Foi investigada a susceptibilidade de sete linhagens isogênicas de camundongos à infecção experimental, primária e secundária, por Strongyloides venezuelensis a fim de servir de base para estudos genéticos sobre a resistência. Foram utilizados 12 camundongos machos, com seis semanas de idade, das seguintes linhagens isogênicas: A/J, BALB/c, CBA/J, C3H/Hepos, C57BL/6, DBA/2 e NIH. Os animais foram inoculados, via sub-cutânea, com 2000 larvas infectantes. As contagens médias (± desvio padrão) de parasitas no intestino delgado dos camundongos seis dias após a infecção, em ordem crescente, foram: 28 (± 19) na linhagem NIH; 647 (± 228) na BALB/c; 709 (± 425) na DBA/2; 731 (± 151) na C3H/Hepos, 801 (± 174) na CBA/J; 1024 (± 267) na C57BL/6 e 1313 (± 483) na A/J. Os camundongos C57BL/6 apresentaram as mais elevadas contagens de ovos de S. venezuelensis por grama de fezes (OPG) e os NIH, as mais baixas. Não foram detectados ovos nos exames de fezes e não foram encontrados parasitas no intestino delgado dos animais re-infectados 14 dias após a infecção primária. A linhagem NIH apresentou elevada resistência contra as infecções primárias por S. venezuelensis. Entre as outras seis linhagens, uma das mais susceptíveis foi a linhagem C57BL/6.

Avaliaram-se o desenvolvimento e a sobrevivência dos estágios de vida livre de Haemonchus contortus em fezes obtidas de ovinos infectados por esse nematódeo, depositadas nas quatro estações do ano, em pastagens de três espécies de gramíneas (Brachiaria decumbens cv Australiana, Cynodon dactylon cv. Coast-cross e Panicum maximum cv. Aruana), com duas alturas (5cm e 30cm) no momento da deposição das fezes. Amostras de fezes e de capim foram coletadas uma, duas, quatro, oito, 12 e 16 semanas após a deposição das fezes e processadas em laboratório, para recuperação de larvas infectantes. O maior número de larvas infectantes foi recuperado das amostras de capim e de fezes nas pastagens com 30cm de altura. As condições climáticas que mais favoreceram o desenvolvimento e a sobrevivência das larvas nas fezes, bem como a sobrevivência e a manutenção das larvas no capim foram aquelas com temperaturas médias em torno de 17ºC, acompanhadas de baixas precipitações pluviométricas. As temperaturas mais altas e as precipitações elevadas, que ocorrem na chamada estação das águas, foram desfavoráveis para a recuperação de larvas infectantes da pastagem. de forma geral, a pastagem de Aruana foi a que possibilitou as maiores recuperações de larvas.

Estudaram-se os efeitos do pastejo alternado de ovinos e bovinos e do pastejo rotacionado sobre o controle da verminose em ovelhas. Utilizou-se uma área experimental composta por três módulos de 1,67ha cada. Os módulos foram subdivididos em oito piquetes. Vinte ovelhas foram colocadas no módulo 1 e quatro bovinos adultos no módulo 2. Os animais permaneceram em cada piquete do módulo por cinco dias, totalizando 40 dias de permanência em cada módulo. Ao final desse período, as ovelhas foram transferidas para o módulo onde estavam os bovinos e estes para o módulo onde estavam os ovinos, mantendo esse esquema até o final do experimento. Um grupo-controle de 20 ovelhas foi mantido, também em sistema rotacionado, em um terceiro módulo, sem compartilhar a pastagem. As ovelhas submetidas ao manejo com bovinos apresentaram o menor grau de infecção por nematódeos gastrintestinais e os maiores valores de volume globular. O pastejo rotacionado de ovinos, sem a utilização de bovinos, não foi eficiente no controle da verminose das ovelhas. A utilização do pastejo rotacionado e alternado de ovinos e bovinos adultos exerceu efeito benéfico significativo no controle da verminose ovina.

Cellular and humoral immune response, as well as cytokine gene expression, was assessed in Nelore cattle with different degrees of resistance to Cooperia punctata natural infection. One hundred cattle (male, weaned, 11-12 months old), kept together on pasture, were evaluated. Faecal and blood samples were collected for parasitological and immunological assays. Based on nematode faecal egg counts (FEC) and worm burden, the seven most resistant and the eight most susceptible animals were selected. Tissue samples of the small intestine were collected for histological quantification of inflammatory cells and analysis of cytokine gene expression (IL-2, IL-4, IL-8, IL-1 2p35, IL-13, TNF-alpha, IFN-gamma, MCP-1, MCP-2, and MUC- 1) using real-time RT-PCR. Mucus samples were also collected for IgA levels determination. Serum IgG1 mean levels against C. punctata antigens were higher in the resistant group, but significant differences between groups were only observed 14 days after the beginning of the experiment against infective larvae (1-3) and 14 and 84 days against adult antigens. The resistant group also presented higher IgA levels against C. punctata (L3 and adult) antigens with significant difference 14 days after the beginning of the trial (P < 0.05). In the small-intestine mucosa, levels of IgA anti-L3 and anti-adult C. punctata were higher in the resistant group, compared with the susceptible group (P < 0.05). Gene expression of both T(H)2 cytokines (IL-4 and IL-13) in the resistant group and T(H)1 cytokines (IL-2, IL-1 2p35, IFN-gamma and MCP-1) in the susceptible group was up-regulated. Such results suggested that immune response to C. punctata was probably mediated by TH2 cytokines in the resistant group and by T(H)1 cytokines in the susceptible group. (C) 2008 Elsevier B.V. The objective of this study was to determine the level of resistance of Haemonchus contortus and Trichostrongylus colubriformis in sheep to levamisole albendazole ivermectin moxidectin closantel and trichlorfon The parasites were isolated from sheep naturally infected by gastrointestinal nematodes and were then kept in monospecifically infected lambs for production of infective larvae (L3) of both species Forty-two lambs at three months of age were simultaneously artificially infected with 4000 L3 of H contortus and 4000 L3 of T colubriformis The animals were allocated Into seven groups with six animals each that received one of the following treatments Group 1-control no treatment Group 2-moxidectin (0 2 mg/kg body weight (BW)) Group 3-closantel (10 mg/kg BW) Group 4-trichlorfon (100 mg/kg BW) Group 5-levamisole phosphate (4 7 mg/kg BW) Group 6-albendazole (5 0 mg/kg BW) and Group 7-ivermectin (0 2 mg/kg BW) Nematode fecal egg counts (FEC) were carried out on the day of treatment and again at 3 7 10 and 14 days post-treatment on the same occasions composite fecal cultures were prepared for each group for production of L3 which were identified into genus The animals were sacrificed for worm counts at 14 days after treatment The efficacy of each treatment was calculated from the arithmetic mean of the FEC or worm burden of the treated group compared with the values of the control group Only trichlorfon and moxidectin treatments resulted in a significant reduction of H contortus recorded at necropsy (73% and 45% respectively) Moxidectin reduced T colubriformis worm burdens by 82% and albendazole by 19% All other anthelmintics resulted in no significant reduction in the numbers of worms found at necropsy In conclusion the Isolates of H contortus and T colubriformis showed multiple resistance to all groups of anthelmintics tested This is the first report based on the controlled efficacy test to show resistance of T colubriformis to macrocyclic lactones in Brazil (C) 2010 Elsevier B.V. Seventy specimens of Hypsiboas albopunctatus (Anura: Hylidae) were collected and surveyed for endoparasites in Brasilia, Brazil at the end of the rainy season (February and March) in 2005. Sixteen males (22.86%) were infected by Rhabdias sp., Aplectana sp., Cosmocerca sp., and Falcaustra mascula. Rhabdias sp., a lung parasite, presented the highest prevalence (8.57%), mean intensity of infestation (1.333), and mean abundance (0.114). Moreover, this is the first report of a rhabdiasid nematode in the Hylidae in Brazil. The intestinal parasites, Cosmocerca sp. and Aplectana sp., had a similar prevalence (5.71%), but the former presented a higher mean intensity of infestation and mean abundance. The behavior and food habits of amphibians are responsible for the differences in their helminthofauna, and this study presents four new records of nematodes in H. albopunctatus.

Anthelmintic resistance is a worldwide concern in small ruminant industry and new plant-derived compounds are being studied for their potential use against gastrointestinal nematodes. Mentha piperita, Cymbopogon martinii and Cymbopogon schoenanthus essential oils were evaluated against developmental stages of trichostrongylids from sheep naturally infected (95% Haemonchus contortus and 5% Trichostrogylus spp.) through the egg hatch assay (EHA), larval development assay (LDA), larval feeding inhibition assay (LFIA), and the larval exsheathment assay (LEA). The major constituent of the essential oils, quantified by gas chromatography for M. piperita oil was menthol (42.5%), while for C. martinii and C. schoenanthus the main component was geraniol (81.4% and 62.5%, respectively). In all in vitro tests C. schoenanthus essential oil had the best activity against ovine trichostrongylids followed by C. martini, while M. piperita presented the least activity. Cymbopogon schoenanthus essential oil had LC(50) value of 0.045 mg/ml in EHA, 0.063 mg/ml in LDA, 0.009 mg/ml in LFIA, and 24.66 mg/ml in LEA. The anthelmintic activity of essential oils followed the same pattern in all in vitro tests, suggesting C. schoenanthus essential oil could be an interesting candidate for nematode control, although in vivo studies are necessary to validate the anthelmintic properties of this oil.
