Complex organization of CTF/NF-I, C/EBP, and HNF3 binding sites within the promoter of the liver-specific vitellogenin gene.

Vitellogenin genes are expressed specifically in the liver of female oviparous vertebrates under the strict control of estrogen. To explain this tissue-specific expression, we performed a detailed analysis of the Xenopus laevis vitellogenin gene B1 promoter by DNase I footprinting and gel mobility-shift assays. We characterized five binding sites for the ubiquitous factor CTF/NF-I. Two of these sites are close to the TATA-box, whereas the others are located on both sides of the estrogen responsive unit formed by two imperfect estrogen response elements. Moreover two liver-enriched factors, C/EBP and HNF3, were found to interact with multiple closely spaced proximal promoter elements in the first 100 base pairs upstream of the TATA-box. To confirm the physiological significance of this in vitro analysis, in vivo DNase I footprinting experiments were carried out using the ligation-mediated polymerase chain reaction technique. The various cis-elements characterized in vitro as binding sites for known transcription factors and more particularly for liver-enriched transcription factors are efficiently recognized in vivo as well, suggesting that they play an important role in the control of the liver-specific vitellogenin gene B1 expression.

Modified SH2 domain to phototrap and identify phosphotyrosine proteins from subcellular sites within cells.

Spatial regulation of tyrosine phosphorylation is important for many aspects of cell biology. However, phosphotyrosine accounts for less than 1% of all phosphorylated substrates, and it is typically a very transient event in vivo. These factors complicate the identification of key tyrosine kinase substrates, especially in the context of their extraordinary spatial organization. Here, we describe an approach to identify tyrosine kinase substrates based on their subcellular distribution from within cells. This method uses an unnatural amino acid-modified Src homology 2 (SH2) domain that is expressed within cells and can covalently trap phosphotyrosine proteins on exposure to light. This SH2 domain-based photoprobe was targeted to cellular structures, such as the actin cytoskeleton, mitochondria, and cellular membranes, to capture tyrosine kinase substrates unique to each cellular region. We demonstrate that RhoA, one of the proteins associated with actin, can be phosphorylated on two tyrosine residues within the switch regions, suggesting that phosphorylation of these residues might modulate RhoA signaling to the actin cytoskeleton. We conclude that expression of SH2 domains within cellular compartments that are capable of covalent phototrapping can reveal the spatial organization of tyrosine kinase substrates that are likely to be important for the regulation of subcellular structures.

En aquest projecte d’electrificació del polígon industrial de Palafrugell s'ha realitzat un disseny complet de tots els aspectes determinants des de l’arribada de la línia d’alta tensió, les ramificacions de baixa tensió, càlcul d’enllumenat, postes a terra, centres de transformació amb els seus respectius transformadors, i per últim un estudi i orientació sobre tarificacions a indústries. De cada apartat s'ha realitzat tota la sèrie de càlculs necessaris per determinar els components i aparellatge dimensionat per les necessitats

Disseny d’una nau industrial amb estructura i fonamentació de formigó prefabricat

L’objectiu d’aquest treball és realitzar un projecte on es buscaran diferents tipus de sabates prefabricada de formigó que hi ha en el mercat, per tal de seleccionar la més adequada a través d’una sèrie de càlculs, tant des del punt de vista tècnic, de facilitat de muntatge i de transport, com econòmic, i d’aquesta manera arribar a una solució. Aquest projecte és un subprojecte d’un altre que correspon a la construcció de la totalitat de la nau industrial prefabricada. Per tant, la part important d’aquest projecte és l’estudi de les sabates prefabricades de formigó, el qual, es realitzarà partint d’una informació addicional, que aparentment sembla poc important, però, en realitat, és imprescindible per dur-lo a terme

Projecte d’una xarxa de distribució elèctrica de mitja tensió, centres de transformació i instal·lacions d'enllaç d’un polígon industrial

Disseny d’una xarxa de distribució d’alta tensió de tercera categoria de 25 kV subterrània, a partir d’ara mitja tensió (MT), amb deu centres de transformació i distribució de 1000 kVA cadascun. La reforma d’un centre de transformació i distribució de 630 kVA i el disseny de la xarxa de baixa tensió. Amb la finalitat de donar subministrament d’energia elèctrica a les industries d’un polígon del terme municipal de Torroella de Montgrí (GIRONA) i obtenir l’autorització administrativa per la posterior posada en servei d’aquesta instal•lació.No queda inclòs en aquest projecte la instal•lació d’enllumenat públic

Una part de l’estructura està formada perelements estructurals prefabricats per una empresa externa mentre, qued’altres elements estructurals, com és el cas de les sabates de fonamentació,es fabriquen a l’obra

Projecte d’ampliació i acondicionament d’una nau industrial destinada a processos metal•lúrgics

El present projecte pretèn ampliar i condicionar una nau industrial existent, destinada aprocessos metal•lúrgics, propietat de l’empresa Muntatges Industrials Palafolls, SCP.L’establiment es troba situat al polígon Industrial de Can Baltasar, en el municipi de Palafolls.Per tal de dur a terme aquestes modificacions, s’enderrocaran els departaments existents i esdimensionaran en una nova ubicació dintre del recinte de l’establiment. Es dimensionarà unanova estructura per a sustentar un pont grua de 10 tn, situada a l’interior de la nau.A més a més, es dimensionaran les instal•lacions elèctriques i contra incendis de l’establiment.El projecte contindrà tota la documentació necessària per a dur a terme l’ampliació iremodelació de la nau industrial i la instal•lació del pont grua.

Global GacA-steered control of cyanide and exoprotease production in Pseudomonas fluorescens involves specific ribosome binding sites.

The conserved two-component regulatory system GacS/GacA determines the expression of extracellular products and virulence factors in a variety of Gram-negative bacteria. In the biocontrol strain CHA0 of Pseudomonas fluorescens, the response regulator GacA is essential for the synthesis of extracellular protease (AprA) and secondary metabolites including hydrogen cyanide. GacA was found to exert its control on the hydrogen cyanide biosynthetic genes (hcnABC) and on the aprA gene indirectly via a posttranscriptional mechanism. Expression of a translational hcnA'-'lacZ fusion was GacA-dependent whereas a transcriptional hcnA-lacZ fusion was not. A distinct recognition site overlapping with the ribosome binding site appears to be primordial for GacA-steered regulation. GacA-dependence could be conferred to the Escherichia coli lacZ mRNA by a 3-bp substitution in the ribosome binding site. The gene coding for the global translational repressor RsmA of P. fluorescens was cloned. RsmA overexpression mimicked partial loss of GacA function and involved the same recognition site, suggesting that RsmA is a downstream regulatory element of the GacA control cascade. Mutational inactivation of the chromosomal rsmA gene partially suppressed a gacS defect. Thus, a central, GacA-dependent switch from primary to secondary metabolism may operate at the level of translation.

The Geographical Scope of Industrial Location Determinants: An Alternative Approach

This paper considers the estimation of the geographical scope of industrial location determinants. While previous studies impose strong assumptions on the weighting scheme of the spatial neighbour matrix, we propose a exible parametrisation that allows for di fferent (distance-based) de finitions of neighbourhood and di fferent weights to the neighbours. In particular, we estimate how far can reach indirect marginal e ffects and discuss how to report them. We also show that the use of smooth transition functions provides tools for policy analysis that are not available in the traditional threshold modelling.
