The mechanism of action and properties of a solid-phase ligand library made of hexapeptides (combinatorial peptide ligand libraries or CPLL, for capturing the "hidden proteome", i.e. the low- and very low-abundance proteins Constituting the vast majority of species in any proteome. as applied to plant tissues, are reviewed here. Plant tissues are notoriously recalcitrant to protein extraction and to proteome analysis, Firstly, rigid plant cell walls need to be mechanically disrupted to release the cell content and, in addition to their poor protein yield, plant tissues are rich in proteases and oxidative enzymes, contain phenolic Compounds, starches, oils, pigments and secondary metabolites that massively contaminate protein extracts. In addition, complex matrices of polysaccharides, including large amount of anionic pectins, are present. All these species compete with the binding of proteins to the CPLL beads, impeding proper capture and identification I detection of low-abundance species. When properly pre-treated, plant tissue extracts are amenable to capture by the CPLL beads revealing thus many new species among them low-abundance proteins. Examples are given on the treatment of leaf proteins, of corn seed extracts and of exudate proteins (latex from Hevea brasiliensis). Acca sellowiana (Berg.) Burr. is a native Myrtaceae from southern Brazil and Uruguay, now the subject of a domestication and breeding program. Biotechnological tools have been used to assist in this program. The establishment of a reliable protocol of somatic embryogenesis has been pursued, with a view to capturing and fixing genetic gains. The rationale behind this work relies on the fact that deepening comprehension of the general metabolism of zygotic embryogenesis may certainly improve the protocol for somatic embryogenesis. Thus, in the present work we studied the accumulation of protein, total sugars, starch, amino acids, polyamines (PAs), IAA and ABA, in different stages of A. sellowiana zygotic embryogenesis. Starch is the predominant storage compound during zygotic embryo development. Increased synthesis of amino acids in the cotyledonary stage, mainly of asparagine, was observed throughout development. Total free PAs showed increased synthesis, whereas total conjugated PAs were mainly observed in the early developmental stages. IAA decreased and ABA increased with the progression from early to late embryogenesis. Besides providing basic information on the morphophysiological and biochemical changes of zygotic embryogenesis, the results here obtained may provide adequate strategies towards the modulation of somatic embryogenesis in this species as well as in other woody angiosperms.

The present paper reports on 22 species collected by the Brazilian Program of Living Resources in the Exclusive Economic Zone (REVIZEE). A new genus and species of Cribrilinidae, Corbuliporina crepida n. gen. et sp., is described, along with seventeen other new species: Chaperia brasiliensis n. sp., Amastigia aviculifera n. sp., Isosecuriflustra pinniformis n. sp., Cellaria subtropicalis n. sp., Melicerita brasiliensis n. sp., Arachnopusia haywardi n. sp., Smittina migottoi n. sp., Hippomenella amaralae n. sp., Rogicka joannae n. sp., Malakosaria atlantica n. sp., Turbicellepora winstonae n. sp., Rhynchozoon coalitum n. sp., Stephanollona angusta n. sp., Stephanollona arborescens n. sp., Aulopocella americana n. sp., Conescharellina cookae n. sp. and Conescharellina bocki n. sp. Chorizopora brongniartii (Audouin, 1826) is recorded for the first time in Brazilian waters and a new combination for Rhynchozoon arborescens Canu & Bassler, 1928 is established. New illustrations and taxonomic remarks are included for two little-known species from Brazil, Rogicka scopae (Canu & Bassler, 1928) and Fenestrulina ampla Canu & Bassler, 1928. A compilation of species recorded from deeper waters of the Brazilian coast is included.

Fatty acids have been used in marine biogeochemistry as food chain biomarkers, but in freshwater these studies are rare. In order to evaluate the fatty acid potential as biomarkers in freshwater, their profile was analyzed during vitellogenesis in two fish species, in both waterfall and reservoir environments of the Paraiba do Sul River Basin. Detrivorous Hypostomus affinis and omnivorous Geophagus brasiliensis seem to elongate and desaturate polyunsaturated fatty acids (PUFA) and transfer them to the ovaries` phospholipids. Waterfall Geophagus brasiliensis have more highly unsaturated fatty acids in the liver, but in the reservoir, accumulation mainly occurs in muscle and ovary triglycerides, suggesting trophic opportunism and a plasticity during vitellogenesis. In Hypostomus affinis, PUFA alteration occurs only in the reservoir, suggesting a high phytoplankton occurrence. Eutrophication and water speed is reflected in Hypostomus affinis ovaries by higher PUFAn3 and bacterial fatty acids. As in marine environments, analysis of mono- and polyunsaturated fatty acids during vitellogenesis can be used as a tool in food chain studies in freshwater.

T-cell immunity has been claimed as the main immunoprotective mechanism against Paracoccidioides brasiliensis infection, the most important fungal infection in Latin America. As the initial events that control T-cell activation in paracoccidioidomycosis (PCM) are not well established, we decided to investigate the role of CD28, an important costimulatory molecule for the activation of effector and regulatory T cells, in the immunity against this pulmonary pathogen. Using CD28-deficient (CD28(-/-)) and normal wild-type (WT) C57BL/6 mice, we were able to demonstrate that CD28 costimulation determines in pulmonary paracoccidioidomycosis an early immunoprotection but a late deleterious effect associated with impaired immunity and uncontrolled fungal growth. Up to week 10 postinfection, CD28(-/-) mice presented increased pulmonary and hepatic fungal loads allied with diminished production of antibodies and pro-and anti-inflammatory cytokines besides impaired activation and migration of effector and regulatory T (Treg) cells to the lungs. Unexpectedly, CD28-sufficient mice progressively lost the control of fungal growth, resulting in an increased mortality associated with persistent presence of Treg cells, deactivation of inflammatory macrophages and T cells, prevalent presence of anti-inflammatory cytokines, elevated fungal burdens, and extensive hepatic lesions. As a whole, our findings suggest that CD28 is required for the early protective T-cell responses to P. brasiliensis infection, but it also induces the expansion of regulatory circuits that lately impair adaptive immunity, allowing uncontrolled fungal growth and overwhelming infection, which leads to precocious mortality of mice.

Toll-like receptors (TLRs) present in innate immune cells recognize pathogen molecular patterns and influence immunity to control the host-parasite interaction. The objective of this study was to characterize the involvement of TLR4 in the innate and adaptive immunity to Paracoccidioides brasiliensis, the most important primary fungal pathogen of Latin America. We compared the responses of C3H/HeJ mice, which are naturally defective in TLR4 signaling, with those of C3H/HePas mice, which express functional receptors, after in vitro and in vivo infection with P. brasiliensis. Unexpectedly, we verified that TLR4-defective macrophages infected in vitro with P. brasiliensis presented decreased fungal loads associated with impaired synthesis of nitric oxide, interleukin-12 (IL-12), and macrophage chemotactic protein 1 (MCP-1). After intratracheal infection with 1 million yeasts, TLR4-defective mice developed reduced fungal burdens and decreased levels of pulmonary nitric oxide, proinflammatory cytokines, and antibodies. TLR4-competent mice produced elevated levels of IL-12 and tumor necrosis factor alpha (TNF-alpha), besides cytokines of the Th17 pattern, indicating a proinflammatory role for TLR4 signaling. The more severe infection of TLR4-normal mice resulted in increased influx of activated macrophages and T cells to the lungs and progressive control of fungal burdens but impaired expansion of regulatory T cells (Treg cells). In contrast, TLR4-defective mice were not able to clear their diminished fungal burdens totally, a defect associated with deficient activation of T-cell immunity and enhanced development of Treg cells. These divergent patterns of immunity, however, resulted in equivalent mortality rates, indicating that control of elevated fungal growth mediated by vigorous inflammatory reactions is as deleterious to the hosts as low fungal loads inefficiently controlled by limited inflammatory reactions.

To study the role of TLR2 in a experimental model of chronic pulmonary infection, TLR2-deficient and wild-type mice were intratracheally infected with Paracoccidioides brasiliensis, a primary fungal pathogen. Compared with control, TLR2(-/-) mice developed a less severe pulmonary infection and decreased NO synthesis. Equivalent results were detected with in vitro-infected macrophages. Unexpectedly, despite the differences in fungal loads both mouse strains showed equivalent survival times and severe pulmonary inflammatory reactions. Studies on lung-infiltrating leukocytes of TLR2(-/-) mice demonstrated an increased presence of polymorphonuclear neutrophils that control fungal loads but were associated with diminished numbers of activated CD4(+) and CD8(+) T lymphocytes. TLR2 deficiency leads to minor differences in the levels of pulmonary type 1 and type 2 cytokines, but results in increased production of KC, a CXC chemokine involved in neutrophils chemotaxis, as well as TGF-beta, IL-6, IL-23, and IL-17 skewing T cell immunity to a Th17 pattern. In addition, the preferential Th17 immunity of TLR2(-/-) mice was associated with impaired expansion of regulatory CD4(+)CD25(+)FoxP3(+) T cells. This is the first study to show that TLR2 activation controls innate and adaptive immunity to P. brasiliensis infection. TLR2 deficiency results in increased Th17 immunity associated with diminished expansion of regulatory T cells and increased lung pathology due to unrestrained inflammatory reactions. Chemotherapy is the basis of treatment of paracoccidioidomycosis in its various forms. Depending on the Paracoccidioides brasiliensis virulence, the status of host immunity, the degree of tissue involvement and fungal dissemination, treatment can be extended for long periods with an alarming frequency of relapses. Association of chemotherapy with a vaccine to boost the cellular immune response seemed a relevant project not only to reduce the time of treatment but also to prevent relapses and improve the prognosis of anergic cases. The candidate immunogen is the gp43 major diagnostic antigen of P. brasiliensis and more specifically its derived peptide P10, carrying the CD4(+) T-cell epitope. Both gp43 and P10 protected Balb/c mice against intratracheal infections with virulent P. brasiliensis strain. P10 as single peptide or in a multiple-antigen-peptide (MAP) tetravalent construction was protective without adjuvant either by preimmunization and intratracheal challenge or as a therapeutic agent in mice with installed infection. P10 showed additive protective effects in drug-treated mice stimulating a Th-1 type immune response with high IFN-gamma and IL-12. P10 and few other peptides in the gp43 were selected by Tepitope algorithm and actually shown to promiscuously bind several prominent HLA-DR molecules suggesting that a peptide vaccine could be devised for a genetically heterogenous population. P10 was protective in animals turned anergic, was effective in a DNA minigene vaccine, and increased the protection by monoclonal antibodies in Balb/c mice. DNA vaccines and peptide vaccines are promising therapeutic tools to be explored in the control of systemic mycoses.

Ajoene has been described as an antithrombotic, anti-tumour, antifungal, antiparasitic and antibacterial agent. This study deals with the efficacy of ajoene to treat mice intratracheally infected with Paracoccidioides brasiliensis. The results indicate that ajoene therapy is effective in association with antifungal drugs (sulfametoxazol/trimethoprim), showing a positive additive effect. Ajoene-treated mice developed Th1-type cytokine responses producing higher levels of IFN-gamma and IL-12 when compared to the infected but untreated members of the control group. Antifungal activity of ajoene involves a direct effect on fungi and a protective pro-inflammatory immune response. Reduction of fungal load is additive to chemotherapy and therefore the combined treatment is mostly effective against experimental paracoccidioidomycosis.

Background and purpose: The present study reports on the preparation and testing of a sustained delivery system for the immunomodulatory peptide P10 aimed at reducing the in vivo degradation of the peptide and the amount required to elicit a protective immune response against paracoccidioidomycosis. Experimental approach: BALB/c mice were infected with the yeast Paracoccidioides brasiliensis to mimic the chronic form of paracoccidioidomycosis. The animals were treated daily with sulfamethoxazole/trimethoprim alone or combined with peptide P10, either emulsified in Freund`s adjuvant or entrapped in poly(lactic acid-glycolic acid) (PLGA) nanoparticles at different concentrations (1 mu g, 5 mu g, 10 mu g, 20 mu g or 40 mu g center dot 50 mu L-1). Therapeutic efficacy was assessed as fungal burden in tissues and the immune response by quantitative determination of cytokines. Key results: Animals given combined chemotherapy and P10 nanotherapy presented a marked reduction of fungal load in the lungs, compared with the non-treated animals. After 30 days of treatment, P10 entrapped within PLGA (1 mu g center dot 50 mu L-1) was more effective than `free` P10 emulsified in Freund`s adjuvant (20 mu g center dot 50 mu L-1), as an adjuvant to chemotherapy. After treatment for 90 days, the higher doses of P10 entrapped within PLGA (5 or 10 mu g center dot 50 mu L-1) were most effective. Treatment with P10 emulsified in Freund`s adjuvant (20 mu g center dot 50 mu L-1) or P10 entrapped within PLGA (1 mu g center dot 50 mu L-1) were accompanied by high levels of interferon-gamma in lung. Conclusions and implications: Combination of sulfamethoxazole/trimethoprim with the P10 peptide entrapped within PLGA demonstrated increased therapeutic efficacy against paracoccidioidomycosis. P10 incorporation into PLGA nanoparticles dramatically reduced the peptide amount necessary to elicit a protective effect.

Garciniaphenone (=rel-(1R,5R,7R)-3-benzoyl-4-hydroxy-8,8-dimethyl-1,7-bis(3-methylbut-2-en-1-yl)bicyclo[3.3.1]non-3-ene-2,9-dione; 1). a novel natural product, was isolated from a hexane extract of Garcinia brasiliensis fruits. The crystal structure of 1 as well as the selected geometrical and Configurational features were compared with those of known related polyprenylated benzophenones. Garciniaphenone is the first representative of polyprenylated benzophenones without a prenyl substituent at C(5). Notably, the absence of a 5-prenyl substituent has an impact on the molecular geometry. The tautomeric form of 1 in the solid state was readily established by a residual-electronic-density map generated by means of a difference Fourier analysis, and there is an entirely delocalized six-membered chelate ring encompassing the keto-enol moiety. The configuration at C(7) was used to rationalize the nature of the keto-enol tautomeric form within 1. The intermolecular array in the network is maintained by nonclassical intermolecular H-bonds.

A robust, direct, rapid and non-destructive X-ray diffraction crystallography method to detect the polyprenylated benzophenones 7-epi-clusianone (1) and guttiferone A (2) in extracts from Garcinia brasiliensis is presented. Powder samples of benzophenones 1 and 2, dried hexane extracts from G. brasiliensis seeds and fruit`s pericarp, and the dried ethanolic extract from G. brasiliensis seeds were unambiguously characterized by powder X-ray diffractometry. The calculated X-ray diffraction peaks from crystal structures of analytes 1 and 2, previously determined by single-crystal X-ray diffraction technique, were overlaid to those of the experimental powder diffractograms, providing a practical identification of these compounds in the analyzed material and confirming the pure contents of the powder samples. Using the X-ray diffraction crystallography method, the studied polyprenylated benzophenones were selectively and simultaneously detected in the extracts which were mounted directly on sample holder. In addition, reference materials of the analytes were not required for analyses since the crystal structures of the compounds are known. High performance liquid chromatography analyses also were comparatively carried out to quantify the analytes in the same plant extracts showing to be in agreement with X-ray diffraction crystallography method. Em 1982 o Brasil participa da Convenção das Nações Unidas para o Direito do Mar. Esta convenção dispõe sobre os usos dos espaços marítimos e oceânicos, buscando a utilização racional e a exploração dos recursos do mar. Devido ao escasso conhecimento da região da plataforma externa e talude continental, surge o programa Avaliação do Potencial Sustentável de Recursos Vivos da Zona Econômica Exclusiva – REVIZEE, tanto para a diversificação da exploração pesqueira quanto para a determinação dos limites sustentáveis de captura. Com base em material coletado em abril de 1998, durante campanha de Bentos concernente ao programa REVIZEE, são fornecidos resultados relativos a Gastropoda encontrados na radial 45, estações hidrográficas 6839 (32o55´70´´S - 50o34´60´´W), 6840 (33o01´45´´S - 50o12´75´´W), 6841 (33o00´16´´S - 50o22´84´´W) e 6842 (32o57´81´´S - 50o29´18´´W), localizadas ao largo de Rio Grande, Rio Grande do Sul, Brasil. Para a avaliação da diversidade bêntica foram utilizados três tipos de amostradores de fundo; Draga de arrasto, Box Corer e Van Veen. Molluscos foram separados dos demais grupos taxonômicos pela equipe do REVIZEE e recebidos pelo laboratório de malacologia da Universidade Federal do Rio Grande do Sul. O material foi triado sob lupa Zeiss Stemi 2000- C ou Wild M-38. O material foi identificado por comparação com os dados da bibliografia e/ou por comparação com exemplares depositados nas coleções do Museu Oceanográfico Eliézer de Carvalho Rios (MORG) e/ou com a coleção particular do Dr. José Carlos Tarasconi São identificados e caracterizados quanto a tamanho, esculturação e área de ocorrência 6.020 exemplares distribuídos em 36 famílias - 80 táxons em nível de espécie, 19 em nível de gênero e 2 em nível de família. A família Turridae é a que apresenta o maior número de táxons com 16 espécies. Turritella hookeri Reeve, 1849 é a espécie mais abundante, com 684 exemplares. Registra-se pela primeira vez para o Rio Grande do Sul as seguintes espécies: Brookula conica (Watson, 1886), Calliostoma echinatum Dall, 1881, Solariela carvalhoi Lopes & Cardoso, 1958, Caecum massambabensis Absalão, 1994, Turritella hookeri Reeve, 1849, Crepidula plana Say, 1822, Natica pusilla Say, 1822, Polinices lacteus (Guilding, 1833), Eulima mulata Rios & Absalão, 1990, Melanella aff. hypsela (Verrill & Bush, 1900), Trachipollia turricula (von Maltzan, 1884), Anachis sparsa Reeve, 1859, Dentimargo janeiroensis (E. A. Smith, 1915), Axelella brasiliensis Verhecken, 1991, Drilliola loprestiana (Calcara, 1841), Mitrolumna biplicata (Dall, 1889), Nannodiella vespuciana (Orbigny, 1842), Stenodrillia goundlachi (Dall & Simpson, 1901), Terebra brasiliensis E. A. Smith, 1873 e Mathilda aff. scitula Dall, 1889. Novos limites de profundidade são registrados para 29 espécies. Com base na lista de espécies encontradas, se discute sobre o limite das Províncias Caribenha e Patagônica e sobre a influência da Laguna dos Patos no Talude Continental ao largo de Rio Grande.

Entre maio de 2001 e janeiro de 2002, foram realizadas coletas sazonais na lagoa Azul, Siderópolis, SC. Esta lagoa formou-se por lavra de mineração de carvão a céu aberto desativada. Foram avaliadas as concentrações de Cr, Mn, Ni, Zn e Fe na água e no sedimento. Estes metais, exceto o Fe, foram também analisados no músculo e no fígado das espécies dos peixes Oreochromis niloticus (tilápia) e Geophagus brasiliensis (acará). O objetivo foi avaliar a qualidade da lagoa, bem como, das espécies de peixes predominantes, devido a preocupação quanto ao consumo potencial das mesmas pela população do entorno. As análises do sedimento, quando comparada a locais não contaminados, indicam concentração elevada para Fe, Mn, Cr e Zn. No compartimento água as concentrações de Fe, Mn e Ni estão acima do estabelecido para a classe 2 da Resolução CONAMA 20/86. Conforme análise estatística, em ambos os compartimentos abióticos ocorrem diferenças significativas entre os 4 pontos amostrados, porém diferença sazonal ocorre somente para o compartimento água. O conteúdo de metais em peixes não apresenta diferença significativa entre as espécies analisadas, todavia entre os diferentes tecidos analisados, o fígado apresentou a maior concentração em ambas as espécies. Através das análises do músculo (parte comestível), foi possível avaliar o risco à saúde humana. Segundo a estimativa da ingestão diária, os peixes analisados não propiciam risco aparente de contaminação à população do entorno, conforme os critérios adotados para o teor dos metais avaliados.

O presente trabalho teve por objetivo avaliar alguns aspectos relacionados à zoogeografia e ecologia dos ostracodes da plataforma interna norte do Rio de Janeiro, próximo à cidade de Cabo Frio. O material de estudo resultou da análise de 43 amostras sedimentológicas coletadas durante a expedição GEOCOSTA RIO II. Este estudo permitiu o reconhecimento de 16 famílias, 49 gêneros, 66 espécies e dois gêneros que permaneceram em nomenclatura indeterminada. A descrição de uma nova espécie, Actinocythereis brasiliensis sp. n., também foi realizada no âmbito do presente trabalho. As famílias mais representativas quanto à abundância e o número de espécies foram Thaerocytheridae e Cytheruridae, respectivamente. A espécie mais constante na área foi Caudites ohmerti e, dentre as com maior dominância estão, Caudites ohmerti, Meridionalicythere? dubia, Callistocythere litoralensis, Paracytheridea bulbosa, Urocythereis dimorphica, Henryhowella inflata, Oculocytheropteron delicatum, Xestoleberis umbonata, Henryhowella macrocicatricosa e Brasilicythere reticulispinosa. As preferências sedimentológicas e batimétricas das espécies são também discutidas neste trabalho. É sugerido que se estenda a distribuição zoogeográfica, um pouco mais ao norte, para sete espécies: Cushmanidea variopunctata, Neocytherideis impudicus, Hemingwayella advena, Hemingwayella sp., Urocythereis dimorphica, Munseyella cornuta e Basslerites costata. A grande maioria das espécies são características da plataforma continental sul, sendo que destas o maior percentual pertence à Subprovíncia Sul-Brasileira. De acordo com a fauna registrada, predominantemente de águas frias, chegamos a conclusão que a sua presença se deve, em grande parte, à influência da ressurgência marinha na área de estudo A proporção sexual foi de 1,19 machos para 1 fêmea. Fêmeas ovígeras encontradas entre abril e setembro e o ingresso de juvenis na população (recrutamento) foi observado em novembro e dezembro de 2000.
