The role of the mitogen-activated protein kinase kinase (MKK)/extracellular-activated protein kinase (ERK) pathway in mitotic Golgi disassembly is controversial, in part because Golgi-localized targets have not been identified. We observed that Golgi reassembly stacking protein 55 (GRASP55) was phosphorylated in mitotic cells and extracts, generating a mitosis-specific phospho-epitope recognized by the MPM2 mAb. This phosphorylation was prevented by mutation of ERK consensus sites in GRASP55. GRASP55 mitotic phosphorylation was significantly reduced, both in vitro and in vivo, by treatment with U0126, a potent and specific inhibitor of MKK and thus ERK activation. Furthermore, ERK2 directly phosphorylated GRASP55 on the same residues that generated the MPM2 phospho-epitope. These results are the first demonstration of GRASP55 mitotic phosphorylation and indicate that the MKK/ERK pathway directly phosphorylates the Golgi during mitosis.

The cation-pi interaction is an important, general force for molecular recognition in biological receptors. Through the sidechains of aromatic amino acids, novel binding sites for cationic ligands such as acetylcholine can be constructed. We report here a number of calculations on prototypical cation-pi systems, emphasizing structures of relevance to biological receptors and prototypical heterocycles of the type often of importance in medicinal chemistry. Trends in the data can be rationalized using a relatively simple model that emphasizes the electrostatic component of the cation-pi interaction. In particular, plots of the electrostatic potential surfaces of the relevant aromatics provide useful guidelines for predicting cation-pi interactions in new systems.

The replication initiator protein pi of plasmid R6K is known to interact with the seven iterons of the gamma origin/enhancer and activate distant replication origins alpha and beta (ori alpha and ori beta) by pi-mediated DNA looping. Here we show that pi protein specifically interacts in vitro with the host-encoded helicase DnaB. The site of interaction of pi on DnaB has been localized to a 37-aa-long region located between amino acids 151 and 189 of DnaB. The surface of pi that interacts with DnaB has been mapped to the N-terminal region of the initiator protein between residues 1 and 116. The results suggest that during initiation of replication, the replicative helicase DnaB is first recruited to the gamma enhancer by the pi protein. In a subsequent step, the helicase probably gets delivered from ori gamma to ori alpha and ori beta by pi-mediated DNA looping.

A 20-mer phosphorothioate oligonucleotide (AS1) was designed to hybridize to the message for the rat kidney sodium phosphate cotransporter NaPi-2 close to the translation initiation site. Single intravenous doses of this oligonucleotide were given to rats maintained on a low phosphorus diet to increase NaPi-2 expression. At 3 days after oligonucleotide infusion, rats receiving 2.5 micromol of AS1 exhibited a reduction in renal NaPi-2 to cyclophilin mRNA ratio by 40% +/- 17%, and rats receiving 7.5 micromol of AS1 exhibited a reduction in NaPi-2 to cyclophilin mRNA ratio by 46% +/- 21%. Reversed-sequence AS1 was without effect. The higher dose of 7.5 micromol of AS1 also reduced the rate of phosphate uptake into renal brush border membrane vesicles and the expression of NaPi-2 protein detected by Western blotting in these vesicles. Reversed sequence AS1 was again without effect on these parameters. These results suggest that systemically infused oligonucleotides can exert antisense effects in the renal proximal tubule.

Este trabalho apresenta uma investigação das propriedades dinâmicas da válvula borboleta eletrônica do sistema de admissão de ar de um motor a combustão interna. O objetivo é utilizar um modelo matemático da válvula borboleta com todos os seus parâmetros identificados para efetuar o controle da válvula e aplica-lo em ambiente real. Tendo o modelo e os parâmetros identificados, compara-se o comportamento real de uma válvula borboleta com o comportamento simulado, utilizando os mesmos sinais de entrada, de tal forma a validar o modelo desenvolvido. Utilizando o controle Proporcional Integral, foi possível mostrar a aplicabilidade da metodologia que pode ser estendida para projetos de controle mais complexos. Com o controle PI, o modelo matemático é validado e pode ser utilizado como base para projetos de controle mais complexos. O modelo aqui desenvolvido representa satisfatoriamente a dinâmica da válvula borboleta, sendo possível utilizá-lo em outros tipos de válvula borboleta.

Hoy día vivimos en la sociedad de la tecnología, en la que la mayoría de las cosas cuentan con uno o varios procesadores y es necesario realizar cómputos para hacer más agradable la vida del ser humano. Esta necesidad nos ha brindado la posibilidad de asistir en la historia a un acontecimiento sin precedentes, en el que la cantidad de transistores era duplicada cada dos años, y con ello, mejorada la velocidad de cómputo (Moore, 1965). Tal acontecimiento nos ha llevado a la situación actual, en la que encontramos placas con la capacidad de los computadores de hace años, consumiendo muchísima menos energía y ocupando muchísimo menos espacio, aunque tales prestaciones quedan un poco escasas para lo que se requiere hoy día. De ahí surge la idea de comunicar placas que se complementan en aspectos en las que ambas se ven limitadas. En nuestro proyecto desarrollaremos una interfaz s oftware/hardware para facilitar la comunicación entre dos placas con distintas prestaciones, a saber, una Raspberry Pi modelo A 2012 y una FPGA Spartan XSA3S1000 con placa extendida XStend Board V3.0. Dicha comunicación se basará en el envío y recepción de bits en serie, y será la Raspberry Pi quien marque las fases de la comunicación. El proyecto se divide en dos partes: La primera parte consiste en el desarrollo de un módulo para el kernel de Linux, que se encarga de gestionar las entradas y salidas de datos de la Raspberry Pi cuando se realizan las pertinentes llamadas de write o read. Mediante el control de los GPIO y la gestión de las distintas señales, se realiza la primera fase de la comunicación. La segunda parte consiste en el desarrollo de un diseño en VHDL para la FPGA, mediante el cual se pueda gestionar la recepción, cómputo y posterior envío de bits, de forma que la Raspberry Pi pueda disponer de los datos una vez hayan sido calculados. Ambas partes han sido desarrolladas bajo licencias libres (GPL) para que estén disponibles a cualquier persona interesada en el desarrollo y que deseen su reutilización.
