Un déséquilibre de la balance énergétique constitue la principale cause du développement des pathologies métaboliques telles que l’obésité et le diabète de type 2. Au sein du cerveau, l’hypothalamus joue un rôle primordial dans le contrôle de la prise alimentaire et du métabolisme périphérique via le système nerveux autonome. Ce contrôle, repose sur l’existence de différentes populations neuronales au sein de l’hypothalamus médio-basal (MBH), neurones à neuropeptide Y (NPY)/Agouti-related peptide (AgRP), et neurones a proopiomelanocortine (POMC), dont l’activité est directement modulée par les variations des taux circulants des nutriments tels que le glucose et les acides gras (FA). Alors que les mécanismes de détection et le métabolisme intracellulaire du glucose ont été largement étudiés, l’implication du métabolisme intracellulaire des FA dans leurs effets centraux, est très peu comprise. De plus, on ignore si le glucose, module le métabolisme intracellulaire des acides gras à longue chaine (LCFA) dans le MBH. Le but de notre première étude est, de déterminer l'impact du glucose sur le métabolisme des LCFA, le rôle de l’AMP-activated protein kinase (AMPK), kinase détectrice du statut énergétique cellulaire, et d'établir s’il y a des changements dans le métabolisme des LCFA en fonction de leur structure, du type cellulaire et de la région cérébrale. Nos résultats montrent que le glucose inhibe l'oxydation du palmitate via l’AMPK dans les neurones et les astrocytes primaires hypothalamiques, in vitro, ainsi que dans les explants du MBH, ex vivo, mais pas dans les astrocytes et les explants corticaux. De plus, le glucose augmente l'estérification du palmitate et non de l’oléate dans les neurones et les explants du MBH, mais pas dans les astrocytes hypothalamiques. Ces résultats décrivent le devenir métabolique de différents LCFA dans le MBH, ainsi que, la régulation AMPK - dépendante de leur métabolisme par le glucose dans les astrocytes et les neurones, et démontrent pour la première fois que le métabolisme du glucose et des LCFA est couplé spécifiquement dans les noyaux du MBH, dont le rôle est critique pour le contrôle de l'équilibre énergétique. Le deuxième volet de cette thèse s’est intéressé à déterminer les mécanismes intracellulaires impliqués dans le rôle de la protéine de liaison ACBP dans le métabolisme central des FA. Nous avons démontré que le métabolisme de l’oléate et non celui du palmitate est dépendant de la protéine ACBP, dans les astrocytes hypothalamiques ainsi que dans les explants du MBH. Ainsi, nos résultats démontrent qu’ACBP, protéine identifiée originellement au niveau central, comme un modulateur allostérique des récepteurs GABA, agit comme un régulateur du métabolisme intracellulaire des FA. Ces résultats ouvrent de nouvelles pistes de recherche liées à la régulation du métabolisme des acides gras au niveau central, ainsi que, la nouvelle fonction de la protéine ACBP dans la régulation du métabolisme des FA au niveau du système nerveux central. Ceci aiderait à identifier des cibles moléculaires pouvant contribuer au développement de nouvelles approches thérapeutiques de pathologies telles que l’obésité et le diabète de type 2.

Introducción: El programa Centros Infantiles de Cultura Productiva (CICP), es propio de Tocancipá, a cargo de la Gerencia de Desarrollo Social Municipal, consiste en centros de atención a niños de 5 a 12 años, SISBEN 1 y 2 que reciben apoyo nutricional y en tareas escolares, dentro de las iniciativas mundiales de desarrollo del Milenio, el Plan Nacional de Desarrollo 2006-2010 y el Plan de Desarrollo Municipal 2008-2011. Metodología: Estudio pre-experimental postest con grupo de comparación para evaluar estado nutricional, de niños del programa CICP en la zona centro del municipio en 2010, evaluó 286 escolares con indicadores antropométricos Talla/Edad, IMC/Edad según los patrones OMS 2007. Clasificación nutricional determinada mediante z score con el software Anthro Plus, prueba t para poblaciones independientes, en software SPSS 15. Resultados: Prevalencia de riesgo de talla baja para la edad 28,7%, de talla baja para la edad 12,9%, sobrepeso 12,6%, obesos 2,4%, riesgo de delgadez 15% p y delgadez el 2,8%, en el total de las observaciones. Análisis de menú ofrecido en el programa CICP en un día de observación evidencia aporte del 100% de requerimientos diarios, sin considerar el consumo de alimentos de cada niño en el hogar. Conclusiones: No existe diferencia estadísticamente significativa entre los grupos. La prevalencia de obesidad en el grupo beneficiario del CICP es 3,9% vs. 1,3% en grupo control, relacionado con aportes nutricionales, lo que contrasta con la proporción de delgadez del 4,7% en grupo CICP vs 1,3% en grupo control. Es necesario realizar un estudio de seguimiento para confirmar hallazgos del presente estudio.

The seeds are excellent sources of proteinase inhibitors and have been highlighted owing to various applications. Among these applications are those in effect on food intake and weight gain that stand out because of the increasing number of obese individuals. This study evaluated the effects of trypsin inhibitor present in the seed of tamarind (Tamarindus indica L.) reduction in weight gain, biochemical and morphological alterations in Wistar rats. For this, we partially purified a trypsin inhibitor tamarind seed. This inhibitor, ITT2 at a concentration of 25 mg / kg body weight, over a period of 14 days was able to reduce food intake in rats (n = 6) by approximately 47%, causing a reduction in weight gain approximately 70% when compared with the control group. With the evaluation of the in vivo digestibility was demonstrated that the animals lost weight due to satiety, presented by the reduction of food intake, since there were significant differences between true digestibility for the control group (90.7%) and the group treated with inhibitor (89.88%). Additionally, we checked the deeds of ITT2 on biochemical parameters (glucose, triglycerides, total cholesterol, high-density lipoprotein, low-density lipoprotein, glutamic-pyruvic transaminase, glutamic oxaloacetic transaminase, gamma glutamyl transferase albumin, globulin, total protein and C-reactive protein) and these, when assessed in the study groups showed no statistically significant variations. We also evaluate the histology of some organs, liver, stomach, intestine, and pancreas, and showed no changes. And to evaluate the effect of trypsin inhibitor on food intake due to the satiety is regulated by cholecystokinin (CCK) were measured plasma levels, and it was observed that the levels of CCK in animals receiving ITT2 were significantly higher ( 20 + 1.22) than in animals receiving only solution with casein (10.14 + 2.9) or water (5.92 + 1.15). Thus, the results indicate that the effect caused ITT2 satiety, reducing food intake, which in turn caused a reduction in weight gain in animals without causing morphological and biochemical changes, this effect caused by the elevation of plasma levels CCK

The experiment was developed in Marta Farm, Tremembe city, located in the State of São Paulo - Brazil, using a frogculture commercial project. The aim of this study was evaluate the performance (growth, survival and feed conversion) of bullfrogs Lithobates catesbeianus, post-metamorphosed fed with diets containing different amounts of vitamin C and your performance with the temperature. The animals were kept in 24 boxes of (2.0 x 1.0 m) with an initial density of 50 animals per square meter. We tested the following levels of supplementation in the diet: 0, 250, 500, 750, 1,000 and 2,000 mg vitamin C kg(-1) of diet. The experimental design was completely randomized with six treatments and four replicates. The parameters evaluated were: feed intake, weight gain, feed conversion and survival tax. The animals were weighed at the beginning of the experiment and every 30 days and recorded daily to room temperature. The percentage of food intake decreased with the growth of the frogs, from 3.85 to 1.2% of the live weight, and the mean of temperature decreased from 28 to 20.1 degrees C. It was concluded that there was no influence of vitamin C on the performance (growth, survival and feed conversion) of bullfrogs in the growing stage.

A prevalência da obesidade está aumentando e estudos prospectivos mostram que, em 2025, o Brasil será o quinto país do mundo a apresentar problemas de obesidade em sua população. A etiologia da obesidade não é de fácil identificação, uma vez que a mesma é caracterizada como uma doença multifatorial, ou seja, diversos fatores estão envolvidos em sua gênese, incluindo fatores genéticos, psicológicos, metabólicos e ambientais. Pesquisas recentes na área de metabolismo mostram que o adipócito é capaz de sintetizar várias substâncias e, diferentemente do que se supunha anteriormente, o tecido adiposo não é apenas um sítio de armazenamento de triglicérides, é hoje considerado um órgão endócrino. Dentre as diversas substâncias sintetizadas pelo adipócito, destacam-se a adiponectina, a angiotensina e a leptina. A leptina é um petídeo que desempenha importante papel na regulação da ingestão alimentar e no gasto energético, gerando um aumento na queima de energia e diminuindo a ingestão alimentar. Além dos avanços no estudo da célula adiposa, um novo hormôrnio relacionado ao metabolismo foi descoberto recentemente, a grelina. A grelina é um peptídeo produzido nas células do estômago, e está diretamente envolvida na regulação do balanço energético a curto prazo. Assim, este artigo abordará o papel da leptina e da grelina no controle do peso corporal e as limitações que ainda existem para tratar a obesidade em humanos.

Objectives: To evaluate the association between the consumption of different dietary fats with the quality of the diet, insulin resistance, and hyperhomocysteinemia in adults. Methods: Cross-sectional study conducted with 624 overweight subjects (73.7% females). Assessments of food intake (24h food recall and health eating index-HEI), anthropometry, and biochemical assays of fasting glucose, insulin (HOMA-IR and β calculus) and homocysteinemia were performed. Results: The low quality of diet was associated with the vegetable oil at 3rd quintile (≥1.5-2.0 servings) showed risk 2.9 times and cholesterol at quintiles 2nd, 3rd, and 4th was 2.0 times. HOMA-IR was higher at 5th quintile of saturated fat (≥10,7% - total caloric value) with risk of 60% and hyperhomocysteinemia the vegetable oil at 3rd quintile (>1.5-2.0 servings) with risk of 12.0 times and 5th (≥3.5 servings) 7.1 times. However, significance disappeared when adjusted for anthropometric variables. Conclusion: Dietary fats were associated with the harm diet quality, insulin resistance, and hyperhomocysteinemia. However, associations are dependant of demographic variables, dietetic, and nutritional state. © 2011 CELOM.

Brain insulin has had widespread metabolic, neurotrophic, and neuromodulatory functions and has been involved in the central regulation of food intake and body weight, learning and memory, neuronal development, and neuronal apoptosis. Purpose: The present study investigated the role of swimming training on cerebral metabolism on insulin concentrations in cerebellum and the body balance performance of diabetic rats. Methods: Forty Male Wistar rats were divided in four groups: sedentary control (SC), trained control (TC), sedentary diabetic (SD), and trained diabetic (TD). Diabetes was induced by alloxan (32 mg kg b.w.), single dose injection. The mean blood glucose of diabetic groups was 367 ± 40 mg/dl. Training program consisted in swimming 5 days/week, 1 h/day, 8 weeks, supporting a workload corresponding to 90% of maximal lactate steady state (MLSS). For the body balance testing rats were trained to traverse for 5 min daily for 5-7 days. All dependent variables were analyzed by one-way analysis of variance (ANOVA) and a significance level of p < 0.05 was used for all comparisons. Results: The body balance testing scores were different between groups. Insulin concentrations in cerebellum were not different between groups. Conclusion: It was concluded that in diabetic rats, aerobic training does not induce alterations on cerebellum insulin but induces important metabolic, hormonal and behavioral alterations which are associated with an improvement in glucose homeostasis, serum insulin concentrations and body balance. © 2013 Elsevier Inc.

Este estudo investigou os efeitos nutricionais resultantes da administração crônica via oral do óleo de Plukenetia polyadenia em ratos sobre o peso, a ingesta de ração, a composição lipídica do sangue, e a histologia dos órgãos, além de investigar a toxicidade e a atividade antinociceptiva após administração de diferentes dosagens via oral em camundongos. A análise nutricional de peso e ingesta de ração mostrou que a administração crônica do óleo não interferiu nutricionalmente na homeostasia destas variáveis, independente das dosagens utilizadas (100 e 200mg/kg). As frações lipídicas do sangue como colesterol total, LDL, HDL e triglicerídeos também não sofreram alterações nos animais suplementados. Não foi encontrado edema, morte celular ou resposta inflamatória nas células hepáticas, cardíacas, pulmonares, gástricas, intestinais, pancreáticas e renais após a suplementação crônica em ambas as dosagens. No teste de contorções abdominais induzidas por ácido acético, o óleo administrado via oral (25, 50 e 100mg/kg) reduziu significantemente o número de contorções em comparação com o controle. No teste da placa quente, o tratamento com o óleo na dosagem de 200mg.kg/peso não induziu alterações no tempo de latência ao estímulo térmico, quando comparado com o controle. No teste da formalina, o tratamento oral nas doses de 50 e 100mg.kg/peso, mostrou um efeito antinociceptivo significativo, reduzindo o tempo de lambida somente na segunda fase (inflamatória). O óleo na dosagem de 200mg/kg não afetou a atividade comportamental dos ratos submetidos ao teste de campo aberto e não apresentou efeitos tóxicos. Nossos resultados demonstram que o óleo de Plukenetia polyadenia, apresentou atividade antinociceptiva sem promover toxicidade.

The obesity along with overweight has been considered one of the most serious public health problems in the world, especially because they are the main risk factors for many chronic diseases such as coronary heart disease, diabetes mellitus, hypertension and some types of tumors, which are associated with high mortality rates. The use of functional foods and appropriate diets to promote health has grown as a prevention mechanism. Previous studies suggested Bidens pilosa L. for the treatment of disorders associated with obesity, with one important attraction of this regiment is perceived as safer and more effective health than the traditional anorexigen .The aim of this project is to standardize a new model of obesity induced by hipercaloric diet in Swiss mice, and also to evaluate the effect of Bidens pilosa L. extract in the reduction of food intake, weight and toxicity. After induction of obesity, the animals were treated for 21 days with the extract. Then the animals were killed and samples were collected for further analysis. Preliminary, the effect of this product has being evaluated on the daily food intake and body weight. In dose of 100mg/Kg, there were no significant changes in food intake and body weight. Thus, this concentration of Bidens pilosa was not effective in reducing body weight of obese animals, and did not reduce food intake

Protease inhibitors (PIs), part of HAART (Highly Active Antiretroviral Therap) are selective, competitive inhibitors of protease, a crucial enzyme to viral maturation, infection and replication. A lipodystrophic syndrome has been reported in individuals treated with HAART, and associated to hyperglycemia, hypercholesterolemia, hypertrigliceridemia, hyperlipidemia, hypertension and hypreinsulinemia. The HAART-associated metabolic abnormalities were first associated with protease inhibitors, Ritonavir mostly, but the mechamisns that underlie these metabolic alterations are to date, not completely understood. Since Pis are candidate to be the drug of choice for other diseases treatment, such as the Hepatitis C, malaria and some types of cancer, it seems to be important to clarify the metabolic alterations associated to PIs. Wistar rats were treated twice a week with 30mg/kg Ritonavir for 4 and 8 weeks. Total cholesterol, HDL, LDL, VLDL, triglycerides and glycemic levels were measured by the end of each period of time selected. To avoid confunding effects of food intake, the animals were fasted 16 hours before. Our results showed rapid increase in serum triglycerides, total cholesterol, LDL-C and glycemic levels. No significant differences were observed for HDL-C or VLDL serum levels. Our study addresses the importance to observe the possible family history of dyslipidemia or diabetes, and control any other cardiovascular and diabetes risk factors when using protease inhibitors
