Palmer Deep is a series of three glacially overdeepened basins on the Antarctic Peninsula shelf, ~20 km southwest of Anvers Island. Site 1098 (64°51.72'S, 64°12.48'W) was drilled in the shallowest basin, Basin I, at 1012 m water depth. The sediment recovered was primarily laminated, siliceous, biogenic, pelagic muds alternating with siliciclastic hemipelagic sediments (Barker, Camerlenghi, Acton, et al., 1999). Sedimentation rates of 0.1725 cm/yr in the upper 25 m and 0.7-0.80 cm/yr in the lower 25 m of the core have been estimated from 14C (Domack et al., 2001). The oldest datable sediments have an age of ~13 ka and were underlain by diamicton sediments of the last glacial maximum (Domack et al., 2001). The large-scale water-mass distribution and circulation in the vicinity of Palmer Deep is dominated by Circumpolar Deep Water (CDW) below 200 m (Hofmann et al., 1996). Palmer Deep is too far from the coast to be influenced by glacial meltwater and cold-tongue generation associated with it (Domack and Williams, 1990; Dixon and Domack, 1991). Circulation patterns in the Palmer Deep area are not well understood, but evidence suggests southward flow across Palmer Deep from Anvers Island to Renaud Island (Kock and Stein, 1978). The water south of Anvers Island is nearly open with loose pack ice from February through May. The area is covered with sea ice beginning in June (Gloersen et al., 1992; Leventer et al., 1996). Micropaleontologic data from the work of Leventer et al. (1996) on a 9-m piston core has revealed circulation and climate patterns for the past 3700 yr in the Palmer Deep. The benthic foraminifer assemblage is dominated by two taxa, Bulimina aculeata and Bolivina pseudopunctata, which are inversely related. High relative abundances of B. aculeata occur cyclically over a period of ~230 yr. The assemblage associated with high abundance of B. aculeata in Palmer Deep resembles that from the Bellingshausen shelf, which is associated with CDW. In addition to the faunal evidence, hydrographic data indicate incursions of CDW into Palmer Deep (Leventer et al., 1996). A distinctive diatom assemblage dominated by a single genus was associated with peaks in B. aculeata, whereas a few different assemblages were associated with lows in B. aculeata. Leventer et al. (1996) interpreted the variability in diatom assemblages as an indication of changes in productivity associated with changes in water column stability. Abelmann and Gowing (1997) studied the horizontal and vertical distributions of radiolarians in the Atlantic sector of the Southern Ocean. They show that the spatial distribution of radiolarian assemblages reflects hydrographic boundaries. In a transect from the subtropical Atlantic to polar Antarctic zones, radiolarians in the upper 1000 m of the water column occurred in distinct surface and deep-living assemblages related to water depth, temperature, salinity, and nutrient content. Living assemblages resembled those preserved in underlying surface sediments (Abelmann and Gowing, 1997). Circumantarctic coastal sediments from neritic environments contained a distinctive assemblage dominated by the Phormacantha hystrix/Plectacantha oikiskos group and Rhizoplegma boreale (Nishimura et al., 1997). Low diversity and species compositions distinguished the coastal sediments from the typical pelagic Antarctic assemblages. Factors that controlled the assemblages were water depth, proximity to the coast, occurrence of sea ice, and steepness of topography, rather than temperature and salinity. Nishimura et al. (1997) found a gradient of sorts from deep-water sites containing diverse assemblages typical of pelagic environments to coastal sites with low diversity assemblages dominated by P. hystrix/P. oikiskos group and R. boreale. In general, sites between these two extremes had increased proportions of the coastal assemblage with decreasing water depth (Nishimura et al., 1997). At a site near Hole 1098 (GC905), they showed that the relative abundance of the coastal assemblage increased downcore (Nishimura et al., 1997). The purpose of the research presented here was to make a cursory investigation into the radiolarian assemblages as possible paleoenvironmental indicators.

Concentrations of Ir have been measured in 87 sediment samples from Ocean Drilling Program Site 1096 in search of evidence of fallout from the impact of the Eltanin asteroid, which occurred at 2.15 Ma, ~1300 km northwest of the site. An additional six samples were measured from a unique sand layer and adjacent sediments that are dated at ~1.6 Ma. These 93 sediment samples are all silts and muds that were deposited on a continental rise drift of the Antarctic Peninsula. No evidence of the Eltanin impact deposit was found in this study.

Background: Lifetimes of nuclear excited states in fission fragments have been studied in the past following isotope separation, thus giving access mainly to the fragments' daughters and only to long-lived isomeric states in the primary fragments. For the first time now, short-lived excited states in the primary fragments, produced in neutron-induced prompt fission of U-235 and Pu-241, were studied within the EXILL&FATIMA campaign at the intense neutron-beam facility of the Institute Laue-Langevin in Grenoble. Purpose: We aim to investigate the quadrupole collective properties of neutron-rich even-even Xe-138,Xe-140,Xe-142 isotopes lying between the double shell closure N = 82 and Z = 50 and a deformed region with octupole collectivity. Method: The gamma rays emitted from the excited fragments were detected with a mixed array consisting of 8 HPGe EXOGAM Clover detectors (EXILL) and 16 LaBr3(Ce) fast scintillators (FATIMA). The detector system has the unique ability to select the interesting fragment making use of the high resolution of the HPGe detectors and determine subnanosecond lifetimes using the fast scintillators. For the analysis the generalized centroid difference method was used. Results: We show that quadrupole collectivity increases smoothly with increasing neutron number above the closed N = 82 neutron shell. Our measurements are complemented by state-of-the-art theory calculations based on shell-model descriptions. Conclusions: The observed smooth increase in quadrupole collectivity is similar to the evolution seen in the measured masses of the xenon isotopic chain and is well reproduced by theory. This behavior is in contrast to higher Z even-even nuclei where abrupt change in deformation occurs around N = 90.

Numerose osservazioni astrofisiche e cosmologiche compiute a partire dagli anni '30 confermano che circa il 26% dell'Universo è costituito da materia oscura. Tale materia ha la particolarità di interagire solo gravitazionalmente e, forse, debolmente: essa si presenta massiva e neutra. Tra le numerose ipotesi avanzate riguardanti la natura della materia oscura una delle più accreditate è quella delle WIMP (Weakly Interacting Massive Particle). Il progetto all'avanguardia nella ricerca diretta delle WIMP è XENON presso i Laboratori Nazionali del Gran Sasso (LNGS). Tale esperimento è basato sulla diffusione elastica delle particelle ricercate su nuclei di Xeno: il rivelatore utilizzato è una TPC a doppia fase (liquido-gas). La rivelazione diretta di materia oscura prevede l'impiego di un rivelatore molto grande, a causa della piccola probabilità di interazione, e di ambienti a bassa radioattività naturale, per ridurre al minimo il rumore di fondo. Risulta necessario inoltre l'utilizzo di uno schermo attivo che individui particelle di alta energia, in particolare muoni cosmici, che possono produrre falsi segnali. È stato realizzato a tale scopo un sistema di Muon Veto composto da un grande cilindro d'acqua posto attorno alla TPC, equipaggiato con 84 fotorivelatori atti ad osservare i fotoni ottici emessi per effetto Čherenkov dai raggi cosmici. Il presente lavoro di tesi si colloca nell'ambito di un programma di simulazione Monte Carlo, creato per realizzare virtualmente l'esperimento XENON1T e per effettuare studi preliminari. Lo scopo di tale lavoro è stato quello di contribuire alla scrittura e alla verifica del codice di simulazione e allo studio di eventi di muoni cosmici da esso generati. L'analisi dati è stata effettuata scrivendo un programma in C++ in grado di analizzare i risultati forniti dal simulatore e di generare degli Event Display statici e dinamici per una visualizzazione efficace degli eventi.

Early detection of right ventricular (RV) involvement in chronic pulmonary hypertension (PH) is essential due to prognostic implications. T1 mapping by cardiac magnetic resonance (CMR) has emerged as a noninvasive technique for extracellular volume fraction (ECV) quantification. We assessed the association of myocardial native T1 time and equilibrium contrast ECV (Eq-ECV) at the RV insertion points with pulmonary hemodynamics and RV performance in an experimental model of chronic PH. Right heart catheterization followed by immediate CMR was performed on 38 pigs with chronic PH (generated by surgical pulmonary vein banding) and 6 sham-operated controls. Native T1 and Eq-ECV values at the RV insertion points were both significantly higher in banded animals than in controls and showed significant correlation with pulmonary hemodynamics, RV arterial coupling, and RV performance. Eq-ECV values also increased before overt RV systolic dysfunction, offering potential for the early detection of myocardial involvement in chronic PH.

O trabalho apresentado, foi realizado no âmbito do mestrado em Química para o ensino, da Universidade de Ãvora e tem como tema " Moléculas Simples de Aplicação na Medicina". A abordagem deste tema deve-se à necessidade cada vez maior de um ensino que motive os alunos e lhes proporcione um gosto maior pelo estudo das ciências, uma vez que cada vez mais os jovens estudantes revelam pouca motivação e gosto pelo estudo das áreas científicas quer ao nível do ensino secundário quer ao nível do ensino superior. Pretende-se também tentar compreender e explicar o porquê das suas desmotivações e tentar adaptar novas abordagens a temas que suscitem nos jovens um maior interesse, uma melhor compreensão da importância das ciências, da tecnologia na vida quotidiana de todos nós no que respeita ao nosso bem-estar e à nossa saúde. Numa primeira parte é feita uma revisão sobre os principais conteúdos propostos a desenvolver em sala de aula, conteúdos esses que sustentam o estudo do xénon como anestésico e dos perfluorocarbonetos como substitutos do sangue. A segunda parte integra um programa orientador, seguindo as metodologias e estrutura do programa homologado pelo ME para o 12º ano de escolaridade, e protocolos de algumas actividades de carácter experimental laboratorial propostas. Estas actividades têm como finalidade estimular a curiosidade e facilitar a consolidação das matérias. ABSTRACT: The present study was carried out under the Master's Degree in Chemistry for Teaching, taught at the University of Ãvora under the theme "lmplementation of Simple Molecules in Medicine". The approach to this issue is due to an increasing need for teaching that motivates students and sharpen their senses for the study of science because, increasingly, young students show little motivation and passion for the study of science, both at secondary school and higher education terms. The aim is also trying to understand and explain the reason of their discouragement and try new approaches to issues that raise young in a better understanding of the importance of science and technology in everyday life for all of us in what concerns to our welfare and our health. ln the first part there is an overview of the major content areas to develop in the classroom. These contents support the study of xenon as an anesthetic and perfluorocarbons as blood substitutes. The second part includes the syllabus, following the methodologies and structure of the curriculum approved by the ME to the 12th grade and protocols of some activities for an experimental laboratory. These activities intend to stimulate curiosity and facilitate the consolidation of materials.

This work thesis focuses on the Helicon Plasma Thruster (HPT) as a candidate for generating thrust for small satellites and CubeSats. Two main topics are addressed: the development of a Global Model (GM) and a 3D self-consistent numerical tool. The GM is suitable for preliminary analysis of HPTs with noble gases such as argon, neon, krypton, and xenon, and alternative propellants such as air and iodine. A lumping methodology is developed to reduce the computational cost when modelling the excited species in the plasma chemistry. A 3D self-consistent numerical tool is also developed that can treat discharges with a generic 3D geometry and model the actual plasma-antenna coupling. The tool consists of two main modules, an EM module and a FLUID module, which run iteratively until a steady state solution is converged. A third module is available for solving the plume with a simplified semi-analytical approach, a PIC code, or directly by integration of the fluid equations. Results obtained from both the numerical tools are benchmarked against experimental measures of HPTs or Helicon reactors, obtaining very good qualitative agreement with the experimental trend for what concerns the GM, and an excellent agreement of the physical trends predicted against the measured data for the 3D numerical strategy.

La Materia Oscura (Dark Matter, DM) deve il suo nome al fatto che non interagisce elettromagneticamente, ma solo gravitazionalmente e debolmente (proprietà che ne complica particolarmente la rivelazione). Molti sforzi, sia sperimentali che teorici, sono stati dedicati alla sua ricerca a tal punto da essere considerata uno dei più grandi misteri della fisica moderna. I candidati più promettenti a costituire la materia oscura sono da ricercarsi in teorie oltre il Modello Standard e fra essi figurano le WIMPs (Weakly Interacting Massive Particles). Poiché le particelle di DM sono caratterizzate da sezioni dâurto molto piccole, per poterle osservare si necessita di grandi rivelatori, ultrapuri e situati in ambienti a bassa radioattività. XENONnT è attualmente uno degli esperimenti più sensibili al mondo per la ricerca diretta di WIMPs, grazie allâutilizzo di una camera di proiezione temporale (TPC) a doppia fase (liquido-gas) che presenta una massa bersaglio di 5.9 t di xenon liquido (LXe). XENONnT mira a rilevare lo scattering di WIMPs contro i nuclei bersaglio, sfruttando i segnali osservabili di luce e carica prodotti dai rinculi nel LXe. Per raggiungere tale risultato sono fondamentali il sistema di veto di neutroni (Neutron Veto, NV) ed il sistema di veto di muoni (Muon Veto, MV): il NV ha lo scopo di rivelare i neutroni radiogenici provenienti dai materiali dellâesperimento mentre il MV (già impiegato in XENON1T) ha la finalità di eliminare il rumore di fondo dovuto ai raggi cosmici. L'obiettivo di questa tesi è lo studio degli eventi muone osservati dal MV ed il NV di XENONnT; misurarne il rate e verificare la congruenza dei risultati ottenuti con quanto atteso da simulazioni Monte Carlo. Questa prova ha costituito un'ulteriore verifica del corretto funzionamento dei sistemi di veto, ed ha permesso di approfondire la conoscenza sulla risposta del Neutron Veto al passaggio di un muone allâinterno del suo volume attivo.

This study aimed to identify novel biomarkers for thyroid carcinoma diagnosis and prognosis. We have constructed a human single-chain variable fragment (scFv) antibody library that was selected against tumour thyroid cells using the BRASIL method (biopanning and rapid analysis of selective interactive ligands) and phage display technology. One highly reactive clone, scFv-C1, with specific binding to papillary thyroid tumour proteins was confirmed by ELISA, which was further tested against a tissue microarray that comprised of 229 thyroid tissues, including: 110 carcinomas (38 papillary thyroid carcinomas (PTCs), 42 follicular carcinomas, 30 follicular variants of PTC), 18 normal thyroid tissues, 49 nodular goitres (NG) and 52 follicular adenomas. The scFv-C1 was able to distinguish carcinomas from benign lesions (P=0.0001) and reacted preferentially against T1 and T2 tumour stages (P=0.0108). We have further identified an OTU domain-containing protein 1, DUBA-7 deubiquitinating enzyme as the scFv-binding antigen using two-dimensional polyacrylamide gel electrophoresis and mass spectrometry. The strategy of screening and identifying a cell-surface-binding antibody against thyroid tissues was highly effective and resulted in a useful biomarker that recognises malignancy among thyroid nodules and may help identify lower-risk cases that can benefit from less-aggressive management.

Nearly 50% of patients with heart failure (HF) have preserved LV ejection fraction, with interstitial fibrosis and cardiomyocyte hypertrophy as early manifestations of pressure overload. However, methods to assess both tissue characteristics dynamically and noninvasively with therapy are lacking. We measured the effects of mineralocorticoid receptor blockade on tissue phenotypes in LV pressure overload using cardiac magnetic resonance (CMR). Mice were randomized to l-nitro-Ï-methyl ester (l-NAME, 3 mg/mL in water; n=22), or l-NAME with spironolactone (50 mg/kg/day in subcutaneous pellets; n=21). Myocardial extracellular volume (ECV; marker of diffuse interstitial fibrosis) and the intracellular lifetime of water (Ïic; marker of cardiomyocyte hypertrophy) were determined by CMR T1 imaging at baseline and after 7 weeks of therapy alongside histological assessments. Administration of l-NAME induced hypertensive heart disease in mice, with increases in mean arterial pressure, LV mass, ECV, and Ïic compared with placebo-treated controls, while LV ejection fraction was preserved (>50%). In comparison, animals receiving both spironolactone and l-NAME (l-NAME+S) showed less concentric remodeling, and a lower myocardial ECV and Ïic, indicating decreased interstitial fibrosis and cardiomyocyte hypertrophy (ECV: 0.43 ± 0.09 for l-NAME versus 0.25 ± 0.03 for l-NAME+S, P<0.001; Ïic: 0.42 ± 0.11 for l-NAME groups versus 0.12 ± 0.05 for l-NAME+S group). Mice treated with a combination of l-NAME and spironolactone were similar to placebo-treated controls at 7 weeks. Spironolactone attenuates interstitial fibrosis and cardiomyocyte hypertrophy in hypertensive heart disease. CMR can phenotype myocardial tissue remodeling in pressure-overload, furthering our understanding of HF progression.
