Despite important progress on Amazonian floodplain research, the flooded forest of the Negro River igapó has been little investigated. In particular, no study has previously focused the linkage between fluvial geomorphology and the floristic variation across the course of the river. In this paper we describe and interpret relations between igapó forest, fluvial geomorphology and the spatial evolution of the igapó forest through the Holocene. Therefore, we investigate the effect of geomorphological units of the floodplain and channel patterns on tree diversity, composition and structural parameters of the late-successional igapó forest. Our results show that sites sharing almost identical flooding regime, exhibit variable tree assemblages, species richness and structural parameters such as basal area, tree density and tree heights, indicating a trend in which the geomorphologic styles seem to partially control the organization of igapó's tree communities. This can be also explained by the high variability of well-developed geomorphologic units in short distances and concentrated in small areas. In this dynamic the inputs from the species pool of tributary rivers play a crucial role, but also the depositional and erosional processes associated with the evolution of the floodplain during the Holocene may control floristic and structural components of the igapó forests. These results suggest that a comprehensive approach integrating floristic and geomorphologic methods is needed to understand the distribution of the complex vegetation patterns in complex floodplains such as the igapó of the Negro River. This combination of approaches may introduce a better comprehension of the temporal and spatial evolutionary analysis and a logic rationale to understand the vegetation distribution and variability in function of major landforms, soil distributions and hydrology. Thus, by integrating the past into macroecological analyses will sharpen our understanding of the underlying forces for contemporary floristic patterns along the inundation forests of the Negro River. © 2013 Elsevier Ltd.

Este estudo apresenta a estimativa dos parâmetros florísticos e estruturais (determinação da espécie, altura, diâmetro a altura do Peito - DAP e biomassa) do mangue a partir de informações da superfície adquiridas remotamente com os sensores Laser Detection and Range (LIDAR), Shuttle Radar Topography Mission (SRTM) e ortofotos na Ilha dos Guarás, conjunto de arquipélagos localizado a 30 km da desembocadura do rio amazonas. Para esse trabalho foram utilizadas informações do SRTM, LIDAR e fotografias aéreas processadas e ortorretificadas durante dois sobrevôos realizados entre o mês de julho e agosto de 2011. Com a ortofoto foi feito o mapa do reconhecimento de unidades geobotânicas que delimitou apenas a classe mangue. Em seguida, foi realizada a correção da altura elipsoidal para a altura ortométrica, onde a nuvem de pontos foi interpolada pelo método vizinho mais próximo, gerando Modelo Digital de Elevação (MDE) LIDAR (full points) com RMSE de 0,88 cm e por meio de uma linguagem macro foi estatisticamente separadas as informações do último pulso da superfície, conhecido também por ground points. Em seguida, os dados foram interpolados pelo método de krigeagem que gerou o valor de Modelo Digital de Superfície (MDS), o qual foi subtraído do MDE. Com base no Modelo Digital de Vegetação (MDV) foram definidos os sítios de coleta e selecionadas as árvores ascendentes, intermediárias e emergentes, porte no qual foi medido o DAP e altura. No total foram coletadas 212 amostras individuais de mangue e para assegurar o nível de acurácia do conjunto coletado, foi realizado o cálculo de RMSE entre as alturas do LIDAR e Campo, que resultou em RMSE= 1,10 m. Os modelos escolhidos para calibração LIDAR e altura de campo foi do tipo linear, com R2 = 91% e RMSE= 0,98 cm e para calibração da DAP e altura de campo foi escolhido o modelo Logarítmico R2 = 74,1%. Nos resultados da calibração do SRTM o modelo logarítmico também foi o mais adequado para a relação entre altura média e SRTM com R2 = 91% e RMSE de 2,2 m e DAP Médio e SRTM, com R2 = 88% e RMSE 2,2 cm. A partir de um inventário foi realizada a estimativa da biomassa por espécie por meio das equações alométricas de Fromard e posteriormente os resultados foram espacializados em forma de mapas com alto nível de detalhamento oriundo das informações LIDAR e SRTM corrigido e ortofotos.

The aim of this research was to analyze the composition and structure of the plant community in four phytophysiognomies at Pantanal Poconé-Mato Grosso, assuming that each sample group is a type of community, and there would be a corresponding group of species that characterize that community. We set up five plots of 50 × 50 m and subdivided each one into subplots of 10 × 10 m. In each subplot, individuals with CAP (circunference at breast height) > 10 cm were sampled. To check the structural and floristic similarity of the five vegetation types the unweighted averages grouping method was used - UPGMA, with Jaccard's coefficient and Bray-Curtis, respectively. DCA ordination of plots was used for inundation gradient analysis and TWINSPAN indicator species groups for the three strata. In the four areas 55 species belonging to 30 families were sampled. The UPMGA indicated floristic and structural differences between cambarazal, savanna types and campo de murundu and similarity between the semideciduous formations. The DCA separated the vegetation types according to the level of flooding and TWINSPAN indicated the existence of typical species for each vegetation type. In the Pantanal, some species may be restricted to certain types of vegetation, and these species can be used as indicators of the effect of flooding and possible changes in flood pulse dynamics in plant communities.

The present study aimed to analyze the floristic and structural descriptors of tree species natural regeneration in a forest sector with synchronized bamboo (Merostachys multiramea Hackel) die-off (CT) and an adjacent area with continuous canopy cover (ST) in an araucaria forest fragment in the municipality of Lages, Santa Catarina state. A total of 14, 5x5m, plots (six plots in CT sector and eight in ST sector) were allocated, where all tree species regenerative individual with circumference at breast height smaller than 15cm and height higher than 25cm was measured (diameter at soil level) and identified. The richest families were: Myrtaceae (nine), Solanaceae (six) and Aquifoliaceae (four). The Shannon Diversity Index in ST and CT sectors were respectively 2.73 and 2.31. The species with the highest importance values in CT sector were Solanum variabile, Piptocarpha angustifolia, Mimosa scabrella, Jacaranda puberula and Solanum pseudoquina. In ST sector, the species with highest importance values were Myrsine lorentziana, Casearia decandra, Cinnamodendron dinisii, Drimys brasiliensis and Ilex paraguariensis. The results showed that the synchronized bamboo die-off influenced the spatial variation in the floristic and structural descriptors of the tree species natural regeneration.

Glucose dehydrogenase (EC 1.1.1.47) from the halophilic Archaeon Haloferax mediterranei belongs to the medium-chain alcohol dehydrogenase superfamily and requires a zinc ion for catalysis. The zinc ion is coordinated by a histidine, a water molecule and two other ligands from the protein or the substrate, which vary during the catalytic cycle of the enzyme. In many enzymes of this superfamily one of the zinc ligands is commonly cysteine, which is replaced by an aspartate residue at position 38 in the halophilic enzyme. This change has been only observed in glucose dehydrogenases from extremely halophilic microorganisms belonging to the Archaea Domain. This paper describes biochemical studies and structural comparisons to analyze the role of sequence differences between thermophilic and halophilic glucose dehydrogenases which contain a zinc ion within the protein surrounded by three ligands. Whilst the catalytic activity of the D38C GlcDH mutant is reduced, its thermal stability is enhanced, consistent with the greater structural similarity between this mutant and the homologous thermophilic enzyme from Thermoplasma acidophilum.

O presente estudo objetivou conhecer os padrões da composição florística e estrutural do componente arbóreo de um trecho de remanescente de Floresta Ombrófila Mista Montana em Campos Novos - SC e determinar as variáveis ambientais que influenciam estes padrões. Para isso, foi amostrado 1 ha de floresta por meio de 50 parcelas de 10 × 20 m dispostas de forma sistemática, distanciada 30 m entre si, no remanescente florestal. Dentro das parcelas foram identificados e mensurados (circunferência medida a altura do peito, CAP, e altura total) CAP, e altura total) todos os indivíduos arbóreos vivos com CAP ≥ 15,7 cm. Os dados ambientais relacionados às propriedades químicas e físicas dos solos e à topografia também foram coletados em cada parcela. Foram calculados o índice de Shannon-Wiener (H’), a equabilidade de Pielou (J’) e os estimadores fitossociológicos. A organização florístico-estrutural do fragmento foi analisada por meio de uma NMDS (Nonmetric multidimensional scalling). As variáveis ambientais foram ajustadas a posteriori à ordenaçã o produzida, sendo aquelas significativas (p < 0,05) plotadas na forma de vetores. Foram amostrados 1.027 indivíduos, que totalizaram uma área basal de 43,57 m2, distribuídos em 88 espécies e 41 famílias botânicas. A diversidade do remanescente estudado foi relativamente alta (H’=3,59) e a dominância baixa (J’=0,80). A espécie de maior VI foi Araucaria angustifolia (Bertol.) Kuntze (14,44%). A análise multivariada NMDS indicou um gradiente florístico-estrutural relacionado à cota média (altitude), saturação de bases, pH e teores de P nos solos.

We examined large-scale spatial variation of structural parameters and floristic composition in open Clusia scrub, a vegetation type of the Brazilian "restingas" (sandy coastal plain vegetation). This vegetation is organized in islands separated by sandy stretches with sparse herbaceous vegetation. We located 12 sample areas on three consecutive beach ridges, lying parallel to the coastline and at different distances from the ocean, in close proximity to two lagoons (Cabiúnas and Comprida). Each sample area was divided into three strips. We used the line intercept method to sample all woody plants ³ 50 cm tall. We used nested ANOVA to verify structural variation between different sampling scales. TWINSPAN analysis was performed to examine the variation in floristic composition between areas. The overall diversity index was 3.07. Six species are repeatedly dominant throughout the entire sampling area. There was homogeneity in relation to diversity and species richness between beach ridges but not within beach ridges. Floristic composition and structural parameters did not vary in relation to distance from the sea but floristic composition did vary as a function of proximity to Cabiúnas or Comprida lagoon. Differences in plant cover between sample areas may be related to the paleoformation of this sandy coastal plain.

The enzymatic activity of thioredoxin reductase enzymes is endowed by at least two redox centers: a flavin and a dithiol/disulfide CXXC motif. The interaction between thioredoxin reductase and thioredoxin is generally species-specific, but the molecular aspects related to this phenomenon remain elusive. Here, we investigated the yeast cytosolic thioredoxin system, which is composed of NADPH, thioredoxin reductase (ScTrxR1), and thioredoxin 1 (ScTrx1) or thioredoxin 2 (ScTrx2). We showed that ScTrxR1 was able to efficiently reduce yeast thioredoxins (mitochondrial and cytosolic) but failed to reduce the human and Escherichia coli thioredoxin counterparts. To gain insights into this specificity, the crystallographic structure of oxidized ScTrxR1 was solved at 2.4 angstrom resolution. The protein topology of the redox centers indicated the necessity of a large structural rearrangement for FAD and thioredoxin reduction using NADPH. Therefore, we modeled a large structural rotation between the two ScTrxR1 domains (based on the previously described crystal structure, PDB code 1F6M). Employing diverse approaches including enzymatic assays, site-directed mutagenesis, amino acid sequence alignment, and structure comparisons, insights were obtained about the features involved in the species-specificity phenomenon, such as complementary electronic parameters between the surfaces of ScTrxR1 and yeast thioredoxin enzymes and loops and residues (such as Ser(72) in ScTrx2). Finally, structural comparisons and amino acid alignments led us to propose a new classification that includes a larger number of enzymes with thioredoxin reductase activity, neglected in the low/high molecular weight classification.

Spider venom sphingomyelinases D catalyze the hydrolysis of sphingomyelin via an Mg2+ ion-dependent acid-base catalytic mechanism which involves two histidines. In the crystal structure of the sulfate free enzyme determined at 1.85 angstrom resolution, the metal ion is tetrahedrally coordinated instead of the trigonal-bipyramidal coordination observed in the sulfate bound form. The observed hyperpolarized state of His47 requires a revision of the previously suggested catalytic mechanism. Molecular modeling indicates that the fundamental structural features important for catalysis are fully conserved in both classes of SMases D and that the Class II SMases D contain an additional intra-chain disulphide bridge (Cys53-Cys201). Structural analysis suggests that the highly homologous enzyme from Loxosceles bonetti is unable to hydrolyze sphingomyelin due to the 95G1y -> Asn and 134Pro -> Glu mutations that modify the local charge and hydrophobicity of the interfacial face. Structural and sequence comparisons confirm the evolutionary relationship between sphingomyelinases D and the glicerophosphodiester phosphoesterases which utilize a similar catalytic mechanism.
