Background: The aim of this study was to assess clinical and inflammatory markers in nonalcoholic fatty liver disease (NAFLD) in postmenopausal women with metabolic syndrome.Methods: This cross-sectional study included 180 Brazilian women (age >= 45 years and amenorrhea >= 12 months). Metabolic syndrome was diagnosed by the presence of at least three of the following indicators: Waist circumference (WC) > 88 cm, triglycerides (TGs) >= 150 mg/dL, high-density lipoprotein (HDL) < 50 mg/dL; blood pressure >= 130/85 mmHg; and glucose >= 100 mg/dL. NAFLD was diagnosed by abdominal ultrasound. Participants were divided into three groups: Metabolic syndrome alone (n = 53); metabolic syndrome + NAFLD (n = 67); or absence of metabolic syndrome or NAFLD (control, n = 60). Clinical, anthropometric, and biochemical variables were quantified. The inflammatory profile included adiponectin, interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-alpha). Data were submitted to statistical analysis using a Tukey test, analysis of variance (ANOVA), chi-squared, Pearson correlation, and logistic regression (odds ratio, OR).Results: Women with metabolic syndrome + NAFLD, abdominal obesity, high glucose, and insulin resistance by HOMA-IR were compared to women with metabolic syndrome alone and controls (P < 0.05). High values of IL-6 and TNF-alpha and low values of adiponectin were observed among women with metabolic syndrome alone or metabolic syndrome + NAFLD when compared to controls (P < 0.05). In multivariate analysis, the variables considered as risk of NAFLD development were: High systolic blood pressure (SBP) [(OR 1.02, 95% confidence interval (CI) 1.0-1.04]; large WC (OR 1.07, 95% CI 1.01-1.13); insulin resistance (OR 3.81, 95% CI 2.01-7.13); and metabolic syndrome (OR 8.68, 95% CI 3.3-24.1). Adiponectin levels reduced NAFLD risk (OR 0.88, 95% CI 0.80-0.96).Conclusion: In postmenopausal women, metabolic syndrome, abdominal obesity, and insulin resistance were risk markers for the development of NAFLD, whereas higher adiponectin values indicated a protection marker.

The over-production of reactive oxygen species (ROS) can cause oxidative damage to a large number of molecules, including DNA, and has been associated with the pathogenesis of several disorders, such as diabetes mellitus (DM), dyslipidemia and periodontitis (PD). We hypothesise that the presence of these diseases could proportionally increase the DNA damage. The aim of this study was to assess the micronucleus frequency (MNF), as a biomarker for DNA damage, in individuals with type 2 DM, dyslipidemia and PD. One hundred and fifty patients were divided into five groups based upon diabetic, dyslipidemic and periodontal status (Group 1 - poor controlled DM with dyslipidemia and PD; Group 2 - well-controlled DM with dyslipidemia and PD; Group 3 - without DM with dyslipidemia and PD; Group 4 - without DM, without dyslipidemia and with PD; and Group 5 - without DM, dyslipidemia and PD). Blood analyses were carried out for fasting plasma glucose, HbA1c and lipid profile. Periodontal examinations were performed, and venous blood was collected and processed for micronucleus (MN) assay. The frequency of micronuclei was evaluated by cell culture cytokinesis-block MN assay. The general characteristics of each group were described by the mean and standard deviation and the data were submitted to the Mann-Whitney, Kruskal-Wallis, Multiple Logistic Regression and Spearman tests. The Groups 1, 2 and 3 were similarly dyslipidemic presenting increased levels of total cholesterol, low density lipoprotein cholesterol and triglycerides. Periodontal tissue destruction and local inflammation were significantly more severe in diabetics, particularly in Group 1. Frequency of bi-nucleated cells with MN and MNF, as well as nucleoplasmic bridges, were significantly higher for poor controlled diabetics with dyslipidemia and PD in comparison with those systemically healthy, even after adjusting for age, and considering Bonferroni's correction. Elevated frequency of micronuclei was found in patients affected by type 2 diabetes, dyslipidemia and PD. This result suggests that these three pathologies occurring simultaneously promote an additional role to produce DNA impairment. In addition, the micronuclei assay was useful as a biomarker for DNA damage in individuals with chronic degenerative diseases.

Objective To evaluate the prevalence and risk factors of non-alcoholic fatty liver disease (NAFLD) in postmenopausal women.Methods A cross-sectional study was carried involving 188 women (age >= 45 years and amenorrhea >= 12 months) attending the outpatient unit in south-eastern Brazil. Exclusion criteria were liver disease (hepatitis B and C, cholestatic disease, liver insufficiency), use of drugs that affect liver metabolism; alcoholics; AIDS or cancer history; and morbid obesity. NAFLD was diagnosed by abdominal ultrasound. Clinical, anthropometric (body mass index, waist circumference) and biochemical variables were measured.Results Of the 188 women, 73 (38.8%) had NAFLD. Blood pressure, waist circumference, body mass index, LDL cholesterol, triglycerides and glucose were significantly higher in NAFLD patients when compared with women without NAFLD (control group) (p < 0.05). HOMA-IR values indicated insulin resistance only in the NAFLD group (6.1 +/- 4.6 vs. 2.4 +/- 1.4 in control group, p < 0.05). Metabolic syndrome was detected in 93.1% of the women affected by NAFLD, and 46.1% of the control group (p < 0.05). In multivariate analysis, adjusted for age and weight, the variables considered at risk for the development of NAFLD, were: high waist circumference (odds ratio (OR) 1.07, 95% confidence interval (CI) 1.01-1.13), insulin resistance (OR 3.81, 95% CI 2.01-7.13), and presence of metabolic syndrome (OR 8.68, 95% CI 3.3-24.1).Conclusion NAFLD showed a high prevalence among postmenopausal women. The presence of metabolic syndrome, abdominal obesity and IR were indicators of risk for the development of NAFLD.

Hemolysis is the main cause of biochemical analysis rejection's in veterinary laboratories, however the relative error caused by hemoglobin on serum biochemical profile has not been properly established on several species. In order to establish criteria for aproval and rejection of hemolyzed samples for serum biochemical tests, the hypothesis that hemolysis causes biochemical changes in canine, cattle and horses and that laboratorial error depends on species and hemolysis degree was tested. Thus, non-hemolyzed serum was contaminated with crescent hemoglobin levels and using commercial routine reagents, the serum concentrations of uric acid, albumin, cholesterol, triglycerides and urea, besides the activity of ALT, AST, CK and GUT were quantified in triplicate samples. The relative error was calculated by the comparison between hemolyzed and non-hemolyzed samples. Hemolys is did not cause significant error on the albumin determination in all three species, AST in canine and cattle, ALT in horses, UK and cholesterol in canine. There was a linear increase on uric acid levels in horses and cattle, triglycerides in all three species. A linear increase in serum urea in all species serum, UK and cholesterol in cattle and cholesterol in horses was observed. Serum AST activity on equine serum and ALT in cattle decreased linearly due to hemolysis. It was concluded that hemolysis promotes changes in canine, equine and bovine serum chemistry profile, however the laboratorial error not necessarily compromises the diagnosis in all cases, because the changes depends on species and degree of in vitro hemolysis.

Introduction: Physical exercise are related to high oxygen consumption, leading to increase on reactive oxygen species (ROS) generation and subsequent oxidative stress. Concomitantly, physical training can improve the antioxidant defense systems, reducing the deleterious activity of ROS. The yerba mate (Ilex paraguariensis) has several bioactive compounds in its composition, providing important antioxidant activity in improving defense systems and reducing the damage caused by ROS. Few studies related to yerba mate with antioxidant effects during exercise. Objective: Evaluate whether the consumption-based drink yerba mate (Ilex paraguariensis) is able to increase the total antioxidant performance (TAP) after an exhaustive test on a treadmill. Methods: The sample counted with 15 female soccers players from Botucatu-SP female soccer team with a mean age of 22.1 ± 4.2 years. For laboratory tests , it was evaluated: triglycerides (TG), total cholesterol (TC) and fractions, glucose and gamma-GT were dosed by dry chemistry (Vitros® System, Johnson & Johnson). LDL-cholesterol was obtained by Friedwald formula. Total antioxidant performance (TAP) was obtained by the method of fluorescence assay for the measurement of plasma. Weight, height and body mass index (IMC) were measured, percentage of body fat was obtained by bioeletrical impedance analysis (Biodinâmics, modelo 450, USA). Arterial blood pressure was checked by auscultatory method and cardiorespiratory fitness was determined by ergoespirometric test (Ramp Protocol). Maltodextrin was supplemented (30 g in 400 ml) 30 minutes after M0 with placebo (400 ml) or mate (5 g in 400 ml of water). Statistical analysis: ANOVA for repeated measures followed by Tukey's test set, p<0.05. Physical activity is associated with lower cardiovascular disease risk factors, being cardiorespiratory fitness a major component of physical activity health related. Body fatness and sarcopenia are related to sedentary lifestyle leading to proinflammatory stress and lower cadiorespiratory capacity. This study aimed correlates C-reactive protein with cardiorespiratory fitness, analyzing the influences of anthropometrics variables and metabolic syndrome (MS) presence. the cross-sectional retrospective study included baseline data of 194 adults (62 male and 132 female), 53,74 ± 8,77 years, clinically and ethically selected for a lifestyle modification program. Total cholesterol (TC) and cholesterol lipoprotein fractions, triglycerides (TG) and glucose was dosed by dry chemistry (Vitros® system, Johnson & Johnson). Blood leukocytes was quantified by automatic cell counter (Coulter ABX®, Horiba). LDL-cholesterol was obtained by Friedwald formula. Serum ultrasensitive C-reactive protein (US-CRP) was accessed by the immunochemoluminescence method (Immulite 2000®, DPC Medlab). Weight, height, body mass index (BMI) and waist circumference (WC) were measured. Muscular mass and fat mass were obtained by bioelectrical impedance analysis (impedancemeter Quantum BIA-101Q®, Clinton Township). Arterial blood pressure was checked by auscultatory method and cardiorespiratory fitness was determined by ergoespirometric test (Balke protocol). The metabolic syndrome was diagnosed according NCEP – ATP III (2001), following recommendations of American Diabetes Association (2004). Pearson’s correlation crude and adjusted for confounders variables with p<0,05. The prevalence of MS was 30.4%.
