) while the expression of GR increased in FR50 (36090 ± 781.5 vs. 4446 ± 364.5 in NF, P=0.0007). The expression of MCR did not change significantly. We also verified a pronounced decrease in fractional urinary sodium excretion in FR50 offspring (0.03 ± 0.02 vs. 0.06 ± 0.04 in NF, p=0.03). Introduction: Physical exercise are related to high oxygen consumption, leading to increase on reactive oxygen species (ROS) generation and subsequent oxidative stress. Concomitantly, physical training can improve the antioxidant defense systems, reducing the deleterious activity of ROS. The yerba mate (Ilex paraguariensis) has several bioactive compounds in its composition, providing important antioxidant activity in improving defense systems and reducing the damage caused by ROS. Few studies related to yerba mate with antioxidant effects during exercise. Objective: Evaluate whether the consumption-based drink yerba mate (Ilex paraguariensis) is able to increase the total antioxidant performance (TAP) after an exhaustive test on a treadmill. Methods: The sample counted with 15 female soccers players from Botucatu-SP female soccer team with a mean age of 22.1 ± 4.2 years. For laboratory tests , it was evaluated: triglycerides (TG), total cholesterol (TC) and fractions, glucose and gamma-GT were dosed by dry chemistry (Vitros® System, Johnson & Johnson). LDL-cholesterol was obtained by Friedwald formula. Total antioxidant performance (TAP) was obtained by the method of fluorescence assay for the measurement of plasma. Weight, height and body mass index (IMC) were measured, percentage of body fat was obtained by bioeletrical impedance analysis (Biodinâmics, modelo 450, USA). Arterial blood pressure was checked by auscultatory method and cardiorespiratory fitness was determined by ergoespirometric test (Ramp Protocol). Maltodextrin was supplemented (30 g in 400 ml) 30 minutes after M0 with placebo (400 ml) or mate (5 g in 400 ml of water). Physical activity is associated with lower cardiovascular disease risk factors, being cardiorespiratory fitness a major component of physical activity health related. Body fatness and sarcopenia are related to sedentary lifestyle leading to proinflammatory stress and lower cadiorespiratory capacity. This study aimed correlates C-reactive protein with cardiorespiratory fitness, analyzing the influences of anthropometrics variables and metabolic syndrome (MS) presence. the cross-sectional retrospective study included baseline data of 194 adults (62 male and 132 female), 53,74 ± 8,77 years, clinically and ethically selected for a lifestyle modification program. Total cholesterol (TC) and cholesterol lipoprotein fractions, triglycerides (TG) and glucose was dosed by dry chemistry (Vitros® system, Johnson & Johnson). Blood leukocytes was quantified by automatic cell counter (Coulter ABX®, Horiba). LDL-cholesterol was obtained by Friedwald formula. Serum ultrasensitive C-reactive protein (US-CRP) was accessed by the immunochemoluminescence method (Immulite 2000®, DPC Medlab). Weight, height, body mass index (BMI) and waist circumference (WC) were measured. Muscular mass and fat mass were obtained by bioelectrical impedance analysis (impedancemeter Quantum BIA-101Q®, Clinton Township). Arterial blood pressure was checked by auscultatory method and cardiorespiratory fitness was determined by ergoespirometric test (Balke protocol). The metabolic syndrome was diagnosed according NCEP – ATP III (2001), following recommendations of American Diabetes Association (2004). Pearson’s correlation crude and adjusted for confounders variables with p<0,05. The prevalence of MS was 30.4%. Plumbism is considered the oldest occupational disease. Among the pathophysiological effects associated with lead (Pb) are cardiovascular disorders. Many diseases that develop later in life are determined during the early stages of life, under the influence of exposure and preferred diet of the mother. Still, one should consider that many environmental contaminants at levels not harmful can determine pathophysiological processes if physical or chemical stressors and/or pathological conditions are present. In this context, the intrauterine malnutrition may represent an additional risk factor in exposure to Pb during pregnancy and lactation. For these reasons, the objective of this study was to evaluate the cardiovascular risk of weaned rats that have suffered perinatal exposure to Pb and intrauterine malnutrition, alone or in combination. After mating, female rats were divided into control (ctrl, ad libitum), food restriction (RA, the same diet 50% of consumption in the control group during pregnancy), exposed to Pb (500 ppm Pb in drinking water during pregnancy and lactation) and association (As, received the last two procedures in combination). Cumulative concentration-effect curves (CECs) to CaCl2 and noradrenaline (NA) were obtained in rings with and without endothelium of the same thoracic aorta from male weaned rats (23-25 days old). Maternal weight, litter weight, weight and number of pups at birth, anogenital distance, arterial blood pressure (ABP) and weight of tissues (kidney, liver, aorta, left ventricle) were evaluated.
