This study was designed to observe the effect of tumor conditioned medium (TCM) on the proliferation and apoptosis of human umbilical vein endothelial cells (HUVECs). HUVECs were exposed to TCM from breast carcinoma cell line MDA-MB-231, then we measured their proliferation, apoptosis and cell cycle distribution by MTT and flow cytometery (FCM). Following the stimulation of TCM, HUVECs showed higher pro-mitogenic and anti-apoptotic ability than did the negative control group (ECGF-free medium with 20% FBS), but a similar ability to the positive control group (medium with ECGF and 20% FBS). From these results, we can conclude that breast carcinoma cell line MDA-MB-231 could secret soluble pro-angiogenic factors that induce HUVEC angiogenic switching, including cell cycle progression, proliferation and growth. The role and character of these factors remain to be further studied.

ENGLISH: The Inter-American Tropical Tuna Commission, in cooperation with the Tuna Oceanography Research program of the Scripps Institution of Oceanography, is studying in the Eastern Tropical Pacific Ocean methods of identifying waters of different characteristics that may influence the distribution and behavior of the tropical tunas. One method of attacking the problem has been to attempt to use zooplankton species as biological indicators of water masses. It has been demonstrated that certain zooplankters have ecological affinities that make them useful for identifying and tracing the movements of water masses. In the Eastern Pacific Ocean, Bieri (1957), Lea (1955), Le Brasseur (1959), Sund (1959), and Sund and Renner (1959) have presented evidence that certain species of Chaetognatha possibly can serve as indicators. The present work reports on a study of the distributions of species of Chaetognatha, obtained from various depths by means of horizontal closing-net hauls, in relation to concurrent measurements of temperature, salinity, and dissolved oxygen. Analyses of these data have provided a basis for determining which species are of potential use as biological indicators within the area of the Eastern Pacific considered in this study. SPANISH: La Comisión Interamericana del Atún Tropical, en cooperación con el programa de la "Tuna Oceanography Research" de la Institución Scripps de Oceanografía, viene estudiando en el Océano Pacífico Oriental Tropical métodos para identificar aguas de características diferentes que podrían influir en la distribución y en el comportamiento de los atunes tropicales. Uno de los métodos para abordar el problema ha sido el de intentar la utilización de especies zooplanctónicas como índices biológicos de masas de agua. Se ha demostrado que ciertos organismos del zooplancton tienen afinidades ecológicas, merced a las cuales son útiles para identificar y trazar los movimientos de las masas de agua. Bieri (1957), Lea (1955), Le Brasseur (1959), Sund (1959) y Sund y Renner (1959) presentaron evidencia de que ciertas especies de quetognatos pueden servir, posiblemente, como tales índices en el Océano Pacífico Oriental. El presente trabajo informa sobre un estudio de la distribución de las especies de quetognatos obtenidos de distintas profundidades por medio de la red de plancton que se cierra en lanzamientos horizontales y en relación con mediciones concomitantes de la temperatura, la salinidad y el oxígeno disuelto. El análisis de estos datos ofreció una base para la determinación de las especies que son potencialmente aptas para ser usadas como índices biológicos dentro del área del Pacífico Oriental a la cual se refiere este estudio.

After reviewing the rather thin literature on the subject, we investigate the relationship between aquaculture and poverty based on a case study of five coastal communities in the Philippines. The analysis relies on a data set collated through a questionnaire survey of 148 households randomly selected in these five communities. The methodological approach combines the qualitative analysis of how this relationship is perceived by the surveyed households and a quantitative analysis of the levels and determinants of poverty and inequality in these communities. There is overwhelming evidence that aquaculture benefits the poor in important ways and that it is perceived very positively by the poor and non-poor alike. In particular, the poor derive a relatively larger share of their income from aquaculture than the rich, and a lowering of the poverty line only reinforces this result. Further, a Gini decomposition exercise shows unambiguously that aquaculture represents an inequality-reducing source of income. We believe that the pro-poor character of brackish water aquaculture in the study areas is explained by the fact that the sector provides employment to a large number of unskilled workers in communities characterized by large surpluses of labour. Our results also suggest that the analysis of the relationship between aquaculture and poverty should not focus exclusively on the socio-economic status of the farm operator/owner, as has often been the case in the past. The goal of our study was to understand the spatial and temporal variation in spawning and settlement of gray snapper (Lutjanus griseus) along the West Florida shelf (WFS). Juvenile gray snapper were collected over two consecutive years from seagrass meadows with a benthic scrape and otter trawl. Spawning, settlement, and growth patterns were compared across three sampling regions (Panhandle, Big bend, and Southwest) by using otolith microstructure. Histology of adult gonads was also used for an independent estimate of spawning time. Daily growth increments were visible in the lapilli of snapper 11–150 mm standard length; ages ranged from 38 to 229 days and estimated average planktonic larval duration was 25 days. Estimated growth rates ranged from 0.60 to 1.02 mm/d and did not differ among the three sampling regions, but did differ across sampling years. Back-calculated fertilization dates from otoliths indicated that juveniles in the Panhandle and Big Bend were mainly summer spawned fish, whereas Southwest juveniles had winter and summer fertilization dates. Settlement occurred during summer both years and in the winter of 1997 for the southern portion of the WFS. Moon phase did not appear to be strongly correlated with fertilization or settlement. Histological samples of gonads from adults collected near the juvenile sampling areas indicated a summer spawning period.

In this study, a detailed analysis of both previously published and new data was performed to determine whether complete, or almost complete, mtDNA sequences can resolve the long-debated issue of which Asian mtDNAs were founder sequences for the Native American mtDNA pool. Unfortunately, we now know that coding region data and their analysis are not without problems. To obtain and report reasonably correct sequences does not seem to be a trivial task, and to discriminate between Asian-and Native American mtDNA ancestries may be more complex than previously believed. It is essential to take into account the effects of mutational hot spots in both the control and coding regions, so that the number of apparent Native American mtDNA founder sequences is not erroneously inflated. As we report here, a careful analysis of all available data indicates that there is very little evidence that more than five founder mtDNA sequences entered Beringia before the Last Glacial Maximum and left their traces in the current Native American mtDNA pool.

Protein powders were prepared from processing waste of prawns either by mechanically squeezing the shell and freeze drying the resultant aqueous extract or by treating the shell with 0.5% sodium hydroxide, filtering it and freeze drying the filtrate. Comparative studies on the proximate composition, amino acid profile, consumer acceptability and nutritional quality of the protein powders showed that the product prepared by freeze drying of the press liquor obtained by passing the waste through a hand operated expeller is better in all aspects studied than the product prepared by mild alkali extraction.

An investigation on growth, production and fishery of three Indian major carps: rohu, Labeo rohita, catla, Catla catla and mrigal, Cirrhinus mrigala and three exotic carps: silver carp, Hypophthalmichthys molitrix, grass carp, Ctenopharyngodon idella, and common carp, Cyprinus carpio was carried out in Nasti baor during February to April months. In catch per unit effort (CPUE) study the highest catch/day/person (3.13 kg) and catch/day/gear (40.65 kg) was recorded in the month of March for kochal fishing. In komar fishing catch/day/person (15.08 kg) and catch/day/gear (1206 kg) was also found higher in March. Komar fishing was done only in March and April and its CPUE was greater in both the months than that of kochal. The average recovery rate (combination of all six species) was 37.80 considering the stocking from July month of the previous year. The recovery rate of common carp (54.1) was the highest and lowest (13.90) in case of silver carp. When the recovery was calculated on the basis of one year data and stocking, it was 55.6%. Analysis of production model revealed that the present production (54,806 kg/year) is less than both theoretical production (model I- 85,285 kg/year and model II -75,952 kg/year) estimated. Therefore, it may be concluded that the fish production from Nasti baor could still be increased from the present level of production.

The water and bottom sediments of Lake Victoria (Kenya) were analysed for A1, Fe, Mn, Zn, Pb, Cu, Cr and Cd. The total metal concentrations were determined and their mean variations and distributions discussed. The bottom lake waters showed higher concentration levels than the surface waters. The range of values (in mg/l) in the bottom and surface lake waters were as follows: Surface Waters: A1(0.08 - 3.98), Fe(0.09 - 4.01), Mn(0.02 - 0.10). Zn(0.01 -0.07), Pb(0.001- 0.007), Cu(not detected - 0.006), Cr(not detected - 0.004). Bottom Waters: A1(0.1 0 - 6.59), Fe(0.23 - 9.64), Mn(0.04 - 0.39), Zn(0.01- 0.08), Pb(0.002 - 0.009), Cu(not detected - 0.03). Cr(not detected -0.002). River mouths and shallow areas in the lake showed higher total metal concentrations than offshore deeper areas. Apart from natural metal levels, varied urban activities and wastes greatly contribute to the lake metal pollution as shown by high Pb and Zn levels in sediments, around Kisumu and Homa Bay areas. Other comparatively high values and variations could be attributed to the varied geological characteristics of the lake and its sediments. Compared to the established W.H.O (1984) drinking water standards manganese, aluminium and iron levels were above these limits whereas zinc, lead, chromium, copper and cadmium were below.

We report the effects of accumulated strain by stacking on the surface and optical properties of stacked 1.3 mu m InAs/GaAs quantum dot (QD) structures grown by MOCVD. It is found that the surface of the stacked QD structures becomes more and more undulated with stacking, due to the increased strain in the stacked QD structures with stacking. The photoluminescence intensity from the QD structures first increases as the stacking number increases from 1 to 3 and then dramatically decreases as it further increases, implying a significant increase in the density of crystal defects in the stacked QD structures due to the accumulated strain. Furthermore, we demonstrate that the strain can be reduced by simply introducing annealing steps just after growing the GaAs spacers during the deposition of the stacked QD structures, leading to significant improvement in the surface and optical properties of the structures. Si1-xCx alloys of carbon (C) concentration between 0.6%-1.0% were grown in Si by ion implantation and high temperature annealing. The formation of Si1-xCx alloys under different ion doses and their stability during annealing were studied. If the implanted dose was less than that for amorphizing Si crystals, the implanted C atoms would like to combine with defects produced during implantation and it was difficult to form Si1-xCx alloys after being annealed at 850 degreesC. With the increment of implanted C ion doses, the lattice damage increased and it was easier to form Si1-xCx alloys. But the lattice strain would become saturate and only part of implanted carbon atoms would occupy the substitutional positions to form Si1-xCx alloys as the implanted carbon dose increased to a certain degree. Once Si1-xCx alloys were formed, they were stable at 950 degreesC, but part of their strain would release as the annealing temperature increased to 1 000 degreesC. Stability of the alloys became worse with the increment of carbon concentration in the alloys.

This work represents the nucleotide sequence of the core histone gene cluster from scallop Chlamys farreri. The tandemly repeated unit of 5671 bp containing a copy of the four core histone genes H4, H2B, H2A and H3 was amplified and identified by the techniques of homology cloning and genomic DNA walking. All the histone genes in the cluster had the structures in their 3' flanking region which related to the evolution of histone gene expression patterns throughout the cell cycle, including two different termination signals, the hairpin structure and at least one AATAAA polyadenylation signal. In their 5' region, the transcription initiation sites with a conserved sequence of 5'-PyATTCPu-3' known as the CAP site were present in all genes except to H2B, generally 37-45 bp upstream of the start code. Canonical TATA and CAAT boxes were identified only in certain histone genes. In the case of the promoters of H2B and H2A genes, there was a 5'-GATCC-3' element, which had been found to be essential to start transcription at the appropriate site. After this element, in the promoter of H2B, there was another sequence, 5'-GGATCGAAACGTTC-3', which was similar to the consensus sequence of 5'-GGAATAAACGTATTC-3' corresponding to the H2B-specific promoter element. The presence of enhancer sequences (5'-TGATATATG-3') was identified from the H4 and H3 genes, matching perfectly with the consensus sequence defined for histone genes. There were several slightly more complex repetitive DNA in the intergene regions. The presence of the series of conserved sequences and reiterated sequences was consistent with the view that mollusc histone gene cluster arose by duplicating of an ancestral precursor histone gene, the birth-and-death evolution model with strong purifying selection enabled the histone cluster less variation and more conserved function. Meanwhile, the H2A and the H2B were demonstrated to be potential good marks for phylogenetic analysis. All the results will be contributed to the characterization of repeating histone gene families in molluscs.

Adsorption and activation of small molecules NO, CO and CO+ NO on LaSrCuO4, La2CuO4 and La1.7Th0.3CuO4 which are typical samples in the sence of nonstoichiometric oxygen(lambda) anrong the series of La2-x(SrTh)(x)CuO4 +/-lambda mixed oxide catalysts were studied by means of MS- TPD (TPSR ), XRD, chemical analysis and so on. It was shown that the adsorption amount of NO can be correlated with the content of oxygen vacancy while the types and strength of adsorption of NO could be related to the oxidation state of the metallic ion. It was also found that CO molecule was first converted into CO32- and then desorbed in the form of CO2 at high temperature during the adsorption and desorption of CO on the mixed oxide with oxygen vacancy. The fact that the profiles of TPD(TPSR) of NO in co-adsorption of NO+CO and in single NO adsorption are similar shows that the adsorption of NO molecule not only has some priority to that of CO but also is stronger than that of CO. It seems that the adsorption of NO plays a dominate role in the activation and decomposition of NO.

Edwardsiella tarda is an important Gram-negative enteric pathogen affecting both animals and humans. It possesses a type III secretion system (T3SS) essential for pathogenesis. EseB, EseC and EseD have been shown to form a translocon complex after secretion, while EscC functions as a T3SS chaperone for EseB and EseD. In this paper we identify EscA, a protein required for accumulation and proper secretion of another translocon component, EseC. The escA gene is located upstream of eseC and the EscA protein has the characteristics of T3SS chaperones. Cell fractionation experiments indicated that EscA is located in the cytoplasm and on the cytoplasmic membrane. Mutation with in-frame deletion of escA greatly decreased the secretion of EseC, while complementation of escA restored the wild-type secretion phenotype. The stabilization and accumulation of EseC in the cytoplasm were also affected in the absence of EscA. Mutation of escA did not affect the transcription of eseC but reduced the accumulation level of EseC as measured by using an EseC-LacZ fusion protein in Ed. tarda. Co-purification and co-immunoprecipitation studies demonstrated a specific interaction between EscA and EseC. Further analysis showed that residues 31-137 of EseC are required for EseC-EscA interaction, Mutation of EseC residues 31-137 reduced the secretion and accumulation of EseC in Ed. tarda. Finally, infection experiments showed that mutations of EscA and residues 31-137 of EseC increased the LD50 by approximately 10-fold in blue gourami fish. These results indicated that EscA functions as a specific chaperone for EseC and contributes to the virulence of Ed. tarda.
