Syfte - Syftet med studien är att få en djupare insikt i sambandet mellan ledarskap och de faktorer som bidrar till ökat engagemang hos medarbetarna i arbetet med ständiga förbättringar. Studien riktar sig mot ett mellanstort industriföretag som implementerat lean production. Design/ metod - I vår undersökning valde vi att göra en kvalitativ fallstudie. Data samlades in i ett svenskt industriföretag. Totalt har 7 intervjuer genomförts. Undersökningsresultat - Resultatet vi kom fram till är att medarbetarna är i låg utsträckning delaktiga i förbättringsarbetet. Tidigare forskning visar att medarbetare som är delaktiga i beslut är också mer engagerade i arbetet. Metodkritik – Resultatet speglar enbart subjektiva bedömningar och de studerade variablerna bygger på respondenternas uppfattningar. Framtida forskning bör inkludera mer objektiva mått. Praktiska konsekvenser - Studien ger belägg till att medarbetarnas engagemang och motivation i förbättringsarbetet kan ökas genom att göra dem mer delaktiga. Forskning visar att medarbetare som får information och får delta i beslut känner större delaktighet i arbetet. Kunskapsbidrag - I den befintliga litteraturen hävdas det att ledarskapet är en viktig komponent för att öka medarbetarnas engagemang. Lite har dock skrivits om hur ledarskapet kan motivera medarbetarna till ständiga förbättringar i en organisation som arbetar i lean production. Nyckelord:

Denna rapport ska ses som en förstudie kring metoder för insamling av information över besökarna vid Svenska Skidspelen samt för att uppskatta Skidspelens ekonomiska betydelse. Resultaten baseras på insamlat material via enkäter till besökare till Skidspelen 2012. Totalt 404 svar i enkätform från besökare inom området samt från bil och bussåkande besökare har använts i analysen. Därutöver intervjuades 718 besökare vid entrén om ålder, bostadsort, vistelselängd samt färdmedel till evenemanget.   Rapporten innehåller uppskattningar av Skidspelens ekonomiska betydelse med hjälp av flera typer av modeller; turism-, boende- samt ålderssegmenteringsmodell. Resultat från studien visar att engångs­kostnader för boende, resor och biljett kunde uppskattas relativt bra, medan konsumtion under dagen inte fångades tillräckligt bra i någon modell. Resultaten från studien visar också att en försiktig uppskattning av Skidspelens ekonomiska betydelse mätt som direkt konsumtion kan sägas ligga mellan 14 - 18 miljoner kr för år 2012.   För att få ett mer korrekt resultat vid eventuella vidare studier eller vid studier av Skid-VM 2015 i Falun så bör en samhällsekonomisk välfärds analys göras för att inkludera samtliga effekter. Beräkningen av Skidspelens ekonomiska effekt skulle kunna göras mer precis genom att tillfråga ett större urval av besökare samt utveckla insamlingen av data över besökarnas totala konsumtion kopplat till besöket vid Svenska Skidspelen.

In present study interviews with five officials and the gathering of documentation carried out in a reception center for newly arrived refugees located in a medium-sized municipality in central Sweden. The aim has been on the basis of the collected empirical data to evaluate how the municipality worked to promote the integration of newcomers, what measures were available, whether there were existing goals, and how they were formulated, and the extent to which they were implemented. This constituted, together with earlier research and systems theory based framework for the study's conclusions. The study showed that communication between and within the investigated unit was partly flawed. It has also showed that there was a degree of conflict in organizational approaches within the examined entity which could cause bottlenecks in the activity. The current study also showed the difficulty in estimating the resources currently needed to cover the need of, among others, Swedish for immigrants.

A acidemia glutárica tipo I (AG I) é um erro inato do metabolismo de herança autossômica recessiva que se caracteriza bioquimicamente pela deficiência da atividade da enzima glutaril-CoA desidrogenase da rota de degradação dos aminoácidos lisina, hidroxilisina e triptofânio. O bloqueio da rota na conversão de glutaril-CoA à crotonil-CoA e resultante da deficiência enzimática leva ao acúmulo tecidual e aumento da concentração nos líquidos biológicos predominantemente dos ácidos glutárico e 3-hidroxiglutárico e, em alguns casos, do ácido glutacônico. Os pacientes afetados apresentam sintomas neurológicos especialmente após crises encefalopáticas, comuns no primeiro ano de vida, em que ocorre necrose bilateral dos gânglios da base, degeneração do globo pálido e da substância branca. Tendo em vista que a patogênese do dano cerebral na AG I é pouco conhecida e que as lesões do SNC dos pacientes muito se assemelham às lesões devido à excitotoxicidade, no presente estudo investigamos a ação dos ácidos glutárico e 3-hidroxiglutárico sobre alguns parâmetros neuroquímicos envolvendo o sistema glutamatérgico em córtex cerebral e astrócitos cultivados de córtex cerebral de ratos. Alguns desses parâmetros já haviam sido testados previamente com o ácido glutárico. Observamos que o ácido 3-hidroxiglutárico, em concentrações de 10 µM a 1 mM, não alterou a captação de L-[3H]glutamato por preparações sinaptossomais. Verificamos também que os ácidos glutárico e 3- hidroxiglutárico, nas mesmas concentrações, não alteraram a liberação do L- [3H]glutamato por estas estruturas em condições basais ou estimuladas (despolarizadas) por potássio (20 mM e 40 mM). Já nos ensaios de união do L- [3H] glutamato a membranas plasmáticas, o ácido 3-hidroxiglutárico inibiu a união do neurotransmissor em meio de incubação sem sódio e cloreto (sítios receptores), quando em baixas concentrações (1 µM a 100 µM), enquanto não alterou esta união em altas concentrações (500 µM a 2 mM). Na presença de alto sódio (sítios transportadores), ambos os ácidos inibiram a união de L- [3H]glutamato em todas as concentrações testadas (10 µM a 1 mM). Outros experimentos demonstraram que o ácido 3-hidroxiglutárico não alterou a captação vesicular de L-[3H]glutamato. Avaliamos também a influência dos ácidos glutárico e 3-hidroxiglutárico sobre a captação de L-[3H]glutamato por astrócitos cultivados de córtex cerebral. Inicialmente verificamos a viabilidade destas células na presença dos ácidos pelo teste do MTT que reflete a atividade das desidrogenases mitocondriais e também pela liberação de lactato desidrogenase para o meio de incubação. Na concentração de 1 mM , os ácidos glutárico e 3- hidroxiglutárico não se mostraram citotóxicos para estas células. Por outro lado, quando incubados por 60 minutos, nas concentrações de 50 µM e 1mM, aumentaram significativamente a captação de L-[3H]glutamato pelos astrócitos.

As doenças cerebrovasculares, popularmente conhecidas como derrames, são uma das principais causas de morbidade e mortalidade entre adultos e idosos. Contudo, muitos pacientes que sofrem derrame sobrevivem e experimentam as conseqüências do insulto por muitos anos, muitas vezes a nível emocional, motor ou intelectual. Dentre estas lesões, destaca-se a isquemia cerebral. Existem modelos experimentais de isquemia cerebral in vivo e in vitro. Os modelos in vitro são realizados em culturas ou fatias de tecido cerebral submetidas à Privação de Oxigênio e Glicose (POG), que mimetizam condições traumáticas similares, mas não idênticas às produzidas in vivo. A investigação da atividade de substâncias potencialmente neuroprotetoras a partir da comparação da morte celular entre culturas ou fatias de tecido cerebral controle e tratadas é facilitada neste tipo de modelo experimental. Após a injúria, as culturas são expostas a métodos de avaliação da viabilidade ou dano celular como, por exemplo, o corante fluorescente iodeto de propídeo, que marca seletivamente células mortas ou em curso de morte, ou MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide) que mede a viabilidade mitocondrial possibilitando uma posterior quantificação. As plantas são uma fonte importante de produtos naturais biologicamente ativos, muitos dos quais se constituem em modelos para a síntese de um grande número de fármacos. Um exemplo é a planta kava-kava ou somente kava (Piper Methysticum) a qual chamou a atenção dos pesquisadores devido à sua utilização nas ilhas do Pacífico sul. Foi demonstrada a possibilidade da kava possuir uma variedade de atividades farmacológicas importantes, entre elas a atividade de neuroproteção Esse trabalho tem como objetivo avaliar a atividade neuroprotetora do extrato de kava-kava (Piper methysticum) em modelos in vitro de morte neuronal em fatias de hipocampo de ratos, investigar o envolvimento da proteína de choque térmico HSP27 (Heat Shock Protein) no processo de morte e neuroproteção induzida pela kava, bem como investigar o efeito da lesão induzida por POG sobre o imunoconteúdo da proteína Oxido Nítrico Sintase induzível (iNOS). Os resultados dos experimentos por nós realizados nas culturas organotípicas submetidas à POG por 40 minutos e tratadas com extrato de kava (7µg/ml) demonstraram uma significativa redução, na ordem de 58% na intensidade da morte neuronal na região CA1 do hipocampo em resposta à injúria. Em fatias hipocampais submetidas à POG por 60 minutos a adição do extrato de kava 7µg/ml aumentou em 15% a viabilidade celular, confirmando a atividade neuroprotetora sugerida para esta planta. As culturas expostas à POG e tratadas com kava apresentaram um aumento significativo no imunoconteúdo da proteína HSP27, esse aumento é acompanhado de um aumento da fosforilação, uma vez que a percentagem de proteína fosforilada se mantém igual. As fatias lesionadas tratadas com kava apresentaram uma diminuição significativa no imunoconteúdo da iNOS na ordem de 35 % em relação as fatias lesionadas tratadas com DMSO (Dimetilsulfoxido). Estes dados podem sugerir que um dos mecanismos da neuroproteção observada para o extrato de kava, possa envolver essas proteínas.

Fungal polysaccharides have received a great deal of attention due to itsbecause of their potential use in a wide rangegreat variety fromof industries. Some studies have demonstrated that polysaccharides extracted offrom basidiomycetes they have presented significant properties as anti-inflammatory, antimicrobial, antioxidant and anti-tumoral properties. In spite of thisDespite these potential properties, these mushrooms have not been insufficiently investigated, and the great number of antibiotics number produced forby these organisms suggests that they canmay be a new source of bioactives composites source. In tThe present work, reports onlated the chemical composition, potential antioxidant, antiinflammatory and citotoxycity of extracted polymers extracted offrom the fruits bodies of the fungiius Geastrum saccatum and Polyporus dermoporus, native mushrooms of the Atlantic forest inof the state of the Rio Grande do Norte, Brazil. The Cchemical analyses had revealed ademonstrated text of total sugar rates of 65% and 49%, and proteins of 7.0% for in extracts of G. saccatum and P. dermoporus extracts, respectively. The analyses ofNMR spectroscopy of RMN had demonstrated that these extracts are composites forof a complex involving β- glucans and- proteins complex. The inhibition of the formation of superoxide radicals formation was of 88.4% in G. saccatum and 83.3% in P. dermoporus, and 75 and 100% for inhibition of hydroxyls radicals inhibition. The cytotoxic action of these polymers was analyzed throughthrough the mitochondrial function (MTT). The incubation of the glucans with mononuclear cells of the peripheral blood demonstrated that the extracted glucans extracted fromof G. saccatum havepossess a moderate cytotoxic action. These results suggest that these mushrooms possess polymers formed byfor a complex glucana-protein complex, with antiinflammatory and antioxidant actions

The present study examines the chemical composition and their effects on free radicals, inflammation, angiogenesis, coagulation, VEGF effects and cellular proliferation of a polysaccharides from alga Sargassum vulgare. The sulfated polysaccharide was extracted from brown seaweed by proteolysis with enzymes maxataze. The presence of proteins and sugars were observed in crude polysaccharides. Fractionation of this crude extract was made with growing concentration of acetone (0.3-1.5 v) and produced four groups of polysaccharides. Anionic polysaccharides from brown seaweed Sargassum vulgare, SV1and PSV1 were fractionated (SV1) and purified (PSV1), and displayed with high total sugars and sulfate content and very low level of protein. This fucan SV1 contains low levels of protein and high carbohydrate and sulfate content. This polysaccharides prolonged activated partial thromboplastin time (aPTT) at 50 μg (>240 s). SV1 was found to have no effect on prothrombin time (PT), corresponding to the extrinsic pathway of coagulation. SV1 exhibits high antithrombotic action in vivo, with a concentration ten times higher than heparin. Polysaccharides from S. vulgare promoted direct inhibition enzymatic activity of thrombin and stimulated enzymatic activity of FXa. SV1 showed optimal inhibitory activity of thrombin (50.2±0.28%) at a concentration of 25 μg/mL. Its antioxidant action on scavenging radicals by DPPH was (22%), indicating the polymer has no cytotoxic action (hemolytic) on ABO and Rh blood types in different erythrocyte groups and displays strong anti-inflammatory action on all concentrations tested in the carrageenan-induced paw edema model, demonstrated by reduced edema and cellular infiltration. Angiogenesis is a dynamic process of proliferation and differentiation. It requires endothelial proliferation, migration, and tube formation. In this context, endothelial cells are a preferred target for several studies and therapies. The antiangiogenic efficacy of polysaccharides was examined in vivo in the chick chorioallantoic membrane (CAM) model by using fertilized eggs. Decreases in the density of the capillaries were assessed and scored. The results showed that SV1 and PSV1 have an inhibitory effect on angiogenesis. These results were also confirmed by inhibition tubulogenesis in rabbit aorta endothelial cell (RAEC) in matrigel. These compounds were assessed in Apoptosis assay (Annexin V - FITC / PI) and cell viability by MTT assay of RAEC. These polysaccharides do not affect the viability and do not have apoptotic or necrotic action. RAEC cell when incubated with SV1 and PSV1showed inhibition of VEGF secretion, observed when compounds were incubated at 25, 50 and 100 μg/μL. The VEGF secretion with the RAEC cell line for 24 h, was more effective for PSV1 at 50 μg/μL(71.4%) than SV1 100 μg/μL (75.9%). SV1 and PSV1 had an antiproliferative action (47%) against tumor cell line HeLa. Our results indicate that these sulfated polysaccharides have antiangiogenic and antitumoral actions

Fucan is a term used to denominate L-fucose rich sulfated polysaccharides. The fucans have been studied due their pharmacological activities like antithrombotic, antiproliferative and antioxidant. We have extracted three fucan fractions from the brown seaweed Spatoglossum schröederi. These fucans were denominated Fuc B 1, Fuc B 1.5 and Fuc B 2. The chemical analyzes show that the fucans have very similar composition as demonstrated by agarose electrophoresis gel, sugar and sulfate content. The antiproliferative effect was determined by MTT and BrdU methodologies in CHO cells. The inhibition of proliferation effect of the three fractions was about 40%. Therefore this we proceed just with the Fuc B 2 due the higher yield. There is no apoptosis indication using the anexin V/propidium iodide test. We found a cell cycle phase G1 arrest. The western blotting show that the PKC; pFAK; pERK 1/2 are activated when the cells were treated with fucans. The treatement with inhibitor of MAPK PD98059 extinguished the fucan effect. These results indicates that fucan act by the ERK pathway inducing the cell death.

The Brazil is the third largest producer of cashew nuts in the world. Despite the social and economic importance of the cashew nut, its production is still carried out artisanally. One of the main problems encountered in the cashew production chain are the conditions under which the roasting of the nut occurs to obtain the kernel from the shell. In the present study was conducted a biomonitoring of the genotoxic and cytotoxicity effects associated with the elements from the cashew nut roasting in João Câmara - RN, semi-arid region of Brazil. To assess the genotoxic was used the bioassay of micronucleus (MN) in Tradescantia pallida. In addition, it was performed a comparative between the Tradescantia pallida and KU-20 and other biomarkers of DNA damage, such as the nucleoplasmic bridges (NBP) and nuclear fragments (NF) were quantified. The levels of particulate matter (PM1.0, PM2.5, PM10) and black carbon (BC) were also measured and the inorganic chemical composition of the PM2.5 collected was determined using X-ray fluorescence spectrometry analysis and the assessment of the cytotoxicity by MTT assay and exclusion method by trypan blue. . For this purpose, were chosen: the Amarelão community where the roasting occurs and the Santa Luzia farm an area without influence of this process. The mean value of PM2.5 (Jan 2124.2 μg/m3; May 1022.2 μg/m3; Sep 1291.9 μg/m3) and BC (Jan 363.6 μg/m3; May 70.0 μg/m3; Sep 69.4 μg/m3) as well as the concentration of the elements Al, Si, P, S, Cl, K, Ca, Ti, Cr, Mn, Fe, Ni, Cu, Zn, Se, Br and Pb obtained at Amarelão was significantly higher than at Santa Luzia farm. The genotoxicity tests with T. pallida indicated a significant increase in the number of MN, NBP and NF and it was found a negative correlation between the frequency of these biomarkers and the rainfall. The concentrations of 200 μg/mL and 400 μg/mL of PM2.5 were cytotoxic to MRC-5 cells. All together, the results indicated genotoxicity and citotoxicity for the community of Amarelão, and the high rates of PM2.5 considered a potential contributor to this effect, mainly by the high presence of transition metals, especially Fe, Ni, Cu, Cr and Zn, these elements have the potential to cause DNA damage. Other nuclear alterations, such as the NPBs and NFs may be used as effective biomarkers of DNA damage in tetrads of Tradescantia pallida. The results of this study enabled the identification of a serious occupational problem. Accordingly, preventative measures and better practices should be adopted to improve both the activity and the quality of life of the population. These measures are of fundamental importance for the sustainable development of this activity.

The use of medicinal plants to cure and treat various diseases is a common practice in the world and in Brazil. In several regions of the Brazil´s Northeast, the cactus Cereus jamacaru, known as mandacaru, is used popularly as a treatment to many diseases, including those related to heart respiratory diseases, gastric ulcers, scurvy, and kidney diseases. However, there is a scarcity in the scientific literature that proves scientifically the popular application of this cactus. Like other plants, Cereus jamacaru synthesizes several potentially bioactive molecules, like as polysaccharides. In this work, three polysaccharides-rich aqueous extracts, MCA80, MPM and MCP60, were obtained from this plant and analyzed chemically, as well as their cytotoxic and antioxidant potential. The data showed that all extracts consist mainly of polysaccharides (89.42 to 95.76%), but also protein (> 2%) and phenolic (3 to 8.87%) contaminants were detected. All extracts are rich in galactose, glucose and mannose. In addition, glucuronic acid was found in MCA80 and MCP60. The extracts showed total antioxidant capacity ranged from 55.21 to 68.13 of ascorbic acid equivalents (AAE). Besides, they exhibited reducer power and cupric chelation in a dose-dependent manner. None of the extracts inhibited the MTT reduction in the presence of prostate tumor cells (PC-3). However, MCP60 was the most effective extract by preventing the reduction of MTT by about 80% in the presence of cells 786. Nuclear fragmentation tests showed that this extract induces cell death. The data indicated that mandacaru synthesizes bioactive polysaccharides with potential as antioxidant and antitumor agents. For future studies, it is intended to purify and characterize these polysaccharides and its antioxidant and antitumor mechanisms

Seaweeds are a major source of biologically active compounds . In the extracellular matrix of these organisms are sulfated polysaccharides that functions as structural components preventing it against dehydration. The fraction 0.9 (FucB) rich in sulfated fucans obtained from brown seaweed Dictyota menstrualis was chemical characterized and evaluated for pharmacological activity by testing anticoagulant activity, stimulatory action on the synthesis of an antithrombotic heparan sulfate, antioxidant activity and its effects in cell proliferation. The main components were FucB carbohydrates (49.80 ± 0.10 %) and sulfate (42.30 ± 0.015 %), with phenolic compounds ( 3.86 ± 0.016 %) and low protein contamination ( 0.58 ± 0.001 % ) . FucB showed polydisperse profile and analysis of signals in the infrared at 1262, 1074 and 930 cm -1 and 840 assigned to S = O bonds sulfate esters , CO bond presence of 3,6- anhydrogalactose , β -D- galactose non- sulfated sulfate and the axial position of fucose C4 , respectively. FucB exhibited moderate anticoagulant activity , the polysaccharides prolonged time (aPTT ) 200 ug ( > 90s ) partial thromboplastin FucB no effect on prothrombin time (PT), which corresponds to the extrinsic pathway of coagulation was observed. This stimulation promoted fraction of about 3.6 times the synthesis of heparan sulfate (HS) by endothelial cells of the rabbit aorta ( RAEC ) in culture compared with cells not treated with FucB . This has also been shown to compete for the binding site with heparin. The rich fraction sulfated fucans exhibited strong antioxidant activity assays on total antioxidant (109.7 and 89.5 % compared with BHT and ascorbic acid standards ) , reducing power ( 71 % compared to ascorbic acid ) and ferric chelation ( 71 , comparing with 5 % ascorbic acid). The fraction of algae showed cytostatic activity on the RAEC cells revealed that the increase of the synthesis of heparan sulfate is not related to proliferation. FucB showed antiproliferative action on cell lines modified as Hela and Hep G2 by MTT assay . These results suggest that FucB Dictyota menstrualis have anticoagulant , antithrombotic , antioxidant potential as well as a possible antitumor action, promoting the stimulation of the synthesis of antithrombotic HS by endothelial cells and is useful in the prevention of thrombosis, also due to its inhibitory action on species reactive oxygen ( ROS ) in some in vitro systems , being involved in promoting a hypercoagulable state

Chitosan is a natural polymer, biodegradable, nontoxic, high molecular weight derived from marine animals, insects and microorganisms. Oligomers of glucosamine (GlcN) and N-acetylglucosamine (GlcNAc) have interesting biological activities, including antitumor effects, antimicrobial activity, antioxidant and others. The alternative proposed by this work was to study the viability of producing chitooligosaccharides using a crude enzymes extract produced by the fungus Metarhizium anisopliae. Hydrolysis of chitosan was carried out at different times, from 10 to 60 minutes to produce chitooligosaccharides with detection and quantification performed by High Performace Liquid Chromatography (HPLC). The evaluation of cytotoxicity of chitosan oligomers was carried out in tumor cells (HepG2 and HeLa) and non-tumor (3T3). The cells were treated for 72 hours with the oligomers and cell viability investigated using the method of MTT. The production of chitosan oligomers was higher for 10 minutes of hydrolysis, with pentamers concentration of 0.15 mg/mL, but the hexamers, the molecules showing greater interest in biological properties, were observed only with 30 minutes of hydrolysis with a concentration of 0.004 mg/mL. A study to evaluate the biological activities of COS including cytotoxicity in tumor and normal cells and various tests in vitro antioxidant activity of pure chitosan oligomers and the mixture of oligomers produced by the crude enzyme was performed. Moreover, the compound with the highest cytotoxicity among the oligomers was pure glucosamine, with IC50 values of 0.30; 0.49; 0.44 mg/mL for HepG2 cells, HeLa and 3T3, respectively. Superoxide anion scavenging was the mainly antioxidant activity showed by the COS and oligomers. This activity was also depending on the oligomer composition in the chitosan hydrolysates. The oligomers produced by hydrolysis for 20 minutes was analyzed for the ability to inhibit tumor cells showing inhibition of proliferation only in HeLa cells, did not show any effect in HepG2 cells and fibroblast cells (3T3)

The titanium and titanium alloys are widely used as biomaterial in biomedical device and so research have been developed aiming to improve and/or better to understand interaction biomaterial/biological environment. The process for manufacturing of this titanium implants usually involves a series of thermal and mechanical processes which have consequence on the final product. The heat treatments are usually used to obtain different properties for each application. In order to understand the influence of these treatments on the biological response of the surface, it was done, in this work, different heat treatments in titanium and analyzed their influence on the morphology, adhesion and proliferation of the pre-osteoblastic cells (MC3T3-E1). For such heat-treated titanium disks were characterized by optical microscopy, contact angle, surface energy, roughness, microhardness, X-ray diffraction and scanning through the techniques (BSE, EDS and EBSD). For the analysis of biological response were tested by MTT proliferation, adhesion by crystal violet and β1 integrin expression by flow cytometry. It was found that the presence of a microstructure very orderly, defined by a chemical attack, cells tend to stretch in the same direction of orientation of the material microstructure. When this order does not happen, the most important factor influencing cell proliferation is the residual stress, indicated by the hardness of the material. This way the disks with the highest level state of residual stress also showed increased cell proliferation
