Vitamin A is required for reproduction and normal embryonic development. We have determined that all-trans-retinoic acid (atRA) can support development of the mammalian embryo to parturition in vitamin A-deficient (VAD) rats. At embryonic day (E) 0.5, VAD dams were fed purified diets containing either 12 μg of atRA per g of diet (230 μg per rat per day) or 250 μg of atRA per g of diet (4.5 mg per rat per day) or were fed the purified diet supplemented with a source of retinol (100 units of retinyl palmitate per day). An additional group was fed both 250 μg of atRA per g of diet in combination with retinyl palmitate. Embryonic survival to E12.5 was similar for all groups. However, embryonic development in the group fed 12 μg of atRA per g of diet was grossly abnormal. The most notable defects were in the region of the hindbrain, which included a loss of posterior cranial nerves (IX, X, XI, and XII) and postotic pharyngeal arches as well as the presence of ectopic otic vesicles and a swollen anterior cardinal vein. All embryonic abnormalities at E12.5 were prevented by feeding pharmacological amounts of atRA (250 μg/g diet) or by supplementation with retinyl palmitate. Embryos from VAD dams receiving 12 μg of atRA per g of diet were resorbed by E18.5, whereas those in the group fed 250 μg of atRA per g of diet survived to parturition but died shortly thereafter. Equivalent results were obtained by using commercial grade atRA or atRA that had been purified to eliminate any potential contamination by neutral retinoids, such as retinol. Thus, 250 μg of atRA per g of diet fed to VAD dams (≈4.5 mg per rat per day) can prevent the death of embryos at midgestation and prevents the early embryonic abnormalities that arise when VAD dams are fed insufficient amounts of atRA.

To determine the role of PTHrP in fetal calcium metabolism, blood calcium was measured in mice homozygous (HOM) for deletion of the PTHrP gene. On day 18.5 of gestation, ionized calcium and the maternal–fetal calcium gradient were significantly reduced in HOM PTHrP-ablated fetuses compared with that of their littermates. To assess the placental contribution to the effect of PTHrP, 45Ca and 51Cr-EDTA (as a blood diffusional marker) were administered by intracardiac injection to pregnant, heterozygous dams on day 17.5 of gestation. Five minutes after the injection, whole fetal 45Ca accumulation was significantly decreased in HOM PTHrP-ablated fetuses compared with that of their littermates. Next, two fetuses from each litter were injected in utero with fragments of PTHrP, PTH, or diluent 1 h before administering 45Ca and 51Cr to the dam. PTHrP-(1–86) and PTHrP-(67–86) significantly increased relative 45Ca accumulation in HOM PTHrP-ablated fetuses, but PTHrP(1–34), PTH-(1–84), and the diluent had no effect. Finally, similar studies were performed on fetal mice that lacked the PTH/PTHrP receptor gene. Ionized calcium was significantly reduced in HOM PTH/PTHrP receptor-ablated fetuses. However, 5 min after maternal injection of 45Ca and 51Cr, relative accumulation of 45Ca was significantly increased in these fetuses. It was concluded that PTHrP is an important regulator of fetal blood calcium and placental calcium transport. In addition, the bioactivity of PTHrP for placental calcium transport is specified by a mid-molecular region that does not use the PTH/PTHrP receptor.

Group B streptococci (GBS) cause sepsis and meningitis in neonates and serious infections in adults with underlying chronic illnesses. Specific antibodies have been shown to be an important factor in protective immunity for neonates, but the role of serum complement is less well defined. To elucidate the function of the complement system in immunity to this pathogen, we have used the approach of gene targeting in embryonic stem cells to generate mice totally deficient in complement component C3. Comparison of C3-deficient mice with mice deficient in complement component C4 demonstrated that the 50% lethal dose for GBS infection was reduced by approximately 50-fold and 25-fold, respectively, compared to control mice. GBS were effectively killed in vitro by human blood leukocytes in the presence of specific antibody and C4-deficient serum but not C3-deficient serum. The defective opsonization by C3-deficient serum in vitro was corroborated by in vivo studies in which passive immunization of pregnant dams with specific antibodies conferred protection from GBS challenge to normal and C4-deficient pups but not C3-deficient pups. These results indicate that the alternative pathway is sufficient to mediate effective opsonophagocytosis and protective immunity to GBS in the presence of specific antibody. In contrast, the increased susceptibility to infection of non-immune mice deficient in either C3 or C4 implies that the classical pathway plays an essential role in host defense against GBS infection in the absence of specific immunity.

Introdução: Diversos estudos indicaram consequências de alterações na nutrição materna durante a gestação sobre a saúde da prole adulta, tais como: hipertensão, doenças cardiovasculares, resistência à insulina, diabete melito e doença renal. No entanto, a literatura é pobre em avaliações decorrentes de modificações nutricionais maternas sobre a prole logo após o nascimento. Métodos: Ratas Wistar durante o período gestacional foram alimentadas com dieta hipossódica (HO - 0,15% de NaCl), normossódica (NR - 1,3% de NaCl) ou hipersódica (HR - 8% de Na Cl). Após o nascimento, nas primeiras vinte e quatro horas foram coletados rins e coração dos neonatos machos e fêmeas (n=6- 8/grupo) para verificar as possíveis alterações na estrutura cardíaca e renal pelo método de estereologia. Também foi avaliada a expressão proteica e gênica dos componentes do sistema renina angiotensina (SRA) no coração e rins através do método ELISA indireto e RT-qPCR. Resultados: O peso ao nascimento foi menor em machos e fêmeas da prole de mães alimentadas com dieta hipossódica durante a gestação quando comparado NR e HR. Não houve diferença no volume renal, volume de seus compartimentos (córtex, medula e pelve) e número de glomérulos entre os grupos experimentais (HO, NR e HR). No entanto, o número de glomérulos foi maior em fêmeas comparado aos machos nos três grupos experimentais. O diâmetro transverso do núcleo dos cardiomiócitos no ventrículo esquerdo e no ventrículo direito de machos da prole HR foi maior do que na prole NR. A expressão proteica do receptor AT1 no rim de machos da prole foi menor no grupo HO do que no grupo NR e HR. A expressão proteica do receptor AT2 também foi menor em machos do grupo HO do que no grupo NR. Não houve diferença entre os grupos na expressão proteica dos receptores AT1 e AT2 no rim das fêmeas. Conclusão: O presente estudo detectou alterações na estrutura cardíaca de neonatos machos, mas não em neonatos fêmeas decorrentes de sobrecarga de sal durante a gravidez. As alterações observadas na expressão dos receptores AT1 e AT2 no rim de neonatos machos podem ser responsáveis por alterações na função renal

Em virtude da perspectiva de exploração do potencial em Pequenas Centrais Hidrelétricas como complemento energético a ser utilizado como alternativa para atendimento de demandas localizadas, discute-se neste trabalho a viabilidade ambiental de sua implantação, bem como dos fatores ambientais intervenientes nesse processo. Partindo-se do pressuposto que, no caso das PCHs, as implicações ambientais apresentam especificidade própria e que não podem ser tratadas da mesma forma que os grandes aproveitamentos, busca-se com esse estudo contemplar os principais fatores a serem considerados no processo de tomada de decisão para que haja uma melhor integração entre o empreendimento e o meio onde será inserido. Nesse contexto são produzidos cartas e documentos com a identificação dos fatores ambientais preponderantes, bem como a análise de alternativas locacionais e os impactos decorrentes pela construção de barragens com base em suas diferentes alturas.

Water in sufficient quantities throughout the Colorado Front Range is becoming increasingly limited. This paper examines the consequences for continued unsustainable use of water for communities of the Denver metropolitan area. This paper also looks at the effect that water law in the West has for otherwise optimum distributions of water. In addition, four regional and state water studies are reviewed for their contribution to sustainable water. Finally, the Final Environmental Impact Statement of the Rueter-Hess dam and reservoir project in Parker, Colorado is explored. Key findings conclude that the Rueter-Hess project may not, by itself, provide sustainable water for Parker; but the project will create incentive and opportunity for communities throughout the region to address the question of sustainable water.

As threatened and endangered species, wild Pacific salmon are in peril. This paper discusses the differences of the five species of wild Pacific salmon. As salmon go through several stages of their lifecycles, they face a myriad of threats to their existence. Threats from humans in the form of hydropower dams, habitat destruction, harvesting issues, and hatcheries are explained. A draft recovery plan for salmon in the Puget Sound area of Washington State is used as a case study. Strengths and weaknesses of this plan are discussed. The paper then discusses the need for growth management laws supporting salmon habitat and a change in individual behaviors if wild Pacific salmon sustainability is to become a reality.

En el presente trabajo se describen y analizan las principales obras hidráulicas del sistema de riego tradicional de la comarca de la Vega Baja del Segura, concretamente, los azudes y los acueductos. Los azudes de toma, construidos en el cauce del río Segura, representan el inicio del sistema, desde donde parte una extensa y compleja red de acueductos constituida por acequias y azarbes, como elementos fundamentales, que distribuyen el agua para el regadío. En este estudio se ha llevado a cabo una amplia y profunda investigación documental, tomado datos de campo y captado imágenes in situ que, junto con los diagramas aportados, resultan imprescindibles para comprender la magnitud del sistema. Los resultados obtenidos han servido no sólo para catalogar y caracterizar los elementos que integran el sistema hidráulico del regadío tradicional de la Vega Baja del Segura, sino también para poner de manifiesto un ejemplo real de optimización de recursos hídricos en una zona del sureste peninsular español de escasa e irregular pluviometría. Asimismo, el estudio efectuado revela un sistema de riego sumamente eficiente desde un punto de vista cuantitativo y no tanto desde el cualitativo, debido a la presencia, entre otros contaminantes, de altas concentraciones de sales.

Se ha localizado en la Rambla de la Gavarnera en Ibi, Alicante una pequeña población de Ulmus laevis Pall., que no había sido citado expresamente en localidades valencianas, ni en las floras locales, regionales, ni nacionales, en unos casos por no haberse localizado, en otros por considerar a este árbol alóctono en España. Desde hace una década un equipo de la ETSIA de Montes de la Universidad Politécnica de Madrid, viene realizando diversos proyectos y tesis doctorales que han dado como resultado la puesta en valor de esta especie, a través de su localización a nivel nacional, estudio ecológico, fisiológico y genético, que arrojan numerosas pruebas a favor de la autoctonía de esta especie en España. Según los autores de este dilatado proyecto, U. laevis habría tenido en España uno de su principales refugios cuaternarios, aunque en la actualidad se encuentre fragmentado en poblaciones dispersas, con pocos individuos y problemas para su conservación y supervivencia. No obstante esta especie ha demostrado mayores tasas de supervivencia a la enfermedad de los olmos, que otros de sus congéneres como Ulmus minor y U. glabra. En cuanto a su presencia en Alicante, son precisamente estos autores los que localizan un primer ejemplar en Ibi, al que ahora añadimos dos ejemplares más repartidos en tres puntos muy próximos y situados a lo largo de la Rambla de la Gavarnera. Este es un cauce irregular pero con nivel freático permanente, como lo atestiguan la existencia de pozos, azudes, albercas que abastecieron a los pequeños regadíos de la zona, muchos de ellos abandonados en la actualidad. Dada la rareza de esta especie a nivel nacional y la excepcionalidad de su presencia en Ibi, consideramos que el ejemplar de mayor porte merece la consideración de árbol singular para la Comunidad Valenciana y por otra parte, los otros ejemplares merecen su protección y multiplicación, para incrementar la población existente. Es necesario desarrollar un mayor esfuerzo de localización en este paraje y zonas próximas de la Comarca, para conocer su potencial distribución en la provincia y determinar un estatus de protección adecuado.
