Systemic lupus erythematosus (SLE) is a prototypical autoimmune disease characterized by polyclonal B cell activation and by the production of anti-double-stranded (ds) DNA antibodies. Given the inhibitory effects of IL-12 on humoral immune responses, we investigated whether IL-12 displayed such an activity on in vitro immunoglobulin production by SLE PBMC. Spontaneous IgG, IgG1, IgG2, IgG3 and IgM antibody production was dramatically reduced by addition of IL-12. These results were confirmed by Elispot assays detecting IgG- and anti-dsDNA-secreting cells. While IL-6 and TNF titres measured in PBMC supernatants were not modified by addition of IL-12, interferon-gamma (IFN-gamma) titres were up-regulated and IL-10 production down-regulated. Since addition of IFN-gamma did not down-regulate immunoglobulin production and since the inhibitory activity of IL-12 on immunoglobulin synthesis was not suppressed by anti-IFN-gamma antibody, we concluded that the effect of IL-12 on immunoglobulin production was not mediated through IFN-gamma. Our data also argue against the possibility that down-regulation of endogenous IL-10 production was responsible for the effect of IL-12. Thus, inhibition of IL-10 production by IFN-gamma was not accompanied by inhibition of immunoglobulin production, and conversely, restoration of IL-10 production by anti-IFN-gamma antibody did not suppress the inhibitory activity exerted by IL-12 on immunoglobulin production. Taken together, our data indicate that reduction of excessive immunoglobulin and anti-dsDNA antibody production by lupus PBMC can be achieved in vitro by IL-12, independently of IFN-gamma and IL-10 modulation.

Prostate cancer (PC) is the second leading cause of cancer death in men. Recent reports suggest that excess of nutrients involved in the one-carbon metabolism pathway increases PC risk; however, empirical data are lacking. Veteran American men (272 controls and 144 PC cases) who attended the Durham Veteran American Medical Center between 2004-2009 were enrolled into a case-control study. Intake of folate, vitamin B12, B6, and methionine were measured using a food frequency questionnaire. Regression models were used to evaluate the association among one-carbon cycle nutrients, MTHFR genetic variants, and prostate cancer. Higher dietary methionine intake was associated with PC risk (OR = 2.1; 95%CI 1.1-3.9) The risk was most pronounced in men with Gleason sum <7 (OR = 2.75; 95%CI 1.32- 5.73). The association of higher methionine intake and PC risk was only apparent in men who carried at least one MTHFR A1298C allele (OR = 6.7; 95%CI = 1.6-27.8), compared to MTHFR A1298A noncarrier men (OR = 0.9; 95%CI = 0.24-3.92) (p-interaction = 0.045). There was no evidence for associations between B vitamins (folate, B12, and B6) and PC risk. Our results suggest that carrying the MTHFR A1298C variants modifies the association between high methionine intake and PC risk. Larger studies are required to validate these findings.

En este trabajo se presenta el resultado obtenido del análisis de un proceso de razonamiento inductivo desarrollado por 12 estudiantes de secundaria en un contexto de resolución de problemas. Se plantea un problema, en el transcurso de una entrevista, que consiste en determinar el número máximo de regiones que se obtienen al trazar rectas sobre un plano. Durante la resolución del problema los estudiantes, y a través del dialogo con el entrevistador, han de explicar y justificar sus decisiones. Centrándonos en el trabajo de Pólya y en otras investigaciones previas relacionadas sobre este tema, se define un sistema de categorías mediante las cuales se organizan los datos para su análisis.

Esta investigación presenta la puesta en práctica de una propuesta pedagógica para apoyar la enseñanza del Cálculo mediante la resolución de problemas a nivel preuniversitarioen Costa Rica. El proyecto tiene su origen en las dificultades que presentan los estudiantes en la comprensión de conceptos básicos de Cálculo, específicamente el de límite y derivada. Esta experiencia se fundamentó en la elaboración de una “situación problema” que provocó un conflicto intelectual en los estudiantes, mientras que el docente fungió como mediador y aprovechó los descubrimientos hechos por los estudiantes para fundamentar teóricamente los diferentes conceptos luego de la aplicación de la propuesta. Los resultados obtenidos son muy positivos y justifican la necesidad de un cambio en las estrategias metodologías utilizadas para enseñar el Cálculo. Sin embargo, es necesario un acercamiento de los docentes hacia la Teoría de Resolución de problemas para aplicar con éxito este tipo de actividades.

Fue el último en poseerla quien, instantes antes de perderla para siempre y movido por una presunción ya del todo instintiva, bautizó la ciudad con el nombre de Memoria grafiando esa palabra en las fachadas de las avenidas. Presumió que en un futuro, muy lejano quizá pero alcanzable, y a pesar de los trazos ya inseguros de su escritura afectada, alguien podría leerla y recobrar la lucidez.

We study two marked point process models based on the Cox process. These models are used to describe the probabilistic structure of the rainfall intensity process. Mathematical formulation of the models is described and some second-moment characteristics of the rainfall depth, and aggregated processes are considered. The derived second-order properties of the accumulated rainfall amounts at different levels of aggregation are used in order to examine the model fit. A brief data analysis is presented.
