The study of the dental pulp can be extended from factors related to its aggression to those related to new concepts of regeneration. The purpose of this compilation of studies is to present the evolution of a research subject from damage to repair. Innitially, studies will demonstrate the ability of dental procedures to generate heat and consequently affect the dental pulp. In sequence, studies will also present some effects of different pulp capping materials on dental pulp cells, related to the cytotoxicity of these materials and inflammatory potential. Finally, as the subject is emmerging and gaining importance in the literature, this compilation will present data from recent studies on the role of dental pulp progenitor cells in the regeneration and repair of dental pulp, as well as an alternative for a scaffold that could be used for clinical translation of research in the field. In summary, dentists must be aware of these different aspects and that the knowledge on factors and mechanisms involved in the aggression of the dental pulp can also serve as basis for understanding aspects for regeneration.

Casearia sylvestris Swartz (Salicaceae), conhecida como guaçatonga, é uma espécie vegetal distribuída amplamente pela América do Sul, ocorrendo em 22 estados brasileiros. Na medicina popular é utilizada no tratamento de picadas de cobra, úlceras gástricas, como cicatrizante, anti-inflamatório e antisséptico tópico. Extratos de suas folhas demonstraram atividade antiulcerogênica, anti-inflamatória e citotóxica em células tumorais, dentre outras. Diversos metabólitos secundários foram isolados e/ou identificados principalmente de suas folhas, incluindo monoterpenos, sesquiterpenos, diterpenos, compostos fenólicos, etc. Apesar do extenso conhecimento sobre ações farmacológicas e sobre a fitoquímica das folhas, há apenas um trabalho publicado sobre metabólitos secundários de suas flores. Portanto, foi proposto neste trabalho realizar análises cromatográficas e espectrométricas de extratos de folhas e flores de C. sylvestris Swartz com o objetivo de comparar os perfis químicos destes órgãos com ênfase em terpenos. Os extratos produzidos com diferentes solventes (hexano: acetato de etila: isopropanol; hexano e etanol 70 %) de folhas e flores foram obtidos por sonicação com rendimentos variando entre 0,03 e 23,39% (m/m), sendo os maiores rendimentos obtidos com etanol 70 %. Os óleos essenciais de folhas e flores foram obtidos por extração por arraste a vapor d`água e foram analisados por cromatografia em fase gasosa e o componente majoritário apresentado foi o biciclogermacreno, tanto em folhas como em flores; o óleo essencial de flores demonstrou ser mais complexo. Os extratos foram analisados por cromatografia em camada delgada e cromatografia líquida de alta eficiência com detector de arranjo de diodos. Os dados de cromatografia em camada delgada sugerem um perfil químico praticamente idêntico para folhas e flores, o que não foi confirmado pelas análises por cromatografia líquida de alta eficiência que ...

Inflammation is an immune complex-related tissue damage and / or cell caused by chemical, physical, immunological or microbial. The inflammatory process involves a complex cascade of biochemical and cellular events, including awareness and receptor activation, lysis and tissue repair. In general, tissue damage trigger a local inflammatory response by recruiting leukocytes, which release inflammatory mediators. These substances are able to sensitize nociceptors. After synaptic transmission and signal modulation by nociceptive sensory neurons, these signals are perceived as pain. Pain is an experience that involves multiple factors. The route of the supraspinal pain control originates in many brain regions, such as substance periarquedutal gray (PAG), median raphe nucleus and rostral ventromedial medulla (RVM) and have a critical role in determining the chronic and acute pain. Anti-inflammatory drugs (NSAIDs) are used to control inflammation, which inhibit the inflammatory mediators, but can cause side effects such as stomach ulcers and cardiovascular damage. An alternative for the treatment of pain and inflammation is the use of plant species. The genus Eugenia belongs to the family Myrtaceae, one of the largest botanical families of expression in the Brazilian ecosystems. From the pharmacological point of view, studies of similar species crude extracts showed the presence of anti-inflammatory, analgesic, antifungal, hypotensive, antidiabetic and antioxidant activity of some species. As a class of importance in therapeutic phytochemical, the flavonoids has represented an important group with significant anti-inflammatory and gastroprotective, and are present in a significant way in the chemical composition of genus Eugenia. The intervertebral disc disease is considered a major neurological disorder involving the spinal cord, being a disease commonly seen in dogs in clinical practice of small animals. The pathophysiology of most discopathies have not been fully elucidated, but it is known that the symptoms observed will depend on the severity of spinal cord compression and location of spinal injury, ranging from localized pain to complete paralysis. Through a detailed clinical examination and laboratory tests, we choose the most appropriate treatment for each patient, which can be conservative, through the use of anti-inflammatories, analgesics and acupuncture or surgical, through decompression techniques. Acupuncture, therapeutic method of ancient Chinese medicine, uses the stimulation of specific points on the body in order to balance the body's physiological. The action mechanism of acupuncture in discopathies has not been fully elucidated, but its analgesic effects, anti-inflammatory and restoration of sensory and motor function are known, showing results as good as surgical therapy

Silicosis, a common type of pneumoconiosis, is an occupational lung disease caused by inhalation of silica dust often with mining activity and thus reaches the miners. The fine and ultrafine silica particles deposited in the alveolar epithelium may lead to the development of progressive massive fibrosis. An increased reactive oxygen species (ROS) production has been proposed to explain the mechanism for induction of pulmonary fibrosis in silicosis. In this situation, alveolar macrophages are activated to phagocytes silica particles deposited in the alveoli. The activated macrophages secrete large amounts of ROS that in turn induce synthesis of fibrotic factors. In addition, the activity of antioxidant enzymes is impaired, which results in increased lipid peroxidation, as well as generating a local inflammatory process. Diffuse pulmonary fibrosis progresses with interstitial collagen deposition. Interstitial collagen overlies small pulmonary arteries and arterioles and thus it is associated with pulmonary hypertension in pulmonary fibrotic diseases. In addition, cytokines and silica particles passing through the respiratory membrane can reach the bloodstream. In this context, the increase in the generation of ROS in the circulation may lead to a reduction in the bioavailability of nitric oxide, an important endothelium-derived relaxing factor. A deficiency in the nitric oxide bioavailability can result in vascular endothelial dysfunction. Moreover, pro-inflammatory cytokines could contribute to the impairment of endothelial function. In the airways, pro-inflammatory cytokines can reduce the smooth muscle responsiveness to β- adrenergic agonists as isoproterenol. Inflammatory bowel disease (IBD) is a chronic and relapsing disease caused by exaggerated response of the immune system. It represents a significant health problem by limiting the quality of life and being the main risk factor for colorectal cancer. Despite of its importance, the high worldwide incidence and being the object of research for several decades, the etiology remains unknown. Studies indicates an interaction between genetic and environmental factors which together with the intestinal microbiota, leads to an uncontrolled immune response. One of the aggravating environmental factors often discussed is stress, as the daily life of the population in general is increasingly rushed. In order to demonstrate the influence of stress on IBD, this study aimed to standardize an experimental model of colitis induced by instillation of a trinitrobenzene sulfonic acid (TNBS) noninflammatory concentration plus exposure to stress that intensify the inflammation. Therefore, an experiment was done to determine what would be the noninflammatory concentration. In this step, four different concentrations of TNBS (1, 6, 12.5 or 40mg/ml) were tested and the lowest concentration capable of inducing a noninflammatory response in the gut was defined as 1 mg/ml. Then, a second experiment was performed which induced colitis and exposed the animals to restraint stress. The results, however, showed that this stimulus was not enough to exacerbate the damage caused by the 1 mg/ml concentration of TNBS in the colon. With some changes in the protocol, the third experiment associated cold and restraint, as well as changes on the day of euthanasia, which occurred immediately after the stress session. The results of myeloperoxidase activity measurement were unexpected due to the noninflammatory concentration of TNBS caused an intestinal inflammation similar to the concentration of 40 mg/ml. O uso disseminado do suco de folhas jovens de trigo (Triticum aestivum L.) para diversos fins terapêuticos foi proposto por Dra. Ann Wigmore há mais de 40 anos nos Estados Unidos e popularizado de tal forma que é utilizado para o tratamento alternativo de doenças autoimunes e gastrointestinais, dentre outras. Baseado nessas informações populares, o presente estudo teve como objetivo avaliar a composição química e a atividade anti-inflamatória intestinal do extrato aquoso de folhas jovens de trigo na fase aguda do modelo experimental de doença inflamatória intestinal induzida por TNBS (ácido trinitrobenzenosulfônico) em ratos. Os animais foram tratados durante cinco dias antes da indução do processo inflamatório, sendo testadas as doses de 100, 50, 25 e 5 mg/Kg e azatioprina como droga de referência. Após 48 horas da indução do processo inflamatório, os animais foram mortos e seus cólons extraídos para a realização dos estudos macroscópicos e bioquímicos. Paralelamente foi realizado um estudo para a caracterização da composição química do extrato, através de testes para a determinação das principais classes de substâncias presentes, bem como o doseamento de compostos fenólicos e atividade sequestradora de radicais livres. O estudo demonstrou que o extrato aquoso de folhas jovens de trigo a 5 mg/Kg apresentou uma tendência em melhorar os parâmetros macroscópicos e bioquímicos avaliados, porém sem diferenças estatísticas. A caracterização fitoquímica do extrato mostrou que o suco de trigo é uma fonte importante de compostos antioxidantes, os quais podem estar relacionados com a melhora no processo inflamatório colônico

O transplante de órgãos é um processo em que se transferem tecidos e/ou órgãos sólidos de um doador vivo ou cadavérico para um hospedeiro. A resposta imune aos aloantígenos, antígenos de um membro da mesma espécie, do doador, caracteriza o processo de rejeição de órgãos. As reações de hipersensibilidade são causadas por respostas imunes controladas inadequadamente, como na formação de complexos antígeno-anticorpo circulantes no Lúpus Eritematoso Sistêmico, ou direcionadas ao tecido do paciente, por resposta de anticorpos, como na Febre Reumática Aguda, e por resposta linfocitária, como na Esclerose Múltipla. A terapia farmacológica para transplantes de órgãos e para reações de hipersensibilidade se baseia na utilização de fármacos imunossupressores, como a prednisona, ciclosporina, anticorpos monoclonais, ácido micofenólico e azatioprina. A talidomida, fármaco com propriedades inibidoras da produção de Fator de Necrose Tumoral Alfa em monócitos, atua como imunossupressor e anti-inflamatório, com atividade de interesse para o tratamento destas condições. Nesse contexto, foi sintetizada, purificada e caracterizada a estrutura química de um novo pró-fármaco planejado para atuar com mecanismo duplo de ação a saber: inibição da inosina 5’-monofosfato desidrogenase e da produção do fator de necrose tumoral alfa.

In this study, the drug indomethacin, a non-steroidal anti-inflammatory indoleacetic acid derivative and the complex of indomethacin and lanthanum (III) in solid form were synthesized and characterized by Thermogravimetry (TGA), Differential Thermal Analysis (DTA), Differential Scanning Calorimetry (DSC) and powder X-ray diffractometry (XRD), infrared vibrational spectroscopy by diffuse reflectance (FTIR) and complexometric titration with EDTA. With the TG curves it was possible to determine the stoichiometry of the complex as La(Ind)3·3.5H2O where Ind is the drug indomethacin. The result of thermal analyzes provided information on the thermal stability, enthalpy of dehydration and thermal behavior of the compounds. The infrared spectrum and with the aid of theoretical calculations suggests that the indomethacin is coordinated by the carboxylate group in the bidentate mode
