Esta pesquisa interdisciplinar centrada no segmento da fotografia de acervo de intelectual apresenta o estudo de caso de 84 imagens feitas pelo fotógrafo amador Ulysses Freyre de alguns prédios e ruas das cidades de Olinda e do Recife entre 1923 e 1925. Ulysses fotografou durante passeios de bicicleta aos domingos ao lado do irmão, o sociólogo Gilberto Freyre. Objetiva-se traçar os dois usos dados por Gilberto às fotos de Ulysses: de base aos desenhos de Manoel Bandeira para o \"Livro do Nordeste\", organizado pelo sociólogo em 1925 para o centenário do Diário de Pernambuco; e como parte da concepção de inventário de edificações da arquitetura civil que serviu à Inspetoria de Monumentos Estaduais em 1928 em Pernambuco. Vale-se do campo acerca do circuito fotográfico nestas cidades, que estavam sob reformas urbanas no início do século XX, a fim de situar e revelar a fotografia de Ulysses como artefato de memória propulsor do embrionário projeto político-intelectual de Gilberto neste período. As fotos estão no acervo da Fundação Gilberto Freyre, em Recife, Pernambuco.

In the context of poetic productions of modernity and postmodernity (Hutcheon, 1999), this paper, from the concept of nomadism (Deleuze, 2012), together with the philosopher Derrida's notion of writing (2009), has the objective to study Harmada the novel (1993), written by artist Joao Gilberto Noll, the aspect of Nolliana nomadic scripture and boredom towards deconstruction of the Romanesque style. The narrative focus on introducing a transit fiction, promoted by nomadisms scripture of the wandering narrator who, in the work constitution, will the conduct of language, a plot that invades the body of the characters full of boredom and foreigners themselves, moving in fragmented and fluid spatiality of narrating. In this perspective, the research is limited with theoretical and methodological foundation in poststructuralist discussions in relation to considerations of literary aesthetics and concerning the thinkers-teóricos- critics: Derrida (2009), Deleuze (1995), Foucault (1996, 2001), Barthes (1977), Svendsen (2006). Against the background of critical understanding, the nomadic writing Harmada interlace in three stages: first, in the author's language; Second, the characters, the narrator-protagonist leading, unnamed, living overwhelming crises and painful existential ambiguity, placed through the artist's metaphor failed under the sign of "missing" while searching for other possible artistic ways of being in the world; and, finally, the nomadic reading instance as presentification effect (GUMBRECHT, 2010) for a reader experimentation. Finally, our work addresses the relationship between the nomadic scripture and the experience of boredom as a strategic power in the literature do Noll gear.

A intervenção foi direcionada à saúde dos hipertensos e diabéticos pertencentes à área de abrangência da ESF Dr. Gilberto Wiesel, pois são doenças crônicas responsáveis por gastos expressivos em saúde, além de substancial redução da capacidade de trabalho e da expectativa de vida. Desse modo, a unidade básica de saúde deve ser a porta de entrada preferencial para esses usuários no sistema de saúde, pois é considerada como ponto estratégico para melhor acolher suas necessidades, proporcionando acompanhamento longitudinal e continuado durante toda a doença; o bom manejo na atenção básica evita hospitalizações e mortes por complicações. A Unidade Dr. Gilberto Wiesel está localizada na zona central de Selbach/RS e conta com uma equipe de ESF, com aproximadamente 4500 usuários. O objetivo principal do projeto de intervenção é melhorar a adesão e qualidade da atenção aos hipertensos e diabéticos, residentes na área adstrita, fazendo parte de um projeto mais amplo “Qualificação das Ações Programáticas na Atenção Básica à Saúde”, aprovado pelo Comitê de Ética em Pesquisa da UFPeL, em março/2012.. O protocolo utilizado para amparar a intervenção será o Caderno de Atenção Básica do MS– Estratégia para Cuidado da Pessoa com Doença Crônica - Diabetes Mellitus e Estratégia para Cuidado da Pessoa com Doença Crônica - Hipertensão Arterial Sistêmica, ano 2013. Os dados obtidos através dos registros específicos e prontuários foram semanalmente revisados para análise dos indicadores. Na área adstrita da unidade há 1006 pessoas com 20 anos ou mais com Hipertensão Arterial Sistêmica e 287pessoas com 20 anos ou mais com Diabetes Mellitus, valores estes determinados por estimativa, já que não havia nenhum registro específico para esta faixa etária. Ao término da intervenção foram cadastrados 278 (37,3%) hipertensos e 85 (61,6%) diabéticos. Do total de 289 pacientes cadastradas, na faixa etária de 20 anos ou mais, 272 (97,8%) dos hipertensos e 83 (97,6%) dos diabéticos foram orientados sobre higiene bucal; 273 (98,2%) dos hipertensos e 97,6% (83) diabéticos receberam orientações sobre alimentação saudável, prática regular de atividade física e riscos do tabagismo; 165 (58,3%) dos hipertensos e 55 (64,7%) dos diabéticos tiveram priorizada a prescrição de medicações pertencentes à farmácia popular; 197 (70,9%) de hipertensos e 63 (74,1%) de diabéticos estavam com exames complementares em dia. Apesar de não ter atingido as metas traçadas, principalmente em função da grande demanda e pouco tempo hábil, com a intervenção foi possível identificar as deficiências do serviço, qualificar e uniformizar o atendimento aos usuários, criar registros específicos possibilitando controle contínuo e planejamento de novas ações.

Este trabalho de conclusão de curso para a especialização de saúde da família e comunidade inicia realizando uma introdução e reflexão sobre um ano de experiência em um município do interior do estado do Rio Grande do Sul - Selbach, na Estratégia de Saúde da Família (ESF) Dr. Gilberto Wiesel, onde atuei como médica e obtive uma ampla evolução pessoal e profissional através de um convívio diário com uma equipe multiprofissional. Este trabalho segue elencando alguns fatores principais nesta atuação próxima à comunidade, como a descrição de um caso clínico complexo de uma paciente com sobrepeso, sedentarismo e dislipidemia e seu manejo clínico embasado em evidências; a organização temporária mas efetiva do mecanismo de renovação de receitas de medicamentos controladas; e a organização da implementação de visitas domiciliares de atenção primária antes inexistentes no município. Em anexo ao final deste trabalho consta o Projeto de Intervenção programado e parcialmente implementado no município, visando sempre as melhorias na atenção primária à saúde e a evolução na implementação do acolhimento na Estratégia de Saúde da Família em que atuei.

Knowledge of the major effects governing desorption/ionization efficiency is required for the development and application of ambient mass spectrometry. Although all triacylglycerols (TAG) have the same favorable protonation and cationization sites, their desorption/ionization efficiencies can vary dramatically during easy ambient sonic-spray ionization because of structural differences in the carbon chain. To quantify this somewhat surprising and drastic effect, we have performed a systematic investigation of desorption/ionization efficiencies as a function of unsaturation and length for TAG as well as for diacylglycerols, monoacylglycerols and several phospholipids (PL). Affinities for Na(+) as a function of unsaturation level have also been assayed via comprehensive metadynamics calculations to understand the influence of this phenomenon on the ionization efficiency. The results suggest that dipole-dipole interactions within a carbon chain tuned by unsaturation sites govern ionization efficiency of TAG and PL.

A rapid, sensitive and specific method for quantifying propylthiouracil in human plasma using methylthiouracil as the internal standard (IS) is described. The analyte and the IS were extracted from plasma by liquid-liquid extraction using an organic solvent (ethyl acetate). The extracts were analyzed by high performance liquid chromatography coupled with electrospray tandem mass spectrometry (HPLC-MS/MS) in negative mode (ES-). Chromatography was performed using a Phenomenex Gemini C18 5μm analytical column (4.6mm×150mm i.d.) and a mobile phase consisting of methanol/water/acetonitrile (40/40/20, v/v/v)+0.1% of formic acid. For propylthiouracil and I.S., the optimized parameters of the declustering potential, collision energy and collision exit potential were -60 (V), -26 (eV) and -5 (V), respectively. The method had a chromatographic run time of 2.5min and a linear calibration curve over the range 20-5000ng/mL. The limit of quantification was 20ng/mL. The stability tests indicated no significant degradation. This HPLC-MS/MS procedure was used to assess the bioequivalence of two propylthiouracil 100mg tablet formulations in healthy volunteers of both sexes in fasted and fed state. The geometric mean and 90% confidence interval CI of Test/Reference percent ratios were, without and with food, respectively: 109.28% (103.63-115.25%) and 115.60% (109.03-122.58%) for Cmax, 103.31% (100.74-105.96%) and 103.40% (101.03-105.84) for AUClast. This method offers advantages over those previously reported, in terms of both a simple liquid-liquid extraction without clean-up procedures, as well as a faster run time (2.5min). The LOQ of 20ng/mL is well suited for pharmacokinetic studies. The assay performance results indicate that the method is precise and accurate enough for the routine determination of the propylthiouracil in human plasma. The test formulation with and without food was bioequivalent to reference formulation. Food administration increased the Tmax and decreased the bioavailability (Cmax and AUC).

Teeth are often included in the radiation field during head and neck radiotherapy, and recent clinical evidence suggests that dental pulp is negatively affected by the direct effects of radiation, leading to impaired sensitivity of the dental pulp. Therefore, this study aimed to investigate the direct effects of radiation on the microvasculature, innervation, and extracellular matrix of the dental pulp of patients who have undergone head and neck radiotherapy. Twenty-three samples of dental pulp from patients who finished head and neck radiotherapy were analyzed. Samples were histologically processed and stained with hematoxylin-eosin for morphologic evaluation of the microvasculature, innervation, and extracellular matrix. Subsequently, immunohistochemical analysis of proteins related to vascularization (CD34 and smooth muscle actin), innervation (S-100, NCAM/CD56, and neurofilament), and extracellular matrix (vimentin) of the dental pulp was performed. The morphologic study identified preservation of the microvasculature, nerve bundles, and components of the extracellular matrix in all studied samples. The immunohistochemical analysis confirmed the morphologic findings and showed a normal pattern of expression for the studied proteins in all samples. Direct effects of radiotherapy are not able to generate morphologic changes in the microvasculature, innervation, and extracellular matrix components of the dental pulp in head and neck cancer patients.

To characterize the relaxation induced by BAY 41-2272 in human ureteral segments. Ureter specimens (n = 17) from multiple organ human deceased donors (mean age 40 ± 3.2 years, male/female ratio 2:1) were used to characterize the relaxing response of BAY 41-2272. Immunohistochemical analysis for endothelial and neuronal nitric oxide synthase, guanylate cyclase stimulator (sGC) and type 5 phosphodiesterase was also performed. The potency values were determined as the negative log of the molar to produce 50% of the maximal relaxation in potassium chloride-precontracted specimens. The unpaired Student t test was used for the comparisons. Immunohistochemistry revealed the presence of endothelial nitric oxide synthase in vessel endothelia and neuronal nitric oxide synthase in urothelium and nerve structures. sGC was expressed in the smooth muscle and urothelium layer, and type 5 phosphodiesterase was present in the smooth muscle only. BAY 41-2272 (0.001-100 μM) relaxed the isolated ureter in a concentration dependent manner, with a potency and maximal relaxation value of 5.82 ± 0.14 and 84% ± 5%, respectively. The addition of nitric oxide synthase and sGC inhibitors reduced the maximal relaxation values by 21% and 45%, respectively. However, the presence of sildenafil (100 nM) significantly potentiated (6.47 ± 0.10, P <.05) this response. Neither glibenclamide or tetraethylammonium nor ureteral urothelium removal influenced the relaxation response by BAY 41-2272. BAY 41-2272 relaxes the human isolated ureter in a concentration-dependent manner, mainly by activating the sGC enzyme in smooth muscle cells rather than in the urothelium, although a cyclic guanosine monophosphate-independent mechanism might have a role. The potassium channels do not seem to be involved.

This study proposes to investigate quercetin antitumor efficacy in vitro and in vivo, using the P39 cell line as a model. The experimental design comprised leukemic cells or xenografts of P39 cells, treated in vitro or in vivo, respectively, with quercetin; apoptosis, cell-cycle and autophagy activation were then evaluated. Quercetin caused pronounced apoptosis in P39 leukemia cells, followed by Bcl-2, Bcl-xL, Mcl-1 downregulation, Bax upregulation, and mitochondrial translocation, triggering cytochrome c release and caspases activation. Quercetin also induced the expression of FasL protein. Furthermore, our results demonstrated an antioxidant activity of quercetin. Quercetin treatment resulted in an increased cell arrest in G1 phase of the cell cycle, with pronounced decrease in CDK2, CDK6, cyclin D, cyclin E, and cyclin A proteins, decreased Rb phosphorylation and increased p21 and p27 expression. Quercetin induced autophagosome formation in the P39 cell line. Autophagy inhibition induced by quercetin with chloroquine triggered apoptosis but did not alter quercetin modulation in the G1 phase. P39 cell treatment with a combination of quercetin and selective inhibitors of ERK1/2 and/or JNK (PD184352 or SP600125, respectively), significantly decreased cells in G1 phase, this treatment, however, did not change the apoptotic cell number. Furthermore, in vivo administration of quercetin significantly reduced tumor volume in P39 xenografts and confirmed in vitro results regarding apoptosis, autophagy, and cell-cycle arrest. The antitumor activity of quercetin both in vitro and in vivo revealed in this study, point to quercetin as an attractive antitumor agent for hematologic malignancies.

To assess the completeness and reliability of the Information System on Live Births (Sinasc) data. A cross-sectional analysis of the reliability and completeness of Sinasc's data was performed using a sample of Live Birth Certificate (LBC) from 2009, related to births from Campinas, Southeast Brazil. For data analysis, hospitals were grouped according to category of service (Unified National Health System, private or both), 600 LBCs were randomly selected and the data were collected in LBC-copies through mothers and newborns' hospital records and by telephone interviews. The completeness of LBCs was evaluated, calculating the percentage of blank fields, and the LBCs agreement comparing the originals with the copies was evaluated by Kappa and intraclass correlation coefficients. The percentage of completeness of LBCs ranged from 99.8%-100%. For the most items, the agreement was excellent. However, the agreement was acceptable for marital status, maternal education and newborn infants' race/color, low for prenatal visits and presence of birth defects, and very low for the number of deceased children. The results showed that the municipality Sinasc is reliable for most of the studied variables. Investments in training of the professionals are suggested in an attempt to improve system capacity to support planning and implementation of health activities for the benefit of maternal and child population.

To evaluate the microtensile bond strength (µTBS) of a fluoride-containing adhesive system submitted to a pH-cycling and storage time regimen for primary outcomes. As secondary outcomes the fluoride released amount was evaluated. Twelve dentin surfaces from sound third molar were divided into 2 groups according to adhesive systems: Clearfil SE Protect (PB) and Clearfil SE Bond (SE). Sticks obtained (1.0 mm2) from teeth were randomly divided into 3 subgroups according to storage regimen model: immediate (24h); 5-month deionized water (W); and pH-cycling model (C). All sticks were tested for µTBS in a universal testing machine. Fluoride concentration was obtained from 1-4 days and 30-day in W and 1-4 days in demineralization (DE)/remineralization (RE) solutions from C, using a fluoride-specific electrode. µTBS and fluoride released data were, respectively, submitted to ANOVA in a split plot design and Tukey, and Friedman' tests (a=0.05). There was no significant interaction between adhesive system and storage regimen for µTBS. W showed the lowest µTBS values. There was no significant difference between 24 h and C models for µTBS. There was no significant difference between adhesive systems. Failure mode was predominantly cohesive within composite for the 24 h and W, for the C group it was mixed for SE and cohesive within composite for PB adhesive system. Fluoride concentrations in the DE/RE solutions were less than 0.03125 ppm and not detected in W. In conclusion, the fluoride-containing adhesive system performed similarly to the regular one. Hydrolytic degradation is the main problem with both adhesive systems, regardless of fluoride contents.

To investigate the effects of a specific protocol of undulatory physical resistance training on maximal strength gains in elderly type 2 diabetics. The study included 48 subjects, aged between 60 and 85 years, of both genders. They were divided into two groups: Untrained Diabetic Elderly (n=19) with those who were not subjected to physical training and Trained Diabetic Elderly (n=29), with those who were subjected to undulatory physical resistance training. The participants were evaluated with several types of resistance training's equipment before and after training protocol, by test of one maximal repetition. The subjects were trained on undulatory resistance three times per week for a period of 16 weeks. The overload used in undulatory resistance training was equivalent to 50% of one maximal repetition and 70% of one maximal repetition, alternating weekly. Statistical analysis revealed significant differences (p<0.05) between pre-test and post-test over a period of 16 weeks. The average gains in strength were 43.20% (knee extension), 65.00% (knee flexion), 27.80% (supine sitting machine), 31.00% (rowing sitting), 43.90% (biceps pulley), and 21.10% (triceps pulley). Undulatory resistance training used with weekly different overloads was effective to provide significant gains in maximum strength in elderly type 2 diabetic individuals.

ANKHD1 is highly expressed in human acute leukemia cells and potentially regulates multiple cellular functions through its ankyrin-repeat domains. In order to identify interaction partners of the ANKHD1 protein and its role in leukemia cells, we performed a yeast two-hybrid system screen and identified SIVA, a cellular protein known to be involved in proapoptotic signaling pathways. The interaction between ANKHD1 and SIVA was confirmed by co-imunoprecipitation assays. Using human leukemia cell models and lentivirus-mediated shRNA approaches, we showed that ANKHD1 and SIVA proteins have opposing effects. While it is known that SIVA silencing promotes Stathmin 1 activation, increased cell migration and xenograft tumor growth, we showed that ANKHD1 silencing leads to Stathmin 1 inactivation, reduced cell migration and xenograft tumor growth, likely through the inhibition of SIVA/Stathmin 1 association. In addition, we observed that ANKHD1 knockdown decreases cell proliferation, without modulating apoptosis of leukemia cells, while SIVA has a proapoptotic function in U937 cells, but does not modulate proliferation in vitro. Results indicate that ANKHD1 binds to SIVA and has an important role in inducing leukemia cell proliferation and migration via the Stathmin 1 pathway. ANKHD1 may be an oncogene and participate in the leukemia cell phenotype.
