Red blood cell-derived microparticles isolated from blood units initiate and propagate thrombin generation.

BACKGROUND: Red blood cell-derived microparticles (RMPs) are small phospholipid vesicles shed from RBCs in blood units, where they accumulate during storage. Because microparticles are bioactive, it could be suggested that RMPs are mediators of posttransfusion complications or, on the contrary, constitute a potential hemostatic agent. STUDY DESIGN AND METHODS: This study was performed to establish the impact on coagulation of RMPs isolated from blood units. Using calibrated automated thrombography, we investigated whether RMPs affect thrombin generation (TG) in plasma. RESULTS: We found that RMPs were not only able to increase TG in plasma in the presence of a low exogenous tissue factor (TF) concentration, but also to initiate TG in plasma in absence of exogenous TF. TG induced by RMPs in the absence of exogenous TF was neither affected by the presence of blocking anti-TF nor by the absence of Factor (F)VII. It was significantly reduced in plasma deficient in FVIII or F IX and abolished in FII-, FV-, FX-, or FXI-deficient plasma. TG was also totally abolished when anti-XI 01A6 was added in the sample. Finally, neither Western blotting, flow cytometry, nor immunogold labeling allowed the detection of traces of TF antigen. In addition, RMPs did not comprise polyphosphate, an important modulator of coagulation. CONCLUSIONS: Taken together, our data show that RMPs have FXI-dependent procoagulant properties and are able to initiate and propagate TG. The anionic surface of RMPs might be the site of FXI-mediated TG amplification and intrinsic tenase and prothrombinase complex assembly.

Odorant binding proteins of the red imported fire ant, Solenopsis invicta: an example of the problems facing the analysis of widely divergent proteins.

We describe the odorant binding proteins (OBPs) of the red imported fire ant, Solenopsis invicta, obtained from analyses of an EST library and separate 454 sequencing runs of two normalized cDNA libraries. We identified a total of 18 putative functional OBPs in this ant. A third of the fire ant OBPs are orthologs to honey bee OBPs. Another third of the OBPs belong to a lineage-specific expansion, which is a common feature of insect OBP evolution. Like other OBPs, the different fire ant OBPs share little sequence similarity (∼ 20%), rendering evolutionary analyses difficult. We discuss the resulting problems with sequence alignment, phylogenetic analysis, and tests of selection. As previously suggested, our results underscore the importance for careful exploration of the sensitivity to the effects of alignment methods for data comprising widely divergent sequences.

La Gestión de la Información Personal en usuarios intensivos de las TIC

Se presenta un modelo de análisis del comportamiento informacional global de un colectivo de individuos (estudiantes de la Universitat Oberta de Catalunya) que tienen una percepción positiva sobre el uso de las tecnologías de la información y la comunicación y que realizan un uso intensivo de las mismas.A partir de una aproximación cualitativa, mediante 24 entrevistas y un posterior análisis del contenido, se identifican cuatro perfiles distintos de gestión de la información personal (reactivo, pasivo, exhaustivo y proactivo) en base a diez variables subyacentes (acceso, gestión y usos de la información, competenciasinformacionales, perfil cognitivo, actitud, percepción de las TIC, ámbito académico, profesional y de la vida diaria) y se ponen derelieve las diferencias de comportamiento informacional dependiendo del ámbito en el que se encuentren. La identificación de los perfiles es un estadio básico del diseño centrado en los usuarios que facilita la realización de intervenciones específicas para cada tipo de usuario, respetando requerimientos de herramientasy procesos para que puedan desarrollar su comportamiento informacional de forma eficiente y eficaz.

From institutional repositories to personal collections of learning resources

Institutional digital repositories are a basic piece to provide preservation and reutilization of learning resources. However, their creation and maintenance is usually performed following a top-down approach, causing limitations in the search and reutilization of learning resources. In order to avoid this problem we propose to use web 2.0 functionalities. In this paper we present how tagging can be used to enhance the search and reusability functionalities of institutional learning repositories as well as promoting their usage. The paper also describes the evaluation process that was performed in a pilot experience involving open educational resources.

From rifting to passive margin - The examples of the Red Sea, Central Atlantic and Alpine Tethys

Evolution of the Red Sea/Gulf of Suez and the Central Atlantic rift systems shows that an initial, transtensive rifting phase, affecting a broad area around the future zone of crustal separation, was followed by a pre-oceanic rifting phase during which extensional strain was concentrated on the axial rift zone. This caused lateral graben systems to become inactive and they evolved into rift-rim basins. The transtensive phase of diffuse crustal extension is recognized in many intra-continental rifts. If controlling stress systems relax, these rifts abort and develop into palaeorifts. If controlling stress systems persist, transtensive rift systems can enter the pre-oceanic rifting stage, during which the rift zone narrows and becomes asymmetric as a consequence of simple-shear deformation at shallow crustal levels and pure shear deformation at lower crustal and mantle-lithospheric levels. Preceding crustal separation, extensional denudation of the lithospheric mantle is possible. Progressive lithospheric attenuation entails updoming of the asthenosphere and thermal doming of the rift shoulders. Their uplift provides a major clastic source for the rift basins and the lateral rift-rim basins. Their stratigraphic record provides a sensitive tool for dating the rift shoulder uplift. Asymmetric rifting leads to the formation of asymmetric continental margins, corresponding in a simple-shear model to an upper plate and a conjugate lower plate margin, as seen in the Central Atlantic passive margins of the United States and Morocco. This rifting model can be successfully applied to the analysis of the Alpine Tethys palaeo-margins (such as Rif and the Western Alps).

Automated segmentation of multiple red blood cells with digital holographic microscopy.

We present a method to automatically segment red blood cells (RBCs) visualized by digital holographic microscopy (DHM), which is based on the marker-controlled watershed algorithm. Quantitative phase images of RBCs can be obtained by using off-axis DHM along to provide some important information about each RBC, including size, shape, volume, hemoglobin content, etc. The most important process of segmentation based on marker-controlled watershed is to perform an accurate localization of internal and external markers. Here, we first obtain the binary image via Otsu algorithm. Then, we apply morphological operations to the binary image to get the internal markers. We then apply the distance transform algorithm combined with the watershed algorithm to generate external markers based on internal markers. Finally, combining the internal and external markers, we modify the original gradient image and apply the watershed algorithm. By appropriately identifying the internal and external markers, the problems of oversegmentation and undersegmentation are avoided. Furthermore, the internal and external parts of the RBCs phase image can also be segmented by using the marker-controlled watershed combined with our method, which can identify the internal and external markers appropriately. Our experimental results show that the proposed method achieves good performance in terms of segmenting RBCs and could thus be helpful when combined with an automated classification of RBCs.

Four-corner bone arthrodesis with dorsal rectangular plate: series and personal technique.

Controversy exists about the best method to achieve bone fusion in four-corner arthrodesis. Thirty-five patients who underwent this procedure by our technique were included in the study. Surgical indications were stage II-III SLAC wrist, stage II SNAC wrist and severe traumatic midcarpal joint injury. Mean follow-up was 4.6 years. Mean active flexion and extension were 34 degrees and 30 degrees respectively; grip strength recovery was 79%. Radiological consolidation was achieved in all cases. The mean DASH score was 23 and the postoperative pain improvement by visual analogue scale was statistically significant. Return to work was possible at 4 months for the average patient. Complications were a capitate fracture in one patient and the need for hardware removal in four cases. Four-corner bone wrist arthrodesis by dorsal rectangular plating achieves an acceptable preservation of range of motion with good pain relief, an excellent consolidation rate and minimal complications.

La documentació personal com a eina docent: ensenyar i aprendre amb els fons personals

L"objectiu de la contribució que presentem és força senzill d"entendre: hem volgut donar forma a una activitat viscuda per recordar-la i per aprofitar-la quan l"haguem de menester. Com ho hem fet?, doncs materialitzant uns fets, uns pensaments i unes idees en forma de document, el qual a hores d"ara ja forma part del nostre fons personal. La finalitat del document, però, va més enllà: volem donar a conèixer la vàlua dels fons personals com a recurs estratègic per a l"aprenentatge de l"Arxivística. Hem volgut compartir la nostra experiència en aquest canal de comunicació professional perquè pensem, modestament, que treballs com aquest poden ser noves formes d"avançar en el progrés teòric de l"Arxivística.

The psychometric properties of the WAI in a career counseling setting: Comparison with a personal counseling sample

This study investigated the psychometric properties of the Working Alliance Inventory-Client version (WAI-C) and Working Alliance Inventory-Short and revised (WAI-SR) in a career counseling setting. Moreover, it compared the impact of career versus personal counseling settings based on results obtained using the WAI-SR. Subjects were 188 French-speaking career counseling clients and 95 French-speaking personal counseling clients, mainly students. For the career counseling sample, total reliability was .87 for the WAI-C and .76 for the WAI-SR. The shape of the distribution was normal but the variance was significantly lower for the career counseling sample. Exploratory factor analyses (EFAs) and confirmatory factor analyses (CFAs) confirmed the presence of an overall working alliance factor but indicated a clearer hierarchical structure for the WAI- SR than for the WAI-C. The psychometric properties seemed only slightly affected by the counseling setting, suggesting that this inventory is also relevant for career counseling, especially the WAI-SR, which has a more robust factorial structure.

Test-retest repeatability of the pupil light response to blue and red light stimuli in normal human eyes using a novel pupillometer.

In this study, we evaluated the repeatability of pupil responses to colored light stimuli in healthy subjects using a prototype chromatic pupillometer. One eye of 10 healthy subjects was tested twice in the same day using monochromatic light exposure at two selected wavelengths (660 and 470 nm, intensity 300 cd/m(2)) presented continuously for 20 s. Pupil responses were recorded in real-time before, during, and after light exposure. Maximal contraction amplitude and sustained contraction amplitude were calculated. In addition, we quantified the summed pupil response during continuous light stimulation as the total area between a reference line representing baseline pupil size and the line representing actual pupil size over 20 s (area under the curve). There was no significant difference in the repeated measure compared to the first test for any of the pupil response parameters. In conclusion, we have developed a novel prototype of color pupillometer which demonstrates good repeatability in evoking and recording the pupillary response to a bright blue and red light stimulus.
