Three different cerium citrate-based precursors were used for synthesizing CeO(2) through thermal treatment. Three morphological types of CeO(2) were obtained. Characterization of these oxides was carried out by XRD patterns, SEM microscopy, N(2) adsorption isotherms, Raman spectroscopy, zeta potential, and UV/Vis luminescence. Ozonation of phenol catalyzed by CeO(2) was studied as a representative reaction of environmental interest. The differences on the catalytic activity showed by these three oxides could be correlated to amounts of Ce(3+) on CeO(2) surface and, consequently, to the demand for oxygen needed to burn each precursor.

Background: The expression levels of the clotting initiator protein Tissue Factor (TF) correlate with vessel density and the histological malignancy grade of glioma patients. Increased procoagulant tonus in high grade tumors (glioblastomas) also indicates a potential role for TF in progression of this disease, and suggests that anticoagulants could be used as adjuvants for its treatment. Objectives: We hypothesized that blocking of TF activity with the tick anticoagulant Ixolaris might interfere with glioblastoma progression. Methods and results: TF was identified in U87-MG cells by flow-cytometric and functional assays (extrinsic tenase). In addition, flow-cytometric analysis demonstrated the exposure of phosphatidylserine in the surface of U87-MG cells, which supported the assembly of intrinsic tenase (FIXa/FVIIIa/FX) and prothrombinase (FVa/FXa/prothrombin) complexes, accounting for the production of FXa and thrombin, respectively. Ixolaris effectively blocked the in vitro TF-dependent procoagulant activity of the U87-MG human glioblastoma cell line and attenuated multimolecular coagulation complexes assembly. Notably, Ixolaris inhibited the in vivo tumorigenic potential of U87-MG cells in nude mice, without observable bleeding. This inhibitory effect of Ixolaris on tumor growth was associated with downregulation of VEGF and reduced tumor vascularization. Conclusion: Our results suggest that Ixolaris might be a promising agent for anti-tumor therapy in humans.

The synthesis and characterization of lanthanide(III) citrates with stoichiometries 1:1 and 2:3; [LnL center dot xH(2)O] and [Ln(2)(LH)(3)center dot 2H(2)O], Ln=La, Ce, Pr, Nd, Sm and Eu are reported. L stands for (C6O7H5)(3-) and LH for (C6O7H6)(2-). Infrared absorption spectra of both series evidence coordination of carboxylate groups through symmetric bridges or chelation. X-ray powder patterns show the amorphous character of [LnL center dot xH(2)O]. The compounds [Ln(2)LH(3)center dot 2H(2)O] are crystalline and isomorphous. Emission spectra of Eu compounds suggest C-2v symmetry for the coordination polyhedron of [LnL center dot xH(2)O] and C-4v for [Ln(2)(LH)(3)center dot 2H(2)O]. Thermal analyses (TG-DTG-DTA) were carried out for both series. The thermal analysis patterns of the two series are quite different and both fit in a 4-step model of thermal decomposition, with lanthanide oxides as final products.

This paper reports the preparation and characterization of poly-{trans-[RuCl(2)(vpy)(4)]-styrene-divinylbenzene} and styrene-divinylbenzene-vinylpiridine filled with nanosilver. Theses materials were synthesized by non aqueous polymerization through a chemical reaction using benzoyl peroxide as the initiator. The nanosilver was obtained from chemical reduction using NaBH(4) as reducing agent and sodium citrate as stabilizer. The nanometric dimension of nanosilver was monitored by UV-visible and confirmed through TEM. The morphology was characterized by SEM and the thermal properties were done by TGA and DSC. The antimicrobial action of the polymers impregnated with nanosilver was evaluated using both microorganisms, Staphylococcus aureus and Escherichia coli. The antimicrobial activity of the poly-{trans-[RuCl(2)(vpy)(4)]-styrene-divinylbenzene} filled with nanosilver was confirmed by the presence of an inhibition halo of the bacterial growth in seeded culture media, but was not confirmed to the styrene-divinylbenzene-vinylpiridine. The present work suggest that trans - [RuCl(2)(vpy)(4)] complex facilitate the release of silver ion from the media.

A systematic and comprehensive study of the interaction of citrate-stabilized gold nanoparticles with triruthenium cluster complexes of general formula [Ru(3)(CH(3)COO)(6)(L)](+) [L = 4-cyanopyridine (4-CNpy), 4,4`-bipyridine (4,4`-bpy) or 4,4`-bis(pyridyl)ethylene (bpe)] has been carried out. The cluster-nanoparticle interaction in solution and the construction of thin films of the hybrid materials were investigated in detail by electronic and surface plasmon resonance (SPR) spectroscopy, Raman scattering spectroscopy and scanning electron microscopy (SEM). Citrate-stabilized gold nanoparticles readily interacted with [Ru(3)O(CH(3)COO)(6)(L)(3)](+) complexes to generate functionalized nanoparticles that tend to aggregate according to rates and extents that depend on the bond strength defined by the characteristics of the cluster L ligands following the sequence bpe > 4,4`-bpy >> 4-CNpy. The formation of compact thin films of hybrid AuNP/[Ru(3)O(CH(3)COO)(6)(L)(3)](+) derivatives with L = bpe and 4,4`-bpy indicated that the stability/lability of AuNP-cluster bonds as well as their solubility are important parameters that influence the film contruction process. Fluorine-doped tin oxide electrodes modified with thin films of these nanomaterials exhibited similar electrocatalytic activity but much higher sensitivity than a conventional gold electrode in the oxidation of nitrite ion to nitrate depending on the bridging cluster complex, demonstrating the high potential for the development of amperometric sensors.

This paper investigates the structure of the pro; ducts obtained from the polymerization of aniline with ammonium persulfate in a citrate/phosphate buffer solution at pH 3 by resonance Raman, NMR, FTIR, and UV-vis-NIR spectroscopies. All the spectroscopic data showed that the major product presented segments that were formed by a 1,4-Michael reaction between aniline and p-benzoquinone monoimine, ruling out the formation of polyazane structure that has been recently proposed. The characterization of samples obtained at different stages of the reaction indicated that, as the reaction progressed, phenazine units were formed and 1,4-Michael-type adducts were hydrolyzed/oxidized to yield benzoquinone. Raman mapping data suggested that phenazine-like segments could be related to the formation of the microspheres morphology.

A dosing algorithm including genetic (VKORC1 and CYP2C9 genotypes) and nongenetic factors (age, weight, therapeutic indication, and cotreatment with amiodarone or simvastatin) explained 51% of the variance in stable weekly warfarin doses in 390 patients attending an anticoagulant clinic in a Brazilian public hospital. The VKORC1 3673G>A genotype was the most important predictor of warfarin dose, with a partial R(2) value of 23.9%. Replacing the VKORC1 3673G>A genotype with VKORC1 diplotype did not increase the algorithm`s predictive power. We suggest that three other single-nucleotide polymorphisms (SNPs) (5808T>G, 6853G>C, and 9041G>A) that are in strong linkage disequilibrium (LD) with 3673G>A would be equally good predictors of the warfarin dose requirement. The algorithm`s predictive power was similar across the self-identified ""race/color"" subsets. ""Race/color"" was not associated with stable warfarin dose in the multiple regression model, although the required warfarin dose was significantly lower (P = 0.006) in white (29 +/- 13 mg/week, n = 196) than in black patients (35 +/- 15 mg/week, n = 76).

CuO/CeO(2), CuO/Al(2)O(3) and CuO/CeO(2)-Al(2)O(3) catalysts, with CuO loading varying from 1 to 5 wt.%, were prepared by the citrate method and applied to the preferential oxidation of carbon monoxide in a reaction medium containing large amounts of hydrogen (PROX-CO). The compounds were characterized ex situ by X-ray diffraction, specific surface area measurements, temperature-programmed reduction and temperature-programmed reduction of oxidized surfaces; XANES-PROX in situ experiments were also carried out to study the copper oxidation state under PROX-CO conditions. These analyses showed that in the reaction medium the Cu(0) is present as dispersed particles. On the ceria, these metallic particles are smaller and more finely dispersed, resulting in a stronger metal-support interaction than in CuO/Al(2)O(3) or CuO/CeO(2)-Al(2)O(3) catalysts, providing higher PROX-CO activity and better selectivity in the conversion of CO to CO(2) despite the greater BET area presented by samples supported on alumina. It is also shown that the lower CuO content, the higher metal dispersion and consequently the catalytic activity. The redox properties of the ceria support also contributed to catalytic performance. Hiperoxalúria ocorre após ressecção extensa do intestino delgado na presença de um colon intacto. De 1984 a 1997, 40 pacientes com ressecção de intestino delgado, extensão maior do que 30cm, foram estudados com o objetivo de se avaliar a prevalência de litíase renal nesta população e alterações metabólicas associadas. Sete pacientes, correspondendo a 17,5% da população em estudo, desenvolveram um ou mais cálculos renais durante o período de observação correspondendo a uma incidência de 3,1 casos por 100 pessoas/ano. Os pacientes foram então divididos em dois grupos. Grupo 1 formado por 33 pacientes com ressecção intestinal que não formaram cálculos renais durante o período de observação e o Grupo 2 com 7 pacientes que formaram um ou mais cálculos renais após a ressecção intestinal. A excreção urinária de oxalato foi, em média, maior no grupo 2, que desenvolveram litíase renal, comparado com o grupo 1, não formadores de cálculos renais (49,2±23,8 mg/24horas Vs 30,6±3,4 mg/24h, p=0,004). De forma oposta, o magnésio foi menor nos pacientes formadores de cálculos renais comparado com os pacientes não formadores de cálculos (54,5±17,9 mg/24horas Vs 94,9±8,5 mg/24horas, p=0,028), assim como o citrato também foi menor nos pacientes com cálculo renal, mas sem diferença do ponto de vista estatístico (265,9±55,8 mEq/24horas Vs 404,8±49,7mEq/24horas, P= 0,278). Este estudo mostra que novos cálculos urinários podem ser detectados em um número significativo de pacientes que se submetem à cirurgia de ressecção do intestino delgado, provavelmente associado ao aumento da excreção de oxalato e diminuição de, pelo menos, um dos inibidores da cristalização de oxalato de cálcio, o magnésio. Por isto, se torna importante a monitorização destes pacientes, regularmente, mesmo após vários anos da cirurgia. É importante a monitorização pelo menos do oxalato e magnésio, na urina de 24 horas, já que ambos estão associados a um maior risco de desenvolvimento de cálculos renais.

As endometrites bacterianas são uma das principais causas de infertilidade na égua. Entre os tratamentos utilizados nesta patologia, estão as infusões uterinas de plasma homólogo acrescido de leucócitos e a infusão de leucócitos heterólogos criopreservados. O presente trabalho teve por objetivo realizar testes in vivo e in vitro, que foram descritos em dois artigos. No primeiro artigo, objetivou-se avaliar in vitro a quimiotaxia dos leucócitos eqüinos em relação a diferentes quimioatraentes, bem como sua vitalidade e produção de radicais livres de oxigênio (ROS) pós-descongelamento. No experimento 1, testou-se dextrose em concentrações de 0, 1, 2 e 6%, acrescida ou não de interleucina-8 (IL-8), como quimioatraente para leucócitos eqüinos suspensos em salina fosfatada tamponada (PBS) ou em R3F; no experimento 2, testou-se plasma homólogo ou heterólogo, a 3% (com ou sem IL-8), 10, 30, 60 e 90%, como quimioatraente; no experimento 3, foi testada a quimiotaxia de leucócitos íntegros em relação a diferentes quantidades de leucócitos lisados. O experimento 4 avaliou a vitalidade e a geração de ROS pelos leucócitos após o descongelamento, comparando quatro graus de diluição em NaCl e PBS. Concluiu-se que a dextrose não apresenta bom efeito quimioatraente para leucócitos eqüinos. Dentre as concentrações de plasma utilizadas, concentrações de plasma homólogo entre 10% e 60% apresentam bom efeito quimiotático. Já o plasma heterólogo apresentou boa atração de leucócitos quando em concentrações de 10% e de 30% Na concentração de 10x106/mL, os leucócitos lisados foram capazes de atrair leucócitos em proporção semelhante à da IL-8. Com relação aos testes-pós-descongelamento, células ressuspendidas em PBS ou NaCl apresentam vitalidade e geração de radicais livres de oxigênio semelhantes, quando incubados por até 15 minutos. No segundo artigo, realizaram-se testes in vivo compararando cinco tratamentos em éguas, em estro, experimentalmente infectadas com Streptococcus equi subsp.zooepidemicus. Foram utilizadas 25 éguas, 20 consideradas resistentes, e 5 éguas susceptíveis à endometrite. Após 24 horas, os animais foram submetidos a exame clínico, bacteriológico e citológico. Com a presença de quadro clínico de endometrite, as éguas eram submetidas a um dos seguintes tratamentos: 1-Infusão de 120mL de plasma homológo com de leucócitos frescos; 2-– Infusão de 4 mL contendo 800 x 106 leucócitos íntegros, congelados; 3-Leucócitos lisados - Infusão de 4 mL contendo 800 x 106 leucócitos lisados; 4- IL-8- Infusão de 4 mL contendo 25 ng/mL de IL-8 congelada; 5-Controle – Infusão de 4 mL de meio R3F congelado. Os exames clínico, bacteriológico e citológico foram realizados diariamente até o sétimo dia pós-infecção, ou até a eliminação da bactéria, ou até a ausência de neutrófilos no esfregaço citológico. Após os exames, as éguas receberam o tratamento designado, sendo estes realizados diariamente até a ausência de crescimento bacteriano no exame microbiológico ou por, no máximo, quatro dias. No sétimo dia, todas as éguas foram tratadas com infusão intra-uterina de 5.000.000 UI de penicilina G potássica cristalina. Após o término de um tratamento, aguardava-se cerca de sete dias para a reinfecção, num outro ciclo, quando um outro tratamento era aplicados. Todas as éguas foram submetidas a todos os tratamentos, perfazendo um total de 125 infecções experimentais num delineamento experimental do tipo Quadrado Latino. Não se observaram diferenças significativas no tempo de eliminação bacteriana nas éguas resistentes. Entretanto, nas éguas susceptíveis, observou-se uma cura bacteriana mais rápida quando as éguas foram tratadas com leucócitos frescos, leucócitos congelados e leucócitos lisados, em relação às tratadas com IL-8 e as do grupo controle. Concluiu-se que o efeito bactericida dos leucócitos viáveis ou lisados, associados ou não à presença de fatores de opsonização do plasma, foi o responsável pela cura bacteriológica e que o efeito quimioatraente de neutrófilos, plasma, restos celulares e IL-8 não influiu no tempo de eliminação bacteriana.

A necessidade de se prolongar o período pós-colheita fez com que a utilização de tratamentos em pós-colheita com o uso de defensivos agrícolas aumentasse nas últimas décadas. No entanto, a preocupação de consumidores quanto a resíduos as deficiências de controle observadas incentivaram a procura por tratamentos alternativos, entre os quais os tratamentos com calor vem sendo testados em frutos de várias espécies. No presente trabalho estudou-se o efeito do tratamento com calor no crescimento do micélio e germinação dos conídios de Botryosphaeria dothidea e, avaliou-se a viabilidade da termoterapia com calor, isolada ou em combinação com carbonato de sódio no controle da podridão branca e na manutenção da qualidade das maçãs ‘Fuji’ em diferentes estádios de maturação, mantidas ou não sob armazenamento refrigerado. Foi objetivo também verificar a expressão de genes que são regulados durante a interação maçã-Botryosphaeria dothidea, após tratamento com calor. Os experimentos foram conduzidos testando-se combinações de diferentes temperaturas e períodos de exposição ao calor sobre o patógeno B. dothidea e sobre frutos inoculados e não inoculados artificialmente com o patógeno. Os resultados obtidos sobre o patógeno mostraram que, independente da concentração de caldo batata-dextrose (BD) utilizada na suspensão, 45% dos conídios de B. dothidea germinaram nos frutos após 24h e 91% após 48h de incubação a 26°C. Constatou-se um aumento significativo da germinação dos conídios quando acrescentado 2,4% de caldo BD a suspensão. Sobre o efeito da temperatura no patógeno, as temperaturas inferiores a 62°C, em qualquer período de exposição não foram capazes de inibir o micélio do patógeno. Os conídios submetidos à temperatura de 58°C somente foram inativados quando o tratamento foi estabelecido por 60s. A termoterapia com calor associada ou não ao carbonato de sódio reduziu a incidência e severidade da doença podridão branca. A severidade da doença foi influenciada pelo estádio de maturação dos frutos verificando pelo aumento no número de lesões/fruto quanto mais avançada a maturação das maçãs. Os resultados obtidos no RT-PCR evidenciaram que os protocolos utilizados não produziram RNA de alta qualidade e desta forma inviabilizaram a análise da expressão diferencial dos genes na interação maçã-B.dothidea.

The occurrence of bioactive compounds in marine organisms comes awaking the interest of the pharmaceutical industry. Heparin, a sulfated polysaccharide which presence was already identified in several marine invertebrates, is very attractive due its remarkable functional versatility. Besides to intervene in blood coagulation, this molecule has a great anti-inflammatory potential. However, its strong anticoagulant activity difficult the clinical exploitation of its anti-inflammatory properties. Thus, the aims of this work were to evaluate the effect of a heparin-like compound (heparinoid), isolated from the cephalotorax of the Litopenaeus vannamei shrimp, on the inflammatory response, hemostasia and synthesis of antithrombotic heparan sulfate by endothelial cells, besides studying some aspects concerning its structure. The purified heparinoid was structurally characterized following an analytical boarding, involving electrophoresis and chromatography. The structural analysis have shown that this compound possess a high content of glucuronic acid residues and disulfated disaccharide units. In contrast to mammalian heparin, the heparinoid was incapable to stimulate the synthesis of heparan sulfate by endothelial cells in the tested concentrations, beyond to show reduced anticoagulant activity and hemorrhagic effect. In a model of acute inflammation, the compound isolated from the shrimp reduced more than 50% of the cellular infiltration. Besides reduce the activity of MMP-9 and proMMP-2 of the peritoneal lavage of inflamed animals, the heparinoid also reduced the activity of MMP-9 secreted by activated human leukocytes. These results demonstrate the potential of heparinoid from L. vannamei to intervene in the inflammatory response. For possessing reduced anticoagulant activity and hemorrhagic effect, this compound can serve as a structural model to direct the development of more specific therapeutical agents to the treatment of inflammatory diseases

Sulfated polysaccharides (PS) are biomolecules with a great biotechnological potential. There are few data about PS from high plants. In addition, pharmacological activities of PS from plants have not been carrying out. The aim of this work was extract PS from the angiosperm Halodule wrightii and study their anticoagulant and antioxidant activities. Histological analysis showed the presence of the PS manly in the roots. A polysaccharide-rich extract was obtained from H. wrightii by proteolysis followed by methanol and TCA precipitation. Chemical, infra-red analysis and agarose gel electrophoresis in 1.3 diaminopropane acetate buffer confirmed the presence of sulfated polysaccharides made by glucose, galactose, xylose and sulfate residues in the proportion 1: 0,9: 1: 1. In addition polyacrilamide electrophoresis have shown that extract is mainly compose by 11kDa sulfated polysaccharides. Pharmacological analysis have shown total antioxidant capacity (CAT) that resulted in 15,21 μg for equivalent of ascorbic acid, scavenging activity of the DPPH radical with 41,36 % of scavenging, activity of reducing power with the maximum of 0,290 nm (50 % of vitamin C activity) and scavenging activity superoxide radical (O2-) with a maximum of 32,23 %. Chelating activity of metal less than 4% and scavenging activity of the radical hydroxyl (OH-) less than 2%. Time of activated partial tromboplastin (aPTT) doubling the time of coagulation from 20μg of and protrombin time (PT) was not present. The data indicate that PS from Halodule wrightii could be considered for future applications in medicine, food production or cosmetic industry

In recent years, sulfated polysaccharides (SP) from marine algae have emerged as an important class of natural biopolymers with potential pharmacology applications. Among these, SP isolated from the cell walls of red algae have been study due to their anticoagulant,antithrombotic and anti-inflammatory activities. In the present study, three sulfated polysaccharides fractions denominated F1.5v, F2.0v and F3.0v were obtained from seaweed G. caudate by proteolysis followed to acetone fractionation. Gel electrophoresis using 0.05 M 1,3-diaminopropane-acetate buffer, pH 9,0, stained with 0.1% toluidine blue, showed the presence of SP in all fractions. The chemical analysis demonstrated that all the fractions are composed mainly of galactose. These compounds were evaluated in anticoagulant, antioxidant and antiproliferative activities. In anticoagulant activity evaluated through aPTT and PT tests, no one fractions presented anticoagulant activity at tested concentrations (0.1 mg/mL; 1.0 mg/mL; 2.0 mg/mL).The antioxidant activities of the three fractions were evaluated by the following in vitro systems: Total antioxidant capacity, superoxide and hydroxyl radical scavenging, ferrous chelating activity and reducing power. The fractions were found to have different levels of antioxidant activity in the systems tested. F1.5v shows the highest activity, especially in the ferrous chelating system, with 70% of ferrous inhibiting at 1.0 mg.mL-1. Finally, all the fractions showed dose-dependent antiproliferative activity against HeLa cells. The fractions F1.5v and F2.0v presented the highest antiproliferative activity at 2.0 mg/mL with 42.7% and 37.0% of inhibition, respectively. Ours results suggests that the sulfated polysaccharides from seaweed G. caudata are promising compounds in antioxidant and/or antitumor therapy
