OBJECTIVE: To report on the demographic data from the first 18 months of enrollment to an international registry on autoinflammatory diseases in the context of the Eurofever project. METHODS: A web-based registry collecting baseline and clinical information on autoinflammatory diseases and related conditions is available in the member area of the PRINTO web-site. Anonymised data were collected with standardised forms. RESULTS: 1880 (M:F=916:964) individuals from 67 centers in 31 countries have been entered in the Eurofever registry. Most of the patients (1388; 74%), reside in western Europe, 294 (16%) in the eastern and southern Mediterranean region (Turkey, Israel, North Africa), 106 (6%) in eastern Europe, 54 in Asia, 27 in South America and 11 in Australia. In total 1049 patients with a clinical diagnosis of a monogenic autoinflammatory diseases have been enrolled; genetic analysis was performed in 993 patients (95%): 703 patients have genetically confirmed disease and 197 patients are heterozygous carriers of mutations in genes that are mutated in patients with recessively inherited autoinflammatory diseases. The median diagnosis delay was 7.3 years (range 0.3-76), with a clear reduction in patients born after the identification of the first gene associated with autoinflammatory diseases in 1997. CONCLUSIONS: A shared online registry for patients with autoinflammatory diseases is available and enrollment is ongoing. Currently, there are data available for analysis on clinical presentation, disease course, and response to treatment, and to perform large scale comparative studies between different conditions.

OBJECTIVE: To assess the seasonality of cardiovascular risk factors (CVRF) in a large set of population-based studies. METHODS: Cross-sectional data from 24 population-based studies from 15 countries, with a total sample size of 237 979 subjects. CVRFs included Body Mass Index (BMI) and waist circumference; systolic (SBP) and diastolic (DBP) blood pressure; total, high (HDL) and low (LDL) density lipoprotein cholesterol; triglycerides and glucose levels. Within each study, all data were adjusted for age, gender and current smoking. For blood pressure, lipids and glucose levels, further adjustments on BMI and drug treatment were performed. RESULTS: In the Northern and Southern Hemispheres, CVRFs levels tended to be higher in winter and lower in summer months. These patterns were observed for most studies. In the Northern Hemisphere, the estimated seasonal variations were 0.26 kg/m(2) for BMI, 0.6 cm for waist circumference, 2.9 mm Hg for SBP, 1.4 mm Hg for DBP, 0.02 mmol/L for triglycerides, 0.10 mmol/L for total cholesterol, 0.01 mmol/L for HDL cholesterol, 0.11 mmol/L for LDL cholesterol, and 0.07 mmol/L for glycaemia. Similar results were obtained when the analysis was restricted to studies collecting fasting blood samples. Similar seasonal variations were found for most CVRFs in the Southern Hemisphere, with the exception of waist circumference, HDL, and LDL cholesterol. CONCLUSIONS: CVRFs show a seasonal pattern characterised by higher levels in winter, and lower levels in summer. This pattern could contribute to the seasonality of CV mortality.

The Mediterranean basin is considered a hotspot of biological diversity with a long history of modification of natural ecosystems by human activities, and is one of the regions that will face extensive changes in climate. For 181 terrestrial mammals (68% of all Mediterranean mammals), we used an ensemble forecasting approach to model the future (approx. 2100) potential distribution under climate change considering five climate change model outputs for two climate scenarios. Overall, a substantial number of Mediterranean mammals will be severely threatened by future climate change, particularly endemic species. Moreover, we found important changes in potential species richness owing to climate change, with some areas (e.g. montane region in central Italy) gaining species, while most of the region will be losing species (mainly Spain and North Africa). Existing protected areas (PAs) will probably be strongly influenced by climate change, with most PAs in Africa, the Middle East and Spain losing a substantial number of species, and those PAs gaining species (e.g. central Italy and southern France) will experience a substantial shift in species composition.

Direct evidence confirming the hypothesis that a dysfunction of the mitochondrial respiratory chain (MRC) underlies the pathogenesis of hyperlactatemia associated with highly active antiretroviral therapy (HAART) is scarce. We studied mitochondrial DNA (mtDNA) content and MRC function in the skeletal muscle of an HIV-infected patient during an episode of symptomatic hyperlactatemia. Skeletal muscle biopsy was performed during the episode when the patient was symptomatic and 3 months later when the patient was clinically recovered. Assessment of mitochondria was performed using histological, polarographic, spectrophotometrical, and Southern blot and real time PCR DNA quantification methods. The histological study disclosed extensive mitochondrial impairment in the form of ragged-red fibers or equivalents on oxidative reactions. These findings were associated with an increase in mitochondrial content and a decrease in both mitochondrial respiratory capacity and MRC enzyme activities. Mitochondrial DNA content declined to 53% of control values. Mitochondrial abnormalities had almost disappeared later when the patient became asymptomatic. Our findings support the hypothesis that MRC dysfunction stands at the basis of HAART-related hyperlactatemia.

IT-toimialan järjestelmätoimittajien liittoutuminen alalla toimivien vertikaalitoimittajien kanssa on tuonut mukanaan tarpeita kumppanuustoiminnan kehittämiselle. Kehittämiseen liittyy läheisesti kumppanuustoiminnan mittaaminen ja kumppanuuksien arviointi, jotta voidaan varmistaa kehittämistoimien tuloksellisuus ja kumppanuustoiminnan tehokkuus. Tutkimuksen tarkoituksena olitutkia case-yrityksen kumppanuusmallin ja kumppanuuteen liittyvän toiminnan tehokkuutta ja kehittymiskykyä. Lähtökohtana oli kumppanuuteen vaikuttavista elementeistä, kuten luottamus, sosiaalinen pääoma ja kumppanuusmekanismit, aikaisemmintuotetun tutkimusaineistojen selvittäminen ja hyväksikäyttö. Kerättyä aineistoapeilattiin case-yrityksen kumppanitoiminnan malliin ja menetelmiin toimittajakumppanuuksissa. Vertailulla pyrittiin selvittämään miten case-yritys toteuttaa kumppanuustoimintaansa ja miten toimintaa voisi parantaa. Lopputuloksena arvioitiin case-yrityksen kumppanuustoimintaa esitettyjen teorioiden näkökulmasta ja muodostettiin case-yritykselle kumppanuustoiminnan mittaamista tukeva mittaristo. Tutkimus on kirjallisuustutkimus ja siinä käytettiin lähteinä keskeisimpiä kumppanuudesta kirjoitettua kirjallisuutta ja kumppanuuden hallinnasta tehtyjä tutkimuksia.

Tämä Pro-Gradu –tutkielma tarkastelee etelä- ja itäsuomalaisia kuljetusyrityksiä kannattavuuden, vakavaraisuuden ja maksuvalmiuden näkökulmasta kuljetuslajeittain. Tutkielman pyrkimyksenä on valottaa Etelä- ja Itä-Suomen läänien kilpailuympäristöä numeeristen tunnuslukujen ja tilinpäätöserien perusteella. Päätavoitteena on osoittaa, onko kuljetuslajien välillä kannattavuuseroja. Tutkielma koostuu teoriaosasta sekä empiirisestä osasta, jonka aineisto koostuu etelä- ja itäsuomalaisten kuljetusyritysten tilinpäätöstiedoista ja tunnusluvuista. Tutkimuksen empiirinen aineisto on kerätty haulla Amadeus–tietokannasta ja postikyselynä haun tuottamilta yrityksiltä. Aineistoa on analysoitu kvantitatiivisilla menetelmillä SPSS–ohjelmistolla. Tuloksista selviää, että kuljetuslajilla on yhteys kannattavuuteen ROCE–tunnusluvulla mitattuna. Kannattavuuden osa-alueita, kuten tuottoja, kustannuksia ja sidottua pääomaa analysoitiin erikseen, mutta kannattavuuseroille ei silti löydetty selitystä.

Background: Myotragus balearicus was an endemic bovid from the Balearic Islands (Western Mediterranean) that became extinct around 6,000-4,000 years ago. The Myotragus evolutionary lineage became isolated in the islands most probably at the end of the Messinian crisis, when the desiccation of the Mediterranean ended, in a geological date established at 5.35 Mya. Thus, the sequences of Myotragus could be very valuable for calibrating the mammalian mitochondrial DNA clock and, in particular, the tree of the Caprinae subfamily, to which Myotragus belongs. Results: We have retrieved the complete mitochondrial cytochrome b gene (1,143 base pairs), plus fragments of the mitochondrial 12S gene and the nuclear 28S rDNA multi-copy gene from a well preserved Myotragus subfossil bone. The best resolved phylogenetic trees, obtained with the cytochrome b gene, placed Myotragus in a position basal to the Ovis group. Using the calibration provided by the isolation of Balearic Islands, we calculated that the initial radiation of caprines can be dated at 6.2 ± 0.4 Mya. In addition, alpine and southern chamois, considered until recently the same species, split around 1.6 ± 0.3 Mya, indicating that the two chamois species have been separated much longer than previously thought. Conclusion: Since there are almost no extant endemic mammals in Mediterranean islands, the sequence of the extinct Balearic endemic Myotragus has been crucial for allowing us to use the Messinian crisis calibration point for dating the caprines phylogenetic tree.

Health and inequalities in health among inhabitants of European cities are of major importance for European public health and there is great interest in how different health care systems in Europe perform in the reduction of health inequalities. However, evidence on the spatial distribution of cause-specific mortality across neighbourhoods of European cities is scarce. This study presents maps of avoidable mortality in European cities and analyses differences in avoidable mortality between neighbourhoods with different levels of deprivation. Methods: We determined the level of mortality from 14 avoidable causes of death for each neighbourhood of 15 large cities in different European regions. To address the problems associated with Standardised Mortality Ratios for small areas we smooth them using the Bayesian model proposed by Besag, York and Mollié. Ecological regression analysis was used to assess the association between social deprivation and mortality. Results: Mortality from avoidable causes of death is higher in deprived neighbourhoods and mortality rate ratios between areas with different levels of deprivation differ between gender and cities. In most cases rate ratios are lower among women. While Eastern and Southern European cities show higher levels of avoidable mortality, the association of mortality with social deprivation tends to be higher in Northern and lower in Southern Europe. Conclusions: There are marked differences in the level of avoidable mortality between neighbourhoods of European cities and the level of avoidable mortality is associated with social deprivation. There is no systematic difference in the magnitude of this association between European cities or regions. Spatial patterns of avoidable mortality across small city areas can point to possible local problems and specific strategies to reduce health inequality which is important for the development of urban areas and the well-being of their inhabitants

Many viral diseases, including leafroll, which is of great economic importance, affect grapevines (Vitis spp.). A complex of eight viruses [Grapevine leafroll-associated virus (GLRaV) -1 to 8] is associated with this disease. The objective of this study was to compare the variability of the 3' terminal region of the polymerase gene of three isolates of GLRaV-3 (Grapevine leafroll-associated virus-3), from Submédio do Vale do Rio São Francisco (Petrolina-PE) with that of other isolates available at the GenBank, including an isolate from North America and another from Southern Brazil. The viral RNA was extracted from three infected ELISA reactive plants and a fragment of 340 bp was amplified, by RT-PCR, using primers that recognize that portion of the polymerase gene found between nucleotides 8267 and 8606. The three isolates from Vale do Rio São Francisco named Pet-1, Pet-2 and Pet-3, showed similarities ranging from 98% and 94%, respectively to the isolates from North America (AF037268) and Southern Brazilian (AF438411). Considering the whole genome, the main variation found was one amino acid change at position 2766 (F2766Y). These preliminary data indicate the existence of a natural variation among GLRaV-3 isolates from grapevines. This could be due to the vegetative propagation and long cycle of the plant, associated with the error-prone nature of RNA-dependent RNA polymerase.

Diplomityön tavoitteena oli kehittää Pilomac – konsernin päähankkijayrityksien tuotteiden valmistettavuutta yhteistyössä yrityksien suunnitteluosastojen ja teräsrakenteita valmistavien toimittajayrityksien kanssa. Päähankkija-yritysten, Junttan Oy:n ja Logset Oy:n, osavalmistusten ulkoistamisen myötä toimittajasuhteet ovat vähitellen muuttumassa verkostomaiseen toimintatapaan. Täten on tärkeää muuttaa ja kehittää tuotteiden valmistettavuuden mahdollistavat toimintamallit vastaamaan uutta toimintatapaa. Keskeinen perusmenetelmä valmistettavuuden huomioimisessa on suunnittelun ja valmistuksen välisen tuotekehitysyhteistyön syventäminen rinnakkaissuunnitteluksi, (CE). Verkostokumppaneiden välinen yhteistyö perustuu avoimuuteen, luottamukseen ja molempia osapuolia hyödyttävään jatkuvaan kehitystyöhön. Verkostomainen toimintatapa mahdollistaa luontevasti tuotteen suunnittelu- ja kehitysvastuiden jaottelun yritysten ydinosaamisen mukaan. Onnistuneen työnjaon myötä resurssit lisääntyvät ja valmistusteknologiat, suunnittelu ja laatu kehittyvät ja monipuolistuvat. Työn aihetta tutkittiin soveltavassa osassa benchmarkingin, nykytilakuvauksien sekä toimittajaverkoston analysoinnin avulla. Toimittajaverkoston analysointi tehtiin laajan verkostokumppanikyselyn avulla, josta selvisi Junttan Oy:n toimittajaverkoston resurssit ja kyvykkyys. Kyselyssä selvitettiin myös nykyisten toimittajien näkemyksiä valmistettavuuden kehittämisestä, kumppaneille asetettavista yhteistyövaatimuksista sekä näiden toimintamallien vaikutuksista tuotteiden kilpailukyvyn parantumiseen. Työn tuloksena selvisivät osa-alueet, joita Junttan Oy:n kannattaa hyödyntää tulevaisuudessa enemmän sekä toimintamallit, jotka mahdollistavat tuotteiden valmistettavuuden jatkuvan kehittämisen.

Pneumocystis has been isolated from a wide range of unrelated mammalian hosts, including humans, domestic and wild animals. It has been demonstrated that the genome of Pneumocystis of one host differs markedly from that of other hosts. Also, variation in the chromosome and DNA sequence of Pneumocystis within a single host species has been observed. Since information about the occurrence and nature of infections in wild animals is still limited, the objective of this work was to detect the presence of Pneumocystis sp. in lungs of bats from two states from Brazil by Nested-PCR amplification. The bats, captured in caves and in urban areas, were obtained from the Program of Rabies Control of two States in Brazil, Mato Grosso and Rio Grande do Sul, located in the Mid-Western and Southern regions of the country, respectively. DNAs were extracted from 102 lung tissues and screened for Pneumocystis by nested PCR at the mtLSU rRNA gene and small subunit of mitochondrial ribosomal RNA (mtSSU rRNA). Gene amplification was performed using the mtLSU rRNA, the primer set pAZ102H - pAZ102E and pAZ102X - pAZY, and the mtSSU rRNA primer set pAZ102 10FRI - pAZ102 10R-RI and pAZ102 13RI - pAZ102 14RI. The most frequent bats were Tadarida brasiliensis (25), Desmodus rotundus (20), and Nyctinomops laticaudatus (19). Pneumocystis was more prevalent in the species Nyctinomops laticaudatus (26.3% = 5/19), Tadarida brasiliensis (24% = 6/25), and Desmodus rotundus (20% = 4/20). Besides these species, Pneumocystis also was detected in lungs from Molossus molossus (1/11, 9.1%), Artibeus fimbriatus (1/1, 100%), Sturnira lilium (1/3, 33.3%), Myotis levis (2/3, 66.7%)and Diphylla ecaudata (1/2, 50%). PCR products which could indicate the presence of Pneumocystis (21.56%) were identified in DNA samples obtained from 8 out of 16 classified species from both states (5 bats were not identified). This is the first report of detection of Pneumocystis in bats from Brazil.
