Increased plasma levels of free fatty acids (FFA) occur in states of insulin resistance such as obesity and type 2 diabetes mellitus. These high levels of plasma FFA are proposed to play an important role for the development of insulin resistance but the mechanisms involved are still unclear. This study investigated the effects of saturated and unsaturated FFA on insulin sensitivity in parallel with mitochondrial function. C2C12 myotubes were treated for 24 h with 0.1 mM of saturated (palmitic and stearic) and unsaturated (oleic, linoleic, eicosapentaenoic, and docosahexaenoic) FFA. After this period, basal and insulin-stimulated glucose metabolism and mitochondrial function were evaluated. Saturated palmitic and stearic acids decreased insulin-induced glycogen synthesis, glucose oxidation, and lactate production. Basal glucose oxidation was also reduced. Palmitic and stearic acids impaired mitochondrial function as demonstrated by decrease of both mitochondrial hyperpolarization and ATP generation. These FFA also decreased Akt activation by insulin. As opposed to saturated FFA, unsaturated FFA did not impair glucose metabolism and mitochondrial function. Primary cultures of rat skeletal muscle cells exhibited similar responses to saturated FFA as compared to C2C12 cells. These results show that in muscle cells saturated FFA-induced mitochondrial dysfunction associated with impaired insulin-induced glucose metabolism. J. Cell. Physiol. 222: 187-194, 2010. The use of liposomes to encapsulate materials has received widespread attention for drug delivery, transfection, diagnostic reagent, and as immunoadjuvants. Phospholipid polymers form a new class of biomaterials with many potential applications in medicine and research. Of interest are polymeric phospholipids containing a diacetylene moiety along their acyl chain since these kinds of lipids can be polymerized by Ultra-Violet (UV) irradiation to form chains of covalently linked lipids in the bilayer. In particular the diacetylenic phosphatidylcholine 1,2-bis(10,12-tricosadiynoyl)- sn-glycero-3-phosphocholine (DC8,9PC) can form intermolecular cross-linking through the diacetylenic group to produce a conjugated polymer within the hydrocarbon region of the bilayer. As knowledge of liposome structures is certainly fundamental for system design improvement for new and better applications, this work focuses on the structural properties of polymerized DC8,9PC:1,2-dimyristoyl-sn-glycero-3-phusphocholine (DMPC) liposomes. Liposomes containing mixtures of DC8,9PC and DMPC, at different molar ratios, and exposed to different polymerization cycles, were studied through the analysis of the electron spin resonance (ESR) spectra of a spin label incorporated into the bilayer, and the calorimetric data obtained from differential scanning calorimetry (DSC) studies. Upon irradiation, if all lipids had been polymerized, no gel-fluid transition would be expected. However, even samples that went through 20 cycles of UV irradiation presented a DSC band, showing that around 80% of the DC8,9PC molecules were not polymerized. Both DSC and ESR indicated that the two different lipids scarcely mix at low temperatures, however few molecules of DMPC are present in DC8,9PC rich domains and vice versa. UV irradiation was found to affect the gel fluid transition of both DMPC and DC8,9PC rich regions, indicating the presence of polymeric units of DC8,9PC in both areas, A model explaining lipids rearrangement is proposed for this partially polymerized system.

Samples of Araucaria area soil from Parana state, Brazil, were separated by particle size fractionation and investigated by electron paramagnetic resonance (EPR) in X-Band of 9.5 GHz at room temperature and 77K, infra-red spectroscopy and X-ray diffractometry. The paramagnetic species in the soil samples were identified by comparison with EPR spectra of some minerals studied recently by our group, several soil types and/or soil components investigated in the literature. The value of g = 2.1 (Delta H = 85 mT) indicated the presence of ferrihydrite. Hematite was identified by g = 2.1 (Delta H = 100 mT) and g = 4.3 for Fe(3+) lines of the concentrated dominium and diluted dominium. Kaolinite was identified by IR and EPR with the resonance at g = 4.3 attributed to Fe(3+) ions in isolated sites of tetrahedral and octahedral symmetry with rhombic distortion. The resonances at g = 3.7 and g = 4.9 were attributed to Fe(3+) in more highly symmetrical environment than rhombic symmetry, but not in axial symmetry. Three signals around g = 2 were attributed to radiation defects, plus additional resonances at g = 2.8 and 9.0. Signals less intense than those at g = 2.1, 3.7, and 6.5, observed for clear grains of soil, were attributed to presence of Fe(3+) in quartz which was identified by IR and XDR. (C) 2011 Elsevier B.V. Five zones along a transect of 180 m were selected for study on the Island of Pai Matos (Sao Paulo, Brazil). Four of the zones are colonised by vascular plants (Spartina SP, Laguncularia LG, Avicennia AV and Rhizophora RH) and were denominated soils, and the other zone, which lacks vegetation, was denominated sediment (SD). The geochemical conditions differed significantly in soils and sediment and also at different depths. The soils were oxic (Eh > 350 mV) or suboxic (Eh: 350-100 mV) at the surface and anoxic (Eh < 100 mV) at depth, whereas in the sediment anoxic conditions prevailed at all depths, but with a lower concentration of sulphides in the pore water and pyrite in the solid fraction. Under these geochemical conditions Fe is retained in the soils, while the Mn tends to be mobilized and lost. The most abundant form of iron oxyhydroxide was lepidocrocite (mean concentration for all sites and depths, 45 +/- 19 mu mol g(-1)), followed by goethite (30 19 mu mol g(-1))and ferrihydrite (19 +/- 11 mu mol g(-1)),with significant differences among the mean concentrations. There was a significant decrease with depth in all the types of Fe oxyhydroxides measured, particularly the poorly crystalline forms. The pyrite fraction was an important component of the free Fe pool (non-silicate Fe) in all soils as well as in the sediment, especially below 20 cm depth (mean concentration for all sites and depths, 60 +/- 54 mu mol CI). Furthermore, the mean concentration of Fe-pyrite for all sites and depths was higher than that obtained for any of the three Fe oxyhydroxides measured. The Fe-AVS was a minor fraction, indicating that the high concentrations of dissolved Fe in the soils in the upper area of the transect result from the oxidation of Fe sulphides during low tide. Mossbauer spectroscopy also revealed that most of the Fe (III) was associated with silicates, in this case nontronite. The presence of crystals of pyrite associated with phyllosilicates in samples from the upper layer of the soils may indicate that pyritization of this form of Fe(III) is more rapid than usually reported for ocean bed sediments. The sequential extraction of Mn did not reveal any clearly dominant fraction, with the Mn-carbonate fraction being the most prevalent, followed by exchangeable Mn and oxides of Mn, whereas pyrite-Mn and Mn associated with crystalline Fe-oxides were present at significantly lower concentrations. The high concentration of dissolved Mn found in the soils in the lower part of the transect is consistent with the fact that the solubility is determined by the carbonate fraction. Unlike for Fe, in the soils in the higher zone, which are subject to intense drainage during low tide, there was loss of Mn, as reflected by the concentration of total Mn. (C) 2008 Elsevier B.V. Results of the study of Argiudolls in two localities (Zarate and Veronica) of the Pampean plain, Argentina, are presented in this contribution. This is a typical area covered by loess. The magnetic studies carried out allowed to determine the presence of detrital magnetite and titanomagnetite, as well as maghemite, pedogenic goethite and superparamagnetic particles (SP). In Veronica soils, a depletion of ferromagnetic minerals is recorded The dominant process in these soils has been the reductive loss of detrital magnetite and titanomagnetite. This is associated with a greater degree of evolution of the soil, which is determined by the concentration and type of detected clays. The higher clay concentration in these soils facilitated reducing conditions and a greater loss of detrital magnetic particles. The loss is reinforced under poor drainage conditions. In the poorly drained soil of Zarate, a concentration of magnetic particles is observed in the Bt horizon, which is associated with an illuviation process. The well drained soil of the same locality shows neoformation of SP particles. These particles would have an ephemeral life until a new wet period in the annual cycle occurs. Although some characteristics of the magnetic signal appear reinforced by the conditions of drainage, this aspect does not seem to be too significant, at least in the Pampean region with low topographic gradients.

Determinations of the volatile elements carbon, hydrogen, sulfur and nitrogen in many geological RM, performed with the LECO CHN and SC analysers, are presented. The method allowed the determination of S in concentrations from a few % m/m to 0.001% m/m or less, of C from % m/m to 0.01% m/m and of H from % m/m to 0.004% m/m. Accuracy was usually better than the XRF method (for S). All obtained values passed the Sutarno-Steger test, which establishes that vertical bar(mean(analysed) - mean(certified))vertical bar/ S(certified) < 2, for the cases with an appropriate number of determinations (n > 10 for each element). It was possible to perform routine determination of C, H and S with the instrumentation, coupled with the determination of major and minor elements in geological materials. Determination of nitrogen could also be performed on an exploratory basis, with improvements in the method dependent on the future availability of more reference materials with reliable composition of this element.

There has been little research on metal concentration levels in urban soils of SA o pound Paulo, a city with 19 million inhabitants with severe pollution problems. In the present study, the concentration of As, Ba, Cr, Cu, Pb, Sb and Zn, were determined by INAA and XRF in surface soil samples from 7 public parks located within SA o pound Paulo city. The results obtained showed that soils of SA o pound Paulo public parks present concentration levels of the studied elements higher than the reference values for soils in SA o pound Paulo State. Traffic related elements such as Cu, Pb and Zn presented high concentrations in parks located near avenues of highly dense traffic and may be associated to vehicular sources.

Microbial community composition was examined in two soil types, Anthrosols and adjacent soils, sampled from three locations in the Brazilian Amazon. The Anthrosols, also known as Amazonian dark earths, are highly fertile soils that are a legacy of pre-Columbian settlement. Both Anthrosols and adjacent soils are derived from the same parent material and subject to the same environmental conditions, including rainfall and temperature; however, the Anthrosols contain high levels of charcoal-like black carbon from which they derive their dark color. The Anthrosols typically have higher cation exchange capacity, higher pH, and higher phosphorus and calcium contents. We used culture media prepared from soil extracts to isolate bacteria unique to the two soil types and then sequenced their 16S rRNA genes to determine their phylogenetic placement. Higher numbers of culturable bacteria, by over two orders of magnitude at the deepest sampling depths, were counted in the Anthrosols. Sequences of bacteria isolated on soil extract media yielded five possible new bacterial families. Also, a higher number of families in the bacteria were represented by isolates from the deeper soil depths in the Anthrosols. Higher bacterial populations and a greater diversity of isolates were found in all of the Anthrosols, to a depth of up to 1 m, compared to adjacent soils located within 50-500 m of their associated Anthrosols. Compared to standard culture media, soil extract media revealed diverse soil microbial populations adapted to the unique biochemistry and physiological ecology of these Anthrosols.

Chemical investigation of the crude extract of a marine sponge Dysidea robusta led to the isolation of an inseparable mixture of saturated ceramides. These were identified from spectroscopic data as well as by hydrolysis followed by LC-MS analysis of the sphingosine moieties.

Um equipamento triaxial convencional originalmente utilizado para ensaios em amostras de solos saturados saturada foi modificado para que fosse possível a realização de ensaios triaxiais em solos não saturados. As principais modificações foram a instalação de uma pedra cerâmica de alto valor de entrada de ar (300 kPa) na nova base da câmara triaxial e uma nova linha de aplicação de pressão para aplicação de pressão de ar na amostra. A técnica de translação de eixos é utilizada para evitar cavitação no sistema de aplicação de pressão. O solo estudado nesta pesquisa consiste de um colúvio de arenito localizado nas encostas da Formação Serra Geral, entre os municípios de Timbé do Sul (SC) e São José dos Ausentes (RS). Ensaios triaxiais convencionais foram realizados nas condições drenado e não drenado em amostras indeformadas para tensões de confinamento iniciais variando de 50 até 500 kPa; ensaios triaxiais convencionais foram realizados na condição drenada em amostras remoldadas para as tensões de confinamento iniciais de 50, 100 e 200kPa. Ensaios triaxiais com sucção controlada foram realizados na condição drenado em amostras indeformadas para as tensões normais líquidas de 50, 100, 150 e 200 kPa em níveis de sucção variando de 25 até 150 kPa. A partir dos resultados dos ensaios triaxiais determinou-se os parâmetros de resistência ao cisalhamento: c’ – intercepto coesivo efetivo; f´ - ângulo de atrito interno efetivo; fb – ângulo de atrito interno que quantifica a contribuição da sucção na resistência ao cisalhamento. O comportamento encontrado foi de uma envoltória de resistência ao cisalhamento não saturada bilinear com acréscimo na resistência ao cisalhamento para baixos níveis de sucção até aproximadamente 75 kPa, e após um decréscimo na resistência ao cisalhamento. A tensão normal líquida não influencia no valor de fb e a sucção não provoca alteração no valor de f´. O valor de fb apresenta valores superiores ao valor de f´, quando se utiliza o critério de ruptura clássico de resistência ao cisalhamento. Alguns critérios de determinação dos valores máximos de ruptura foram aplicados para um melhor entendimento do comportamento resistente deste material.

O objetivo deste trabalho foi determinar a mobilidade do herbicida sulfentrazone em duas classes de solos (Nitossolo Vermelho e Neossolo Quartzarênico), em função de índices pluviométricos crescentes, sendo, portanto, influenciado pelas propriedades químicas e físicas dos solos com diferentes teores de ferro. em tubos de PVC de 10 cm de diâmetro por 50 cm de comprimento, preenchidos com os solos e saturados com água a 65% (p/p), foi aplicado o sulfentrazone (800 g ha-1). Na seqüência, foram simuladas chuvas diárias de 10 mm até atingir os índices pluviométricos de 30, 60 e 90 mm. Os tubos de PVC foram então desmontados - semeando-se Sorghum bicolor nas profundidades correspondentes de 2,5; 7,5; 12,5; 17,5; 22,5; e 30,0 cm da superfície do tubo - e mantidos em casa de vegetação por 15 dias, para avaliação da germinação e do crescimento inicial das plântulas. No final do experimento foram avaliadas as alterações morfofisiológicas que caracterizavam os efeitos tóxicos do produto, além de se medir o comprimento da parte aérea até a última lígula visível e a fitomassa seca das plantas. Quando sob precipitação de 90 mm no Neossolo Quartzarênico, o sulfentrazone formou uma banda de arraste de até 12,5 cm e, no Nitossolo Vermelho, até os 7,5 cm.

Este trabalho objetivou determinar a mobilidade do sulfentrazone em duas classes de solos em função de índices pluviométricos, bem como possíveis influências das propriedades químicas e físicas de Latossolo Vermelho e Chernossolo com diferentes teores de ferro na ação do herbicida. Foram utilizados como recipientes 36 tubos de PVC de 10 cm de diâmetro por 50 cm de comprimento. Os recipientes foram preenchidos com os solos e umedecidos a 65% (p/p) da capacidade de saturação, quando se fez a aplicação do sulfentrazone (800 g ha-1 de i.a.) na área exposta dos solos. Na seqüência, foram simuladas chuvas diárias de 10 mm até atingir o índice pluviométrico desejado (30, 60 e 90 mm), sendo, posteriormente, desmontados seis tubos de cada solo (com e sem aplicação). Foram semeadas cinco sementes de sorgo (Sorghum bicolor) nas profundidades de 2,5; 7,5; 12,5; 17,5; 22,5; e 30,0 cm, mantidas em casa de vegetação por 15 dias para avaliação da germinação e do crescimento inicial. Decorrido esse tempo, foi realizada avaliação de possíveis alterações morfofisiológicas que pudessem ser caracterizadas como efeitos tóxicos do produto e mediu-se o comprimento da parte aérea até a última lígula visível. As partes aéreas foram secas em estufa com circulação forçada de ar (70 ºC por 96 horas), para obtenção de matéria seca. No Chernossolo ocorreu uniformidade da distribuição do produto ao longo do tubo, proporcional à precipitação, e no Latossolo Vermelho o sulfentrazone foi pouco móvel, permanecendo na camada superficial, independentemente da precipitação.
