The issues of poverty, famine and malnutrition mainly concern underdeveloped regions. Yet in recent years the proportion of the population threatened with poverty and social exclu- sion has been growing not only in these regions but also in developed areas, including the EU. The issue of poverty is additionally related to demography. In this context, the aging popula- tion of West European countries is worrisome, as the elderly are one of the groups particularly threatened by poverty, and their number continues to grow. Appreciating the gravity of this is- sue, the European Commission considers the fight against poverty to be one of the basic five directions established in the Europe 2020 strategic document.

We propose and evaluate an admission control paradigm for RTDBS, in which a transaction is submitted to the system as a pair of processes: a primary task, and a recovery block. The execution requirements of the primary task are not known a priori, whereas those of the recovery block are known a priori. Upon the submission of a transaction, an Admission Control Mechanism is employed to decide whether to admit or reject that transaction. Once admitted, a transaction is guaranteed to finish executing before its deadline. A transaction is considered to have finished executing if exactly one of two things occur: Either its primary task is completed (successful commitment), or its recovery block is completed (safe termination). Committed transactions bring a profit to the system, whereas a terminated transaction brings no profit. The goal of the admission control and scheduling protocols (e.g., concurrency control, I/O scheduling, memory management) employed in the system is to maximize system profit. We describe a number of admission control strategies and contrast (through simulations) their relative performance.

We propose and evaluate admission control mechanisms for ACCORD, an Admission Control and Capacity Overload management Real-time Database framework-an architecture and a transaction model-for hard deadline RTDB systems. The system architecture consists of admission control and scheduling components which provide early notification of failure to submitted transactions that are deemed not valuable or incapable of completing on time. In particular, our Concurrency Admission Control Manager (CACM) ensures that transactions which are admitted do not overburden the system by requiring a level of concurrency that is not sustainable. The transaction model consists of two components: a primary task and a compensating task. Acute myeloid leukaemia (AML) is the most common form of acute leukaemia in adults. Its treatment has remained largely unchanged for the past 30 years. Chronic myeloid leukaemia (CML) represents a tremendous success story in the era of targeted therapy but significant challenges remain including the development of drug resistance and disease persistence due to presence of CML stem cells. The Aurora family of kinases is essential for cell cycle regulation and their aberrant expression in cancer prompted the development of small molecules that selectively inhibit their activity. Chapter 2 of this thesis outlines the efficacy and mechanism of action of alisertib, a novel inhibitor of Aurora A kinase, in preclinical models of CML. Alisertib possessed equipotent activity against CML cells expressing unmutated and mutated forms of BCR-ABL. Notably, this agent retained high activity against the T315I and E255K BCR-ABL mutations, which confer the greatest degree of resistance to standard CML therapy. Chapter 3 explores the activity of alisertib in preclinical models of AML. Alisertib disrupted cell viability, diminished clonogenic survival, induced expression of the forkhead box O3 (FOXO3a) targets p27 and BCL-2 interacting mediator (BIM), and triggered apoptosis. A link between Aurora A expression and sensitivity to ara-C was established. Chapter 4 outlines the role of the proto-oncogene serine/threonine-protein (PIM) kinases in resistance to ara-C in AML. We report that the novel small molecule PIM kinase inhibitor SGI-1776 disrupted cell viability and induced apoptosis in AML. We establish a link between ara-C resistance and PIM over-expression. Finally, chapter 5 explores how the preclinical work outlined in this thesis may be translated into clinical studies that may lead to novel therapeutic approaches for patients with refractory myeloid leukaemia.

The wave energy industry is progressing towards an advanced stage of development, with consideration being given to the selection of suitable sites for the first commercial installations. An informed, and accurate, characterisation of the wave energy resource is an essential aspect of this process. Ireland is exposed to an energetic wave climate, however many features of this resource are not well understood. This thesis assesses and characterises the wave energy resource that has been measured and modelled at the Atlantic Marine Energy Test Site, a facility for conducting sea trials of floating wave energy converters that is being developed near Belmullet, on the west coast of Ireland. This characterisation process is undertaken through the analysis of metocean datasets that have previously been unavailable for exposed Irish sites. A number of commonly made assumptions in the calculation of wave power are contested, and the uncertainties resulting from their application are demonstrated. The relationship between commonly used wave period parameters is studied, and its importance in the calculation of wave power quantified, while it is also shown that a disconnect exists between the sea states which occur most frequently at the site and those that contribute most to the incident wave energy. Additionally, observations of the extreme wave conditions that have occurred at the site and estimates of future storms that devices will need to withstand are presented. The implications of these results for the design and operation of wave energy converters are discussed. The foremost contribution of this thesis is the development of an enhanced understanding of the fundamental nature of the wave energy resource at the Atlantic Marine Energy Test Site. The results presented here also have a wider relevance, and can be considered typical of other, similarly exposed, locations on Ireland’s west coast.

This thesis details the design and implementation of novel chemical routes towards a series of highly propitious 7-azaindolyl derivatives of the indolocarbazole (ICZ) and bisindolylmaleimide (BIM) families, with subsequent evaluation for use as cancer chemotherapeutic agents. A robust synthetic strategy was devised to allow the introduction of a 7-azaindolyl moiety into our molecular template. This approach allowed access to a wide range of β-keto ester and β-keto nitrile intermediates. Critical analysis identified F-ring modulation as a major theme towards the advancement of ICZ and BIM derivatives in drug therapy. Thus, the employment of cyclocondensation methodology furnished a number of novel aminopyrazole, isoxazolone, pyrazolone and pyrimidinone analogues, considerably widening the scope of the prevalent maleimide functionality. Photochemical cyclisation provided for the first reported aza ICZ containing a six-membered F-ring. Another method towards achieving the aza ICZ core involved use of a Perkin-type condensation approach, with chemical elaboration of the headgroup instigated post-aromatisation. Subsequent use of a modified Lossen rearrangement allowed access to further analogues containing a six-membered F-ring. Extensive screening of the novel aza ICZ and BIM derivatives was carried out against the NCI-60 cancer cell array, with nine prospective candidates selected for continued biological evaluation. From these assays, a number of compounds were shown to inhibit cancer cell growth at concentrations of below 10 nM. Indeed, the most active aza ICZ tested is currently under assessment by the Biological Evaluation Committee of the NCI due to excellent antiproliferative activity demonstrated across the panel of cell lines, with a mean GI50 of 34 nM, a mean total growth inhibition (TGI) of 4.6 μM and a mean cytotoxicity (LC50) of 63.1 μM. Correlation to known topoisomerase I (topo I) inhibitors was revealed by COMPARE analysis, and subsequent topo I-mediated DNA cleavage assays showed inhibitory activity below 1 μM for several derivatives.

An enduring challenge for the policy and political sciences is valid and reliable depiction of policy designs. One emerging approach for dissecting policy designs is the application of Sue Crawford and Elinor Ostrom's institutional grammar tool. The grammar tool offers a method to identify, systematically, the core elements that comprise policies, including target audiences, expected patterns of behavior, and formal modes of sanctioning for noncompliance. This article provides three contributions to the study of policy designs by developing and applying the institutional grammar tool. First, we provide revised guidelines for applying the institutional grammar tool to the study of policy design. Second, an additional component to the grammar, called the oBject, is introduced. Third, we apply the modified grammar tool to four policies that shape Colorado State Aquaculture to demonstrate its effectiveness and utility in illuminating institutional linkages across levels of analysis. The conclusion summarizes the contributions of the article as well as points to future research and applications of the institutional grammar tool. © 2011 Policy Studies Organization.

El efecto de la fecha de siembra sobre la producción y calidad del cultivo de maíz dulce en el norte de la provincia de Buenos Aires no ha sido estudiado y su conocimiento resulta de suma importancia para la planificación y obtención de resultados óptimos y viabilidad comercial para los productores de dicha zona. En base a esta problemática se estudió el efecto de tres fechas de siembra: 28 de octubre, 9 de noviembre y 5 de diciembre 2009 (fecha 1, 2 y 3, respectivamente) sobre el rendimiento y calidad comercial de Maíz Dulce (Zea Mays L., var. saccharata körn). La hipótesis principal que guió este trabajo fue que el atraso de la fecha de siembra reduciría el tamaño y peso de la espiga de maíz dulce afectando sus parámetros comerciales. Para poner a prueba esta hipótesis, se realizó un ensayo a campo en la localidad de Capilla del Señor, provincia de Buenos Aires. En las tres fechas mencionadas se sembró el híbrido Cahill (P) de un ciclo en fecha de siembra óptima de 75 días. El manejo de los cultivos respondió a un manejo convencional de la zona. Durante el ciclo del cultivo se llevaron a cabo determinaciones densidad y área foliar por planta. El momento de la cosecha se determinó por el método de apreciación visual del grano lechoso. Se tomaron muestras de espigas para la determinación de rendimiento y calidad comercial. El área foliar por planta no presentó diferencias significativas entre la fecha 2 y3, sin embargo hubo una tendencia hacia una mayor área foliar en la fecha 2 (2826 cm2 ± 256) que en la fecha 3 (2596 cm2 ± 256). Además, fue la fecha en la que la radiación acumulada incidente fue mayor (1605 mj/m2) en el ciclo de cultivo, ya que ésta fue en aumento desde principios de octubre hasta fines de enero; a partir de ese momento la radiación cayó marcadamente, coincidiendo con el ciclo del cultivo de la fecha 3 (1555 mj/m2). La mayor radiación incidente y la mayor capacidad de captura de la misma (Área Foliar) se tradujo en un mayor peso de la espiga (270 ± 58 gr) para los cultivos sembrados en la fecha 2 respecto de los de la fecha 3 (204 ± 58) y la fecha 1 (190 ± 58). Por otro lado, los cultivos en la fecha 2 presentaron mejor relación grano/marlo (2,57 ± 0,29) y mayor cantidad de granos comerciales (446 ± 45 granos/espiga) diferenciándose significativamente de los cultivos sembrados en la fecha 3 (1,97 ± 0,29) y la fecha 1 (0,92 ± 0,29) para la relación grano/marlo y también para los granos comerciales, 384 ± 45 y 394 ± 45 en los cultivos de la fecha 3 y 1, respectivamente. En vista de los resultados obtenidos se recomienda la fecha de principios de noviembre como fecha óptima de siembra ya que fue en la que se obtuvo el mayor rendimiento y mejor calidad de producto.

Syntheses and NMR studies are reported of two 15N-labelled Pt(II) complexes of anticancer interest: cis-PtCl2(15NH3)(c-C6H1115NH2), a metabolite of the orally-active Pt(IV) complex cis,trans,cis-[PtCl2(acetate)2(c-C6H11NH2)(NH3), and trans-[PtCl2(15NH3)(c-C6H1115NH2), a reduction product of the active Pt(IV) complex trans,trans,trans-[PtCl2(OH)2(c-C6H11NH2). For cis-[PtCl2(15NH3)(c-C6H1115NH2), hydrolysis was faster for the chloride ligand trans to cyclohexylamine, and the pKa values determined by [1H, 15N NMR spectroscopy for the two cis monoaqua isomers were the same (6.73). The trans monoaqua complex was a stronger acid with pKa of 5.4 (determined by 195Pt NMR). For the cis diaqua complex, pKa values of 5.68 and 7.68 were determined.

The monodentate and bidentate pyridyl phosphines, PR3 and R2P(CH2)2PR2, where R=3- or 4-pyridyl can be prepared in high yields by treatment of butyllithium/TMEDA/3- or 4-bromopyridine with PCl3 or Cl2P(CH2)2PCl2 at low temperature. 1,2-Bis(di-2-pyridylphosphino)ethane is conveniently synthesised by an alternative route involving reaction of 1,2-dibromoethane with lithium di-2-pyridylphosphide.
