We have investigated the effects of L-arginine, D-arginine and L-lysine on airway smooth muscle responsiveness to spasmogens in vitro. Both L-arginine and D-arginine (100 mM) significantly reduced the contractile potency and maximal contractile response to histamine but not to methacholine or potassium chloride in guinea-pig epithelium-denuded isolated trachea. Similarly, the contractile response to histamine was significantly reduced by L-arginine (100 mM) in rabbit epithelium-denuded isolated bronchus. The amino acid L-lysine (100 mM) failed to significantly alter the contractile potency of histamine in guinea-pig isolated trachea (P>0.05). In guinea-pig isolated trachea precontracted with histamine, both L-arginine and D-arginine produced a concentration-dependent relaxation which was not significantly altered by epithelium removal or by the presence of the nitric oxide synthase inhibitor, NG-nitro L-arginine methyl ester (L-NAME; 50 µM). Thus, at very high concentrations, arginine exhibit a non-competitive antagonism of histamine-induced contraction of isolated airway preparations that was independent of the generation of nitric oxide and was not dependent on charge. These observations confirm previous studies of cutaneous permeability responses and of contractile responses of guinea-pig isolated ileal smooth muscle. Taken together, the data suggest that high concentrations of arginine can exert an anti-histamine effect.

RACIONAL: A utilização de anel nas derivações gástricas em Y-de-Roux ainda é motivo de polêmica entre os cirurgiões bariátricos. Não há consenso quanto às suas repercussões em relação à perda ponderal e à manutenção do peso em longo prazo. OBJETIVO: Avaliar a influência do anel sobre a evolução do peso corporal no decorrer de quatro anos após operação bariátrica. MÉTODO: Foram analisadas retrospectivamente 143 mulheres submetidas à derivação gástrica em Y-de-Roux videolaparoscópica pareadas pela utilização ou não do anel de Silastic®. O tempo de seguimento foi de até 48 meses. Os critérios de inclusão foram idade superior a 18 anos, operação bariátrica primária e frequência regular à clínica no período de interesse para a pesquisa. A técnica manteve reservatório gástrico de pequena curvatura, volume estimado em 30 ml. A alça alimentar media 150 cm e a biliar 40 cm a partir do ângulo duodenojejunal. O grupo com anel utilizou anel tubular de Silastic® com comprimento de 6,5 cm, colocado à 2 cm da anastomose gastrojejunal. O anel era fechado por cinco nós com fio de polipropileno em seu interior. Na manhã seguinte ao procedimento cirúrgico as pacientes recebiam líquidos isotônicos; no segundo dia dieta líquida salgada sem resíduos e alta hospitalar no terceiro dia. Dieta pastosa iniciava a partir do 20o dia e sólida no 30o, juntamente com uma drágea diária de polivitamínico. RESULTADOS: O emagrecimento do grupo com anel foi maior que o sem anel em todos os períodos analisados a nível de 10% e de 5% apenas no 3o ano pós-operatório. A proporção das operadas que não atingiram perda do excesso de peso de 50% foi significativamente maior no grupo sem anel que no grupo com anel (31% entre as sem anel e 8% das com anel no 4o ano). Não houve diferença entre os grupos na recuperação tardia do peso perdido na operação. CONCLUSÕES: Os resultados foram favoráveis à utilização do anel ao se analisar exclusivamente a perda de peso.

OBJETIVO: Estudar no implante lobar autólogo a eficiência da anastomose brônquica simples, a perfusão pulmonar e as complicações devido a desproporção doador/receptor. MÉTODOS: Estudou-se 15 cães submetidos à pneumonectomia esquerda, e reimplante do lobo caudal. Estudou-se a perfusão pulmonar e a anastomose brônquica, respectivamente, por cintilografia e por broncografia. O sacrifício ocorreu aproximadamente 200 dias após a cirurgia para o estudo das anastomoses e medidas de volumes e pesos pulmonares. RESULTADOS: Quatro cães foram a óbito, um por deiscência precoce de anastomose brônquica, 1 com infecção e outros 2 por infarto pulmonar devido a oclusão da veia pulmonar no local da anastomose. A perfusão relativa do pulmão direito e esquerdo foram em média respectivamente 72,7% e 27,3%. A broncografia não mostrou nem estenose nem outras alterações na anastomose brônquica. No momento do sacrifício o lobo caudal ocupava totalmente a cavidade pleural sem que houvesse evidentes desvios do mediastino. CONCLUSÕES: O estudo mostrou que a sutura brônquica término-terminal desprotegida não levou a complicações anastomóticas. Não houve complicações pelo fato do lobo implantado ter ocupado somente metade da cavidade pleural. As complicações mais importantes foram o infarto pulmonar e deiscência total da parede torácica. Tanto a broncografia como a cintilografia se mostraram eficientes respectivamente, para o estudo da anastomose brônquica e da perfusão relativa do pulmão funcionante in vivo.

PURPOSE:To assess whether late introduction of a specific COX-2 inhibitor (Meloxicam) can treat and/or prevent the progression of tumors in the stomach of rats submitted to duodenogastric reflux. METHODS: Seventy five male Wistar rats, weighing 150 grams, were submitted to the induction of duodenogastric reflux through the pylorus. At 36 weeks of follow-up were established three experimental groups: DGR36 sacrificed immediately, DGR54 and DGR54MLX both sacrificed at 54th week of follow-up . The animals of the latter group were fed with a rat chow premixed with Meloxicam (2.0 mg/ kg feed; 0.3 mg / kg bw / day) and the other two with standard rat chow. The lesions found in the pyloric mucosa and gastrojejunal anastomosis were analyzed macroscopically and histologically. For statistical analysis was adjusted a generalized linear model assuming a binomial distribution with LOGIT link function. RESULTS: No significant differences were found when comparing the incidences of benign tumor lesions (Adenomatous Hyperplasia), p=0.4915, or malignant (Mucinous Adenocarcinoma), p=0.2731, among groups. CONCLUSION: Late introduction of specific COX-2 inhibitor (Meloxicam) did not treat and was not able to prevent the progression of tumoral lesions induced by duodenogastric reflux in the rat stomachs.
Comparação entre dois fios de sutura não absorvíveis na anastomose traqueal término-terminal em cães

Doze cães sem raça definida, com idade variando entre 1 e 6 anos e peso de 6 a 20kg, foram submetidos a ressecção traqueal e anastomose término-terminal, na qual foram testados os fios poliéster trançado não capilar e náilon monofilamento. Seis animais, cada três com um mesmo tipo de fio de sutura, sofreram a excisão equivalente a três anéis traqueais. Com 15 dias foi executada uma nova intervenção onde se ressecou o equivalente a mais seis anéis, perfazendo um total de nove. Ao final de outros 15 dias foram sacrificados. Os outros seis animais, cada três com um mesmo tipo de fio, foram submetidos à excisão equivalente a três anéis traqueais e mantidos por 43 dias. As traquéias foram avaliadas por exames clínicos, radiográficos, macroscópicos e histopatológicos. O fio de náilon monofilamento apresentou menos reação tecidual do que o poliéster trançado não capilar, promoveu uma anastomose segura e com menor chance de formação de granuloma.

A implantação do ureter no cólon descendente advém da impossibilidade de reimplantá-lo na vesícula urinária, devido a perdas substanciais do tecido ureteral. O presente trabalho avaliou as alterações macroscópicas, microscópicas e laboratoriais em cães submetidos à ureterocoloanastomose esquerda. Foram utilizados 8 cães adultos, hígidos, de ambos os sexos. A técnica operatória consistiu na ligadura do coto distal do ureter próximo à bexiga e na implantação de um curto seg- mento do coto proximal, através de um túnel submucoso, na face antimesentérica do cólon. Realizou-se dosagens séricas de uréia, creatinina, sódio e potássio no pré-operatório, 2, 7, 15 e 30 dias após a cirurgia. Um total de 6 animais foram observados por 30 dias, e outros dois foram observados durante 7 e 180 dias, res- pectivamente. Todos apresentaram fezes moles durante o decorrer do experimento. Ao exame macroscópico, todos os animais apresentaram dilatação ureteral e pielonefrites. Azotemia transitória e hidronefrose foram observadas no animal mantido por 7 dias, e em mais quatro animais, mantidos por 30 dias, também se observou hidronefrose. O animal mantido por 180 dias demonstrou hidronefrose ao exame ultrasono gráfico aos 30 dias de evolução, mas a mesma não foi observada quando se realizou a necropsia. em um animal, mantido por 30 dias , houve um aumento nos níveis de creatinina nos dias 7, 15 e 30, entretanto estes permaneceram dentro dos limites fisiológicos. Nenhuma alteração foi observada no exame histológico da mucosa intestinal exposta à drenagem de urina. As poucas alterações encontradas nas análises laboratoriais não comprometeram a saúde dos animais, e, considerando que não foram encontradas lesões na mucosa intestinal analisada, é possível concluir que a ureterocoloanastomose é um procedimento viável para ser usada por um curto período de tempo.

Com o objetivo de avaliar o efeito do fotoperíodo sobre o desenvolvimento de ovinos até o peso vivo em torno de 38-40kg, foram utilizados 26 cordeiros Ile de France x Corriedale, divididos aleatoriamente em 2 grupos: fotoperíodo longo(18h de luz/ 6h de escuridão) e fotoperíodo controle(12h de luz/ 12h de escuridão). Diariamente, dados sobre consumo alimentar foram coletados e, semanalmente, os animais eram pesados e amostras de sangue eram colhidas para dosagem dos níveis plasmáticos dos hormônios Triiodotironina(T3), Tiroxina(T4) e Cortisol. Amostras de jejuno e íleo foram coletadas para análise morfométrica do epitélio intestinal, e foram estudadas características da carcaça. Os cordeiros submetidos ao fotoperíodo longo, apresentaram maior consumo alimentar e maior ganho de peso (P<0,05). Não se verificou diferença significativa nas concentrações plasmáticas dos hormônios estudados, na morfometria intestinal e nas características da carcaça. Concluiu-se que o fotoperíodo longo influencia, positivamente, o desempenho dos cordeiros, sendo esta prática economicamente viável.

Rhizoctonia solani AG-1 IA causes leaf blight on soybean and rice. Despite the fact that R. solani AG-1 IA is a major pathogen affecting soybean and rice in Brazil and elsewhere in the world, little information is available on its genetic diversity and evolution. This study was an attempt to reveal the origin, and the patterns of movement and amplification of epidemiologically significant genotypes of R. solani AG-1 IA from soybean and rice in Brazil. For inferring intraspecific evolution of R. solani AG-1 IA sampled from soybean and rice, networks of ITS-5.8S rDNA sequencing haplotypes were built using the statistical parsimony algorithm from Clement et al. (2000) Molecular Ecology 9: 1657-1660. Higher haplotype diversity (Nei M 1987, Molecular Evolutionary Genetics Columbia University Press, New york: 512p.) was observed for the Brazilian soybean sample of R. solani AG-1 IA (0.827) in comparison with the rest of the world sample (0.431). Within the south-central American clade (3-2), four haplotypes of R. solani AG-1 IA from Mato Grosso, one from Tocantins, one from Maranhao, and one from Cuba occupied the tips of the network, indicating recent origin. The putative ancestral haplotypes had probably originated either from Mato Grosso or Maranhao States. While 16 distinct haplotypes were found in a sample of 32 soybean isolates of the pathogen, the entire rice sample (n=20) was represented by a single haplotype (haplotype 5), with a worldwide distribution. The results from nested-cladistic analysis indicated restricted gene flow with isolation by distance (or restricted dispersal by distance in nonsexual species) for the south-central American clade (3-2), mainly composed by soybean haplotypes.

Heterokaryons were obtained by hyphal anastomosis in Trichoderma pseudokoningii using strains with morphologic and double auxotrophic genetic markers. Stable haploid and unstable hyperhaploid recombinants were detected in monosporic colonies derived from heterokaryons and selected in appropriate media or by an enrichment filtration technique. Diploids were not observed and the recombinant frequencies were higher than those found in fungi with a typical parasexual cycle. The mechanism of recombinant emergence may involve a transient diploid formation followed by chromosomal losses at the hyphal stages, resulting in haploid and hyperhaploid recombinants. The possibility of intrachromosomal rearrangement with the involvement of cytoplasmatic DNA as a heterokaryotic transformation system cannot be excluded.

Digestibility of diets based on corn and soybean meal or soybeans treated by roasting or extrusion, with or without an enzyme supplementation, was measured by true (Sibbald) methods, by analysis of excreta, and by analysis of ileal digesta. Only analysis of ileal digesta was able to consistently measure differences between soybean and enzyme treatments in the digestibility of CP, starch, fat, and ME. The amino acid (AA) digestibility of the diets was measured by analysis of the ileal contents. Whereas enzyme supplementation improved overall CP digestibility by 2.9%, this improvement was not equal for all AA. of the AA most important for broilers fed corn-soybean diets, the digestibilities of Lys, Met, and Arg were not improved or not improved significantly by the enzyme supplementation; however, that of Val was improved by 2.3% and that of Thr was improved by 3.0%. A performance trial demonstrated that enzyme supplementation with equal diet formulation improved BW and the feed conversion ratio by 1.9 and 2.2%, respectively. A second performance trial compared standard diet formulations with formulations using enzyme supplementation and energy levels that were reduced by the amount of improvement provided by the inclusion of enzyme in the first performance trial. No difference was seen between treatments, showing that the improvement of nutrient utilization brought about by enzyme supplementation completely compensated for the reduced energy content. Whereas enzyme supplementation should allow a reduction in CP formulation as well, individual AA were not improved equally by supplementation and should also be balanced.

The study of the influence of motion and initial intra-articular pressure (IAP) on intra-articular pressure profiles in equine cadaver metatarsophalangeal (MTP) joints was undertaken as a prelude to in vivo studies, Eleven equine cadaver MTP joints were submitted to 2 motion frequencies of 5 and 10 cycles/min of flexion and extension, simulating the condition of lower and higher (double) rates of passive motion. These frequencies were applied and pressure profiles generated with initial normal intra-articular pressure (-5 mmHg) and subsequently 30 mmHg intra-articular pressure obtained by injection of previously harvested synovial fluid.The 4 trials performed were 1) normal IAP; 5 cyles/min; 2) normal IAP; 10 cycles/min; 3) IAP at 30 mmHg; 5 cycles/min and 4) IAP at 30 mmHg; 10 cycles/min. The range of joint motion applied (mean +/- s.e.) was 67.6 +/- 1.61 degrees with an excursion from 12.2 +/- 1.2 degrees in extension to 56.2 +/- 2.6 degrees in flexion, Mean pressure recorded in mmHg for the first and last min of each trial, respectively, were 1) -5.7 +/- 0.9 and -6.3 +/- 1.1; 2) -5.3 +/- 1.1 and -6.2 +/- 1.1; 3) 58.8 +/- 8.0 and 42.3 +/- 7.2; 4) 56.6 +/- 3.7 and 40.3 +/- 4.6. Statistical analyses showed a trend for difference between the values for the first and last minute in trial 3 (0.05>P<0.1) with P = 0.1 and significant difference (P = 0.02) between the mean IAP of the first and last min in trial 4. The loss of intra-articular pressure associated with time and motion was 10.5, 16.9, 28.1 and 28.9% for trials 1-4, respectively. As initial intraarticular pressure and motion increased, the percent loss of intra-articular pressure increased.The angle of lowest pressure was 12.2 +/- 1.2

Steers (379 +/- 10 kg) with ruminal, duodenal, and ileal cannulas were used in a 5 x 5 Latin square digestion trial to quantify and evaluate the relationship between intestinal protein supply and intestinal starch disappearance. Treatments were infusions of 0, 50, 100, 150, or 200 g/d of casein along with 1,042 g/d of raw cornstarch. Abomasal infusions were accomplished by passing tubing and a pliable retaining washer through the reticular-omasal orifice into the abomasum. Steers were fed a 93% corn silage, 7% supplement diet that contained 12% crude protein at 1.65% body weight in 12 equal portions/d. Periods lasted 17 d (12 d for adaptation, 2 d of collections, and 3 d of rest). The quantity and percentage of organic matter and protein disappearance from the small intestine increased linearly (P < 0.03) with infused casein. Greater quantities of starch disappeared with increased casein infusion (P < 0.01). The infusion of 200 g/d of casein increased small intestinal starch disappearance by 226 g/d over the control. Casein infusion did not affect the quantity or percent of organic matter, starch, or protein disappearance in the large intestine. Treatments did not change ruminal ammonia N, ruminal pH, or plasma glucose concentrations. Starch disappearance from the small intestine was increased with greater protein flow to the duodenum of steers.

One hundred and twenty-six strains of Campylobacter jejuni isolated from swine with diarrhea were examined for the production of enteroxin, by culture filtrate inoculation into ileal loops in several animal species.Four (3,1 %) of the strains tested produced fluid in ligated ileal loop of albino rabbit, 19 (15 %) in rat and 14(11, 1 %) in swine. By the test of intragastric inoculation in suckling mouse, none of the strains revealed capacity to produce enterotoxin, although two strains were considered suspicious. Twelve (54,5 %) of the strains that induced ileal fluid in rat and swine produced cytotoxic effect in monkey kidney cells (Vero), affecting up to 60 % of the monolayer. However, no alteration was observed in hamster kidney cells (BHK).
