This study investigated the effect of human-animal interaction (HAI) and the stress response on the quality of embryo production in superovulated Nelore (Bos indicus) cattle, under tropical conditions. Thirty-two females underwent a superovulation protocol for 5 days. Cortisol concentrations were determined in blood plasma collected on days 0, 4, and 5. Artificial insemination was performed on days 4 and 5, and nonsurgical embryo flushing on day 11. Embryo production and viability were determined. Human stimulation, animal behaviors, accidents, and handling time were recorded to assess HAI. Cattle age was negatively correlated with accidents, frequency of aversive behaviors, and negative stimuli by stockperson during transit through corral compartments to receive superovulation treatments. The factor analysis revealed two distinct groups. The first group was called stressed and had higher cortisol concentration than the nonstressed group, 16.0 +/- 2.1 and 12.5 +/- 1.0 ng/mL, respectively. Comparisons between these groups showed that the frequency of voice emissions by the stockperson and the number of accidents were higher in the stressed group, and also, the mean handling time was longer in the stressed group than for the nonstressed. As a result, viability rate of the embryos was 19% lower in the stressed group (P < 0.05). This indicates that intensive negative HAI is likely related to stress, which affects embryo production in a superovulation program.

Social behavior depends on the integrity of social brain circuitry. The temporal lobe is an important part of the social brain, and manifests morphological and functional alterations in autism spectrum disorders (ASD). Rats with temporal lobe epilepsy (TLE), induced with pilocarpine, were subjected to a social discrimination test that has been used to investigate potential animal models of ASD, and the results were compared with those for the control group. Rats with TLE exhibited fewer social behaviors than controls. No differences were observed in nonsocial behavior between groups. The results suggest an important role for the temporal lobe in regulating social behaviors. This animal model might be used to explore some questions about ASD pathophysiology. (c) 2008 Elsevier Inc. The neuromuscular disorders are a heterogeneous group of genetic diseases, caused by mutations in genes coding sarcolemmal, sarcomeric, and citosolic muscle proteins. Deficiencies or loss of function of these proteins leads to variable degree of progressive loss of motor ability. Several animal models, manifesting phenotypes observed in neuromuscular diseases, have been identified in nature or generated in laboratory. These models generally present physiological alterations observed in human patients and can be used as important tools for genetic, clinic, and histopathological studies. The mdx mouse is the most widely used animal model for Duchenne muscular dystrophy (DMD). Although it is a good genetic and biochemical model, presenting total deficiency of the protein dystrophin in the muscle, this mouse is not useful for clinical trials because of its very mild phenotype. The canine golden retriever MD model represents a more clinically similar model of DMD due to its larger size and significant muscle weakness. Autosomal recessive limb-girdle MD forms models include the SJL/J mice, which develop a spontaneous myopathy resulting from a mutation in the Dysferlin gene, being a model for LGMD2B. For the human sarcoglycanopahties (SG), the BIO14.6 hamster is the spontaneous animal model for delta-SG deficiency, whereas some canine models with deficiency of SG proteins have also been identified. More recently, using the homologous recombination technique in embryonic stem cell, several mouse models have been developed with null mutations in each one of the four SG genes. All sarcoglycan-null animals display a progressive muscular dystrophy of variable severity and share the property of a significant secondary reduction in the expression of the other members of the sarcoglycan subcomplex and other components of the Dystrophin-glycoprotein complex. Mouse models for congenital MD include the dy/dy (dystrophia-muscularis) mouse and the allelic mutant dy(2J)/dy(2J) mouse, both presenting significant reduction of alpha 2-laminin in the muscle and a severe phenotype. The myodystrophy mouse (Large(myd)) harbors a mutation in the glycosyltransferase Large, which leads to altered glycosylation of alpha-DG, and also a severe phenotype. Other informative models for muscle proteins include the knockout mouse for myostatin, which demonstrated that this protein is a negative regulator of muscle growth. Additionally, the stress syndrome in pigs, caused by mutations in the porcine RYR1 gene, helped to localize the gene causing malignant hypertermia and Central Core myopathy in humans. The study of animal models for genetic diseases, in spite of the existence of differences in some phenotypes, can provide important clues to the understanding of the pathogenesis of these disorders and are also very valuable for testing strategies for therapeutic approaches.

Nonsteroidal antiinflammatory drugs (NSAIDs) have been shown to reduce cell growth in several tumors. Among these possible antineoplastic drugs are cyclooxygenase-2 (COX-2)-selective drugs, such as celecoxib, in which antitumoral mechanisms were evaluated in rats bearing Walker-256 (W256) tumor. W256 carcinosarcoma cells were inoculated subcutaneously (10(7) cells/rat) in rats submitted to treatment with celecoxib (25 mg kg(-1)) or vehicle for 14 days. Tumor growth, body-weight gain, and survival data were evaluated. The mechanisms, such as COX-2 expression and activity, oxidative stress, by means of enzymes and lipoperoxidation levels, and apoptosis mediators were also investigated. A reduction in tumor growth and an increased weight gain were observed. Celecoxib provided a higher incidence of survival compared with the control group. Cellular effects are probably COX-2 independent, because neither enzyme expression nor its activity, measured by tumoral PGE(2), showed significant difference between groups. It is probable that this antitumor action is dependent on an apoptotic way, which has been evaluated by the expression of the antiapoptotic protein Bcl-xL, in addition to the cellular changes observed by electronic microscopy. Celecoxib has also a possible involvement with redox homeostasis, because its administration caused significant changes in the activity of oxidative enzymes, such as catalase and superoxide dismutase. These results confirm the antitumor effects of celecoxib in W256 cancer model, contributing to elucidating its antitumoral mechanism and corroborating scientific literature about its effect on other types of cancer.

There is a need of scientific evidence of claimed nutraceutical effects, but also there is a social movement towards the use of natural products and among them algae are seen as rich resources. Within this scenario, the development of methodology for rapid and reliable assessment of markers of efficiency and security of these extracts is necessary. The rat treated with streptozotocin has been proposed as the most appropriate model of systemic oxidative stress for studying antioxidant therapies. Cystoseira is a brown alga containing fucoxanthin and other carothenes whose pressure-assisted extracts were assayed to discover a possible beneficial effect on complications related to diabetes evolution in an acute but short-term model. Urine was selected as the sample and CE-TOF-MS as the analytical technique to obtain the fingerprints in a non-target metabolomic approach. Multivariate data analysis revealed a good clustering of the groups and permitted the putative assignment of compounds statistically significant in the classification. Interestingly a group of compounds associated to lysine glycation and cleavage from proteins was found to be increased in diabetic animals receiving vehicle as compared to control animals receiving vehicle (N6, N6, N6-trimethyl-L-lysine, N-methylnicotinamide, galactosylhydroxylysine, L-carnitine, N6-acetyl-N6-hydroxylysine, fructose-lysine, pipecolic acid, urocanic acid, amino-isobutanoate, formylisoglutamine. Fructoselysine significantly decreased after the treatment changing from a 24% increase to a 19% decrease. CE-MS fingerprinting of urine has provided a group of compounds different to those detected with other techniques and therefore proves the necessity of a cross-platform analysis to obtain a broad view of biological samples.

Animal traits differ not only in mean, but also in variation around the mean. For instance, one sire’s daughter group may be very homogeneous, while another sire’s daughters are much more heterogeneous in performance. The difference in residual variance can partially be explained by genetic differences. Models for such genetic heterogeneity of environmental variance include genetic effects for the mean and residual variance, and a correlation between the genetic effects for the mean and residual variance to measure how the residual variance might vary with the mean. The aim of this thesis was to develop a method based on double hierarchical generalized linear models for estimating genetic heteroscedasticity, and to apply it on four traits in two domestic animal species; teat count and litter size in pigs, and milk production and somatic cell count in dairy cows. The method developed is fast and has been implemented in software that is widely used in animal breeding, which makes it convenient to use. It is based on an approximation of double hierarchical generalized linear models by normal distributions. When having repeated observations on individuals or genetic groups, the estimates were found to be unbiased. For the traits studied, the estimated heritability values for the mean and the residual variance, and the genetic coefficients of variation, were found in the usual ranges reported. The genetic correlation between mean and residual variance was estimated for the pig traits only, and was found to be favorable for litter size, but unfavorable for teat count.

Anthropomorphism has long been considered a cardinal error when describing animals. Ethicists have feared the consequences of misrepresenting animals in their reasoning. Recent research within human-animal studies, however, has sophisticated the notion of anthropomorphism. It is suggested that avoiding anthropomorphism merely creates other morphisms, such as mechanomorphism. Instead of avoiding anthropomorphism, it is argued that it is a communicative strategy that should be used critically. Instances of anthropomorphism in animal ethics are analyzed in this paper. Some analogies made between people and non-human animals in present theories of animal ethics are clear instances of psychological anthropomorphism. Other analogies are implicit cases of cultural anthropomorphism. It is argued that animal ethics needs to take the wider discourse of critical anthropomorphism into account in order to sophisticate the understanding and use of anthropomorphic projections. Anthropomorphism is an efficient tool of communication, and it may be made an adequate one as well.

Este ensaio pretende, de um lado, identificar e descrever os traços dominantes da literatura e das demais poéticas contemporâneas, e, de outro, relatar o processo de transposição de um texto literário – o romance O Quieto Animal da Esquina, de João Gilberto Noll – para um texto de cinema, o roteiro cinematográfico homônimo, de minha autoria. Menos uma tese acadêmica do que uma tentativa de se constituir como um objeto artístico em si mesmo – tanto em sua condição de roteiro, quanto na de uma aproximação eminentemente subjetiva, não “científica”, de um modo de expressão – este ensaio é uma proposta de discussão sobre a rarefação de limites, sobre o diálogo e a interdependência das artes.

Este trabalho buscou consolidar a hipótese de que nas formações campestres nativas do Rio Grande do Sul, não só a aferta de MS, mas também a estrutura da pastagem, são determinantes do desempenho animal, e que esta estrutura pode ser manipulada através de alterações estacionais na oferta.

A desfolhação e o pisoteio animal podem afetar a cobertura vegetal e alterar a dinâmica do calcário no solo. Este trabalho estudou um sistema de integração lavoura-pecuária com níveis de biomassa de pastagem e seu reflexo sobre o solo, o desenvolvimento da soja e a dinâmica da correção da acidez a partir da superfície do solo. O experimento foi conduzido em um Latossolo Vermelho, em uma pastagem de aveia preta + azevém manejada sob diferentes alturas. O delineamento experimental foi o de blocos ao acaso com quatro tratamentos (10, 20, 30 e 40 cm de altura de manejo) e três repetições. Utilizou-se terneiros jovens em pastejo contínuo, com lotação variável, em dois períodos de pastejo (2001 e 2002). No final do primeiro período aplicou-se 4,5 Mg ha-1 de calcário na área, implantando-se a cultura da soja. As alturas da pastagem afetaram a massa de forragem e o desempenho dos animais. Na média dos dois anos, maiores rendimentos individuais e por área foram obtidos com pastagem manejada nas alturas médias de 25 e 10 cm, respectivamente. Após o pastejo, somente no manejo a 10 cm a densidade e a macroporosidade do solo atingiram valores que podem limitar o desenvolvimento da soja na camada de 0-2,5 cm. Efeitos sobre os atributos químicos manifestaram-se até 5 cm de profundidade, cinco meses após a calagem, avançando com o tempo, independentemente da condição anterior de pastejo. A população de plantas e o rendimento da soja diminuíram com a redução da altura de manejo da pastagem. Houve equivalência de rendimento econômico entre todos os tratamentos, sendo a diminuição do rendimento de soja, em situação de alta intensidade de pastejo, compensada pelo aumento do rendimento animal por unidade de área. O rendimento de soja na área sem pastejo foi similar àquele obtido em áreas com intensidade de pastejo leve.

Esse estudo foi compreendido por dois experimentos de campo, no período de outubro/2003 a abril/2004, na Fundação Estadual de Pesquisa Agropecuária (FEPAGRO), localizado em Tupanciretã, no Planalto Médio, região ecoclimática do RS. O principal objetivo do primeiro experimento foi avaliar o desempenho de novilhas de corte e a dinâmica de três pastos constituídos por Panicum maximum cvs. Gatton e Aruana e por Digitaria diversinervis em um sistema silvipastoril (SSP), associadas com acácia-negra (Acacia mearnsii De Wild.) sob duas densidades arbóreas (833 e 500 árvores/ha). Os resultados mostraram que não houve efeito da densidade arbórea com relação à dinâmica dos pastos e no desempenho animal. As cvs. de P. maximum apresentaram maiores resultados de massa de forragem residual do que a D. diversinervis. A taxa de acúmulo diário de MS, forragem total, oferta de forragem real e relação folha/colmo não diferiram entre as espécies. A cv. Gatton obteve maior resultado de forragem disponível quando comparado com a D. diversinervis. O ganho médio diário, ganho por área, animais.dia/ha, lotação animal e carga animal média também não diferiram com relação as forrageiras. O objetivo do segundo experimento foi avaliar o rendimento de matéria seca total de cinco cultivares de Panicum maximum, crescendo dentro e fora de um bosque de Eucalyptus sp. de 17 anos de idade, estabelecido na densidade de 1111 árvores/ha. A produção media de MS e a taxa de crescimento diário foram significativamente menores (P≤0.05) na condição de sombra (5.529 kg/ha) do que em pleno sol (22.346 kg/ha). Na condição de sombra, os resultados das cvs. não diferiram, enquanto que os resultados em pleno sol apresentaram diferenças significativas (P≤0,01), onde a cv. Tanzânia apresentou menores resultados no rendimento de matéria seca e taxa de crescimento e a cv. Mombaça os maiores resultados Pode-se concluir que as cvs. Mombaça, Tobiatã, Gatton e Vencedor são boas promissoras para o uso em SSP. Esse estudo indicou claramente altos níveis de desempenho animal sob pastejo em SSP com algumas cvs. de P. maximum selecionadas ou D. diversinervis usando Acacia-Negra no Sul do Brasil.
