This work reports on an experimental and finite element method (FEM) parametric study of adhesively-bonded single and double-strap repairs on carbon-epoxy structures under buckling unrestrained compression. The influence of the overlap length and patch thickness was evaluated. This loading gains a particular significance from the additional characteristic mechanisms of structures under compression, such as fibres microbuckling, for buckling restrained structures, or global buckling of the assembly, if no transverse restriction exists. The FEM analysis is based on the use of cohesive elements including mixed-mode criteria to simulate a cohesive fracture of the adhesive layer. Trapezoidal laws in pure modes I and II were used to account for the ductility of most structural adhesives. These laws were estimated for the adhesive used from double cantilever beam (DCB) and end-notched flexure (ENF) tests, respectively, using an inverse technique. The pure mode III cohesive law was equalled to the pure mode II one. Compression failure in the laminates was predicted using a stress-based criterion. The accurate FEM predictions open a good prospect for the reduction of the extensive experimentation in the design of carbon-epoxy repairs. Design principles were also established for these repairs under buckling.

The widespread employment of carbon-epoxy laminates in high responsibility and severely loaded applications introduces an issue regarding their handling after damage. Repair of these structures should be evaluated, instead of their disposal, for cost saving and ecological purposes. Under this perspective, the availability of efficient repair methods is essential to restore the strength of the structure. The development and validation of accurate predictive tools for the repairs behaviour are also extremely important, allowing the reduction of costs and time associated to extensive test programmes. Comparing with strap repairs, scarf repairs have the advantages of a higher efficiency and the absence of aerodynamic disturbance. This work reports on a numerical study of the tensile behaviour of three-dimensional scarf repairs in carbon-epoxy structures, using a ductile adhesive (Araldite® 2015). The finite elements analysis was performed in ABAQUS® and Cohesive Zone Modelling was used for the simulation of damage onset and growth in the adhesive layer. Trapezoidal cohesive laws in each pure mode were used to account for the ductility of the specific adhesive mentioned. A parametric study was performed on the repair width and scarf angle. The use of over-laminating plies covering the repaired region at the outer or both repair surfaces was also tested as an attempt to increase the repairs efficiency. The obtained results allowed the proposal of design principles for repairing composite structures.

Mice immunized with heat or merthiolate-killed culture trypomastigotes of the non-virulent G strain were resistant to the challenge by insect-derived trypomastigotes of the CL strain of Trypanosoma cruzi. No parasitemia was detected, by direct microscopic examination of blood samples, in 90% of immunized mice while all control animals developed a high parasitemia. Trypsinization before heat-inactivation, or fixation with paraformaldehyde, apparently reduced the immunogenicity of the G strain trypomastigotes. Mice immunized with trypomastigotes treated by either of these procedures were not protected against infection by virulent T. cruzi. Analysis of the 13I-labeled surface proteins of G strain trypomastigotes inactivated by the various methods suggests that these components are involved in eliciting protective immunity against T. cruzi infection.

HLA antigens and their relationship with malaria infection were studied in four different ethnic groups in Colombia (South America): two groups of indians (Kunas and Katios), one of negroes and a group of mixed ancestry. A total of 965 persons were studied, 415 with malaria and 550 as controls. HLA-A,B, and C antigen frequencies in the four groups are reported. The association of each HLA antigen with malaria infection due to P. vivax and to P. falciparum was evaluated. Negroes, Kunas and Katios indians variously lack from 6 to 9 of the HLA antigens found in the mixed group. In the designated ethnic groups, antigens B5, B13, B15, Cw2 and Cw4 showed borderline association with malaria infection. However, in the mixed ethnic group, statistically significant associations were found with malaria infection and the presence of A9, Aw19, B17, B35, and Z98 (a B21-B45: crossreacting determinant) with few differences when P. vivax infection and P. falciparum infection were considered individually. This finding may represent a lack of general resistance to malaria in the group that harbors antigens of Caucasian origin. These individuals have been in direct and permanent contact with malaria only in the past 65 years. In contrast, indians, both Kunas and Katios, and Negroes have lived for centuries in malaria endemic areas, and it is possible that a natural selection system has developed through which only those individuals able to initiate an acute immune response to malaria have survived.

São apresentados os resultados de estudos sobre o comportamento do EnterovÃrus-70 (EV-70) na região metropolitana de São Paulo desde sua provável introdução no verão de 1984, determinando extensa epidemia de conjuntivite hemorrágica aguda (C.H.A.), até o final de 1987 abrangendo perÃodo em que este agravo foi pouco notado. Na fase epidêmica ocorrida no primeiro trimestre de 1984 foram estudados 291 indivÃduos divididos em três grupos denominados A, B e C, o primeiro formado por pessoas atingidas pela C.H.A. e os outros dois por indivÃduos não atingidos por este agravo mas que, respectivamente, referiam contato domiciliar com casos de C.H.A. e os que não referiam o citado contato. A demonstração de anticorpos se fez pela técnica de imunofluorescência indireta (IFI) para detectar IgM especÃfico para EV-70 e pela prova de neutralização em cultura de células BHK-21. Verificou-se que 56,7%, 33,3% e 20,6% dos indivÃduos pertencentes, respectivamente, aos grupos A, B e C apresentavam anticorpos especÃficos da classe IgM. No grupo A a faixa etária mais atingida foi a de 10 a 29 anos. No perÃodo que vai do fim da primeira e única epidemia até o final de 1987, identificaram-se três casos esporádicos de C.H.A. e 10 pacientes com afecção neurológica aguda associada à infecção recente pelo EV-70. Nove, destes 10 casos, apresentaram paralisia de nervos cranianos, todos evoluindo sem seqüelas clinicamente discernÃveis. As formas assintomáticas e os casos esporádicos de C.H.A. e de afecções neurológicas mantiveram a circulação do EV-70 no perÃodo não epidêmico.

Aminocarb is a widely applied carbamate insecticide with action of controlling pests such as Lepidoptera and Coleoptera. In this study, subchronic effects on Wistar rats were investigated using hematological, biochemical, and histological techniques. Rats were exposed orally at sublethal levels of 10, 20, or 40 mg/kg body weight (groups A, B, and C, respectively) for 14 d. Hematological results revealed no statistical differences after 1 d of exposure but significant reduction in white blood cells detected after 7 d of exposure in group C, as well as, in all treated groups after 14 d of exposure. Biochemical data showed a decrease of acetylcholinesterase activity in all groups after 1 d of exposure with a return to normal after 7 and 14 d. Significant increase in alkaline phosphatase activity of rats exposed to aminocarb was noted after 7 d of treatment. The levels of triglycerides were also significantly decreased. The present investigation also showed a significant increase in content of serum urea and creatinine in animals from group A (14 d), and from groups B and C (7 and 14 d). Histological results demonstrated hemorrhagic focus on hepatic and renal parenchyma in all exposed groups. Taken together, the attained results were dose dependent and indicated adverse effects of aminocarb on hepatic and renal functions, as well as on immune responsiveness at sublethal tested doses.

A cross-sectional case-control study designed to evaluate the role of malnutrition in the association between the intensity of Schistosoma mansoni infection and clinical schistosomiasis, was conducted in an area with both low frequency of infection and low morbidity of schistosomiasis in Brazil. Cases (256) were patients with a positive stool examination for S. mansoni; their geometrical mean number of eggs/gram of feces was 90. Controls (256) were a random sample of the negative participants paired to the cases by age, sex and length of residence in the area. The clinical signs and symptoms found to be associated with S. mansoni infection, comparing cases and controls, were blood in stools and presence of a palpable liver. A linear trend in the relative odds of these signs and symptoms with increasing levels of infection was detected. Adjusting by the level of egg excretion, the existence of an interaction between palpable liver and ethnic group (white) was suggested. No differences in the nutritional status of infected and non-infected participants were found.

Risk factors for Schistosoma mansoni infection were identified using a 1:1 matched case-control design. The work was conducted in the municipality of Pedro de Toledo, São Paulo State, Brazil, an area where the snail host is Biomphalaria tenagophila. Information on water contact patterns, knowledge, attitudes and pratices (kap), socioeconomic and sanitary conditions were obtained by mean of questionnaires. The crude odds ratio estimates and the adjusted odds ratio estimates using the logistic regression model are presented. Most of the examined individuals admitted recent water contacts (90.6% of the cases). The most frequent reason for contact was swimming, playing and fishing and the preferential site of contact was the river. According to the logistic regression technique, the main risk factors for infection were: a) water contact through swimming, playing and fishing; b) fording; c) bad hygiene. We concluded that recreational activities are the main reasons for schistosomiasis transmission in Pedro de Toledo and leisure alternatives should be offered to the local population.

In the present report the authors discuss the diagnostic difficulties, therapeutic measures and the clinical course of Nocardia infection which occurred among renal transplant recipients at the University Hospital of the Faculty of Medicine of Ribeirão Preto, University of São Paulo (UH-FRP), from 1968 to 1991. Among 500 individuals submitted to renal transplant, 9 patients developed Nocardiosis at varying times after transplant (two months to over two years). All the patients had pulmonary involvement and their most common symptoms were fever, cough and pleural pain. Dissemination of the process is common and three patients presented cutaneous abscesses, four CNS involvement and one had pericarditis due to Nocardia. The diagnostic is quite difficult since there is no specific clinical picture, concomitant infections are frequent and the microorganism presents slow growth in culture (ranging from four to forty days, in our experience). In this report, three cases were only diagnosed by necropsy. The treatment of choice is a combination of Sulfamethoxazole and Trimethoprim (SMX-TMP). In the present series, overall mortality was 77% (7 cases) and in five of the patients who died the diagnosis was late. All the patients who had CNS involvement died.

Neisseria meningitidis are gram-negative diplococci responsible for cases of meningococcal disease all over the world. The epidemic potential of N. meningitidis serogroup B and C is clearly a function of their serotype antigens more than of their capsular polysaccharides. Until recently, hiperimmune sera were used to detect typing antigens on the bacteria. The advent of monoclonal antibodies (MAbs) offered the opportunity to eliminate many of the cross-reactions and have improved the accuracy and reproducibility of meningococcal serotyping. We have produced a MAb to the outer membrane protein of the already existent serotype 17 that have been detected by the use of hiperimmune rabbit sera. The prevalence of this serotype epitope is low in the Brazilian strains. By using the MAb 17 we could not decrease the percentage of nontypeable serogroup C strains. However, there were a decreasing in nontypeable strains to 13% into serogroup B strains and to 25% into the other serogroups.

As micoses estão incluÃdas entre as doenças infecciosas mais ubÃquas em todo o mundo, afectando todos os estratos sociais e todos os grupos etários numa variedade de manifestações superficiais, cutâneas, subcutâneas e sistémicas. Um diagnóstico rápido e eficaz destas doenças, com uma correcta identificação da espécie fúngica responsável pela infecção, é essencial para um planeamento do tratamento mais eficaz para o doente infectado. Tem-se registado um aumento da incidência das infecções fúngicas também em consequência do aumento do número de casos de doentes imunodeprimidos, devido particularmente à crescente utilização de terapêuticas imunossupressivas, procedimentos médicos invasivos, prescrição de tratamentos prolongados, entre outros aspectos. O aparecimento da epidemia da Imunodeficiência Humana no inÃcio da década de 80, causada pelo vÃrus VIH, contribuiu também decisivamente para o aumento das infecções fúngicas oportunistas. A Criptococose é uma infecção fúngica, predominantemente oportunista e com uma distribuição epidemiológica mundial, causada por leveduras encapsuladas do género Cryptococcus. A espécie clinicamente mais relevante é Cryptococcus neoformans, cujas estirpes têm sido tradicionalmente classificadas em cinco serotipos relacionados com os antigénios da respectiva cápsula polissacárida: A (C. neoformans var. grubii); D (C. neoformans var. neoformans); B e C (actualmente reconhecidos como uma espécie distinta mas filogeneticamente próxima, C. gattii); e AD (estirpes hÃbridas). O presente trabalho teve como principal objectivo determinar retrospectivamente os tipos moleculares de uma colecção alargada de estirpes de C. neoformans, isoladas e mantidas durante os últimos 18 anos no Laboratório de Micologia do IHMT/UNL. A maioria das estirpes foi isolada de doentes imunodeprimidos com criptococose, existindo também algumas estirpes de origem ambiental. Foi utilizada a técnica de PCR-RFLP do gene URA5 para diferenciar as estirpes de Cryptococcus neoformans, tendo sido detectados quatro tipos moleculares: VN1 e VN2 (relacionados com C. neoformans var. grubii, serotipo A); VN3 (relacionado com as estirpes hÃbridas de serotipo AD); e VN4 (relacionado com C. neoformans var. neoformans, serotipo D). Não foram encontrados entre os isolados de origem clÃnica perfis de restrição correspondentes aos tipos moleculares VG1, VG2, VG3 e VG4 (relacionados com C. gattii, serotipos B e C). O tipo molecular VN1 foi o mais abundante entre os isolados de origem clÃnica (45% dos isolados), seguindo-se o grupo de estirpes hÃbridas do tipo molecular VN3 (31%). Os tipos moleculares menos abundantes entre os isolados foram o VN2 (12%) e VN4 (12%). A estandardização do método de tipagem molecular utilizado neste trabalho permite comparar os resultados obtidos com os de outros estudos epidemiológicos semelhantes, realizados noutras regiões do globo e publicados em anos recentes, contribuindo para um conhecimento melhorado da epidemiologia global deste importante fungo patogénico. Em Portugal obteve-se uma percentagem mais elevada de isolados dos grupos moleculares VN1 e VN3 em relação a outros paÃses da Europa e América Latina, em que os tipos mais abundantes são VN1 e VN2. Nestas mesmas regiões, o tipo molecular VN4, relacionado com as estirpes de C. neoformans var. neoformans do serotipo D, é muito raro. Esta variedade é mais comum em zonas mediterrâneas e está muito associada a casos clÃnicos de infecções cutâneas associadas a infecções do Sistema Nervoso Central. Este tipo molecular parece ser também significativamente mais abundante no nosso paÃs.
