Electroactive nanostructured membranes have been produced by the layer-by-layer (LbL) technique, and used to make electrochemical enzyme biosensors for glucose by modification with cobalt hexacyanoferrate redox mediator and immobilisation of glucose oxidase enzyme. Indium tin oxide (ITO) glass electrodes were modified with up to three bilayers of polyamidoamine (PAMAM) dendrimers containing gold nanoparticles and poly(vinylsulfonate) (PVS). The gold nanoparticles were covered with cobalt hexacyanoferrate that functioned as a redox mediator, allowing the modified electrode to be used to detect H(2)O(2), the product of the oxidase enzymatic reaction, at 0.0 V vs. SCE. Enzyme was then immobilised by cross-linking with glutaraldehyde. Several parameters for optimisation of the glucose biosensor were investigated, including the number of deposited bilayers, the enzyme immobilisation protocol and the concentrations of immobilised enzyme and of the protein that was crosslinked with PAMAM. The latter was used to provide glucose oxidase with a friendly environment, in order to preserve its bioactivity. The optimised biosensor, with three bilayers, has high sensitivity and operational stability, with a detection limit of 6.1 mu M and an apparent Michaelis-Menten constant of 0.20 mM. The controlled release of drugs can be efficient if a suitable encapsulation procedure is developed, which requires biocompatible materials to hold and release the drug. In this study, a natural rubber latex (NRL) membrane is used to deliver metronidazole (MET), a powerful antiprotozoal agent. MET was found to be adsorbed on the NRL membrane, with little or no incorporation into the membrane bulk, according to energy dispersive X-ray spectroscopy. X-ray diffraction and FTIR spectroscopy data indicated that MET retained its structural and spectroscopic properties upon encapsulation in the NRL membrane, with no molecular-level interaction that could alter the antibacterial activity of MET. More importantly, the release time of MET in a NRL membrane in vitro was increased from the typical 6-8 h for oral tablets or injections to ca. 100 h. The kinetics of the drug release could be fitted with a double exponential function, with two characteristic times of 3.6 and 29.9 h. This is a demonstration that the induced angiogenesis known to be provided by NRL membranes can be combined with a controlled release of drugs, whose kinetics can be tailored by modifying experimental conditions of membrane fabrication for specific applications. We describe the assembly of layer-by-layer films based on the poly(propylene imine) dendrimer (PPID) generation 3 and nickel tetrasulfonated phthalocyanine (NiTsPc) for application as chemically sensitive membranes in sepal alive extended-gate field effect transistor (SEGFET) pH sensors PPID/NiTsPc films wet e adsorbed on quartz, glass. indium tin oxide. or gold (Au)-covered glass substrates Multilayer formation was monitored via UV-vis absorption upon following the increment in the Q-band intensity (615 nm) of NiTsPc The nanostructured membranes were very stable in a pH range of 4-10 and displayed a good sensitivity toward H(+), ca 30 mV/pH for PPID/N(1)TsPc films deposited on Au-covered substrates For films deposited on ITO, the sensitivity was ca 52 4 mV/pH. close to the expected theoretical value for ton-sensitive membranes. The use of chemically stable PPID/NiTsPc films as gate membranes in SEGFETs, as introduced here, may represent an alternative for the fabrication of nanostructured, porous platforms for enzyme immobilization to be used in enzymatic biosensors.

Diacylglycerol acyltransferase 1 (DGAT1) catalyzes the final and dedicated step in the synthesis of triacylglycerol, which is believed to involve the lipids oleoyl coenzyme A (OCoA) and dioleoyl-sn-glycerol (DOG) as substrates. In this work we investigated the interaction of a specific peptide, referred to as SIT2, on the C-terminal of DGAT1 (HKWCIRHFYKP) with model membranes made with OCoA and DOG in Langmuir monolayers and liposomes. According to the circular dichroism and fluorescence data, conformational changes on SIT2 were seen only on liposomes containing OCoA and DOG. In Langmuir monolayers, SIT2 causes the isotherms of neat OCoA and DOG monolayers to be expanded, but has negligible effect on mixed monolayers of OCoA and DOG. This synergistic interaction between SIT2 and DOG + OCoA may be rationalized in terms of a molecular model in which SIT2 may serve as a linkage between the two lipids. Our results therefore provide molecular-level evidence for the interaction between this domain and the substrates OCoA and DOG for the synthesis of triacylglycerol. (C) 2009 Elsevier B.V. This paper describes the first phase of a project attempting to construct an efficient general-purpose nonlinear optimizer using an augmented Lagrangian outer loop with a relative error criterion, and an inner loop employing a state-of-the art conjugate gradient solver. The outer loop can also employ double regularized proximal kernels, a fairly recent theoretical development that leads to fully smooth subproblems. We first enhance the existing theory to show that our approach is globally convergent in both the primal and dual spaces when applied to convex problems. We then present an extensive computational evaluation using the CUTE test set, showing that some aspects of our approach are promising, but some are not. These conclusions in turn lead to additional computational experiments suggesting where to next focus our theoretical and computational efforts.

We describe a simple and efficient strategy to fabricate enzymatic devices based on the deposition of glucose oxidase on aligned and highly oriented CoNiMo metallic nanowires. CoNiMo nanowires with an average diameter of 200 nm and length of 50 mu m were electrodeposited on Au-covered alumina substrates via electrodeposition, using alumina membranes as templates. Enzyme-modified electrodes were fabricated via enzyme immobilization using a cross-linker. To minimize nonspecific reactions in the presence of interfering agents, a permselective membrane composed of poly(vinylsulfonic acid) and polyamidoamine dendrimer was deposited via electrostatic interaction. The formation of hydrogen peroxide as a product of the enzymatic reaction was monitored at low overpotential, 0.0 V (vs Ag/AgCl). The detection limit was estimated at 22 mu M under an applied potential of 0.0 V. The apparent Michaelis-Menten constant determined from the Lineweaver-Burke plot was 2 mM.

Polycarbonate membranes (PCM) of various pores sizes (400, 200, 100 and 50 nm) were used as templates for gold deposition. The electrodeposition from gold ions resulted in the formation of gold nanotubes when large pores size PCMs (400 and 200 nm) were used. On the other hand, gold nanowires were predominant for the PCMs with smaller pores size (100 and 50 nm). Surface-enhanced Raman scattering (SERS) from the probe molecule 4-mercaptopyridine (4-MPy) was obtained from all these nanostructures. The SERS efficiency of the substrates produced using the PC M templates were compared to two commonly used SERS platforms: a roughened gold electrode and gold nanostructures electrodeposited through organized polystyrene spheres (PSS). The SERS signal of the probe molecule increased as the pore diameter of the PCM template decreased. Moreover, the SERS efficiency from the nanostructures produced using 50 nm PCM templates was four and two times better than the signal from the roughened gold electrode and the PSS template, respectively. The SERS substrates prepared using PCM templates were more homogenous over a larger area (ca. 1 cm(2)), presented better spatial and sample to sample reproducibility than the other substrates. These results show that SERS substrates prepared using PCM templates are promising for the fabrication of planar SERS platforms for analytical/bioanalytical applications.

Oroidin was isolated from the marine sponge Agelas sventres and inhibited the activity and function of Pdr5p, an enzyme responsible for the multidrug resistance phenotype in Saccharomyces cerevisiae. This compound may help in the development of new drugs that reverse this dangerous phenotype of pathogenic yeast and fungi.

Este trabalho se propõe a fazer um recorte das interações professor-aluno em ambiente de aprendizagem virtual, qual seja, a ação mediadora na apropriação das ferramentas informáticas junto a alunos com Necessidades Educacionais Especiais. Foram levantados 5 tipos de intervenção efetivadas pelo mediador-professor, através da escolha da metodologia qualitativa do estudo de multicasos, enfocando desta forma, 3 casos de ação pedagógica direcionadas a 3 alunos. Neste sentido, o foco do estudo em si passou a constituir-se nos procedimentos de atuação do mediador no transcurso do processo de apropriação no ambiente de aprendizagem virtual do Núcleo de Informática na Educação Especial (NIEE), da Universidade Federal do Rio Grande do Sul. Os tipos de intervenção identificados foram utilizados pelos mediadores coerentemente aos pressupostos da teoria Sócio-histórica e, em particular, o construto da Zona de Desenvolvimento Proximal (ZDP), os quais permearam sempre a atuação mediacional instaurada como suporte ao aluno, no sentido de mobilizar suas estruturas internas buscando o deslocamento na referida ZDP, materializando desenvolvimento real no sujeito. Os resultados apontaram para a importância da competência pedagógica do professor quanto à qualidade e adequação de seus suportes, dentre os 5 identificados, que devem acompanhar o delineamento e deslocamento do aluno na sua ZDP, suscitando processos internos de aprendizagem e desenvolvimento.

Esta dissertação é apresentada sob a forma de dois artigos. O primeiro foi realizado com o objetivo de comparar medidas verticais e horizontais de imagens adquiridas em estereomicroscópio (E) e scanner de mesa (S). Para isso, 50 dentes posteriores extraídos de humanos e seccionados no sentido mésio-distal foram utilizados. Cada hemi-secção foi fotografada no estereomicroscópio e digitalizada em scanner de mesa, com resolução de 230 dpi. No programa Photoshop foram realizadas medições horizontais (H) e verticais (V) de cada hemi-secção, nas imagens obtidas pelos dois métodos, com repetição aleatória de 20% dos casos da amostra. As medidas iniciais e repetidas apresentaram excelente correlação e o teste t para amostras pareadas não evidenciou diferenças significativas (p = 0,05) entre elas nos dois métodos. As médias das medidas e seus desvios-padrão foram: horizontais E 8,89 mm ? 4,30 mm, S 8,89 mm ? 4,52 mm e verticais E 4,42 mm ? 2,19 mm, S 4,43 mm ? 2,19 mm. O teste t para amostras pareadas com nível de significância de 5% demonstrou não haver diferenças significativas entre os dois métodos, tanto para as medidas horizontais (p=0,685) quanto para as verticais (p=0,299). Conclui-se que o scanner de mesa pode substituir o estereomicroscópio na aquisição de imagens de hemi-secções de dentes, com a finalidade de obtenção de medidas horizontais e verticais. O segundo artigo avaliou, in vitro, a profundidade de lesões de cárie proximal em dentes posteriores, comparando os exames clínico e radiográfico interproximal, com e sem a aplicação de cores, ao exame histológico. No total, 142 superfícies proximais foram avaliadas clinicamente e radiografadas. Para obtenção do padrão-ouro do estudo o exame histológico com cortes seriado foi realizado e as secções adquiridas, assim como as radiografias, digitalizadas com scanner de mesa A partir da criação de um programa digital, foi possível a aplicação de cores nas imagens radiográficas de maneira padronizada, de acordo com a intensidade de pixels de cada superfície. A profundidade das lesões foi aferida por meio de escores e de medidas nas imagens histológicas, radiográficas com e sem o uso de cor em cada superfície da amostra. O programa não foi capaz de identificar superfícies hígidas e lesões em diferentes profundidades no esmalte. Em todas as lesões que apresentavam alteração radiográfica em dentina, os escores atribuídos às imagens radiográficas com aplicação de cores foram 100% concordantes com os escores histológicos. Em relação às lesões que apresentavam histologicamente alteração na metade externa dentina, 55,56% tiveram sua profundidade subestimada, contudo nenhuma era cavidade em dentina. Todas as lesões que apresentavam profundidade histológica na metade interna de dentina apresentavam cavidade em dentina e mostraram a mesma extensão com a aplicação de cor na radiografia, superando os resultados obtidos com a radiografia sem cor. Em relação às medidas de profundidade, quando um desvio-padrão foi subtraído da média de densidade óptica para a aplicação de cores, as medidas resultantes foram ligeiramente menores e não diferiram significativamente daquela do exame histológico de acordo com a Análise de Variância, utilizando o delineamento de blocos casualizados, complementado pelo teste de Comparações Múltiplas de Tukey. O uso de um programa digital que determina de maneira padronizada como cada imagem deve ser colorida auxilia no diagnóstico de lesões de cárie que apresentam extensão histológica em dentina, especialmente aquelas que atinjam a sua porção interna.

Excessive mechanical stress due to caudal sloping of the tibial plateau may result In early breakdown of the cranial cruciate ligament (CrCL). Five dogs with CrCL rupture associated with caudal sloping of the proximal tibial plateau are described. All were small dogs, of between three and six years of age, with a mean bodyweight of 9.3 kg, which had acute hindlimb lameness. Radiographic examination revealed cranial displacement of the tibia, with a tibial angle varying from 58 to 60 degrees. All cases were treated with a lateral fabellotibial suture and cranial cuneiform osteotomy of the proximal tibia. All dogs were using the operated limb three days after surgery, with normal gait re-established after a mean period of 10 days. Excessive tibial plateau sloping is not a frequent cause of hindlimb lameness In small animals, although it Is Important to consider it as a predisposing factor for rupture of the CrCL.

Thermal stability, thermal decomposition process, residual mass, temperature of glass transition (T-g) and temperature dependence of storage modulus (E'), were determined for latex membranes prepared from six clones of Hevea brasiliensis: IAC 331, IAC 332, IAC 333 and IAC 334 grown at experimental plantations of Instituto Agronomico de Campinas (IAC) in Votuporanga, São Paulo State, Brazil. Latex membranes from GT1 and RRIM 600 Asian matrix clones were used as references. The thermal behavior of latex membranes from genetically improved rubber trees was characterized using thermogravimetry/derivative thermogravimetry (TG/DTG), differential scanning calorimetry (DSC) and dynamic mechanical analysis (DMA). The thermal behavior of latex from clones studied in the present work showed similar features of the clones previously reported (IAC 40, IAC 300, IAC 301, IAC 328, IAC 329 and IAC 330), with mass loss in four consecutive steps, except IAC 333, which showed an additional mass loss step.
