Introdução: A reposição volêmica em pacientes traumatizados tem sido controvérsa. O A.T.L.S. recomenda a infusão de um grande volume de fluidos na fase inicial de tratamento, enquanto outros autores recomendam a administração somente quando do controle da hemorragia. O acesso venoso femoral é contra indicado em pacientes com trauma abdominal por temor de aumento de hemorragia. A solução hipertônica de NaCl a 7,5% (SH) possui benefícios consideráveis de logística e de recuperação hemodinâmica com pequenos volumes de infusão, semelhante as vantagens das soluções padrões isotônicas na fase pré-hospitalar. Objetivos: Criar um modelo de choque hemorrágico induzido por trauma venoso. Avaliar a hemodinâmica e o volume de hemorragia abdominal nos animais submetidos a choque hemorrágico e tratados com SH via acesso femoral e jugular. Métodos: Em 18 porcos da raça landrace, divididos em 3 grupos de 6 animais (Controle, Jugular e Femoral), foi induzido um choque hipovolêmico não controlado pela ruptura da veia cava caudal. Os animais do grupo controle (GC) foram observados por 40 minutos quanto ao seu padrão hemodinâmico de Pressão de Artéria Pulmonar (PAP), Pressão Artérial Média (PAM), Débito Cardíaco (DC) e Fluxo de Veia Porta (FVP), porém sem reposição volêmica. Os animais dos grupos Femoral (GC) e Jugular (GJ) foram tratados com 4 ml/Kg de solução hipertônica de NaCl a 7,5% (SH) aos 20 minutos de experimento. Ao final do experimento, o volume de hemorragia abdominal foi mensurado.Resultados: O grupo controle (GC) apresentou queda dos valores hemodinâmicos aos 10 minutos e estes permaneceram estáveis até o final do experimento. Os animais dos grupos tratamento (GF e GJ) apresentaram melhora da hemodinâmica aos 30 minutos, sem aumento da hemorragia abdominal. Conclusão: A solução hipertônica de NaCl (SH) permitiu a melhora parcial da hemodinâmica no modelo de choque hipovolêmico, sem aumento da hemorragia, independentemente do acesso utilizada para a infusão.

[EN] This paper deals with the relief generation in Ourense, an interior territory of the Galicia Country, at NW Spain, after the breakdown of Pangea 200 million years ago. The rupture of supercontinent causes the main effects in the outer part of Galicia, the present coast line and the shelf, but also the inner parts of Galicia where the landscape changes dramatically mainly ruled by fluvial incision connected with uprising, (orogenic, epirogenic, or isostatic origin), or even with eustatic oscillations, that shaped the previous old mesozoic landscape. Various things complicate the correct understanding of Galician geomorphology:1) the prevalent hercynian structure, (presumably reactivated during the Alpine Orogeny), causes that the epigenic processes, (fluvial, glaciar, marine and etching), acting on Galicia from Mesozoic to present times, produce end forms identified erroneously at the previous literature as tectonic and not as etch forms profiting from lithological or structural contrasts. 2) the common morphotectonic model accepted by all previous researchers establishes for the whole of Galicia a blocky pattern, (horst and graben like), due to extensional tectonic regime. This model is proved as no longer valid because the Galician tertiary basins, even were described at the past as graben depressions never have this origin. 3) big differences exist between the north and western sides of Galicia that show contrasted tectonic regime: compressional (with forms as the so called raised platforms), at the northern coast border, and extensional (with forms so typical as the Rias), at the western side. The study area is located at the confluence of two tectonic domains where the above mentioned effects are coincidents and specially well showed through different effects: prominent assimetry of fluvial captures (west facing), pronounced river incision and different kinds of tertiary basins: either strike slipe faults (Maceda, Xinzo de Limia, etc), or overslipped by inverse faults, (Quiroga, A Rúa, etc.), or even corresponding with depressions never, (or anywise passively), affected by tectonic movements, (Monforte). The paper include a detailed inventory of surfaces and terrace levels and their incision sequence which allow stablish a relative chronology of geomorphic evolution at this area of NW Spain during meso-cainozoic times.

[EN] The aortic dissection is a disease that can cause a deadly situation, even with a correct treatment. It consists in a rupture of a layer of the aortic artery wall, causing a blood flow inside this rupture, called dissection. The aim of this paper is to contribute to its diagnosis, detecting the dissection edges inside the aorta. A subpixel accuracy edge detector based on the hypothesis of partial volume effect is used, where the intensity of an edge pixel is the sum of the contribution of each color weighted by its relative area inside the pixel. The method uses a floating window centred on the edge pixel and computes the edge features. The accuracy of our method is evaluated on synthetic images of different hickness and noise levels, obtaining an edge detection with a maximal mean error lower than 16 percent of a pixel.

In questa tesi si è voluta porre l’attenzione sulla suscettibilità alle alte temperature delle resine che li compongono. Lo studio del comportamento alle alte temperature delle resine utilizzate per l’applicazione dei materiali compositi è risultato un campo di studio ancora non completamente sviluppato, nel quale c’è ancora necessità di ricerche per meglio chiarire alcuni aspetti del comportamento. L’analisi di questi materiali si sviluppa partendo dal contesto storico, e procedendo successivamente ad una accurata classificazione delle varie tipologie di materiali compositi soffermandosi sull’ utilizzo nel campo civile degli FRP (Fiber Reinforced Polymer) e mettendone in risalto le proprietà meccaniche. Considerata l’influenza che il comportamento delle resine riveste nel comportamento alle alte temperature dei materiali compositi si è, per questi elementi, eseguita una classificazione in base alle loro proprietà fisico-chimiche e ne sono state esaminate le principali proprietà meccaniche e termiche quali il modulo elastico, la tensione di rottura, la temperatura di transizione vetrosa e il fenomeno del creep. Sono state successivamente eseguite delle prove sperimentali, effettuate presso il Laboratorio Resistenza Materiali e presso il Laboratorio del Dipartimento di Chimica Applicata e Scienza dei Materiali, su dei provini confezionati con otto differenti resine epossidiche. Per valutarne il comportamento alle alte temperature, le indagini sperimentali hanno valutato dapprima le temperature di transizione vetrosa delle resine in questione e, in seguito, le loro caratteristiche meccaniche. Dalla correlazione dei dati rilevati si sono cercati possibili legami tra le caratteristiche meccaniche e le proprietà termiche delle resine. Si sono infine valutati gli aspetti dell’applicazione degli FRP che possano influire sul comportamento del materiale composito soggetto alle alte temperature valutando delle possibili precauzioni che possano essere considerate in fase progettuale.

By the end of the 19th century, geodesy has contributed greatly to the knowledge of regional tectonics and fault movement through its ability to measure, at sub-centimetre precision, the relative positions of points on the Earth’s surface. Nowadays the systematic analysis of geodetic measurements in active deformation regions represents therefore one of the most important tool in the study of crustal deformation over different temporal scales [e.g., Dixon, 1991]. This dissertation focuses on motion that can be observed geodetically with classical terrestrial position measurements, particularly triangulation and leveling observations. The work is divided into two sections: an overview of the principal methods for estimating longterm accumulation of elastic strain from terrestrial observations, and an overview of the principal methods for rigorously inverting surface coseismic deformation fields for source geometry with tests on synthetic deformation data sets and applications in two different tectonically active regions of the Italian peninsula. For the long-term accumulation of elastic strain analysis, triangulation data were available from a geodetic network across the Messina Straits area (southern Italy) for the period 1971 – 2004. From resulting angle changes, the shear strain rates as well as the orientation of the principal axes of the strain rate tensor were estimated. The computed average annual shear strain rates for the time period between 1971 and 2004 are γ˙1 = 113.89 ± 54.96 nanostrain/yr and γ˙2 = -23.38 ± 48.71 nanostrain/yr, with the orientation of the most extensional strain (θ) at N140.80° ± 19.55°E. These results suggests that the first-order strain field of the area is dominated by extension in the direction perpendicular to the trend of the Straits, sustaining the hypothesis that the Messina Straits could represents an area of active concentrated deformation. The orientation of θ agree well with GPS deformation estimates, calculated over shorter time interval, and is consistent with previous preliminary GPS estimates [D’Agostino and Selvaggi, 2004; Serpelloni et al., 2005] and is also similar to the direction of the 1908 (MW 7.1) earthquake slip vector [e.g., Boschi et al., 1989; Valensise and Pantosti, 1992; Pino et al., 2000; Amoruso et al., 2002]. Thus, the measured strain rate can be attributed to an active extension across the Messina Straits, corresponding to a relative extension rate ranges between < 1mm/yr and up to ~ 2 mm/yr, within the portion of the Straits covered by the triangulation network. These results are consistent with the hypothesis that the Messina Straits is an important active geological boundary between the Sicilian and the Calabrian domains and support previous preliminary GPS-based estimates of strain rates across the Straits, which show that the active deformation is distributed along a greater area. Finally, the preliminary dislocation modelling has shown that, although the current geodetic measurements do not resolve the geometry of the dislocation models, they solve well the rate of interseismic strain accumulation across the Messina Straits and give useful information about the locking the depth of the shear zone. Geodetic data, triangulation and leveling measurements of the 1976 Friuli (NE Italy) earthquake, were available for the inversion of coseismic source parameters. From observed angle and elevation changes, the source parameters of the seismic sequence were estimated in a join inversion using an algorithm called “simulated annealing”. The computed optimal uniform–slip elastic dislocation model consists of a 30° north-dipping shallow (depth 1.30 ± 0.75 km) fault plane with azimuth of 273° and accommodating reverse dextral slip of about 1.8 m. The hypocentral location and inferred fault plane of the main event are then consistent with the activation of Periadriatic overthrusts or other related thrust faults as the Gemona- Kobarid thrust. Then, the geodetic data set exclude the source solution of Aoudia et al. [2000], Peruzza et al. [2002] and Poli et al. [2002] that considers the Susans-Tricesimo thrust as the May 6 event. The best-fit source model is then more consistent with the solution of Pondrelli et al. [2001], which proposed the activation of other thrusts located more to the North of the Susans-Tricesimo thrust, probably on Periadriatic related thrust faults. The main characteristics of the leveling and triangulation data are then fit by the optimal single fault model, that is, these results are consistent with a first-order rupture process characterized by a progressive rupture of a single fault system. A single uniform-slip fault model seems to not reproduce some minor complexities of the observations, and some residual signals that are not modelled by the optimal single-fault plane solution, were observed. In fact, the single fault plane model does not reproduce some minor features of the leveling deformation field along the route 36 south of the main uplift peak, that is, a second fault seems to be necessary to reproduce these residual signals. By assuming movements along some mapped thrust located southward of the inferred optimal single-plane solution, the residual signal has been successfully modelled. In summary, the inversion results presented in this Thesis, are consistent with the activation of some Periadriatic related thrust for the main events of the sequence, and with a minor importance of the southward thrust systems of the middle Tagliamento plain.

New concepts on porosity appraisal in ancient and modern construction materials. The role of Fractal Geometry on porosity characterization and transport phenomena. This work studied the potential of Fractal Geometry to the characterization of porous materials. Besides the descriptive aspects of the pore size distribution, the fractal dimensions have led to the development of rational relations for the prediction of permeability coefficients to fluid and heat transfer. The research considered natural materials used in historical buildings (rock and earth) as well as currently employed materials as hydraulic cement and technologically advanced materials such as silicon carbide or YSZ ceramics. The experimental results of porosity derived from the techniques of mercury intrusion and from the image analysis. Data elaboration was carried out according to established procedures of Fractal Geometry. It was found that certain classes of materials are clearly fractal and respond to simple patterns such as Sierpinski and Menger models. In several cases, however, the fractal character is not recognised because the microstructure of the material is based on different phases at different dimensional scales, and in consequence the “fractal dimensions” calculated from porosimetric data do not come within the standard range (less than 3). Using different type and numbers of fractal units is possible, however, to obtain “virtual” microstructures that have the fraction of voids and pore size distribution equivalent with the experimental ones for almost any material. Thus it was possible to take the expressions for the permeability and the thermal conduction which does not require empirical “constants”, these expressions have also provided values that are generally in agreement with the experimental available data. More problematic has been the fractal discussion of the geometry of the rupture of the material subjected to mechanical stress both external and internal applied. The results achieved on these issues are qualitative and prone to future studies. The productivity of agricultural crops is seriously limited by salinity. This problem is rapidly increasing, particularly in irrigated lands. Like almost all the fruit tree species, Pyrus communis is generally considered a salt sensitive species, but only little information is available on its behavior under saline conditions. Previous studies, carried out in the Department of Fruit Tree and Woody Plant Science (University of Bologna), focused their attention on pear and quince salt stress responses to understand which rootstock would be the most suitable for pear in order to tolerate a salt stress condition. It has been reported that pear and quince have different ability in the uptake, translocation and accumulation of chloride (Cl-) and sodium (Na+) ions, when plants were irrigated for one season with saline water (5 dS/m). The aim of the present work was to deepen these aspects and investigate salt stress responses in pear and quince. Two different experiments have been performed: a “short-term” trial in a growth chamber and a “long-term” experiment in the open field. In the short-term experiment, three different genotypes usually adopted as pear rootstocks (MC, BA29 and Farold®40) and the pear variety Abbé Fétel own rooted have been compared under salt stress conditions. The trial was performed in a hydroponic culture system, applying a 90 mM NaCl stress to half of the plants, after five weeks of normal growth in Hoagland’s solution. During the three-weeks of salt stress treatment, physiological, mineral and molecular analyses were performed in order to monitor, for each genotype, the development of the salt stress responses in comparison with the corresponding “unstressed” plants. Farold®40 and Abbé Fétel own rooted showed the onset of leaf necrosis, due to salt toxicity, one week before quinces. Moreover, quinces displayed a significant delay in premature senescence of old leaves, while pears emerged for their ability to regenerate new leaves from apparently dead foliage with the salt stress still running. Physiological measurements, such as shoots length, chlorophyll (Chl) content, and photosynthesis, have been carried out and revealed that pears exhibited a significant reduction in water content and a wilting aspect, while for quinces a decrease in Chl content and a growth slowdown were observed. At the end of the trial, all plants were collected and organs separated for dry weight estimation and mineral analyses (Cu, Fe, Mn, Zn Mg, Ca, K, Na and Cl). Mineral contents have been affected by salinity; same macro/micro nutrients were altered in some organs or relocated within the plant. This plant response could have partially contributed to face the salt stress. Leaves and roots have been harvested for molecular analyses at four different times during stress conditions. Molecular analyses consisted of the gene expression study of three main ion transporters, well known in Arabidopsis thaliana as salt-tolerance determinants in the “SOS” pathway: NHX1 (tonoplast Na+/H+ antiporter), SOS1 (plasmalemma Na+/H+ antiporter) and HKT1 (K+ high-affinity and Na+ low-affinity transporter). These studies showed that two quince rootstocks adopted different responsive mechanisms to NaCl stress. BA29 increased its Na+ sequestration activity into leaf vacuoles, while MC enhanced temporarily the same ability, but in roots. Farold®40, instead, exhibited increases in SOS1 and HKT1 expression mainly at leaf level in the attempt to retrieve Na+ from xylem, while Abbé Fétel differently altered the expression of these genes in roots. Finally, each genotype showed a peculiar response to salt stress that was the sum of its ability in Na+ exclusion, osmotic tolerance and tissue tolerance. In the long-term experiment, potted trees of the pear variety Abbé Fétel grafted on different rootstocks (MC, BA29 and Farold®40), or own rooted and also rootstocks only were subjected to a salt stress through saline water irrigation with an electrical conductivity of 5 dS/m for two years. The purposes of this study were to evaluate salinity effects on physiological (shoot length, number of buds, photosynthesis, etc.) and yield parameters of cultivar Abbé Fétel in the different combinations and to determine the salt amount that pear is able to tolerate over the years. With this work, we confirmed the previous hypothesis that pear, despite being classified as a salt-sensitive fruit tree, can be cultivated for two years under saline water irrigation, without showing any salt toxicity symptoms or severe drawbacks on plant development and production. Among different combinations, Abbé Fétel grafted on MC resulted interesting for its peculiar behaviors under salt stress conditions. In the near future, further investigations on physiological and molecular aspects will be necessary to enrich and broaden the knowledge of salt stress responses in pear.

A novel nanosized and addressable sensing platform based on membrane coated plasmonic particles for detection of protein adsorption using dark field scattering spectroscopy of single particles has been established. To this end, a detailed analysis of the deposition of gold nanorods on differently functionalized substrates is performed in relation to various factors (such as the pH, ionic strength, concentration of colloidal suspension, incubation time) in order to find the optimal conditions for obtaining a homogenous distribution of particles at the desired surface number density. The possibility of successfully draping lipid bilayers over the gold particles immobilized on glass substrates depends on the careful adjustment of parameters such as membrane curvature and adhesion properties and is demonstrated with complementary techniques such as phase imaging AFM, fluorescence microscopy (including FRAP) and single particle spectroscopy. The functionality and sensitivity of the proposed sensing platform is unequivocally certified by the resonance shifts of the plasmonic particles that were individually interrogated with single particle spectroscopy upon the adsorption of streptavidin to biotinylated lipid membranes. This new detection approach that employs particles as nanoscopic reporters for biomolecular interactions insures a highly localized sensitivity that offers the possibility to screen lateral inhomogeneities of native membranes. As an alternative to the 2D array of gold nanorods, short range ordered arrays of nanoholes in optically transparent gold films or regular arrays of truncated tetrahedron shaped particles are built by means of colloidal nanolithography on transparent substrates. Technical issues mainly related to the optimization of the mask deposition conditions are successfully addressed such that extended areas of homogenously nanostructured gold surfaces are achieved. Adsorption of the proteins annexin A1 and prothrombin on multicomponent lipid membranes as well as the hydrolytic activity of the phospholipase PLA2 were investigated with classical techniques such as AFM, ellipsometry and fluorescence microscopy. At first, the issues of lateral phase separation in membranes of various lipid compositions and the dependency of the domains configuration (sizes and shapes) on the membrane content are addressed. It is shown that the tendency for phase segregation of gel and fluid phase lipid mixtures is accentuated in the presence of divalent calcium ions for membranes containing anionic lipids as compared to neutral bilayers. Annexin A1 adsorbs preferentially and irreversibly on preformed phosphatidylserine (PS) enriched lipid domains but, dependent on the PS content of the bilayer, the protein itself may induce clustering of the anionic lipids into areas with high binding affinity. Corroborated evidence from AFM and fluorescence experiments confirm the hypothesis of a specifically increased hydrolytic activity of PLA2 on the highly curved regions of membranes due to a facilitated access of lipase to the cleavage sites of the lipids. The influence of the nanoscale gold surface topography on the adhesion of lipid vesicles is unambiguously demonstrated and this reveals, at least in part, an answer for the controversial question existent in the literature about the behavior of lipid vesicles interacting with bare gold substrates. The possibility of formation monolayers of lipid vesicles on chemically untreated gold substrates decorated with gold nanorods opens new perspectives for biosensing applications that involve the radiative decay engineering of the plasmonic particles.

Membranen spielen eine essentielle Rolle bei vielen wichtigen zellulären Prozessen. Sie ermöglichen die Erzeugung von chemischen Gradienten zwischen dem Zellinneren und der Umgebung. Die Zellmembran übernimmt wesentliche Aufgaben bei der intra- und extrazellulären Signalweiterleitung und der Adhäsion an Oberflächen. Durch Prozesse wie Endozytose und Exozytose werden Stoffe in oder aus der Zelle transportiert, eingehüllt in Vesikel, welche aus der Zellmembran geformt werden. Zusätzlich bietet sie auch Schutz für das Zellinnere. Der Hauptbestandteil einer Zellmembran ist die Lipiddoppelschicht, eine zweidimensionale fluide Matrix mit einer heterogenen Zusammensetzung aus unterschiedlichen Lipiden. In dieser Matrix befinden sich weitere Bausteine, wie z.B. Proteine. An der Innenseite der Zelle ist die Membran über Ankerproteine an das Zytoskelett gekoppelt. Dieses Polymernetzwerk erhöht unter anderem die Stabilität, beeinflusst die Form der Zelle und übernimmt Funktionenrnbei der Zellbewegung. Zellmembranen sind keine homogenen Strukturen, je nach Funktion sind unterschiedliche Lipide und Proteine in mikrsokopischen Domänen angereichert.Um die grundlegenden mechanischen Eigenschaften der Zellmembran zu verstehen wurde im Rahmen dieser Arbeit das Modellsystem der porenüberspannenden Membranen verwendet.Die Entwicklung der porenüberspannenden Membranen ermöglicht die Untersuchung von mechanischen Eigenschaften von Membranen im mikro- bis nanoskopischen Bereich mit rasterkraftmikroskopischen Methoden. Hierbei bestimmen Porosität und Porengröße des Substrates die räumliche Auflösung, mit welcher die mechanischen Parameter untersucht werdenrnkönnen. Porenüberspannende Lipiddoppelschichten und Zellmembranen auf neuartigen porösen Siliziumsubstraten mit Porenradien von 225 nm bis 600 nm und Porositäten bis zu 30% wurden untersucht. Es wird ein Weg zu einer umfassenden theoretischen Modellierung der lokalen Indentationsexperimente und der Bestimmung der dominierenden energetischen Beiträge in der Mechanik von porenüberspannenden Membranen aufgezeigt. Porenüberspannende Membranen zeigen eine linear ansteigende Kraft mit zunehmender Indentationstiefe. Durch Untersuchung verschiedener Oberflächen, Porengrößen und Membranen unterschiedlicher Zusammensetzung war es für freistehende Lipiddoppelschichten möglich, den Einfluss der Oberflächeneigenschaften und Geometrie des Substrates, sowie der Membranphase und des Lösungsmittels auf die mechanischen Eigenschaften zu bestimmen. Es ist möglich, die experimentellen Daten mit einem theoretischen Modell zu beschreiben. Hierbei werden Parameter wie die laterale Spannung und das Biegemodul der Membran bestimmt. In Abhängigkeit der Substrateigenschaften wurden für freitragende Lipiddoppelschichten laterale Spannungen von 150 μN/m bis zu 31 mN/m gefunden für Biegemodulde zwischen 10^(−19) J bis 10^(−18) J. Durch Kraft-Indentations-Experimente an porenüberspannenden Zellmembranen wurde ein Vergleich zwischen dem Modell der freistehenden Lipiddoppelschichten und nativen Membranen herbeigeführt. Die lateralen Spannungen für native freitragende Membranen wurden zu 50 μN/m bestimmt. Weiterhin konnte der Einfluss des Zytoskeletts und der extrazellulä-rnren Matrix auf die mechanischen Eigenschaften bestimmt und innerhalb eines basolateralen Zellmembranfragments kartiert werden, wobei die Periodizität und der Porendurchmesser des Substrates das räumliche Auflösungsvermögen bestimmen. Durch Fixierung der freistehenden Zellmembran wurde das Biegemodul der Membran um bis zu einem Faktor 10 erhöht. Diese Arbeit zeigt wie lokal aufgelöste, mechanische Eigenschaften mittels des Modellsystems der porenüberspannenden Membranen gemessen und quantifiziert werden können. Weiterhin werden die dominierenden energetischen Einflüsse diskutiert, und eine Vergleichbarkeit zurnnatürlichen Membranen hergestellt.rn

La rottura del Legamento Crociato Craniale (LCCr) rappresenta una delle patologie ortopediche di maggiore riscontro clinico nella specie canina. In seguito a rottura del LCCr si presenta un continuo slittamento craniale della tibia il quale esita in un processo osteoartrosico. La risoluzione chirurgica rappresenta la migliore soluzione terapeutica. Le tecniche chirurgiche extra-articolari con sfruttamento dei punti isometrici del ginocchio si presentano come delle procedure molto diffuse e utilizzate. Questa tesi propone di validare l’uso di un nuovo sistema di navigazione computerizzato-assistito per la valutazione cinematica durante la ricostruzione del LCCr nel cane, ma soprattutto di studiare e confrontare il comportamento e l’efficacia dopo ricostruzione TightRope (TR) in due diverse coppie di punti isometrici. Abbiamo effettuato due analisi in parallelo. La prima eseguendo interventi chirurgici con tecnica TR su 18 casi clinici e sfruttando il punto isometrico del femore (F2) e due diversi punti isometrici della tibia (T2 o T3). L’analisi prevedeva dei controlli postoperatori a 1, 3 e 6 mesi. Ad ogni controllo veniva effettuata una visita ortopedica, esami radiografici, un questionario di valutazione clinico e di soddisfazione del proprietario. Mentre nella ricerca Ex-Vivo abbiamo eseguito dei test su 14 preparati anatomici con l’utilizzo di un sistema di navigazione computerizzato per la rilevazione dei dati. L’analisi prevedeva la valutazione dell’articolazione in diversi stadi: LCCr intatto; LCCr rotto; dopo ricostruzione con TR in F2-T2 e tensionato a 22N, 44N e 99N; dopo ricostruzione con TR in F2-T3 e tensionato a 22N, 44N e 99N. Ad ogni stadio si eseguivano cinque test di valutazione, tra cui: Test del Cassetto, Test di compressione tibiale (TCT), Rotazione Interna/Esterna, Flesso/Estensione e Varo/Valgo. Lo scopo di tale studio è quello di confrontare tra loro i punti isometrici del ginocchio e di analizzare l’efficacia della tecnica TR nelle due differenti condizioni di isometria (F2-T2 e F2-T3).

Die Metalloprotease Ovastacin, ein Vertreter der Astacin-Familie, wurde erstmals 2004 beschrieben. Im Ovar von Säugetieren ist Ovastacin-mRNA im Zeitfenster vom Stadium der Sekundärfollikel bis kurz nach der Befruchtung der Eizelle zu finden. Der Expressionsort und -zeitpunkt sowie die Sequenzähnlichkeit von über 60% mit sogenannten „Schlüpfenzymen“ (engl. hatching enzymes), die man in den Eizellen und Zygoten niederer Wirbeltiere und Wirbelloser gefunden hatte, ließen die Vermutung aufkommen, es könnte sich hier um das Säugerhomolog dieser Proteasen handeln. Generell lösen hatching Enzyme die derben embryonalen Hüllstrukturen (bei Säugern die Zona pellucida, ZP) beim Schlüpfvorgang auf. Die essentielle Bedeutung des Ovastacins für die Befruchtung wird durch die um ca. 30% reduzierte Fruchtbarkeit von Ovastacin defizienten Mäusen belegt. Hochinteressant war in diesem Zusammenhang die Entdeckung des Ovastacins in den Cortikalgranula der Oocyten sowie seine Fähigkeit, das Zona pellucida Protein 2 zu schneiden. Die dadurch bewirkte Verhärtung der Zona pellucida verhindert das Eindringen weiterer Spermien, das heißt sie baut eine Barriere gegen Polyspermie auf. Ziel dieser Arbeit war es, Belege für die physiologische Funktion des Ovastacins zu finden. Vor allem galt es, potentielle Aktivatoren zu identifizieren, da das Enzym wie alle Astacine als inaktive Vorstufe gebildet wird, die proteolytisch aktiviert werden muss. Zu diesem Zweck exprimierte ich rekombinantes Pro-Ovastacin in Insektenzellen. Aktivierungsstudien in vitro zeigten, dass ein saures Milieu zu einer Aktivierung führt, ohne die Abspaltung des Propeptids zu bewirken. Sequenzalignments und ein homologes Strukturmodell des Ovastacins wiesen auf Trypsin- oder Elastase-ähnliche Serinproteasen als potentielle Aktivierungsenzyme hin. Tatsächlich konnte mit diesen beiden Proteasetypen zum ersten Mal aktives Ovastacin aus Pro-Ovastacin erzeugt werden. Trypsin kommt als physiologischer Aktivator allerdings nicht in Betracht, da es bisher in keinem der Gewebe nachgewiesen werden konnte, in dem Ovastacin exprimiert wird. Die neutrophile Elastase dagegen konnte in der Leber, im Herz sowie im Blutplasma nachgewiesen werden. Mit Hilfe spezifischer Antikörper konnte das Herz als Expressionsort für Ovastacin bestätigt werden. Somit wäre Elastase ein potentieller physiologischer Aktivator von Ovastacin. Die Identifikation des Ovastacins in Geweben wie Leber, Herz, Nabelschnur und im Blutplasma weist auf eine Rolle der Protease in proteolytischen Netzwerken außerhalb der Spermien-Ei-Interaktion hin. Die Bedeutung der biologischen Kontrolle des Ovastacins bei der Befruchtung der Säugereizelle wird durch die Beobachtung untermauert, dass das Leberprotein Fetuin B als physiologischer Ovastacininhibitor fungiert und dadurch eine vorzeitige Verhärtung der Zona pellucida verhindert, die andernfalls die Penetration von Spermien prinzipiell verhindern würde.

Eine funktionierende Proteinqualitätskontrolle ist essenziell für die Vitalität einer Zelle. Das dynamische Gleichgewicht zwischen Proteinfaltung und -degradation wird von molekularen Chaperonen aufrechterhalten, deren Aktivität wiederum durch die Interaktion mit zahlreichen Cochaperonen moduliert wird. Das Cochaperon CHIP ist ein zentraler Faktor in Proteintriage-Entscheidungsprozessen, da es als Ubiquitinligase Chaperonsubstrate dem Abbau zuführt und somit die Chaperonmaschinerie direkt mit den Systemen der Proteindegradation verbindet. Um Polypeptide vor einem vorzeitigen Abbau zu schützen, wird die destruktive Aktivität von CHIP durch weitere Cochaperone reguliert. rnIn dieser Arbeit konnte die Hemmung der Ligaseaktivität von CHIP durch das Cochaperon BAG2 mechanistisch erstmals in einem zellulären System nachgewiesen werden. Dazu wurde die humane IMR-90 Fibroblasten Zelllinie verwendet. Die Ubiquitinierungsaktivität von CHIP wurde anhand von HSP72 als Modell-CHIP-Substrat untersucht. Durch die verringerte Ubiquitinierung, und damit dem reduzierten Abbau von HSP72, regulierte BAG2 dessen intrazelluläre Proteinspiegel, ohne dabei selbst eine Hitzeschockantwort zu induzieren. Überexprimiertes BAG2 wirkte sich trotz stabilisierter HSP72-Spiegel bei einem appliziertem Hitzestresses negativ auf die Zellvitalität aus, vermutlich da BAG2 durch die Inhibition von CHIP-vermittelter Ubiquitinierung massiv in das Gleichgewicht zwischen Substratfaltung und -degradation eingreift.rnDa sich die Mechanismen der Proteinqualitätskontrolle in der Alterung stark verändern und sich den wandelnden Bedingungen in der Zelle anpassen, wurde in einem zweiten Teil dieser Arbeit mit Hilfe des IMR-90 Zellsystems als etabliertes Modell zellulärer Seneszenz analysiert, inwieweit sich die Aktivität und die Regulation von CHIP durch BAG2 in der zellulären Alterung ändern. In seneszenten Zellen war HSP72 erheblich weniger ubiquitiniert als in jungen Fibroblasten, was auf eine reduzierte CHIP-Aktivität hinweist. Diese blieb jedoch durch BAG2 weiterhin modulierbar. Die Funktion von BAG2 als Inhibitor der Ubiquitinligase CHIP blieb demnach in seneszenten Zellen bestehen. In gealterten Fibroblasten regulierte BAG2 außerdem die Proteinspiegel des CHIP-Substrates und Seneszenzinitiators p53, was BAG2 eine mögliche Rolle in der Etablierung des Seneszenz-Phänotyps zuspricht. Weiterhin unterlagen die Proteinspiegel der beiden funktionell redundanten CHIP-Modulatoren BAG2 und HSPBP1 in der zellulären Alterung einer reziproken Regulation. In gealterten Mäusen trat die gegenläufige Veränderung der beiden Cochaperone gewebsspezifisch in der Lunge auf. Außerdem waren die BAG2-Proteinspiegel im Hippocampus gealterter Tiere signifikant erhöht.rnZusammenfassend konnte anhand der erzielten Ergebnisse die Funktion von BAG2 als Inhibitor von CHIP im zellulären System bestätigt werden. Außerdem durchlaufen die Aktivität und die Regulation von CHIP einen seneszenzspezifischen Adaptationsprozess, welcher für die Erhaltung der Proteostase in der Alterung relevant sein könnte und in welchem die Funktion von BAG2 als CHIP-Modulator möglicherweise eine wichtige Rolle spielt.rnZukünftige Studien könnten die komplexen Mechanismen weiterführend aufklären, mit denen CHIP-Aktivität reguliert wird. Dies kann helfen, der altersbedingten Abnahme an proteostatischer Kontrolle entgegenzuwirken und aberrante Proteinaggregation in altersassoziierten Erkrankungen vorzubeugen.rn

Die Myelinisierung neuronaler Axone ermöglicht eine schnelle und energieeffiziente Weiterleitung von Informationen im Nervensystem. Durch lokale Synthese von Myelinproteinen kann die Myelinschicht, zeitlich und räumlich reguliert, gebildet werden. Dieser Prozess ist abhängig von verschiedensten axonalen Eigenschaften und muss damit lokal reguliert werden. Die Myelinisierung im zentralen sowie im peripheren Nervensystem hängt unter anderem stark von kleinen regulatorischen RNA Molekülen ab. In Oligodendrozyten wird das Myelin Basische Protein (MBP) von der sncRNA715 translational reguliert, indem diese direkt innerhalb der 3’UTR der Mbp mRNA bindet und damit die Proteinsynthese verhindert. Mbp mRNA wird in hnRNP A2‐enthaltenen RNA Granula in die Zellperipherie transportiert, wo in Antwort auf axonale Signale die membranständige Tyrosin‐ Kinase Fyn aktiviert wird, welche Granula‐Komponenten wie hnRNP A2 und F phosphoryliert wodurch die lokale Translation initiiert wird. Während des Transports wird die mRNA durch die Bindung der sncRNA715 translational reprimiert. SncRNAs bilden zusammen mit Argonaut‐Proteinen den microRNA induced silencing complex (miRISC), welcher die translationale Inhibition oder den Abbau von mRNAs vermittelt. In der vorliegenden Arbeit sollte zum einen die Regulation der sncRNA715‐abhängigen translationalen Repression der Mbp mRNA in oligodendroglialen Zellen genauer untersucht werden und im zweiten Teil wurde die Rolle der sncRNA715 in den myelinbildenden Zellen des peripheren Nervensystems, den Schwann Zellen, analysiert. Es konnte in oligodendroglialen Zellen die mRNA‐Expression der vier, in Säugern bekannten Argonaut‐Proteinen nachgewiesen werden. Außerdem konnten die beiden Proteine Ago1 und Ago2 in vitro sowie in vivo detektiert werden. Ago2 interagiert mit hnRNP A2, Mbp mRNA und sncRNA715, womit es als neue Komponente des Mbp mRNA Transportgranulas identifiziert werden konnte. Des Weiteren colokalisiert Ago2 mit der Fyn‐Kinase und alle vier Argonaut‐Proteine werden Fyn‐abhängig Tyrosin‐phosphoryliert. Die Fyn‐abhängige Phosphorylierung der Granula‐Komponenten in Antwort auf axo‐glialen Kontakt führt zum Zerfall des RNA‐Granulas und zur gesteigerten MBP Proteinsynthese. Dies wird möglicherweise durch Abstoßungskräfte der negativ geladenen phosphorylierten Proteine vermittelt, wodurch diese sich voneinander und von der mRNA entfernen. Durch die Ablösung des miRISCs von der Mbp mRNA wird die Translation möglicherweise reaktiviert und die Myelinisierung kann starten. Mit der Identifizierung von Ago2 als neuer Mbp mRNA Transportgranula‐Komponente konnte ein weiterer Einblick in die Regulation der lokalen Translation von MBP gewährt werden. Das Verständnis dieses Prozesses ist entscheidend für die Entwicklung neuer Therapien von demyelinisierenden Erkrankungen, da neue Faktoren als eventuelle Ziele für pharmakologische Manipulationen identifiziert und möglichweise neue Therapiemöglichkeiten entstehen könnten. Im zweiten Teil der Arbeit wurde die translationale Regulation von Mbp mRNA in Schwann Zellen untersucht. Auch Schwann Zell‐Mbp wird als mRNA translational inaktiviert zur axo‐glialen Kontaktstelle transportiert, wo vermutlich auch lokale Translation in Antwort auf spezifische Signale stattfindet. Allerdings bleiben die genauen Mechanismen der mRNA‐Lokalisation und damit verbundenen translationalen Repression bislang ungeklärt. Es konnte hier gezeigt werden, dass auch in Schwann Zellen die sncRNA715 exprimiert wird und die Translation von Mbp reguliert. Überexpression der synthetischen sncRNA715 führt zu einer signifikanten Reduktion der MBP Proteinmengen in differenzierten primären Schwann Zellen. Damit kann vermutet werden, dass die Regulation der lokalen MBP Proteinsynthese in Schwann Zellen der in Oligodendrozyten ähnelt

The spinal column performs important functions in the body, including the support of the entire weight of the human body, the ability to orientate the head in space, bending, flexing and rotating the body. Diseases affecting the spine are manifold: the most frequent is scoliosis, which often affects the female population. It is often treated surgically with a very high percentage of failures. The aim of the thesis is to study the role of instrumentation in mechanical failures encountered 12 months after surgery in the treatment of scoliosis. For the purposes of the study, we analyzed specific biomechanical parameters. The pelvic angles determine the position of the pelvis, while the imbalance parameters the structure of the body. We infer other parameters by analyzing the characteristics of the implanted instrumentation. Initially, the anatomy is described of the spine and vertebrae, the equipment used and the possible failures that may occur after surgery. Subsequently, the materials and methods used for the analysis of the above-mentioned parameters for the 61 patients are reported. All data are obtained by the observation of pre and post-operative x-rays with a special program, by reading reports from operators and by medical records. In the fourth chapter, we report the results: the overall failure rate is 60.9%; the types of failures that occurred are rupture of bars and rupture of bars simultaneously to PJK. The most influential parameters on results of the progress of the surgery are the type of material used and the BMI. It is estimated a high percentage of failures in patients treated with implants of cobalt chromium alloys (90.0%). According to the results obtained, it is possible to understand the aspects that in the future should be studied, in order to find a solution to the most frequent surgical failures.

An unusual case is presented of a tourist who developed fatal cerebral air embolism, pneumomediastinum and pneumopericardium while ascending from low altitude to Europe's highest railway station. Presumably the air embolism originated from rupture of the unsuspected bronchogenic cyst as a result of pressure changes during the ascent. Cerebral air embolism has been observed during surgery, in scuba diving accidents, submarine escapes and less frequently during exposure to very high altitude. People with known bronchogenic cysts should be informed about the risk of cerebral air embolism and surgical removal should be considered. Cerebral air embolism is a rare cause of coma and stroke in all activities with rapid air pressure changes, including alpine tourism, as our unfortunate tourist illustrates.
