Quantification of calcium in the cuticle of the fly larva Exeretonevra angustifrons was undertaken at the micron scale using wavelength dispersive X-ray microanalysis, analytical standards, and a full matrix correction. Calcium and phosphorus were found to be present in the exoskeleton in a ratio that indicates amorphous calcium phosphate. This was confirmed through electron diffraction of the calcium-containing tissue. Due to the pragmatic difficulties of measuring light elements, it is not uncommon in the field of entomology to neglect the use of matrix corrections when performing microanalysis of bulk insect specimens. To determine, firstly, whether such a strategy affects the outcome and secondly, which matrix correction is preferable, phi-rho (z) and ZAF matrix corrections were contrasted with each other and without matrix correction. The best estimate of the mineral phase was found to be given by using the phi-rho (z) correction. When no correction was made, the ratio of Ca to P fell outside the range for amorphous calcium phosphate, possibly leading to flawed interpretation of the mineral form when used on its own.

Relationships between mineral uptake and tobacco shoot organogenesis were investigated during three morphogenic phases: phase 1, days 0-10, pre-meristem formation; phase 2, days 10-20, meristem initiation and formation; and phase 3, days 20-35, growth and differentiation of induced meristems into leafy shoots. The mineral content of both shoot-forming (SF) and non-shoot-forming (NSF) media was examined over the 35-day culture period. Both SF and NSF explants rapidly consumed iron during phase 1. Nitrate uptake in SF explants was high and independent of explant growth during phases 1 and 2, but greatest and strongly correlated with growth during phase 3. Phosphorus uptake was highest in SF explants during phases 2 and 3, and correlated with explant growth. Uptake of potassium, calcium and sulphur was strongly associated with explant growth during phase 3 whereas magnesium uptake was only poorly correlated with growth. Results from this study indicate that particular minerals may have an important role in regulating development as well as generally supporting growth.

Blast fragmentation can have a significant impact on the profitability of a mine. An optimum run of mine (ROM) size distribution is required to maximise the performance of downstream processes. If this fragmentation size distribution can be modelled and controlled, the operation will have made a significant advancement towards improving its performance. Blast fragmentation modelling is an important step in Mine to Mill™ optimisation. It allows the estimation of blast fragmentation distributions for a number of different rock mass, blast geometry, and explosive parameters. These distributions can then be modelled in downstream mining and milling processes to determine the optimum blast design. When a blast hole is detonated rock breakage occurs in two different stress regions - compressive and tensile. In the-first region, compressive stress waves form a 'crushed zone' directly adjacent to the blast hole. The second region, termed the 'cracked zone', occurs outside the crush one. The widely used Kuz-Ram model does not recognise these two blast regions. In the Kuz-Ram model the mean fragment size from the blast is approximated and is then used to estimate the remaining size distribution. Experience has shown that this model predicts the coarse end reasonably accurately, but it can significantly underestimate the amount of fines generated. As part of the Australian Mineral Industries Research Association (AMIRA) P483A Mine to Mill™ project, the Two-Component Model (TCM) and Crush Zone Model (CZM), developed by the Julius Kruttschnitt Mineral Research Centre (JKMRC), were compared and evaluated to measured ROM fragmentation distributions. An important criteria for this comparison was the variation of model results from measured ROM in the-fine to intermediate section (1-100 mm) of the fragmentation curve. This region of the distribution is important for Mine to Mill™ optimisation. The comparison of modelled and Split ROM fragmentation distributions has been conducted in harder ores (UCS greater than 80 MPa). Further work involves modelling softer ores. The comparisons will be continued with future site surveys to increase confidence in the comparison of the CZM and TCM to Split results. Stochastic fragmentation modelling will then be conducted to take into account variation of input parameters. A window of possible fragmentation distributions can be compared to those obtained by Split . Following this work, an improved fragmentation model will be developed in response to these findings.

The Crim1 gene is predicted to encode a transmembrane protein containing six von Willebrand-like cysteine-rich repeats (CRRs) similar to those in the BMP-binding antagonist Chordin (Chrd). In this study, we verify that CRIM1 is a glycosylated, Type I transmembrane protein and demonstrate that the extracellular CRR-containing domain can also be secreted, presumably via processing at the membrane. We have previously demonstrated Crim1 expression at sites consistent with an interaction with bone morphogenetic proteins (BMPs). Here we show that CRIM1 can interact with both BMP4 and BMP7 via the CRR-containing portion of the protein and in so doing acts as an antagonist in three ways. CRIM1 binding of BMP4 and -7 occurs when these proteins are co-expressed within the Golgi compartment of the cell and leads to (i) a reduction in the production and processing of preprotein to mature BMP, (ii) tethering of pre-BMP to the cell surface, and (iii) an effective reduction in the secretion of mature BMP. Functional antagonism was verified by examining the effect of coexpression of CRIM1 and BMP4 on metanephric explant culture. The presence of CRIM1 reduced the effective BMP4 concentration of the media, thereby acting as a BMP4 antagonist. Hence, CRIM1 modulates BMP activity by affecting its processing and delivery to the cell surface

Frequency deviation is a common problem for power system signal processing. Many power system measurements are carried out in a fixed sampling rate assuming the system operates in its nominal frequency (50 or 60 Hz). However, the actual frequency may deviate from the normal value from time to time due to various reasons such as disturbances and subsequent system transients. Measurement of signals based on a fixed sampling rate may introduce errors under such situations. In order to achieve high precision signal measurement appropriate algorithms need to be employed to reduce the impact from frequency deviation in the power system data acquisition process. This paper proposes an advanced algorithm to enhance Fourier transform for power system signal processing. The algorithm is able to effectively correct frequency deviation under fixed sampling rate. Accurate measurement of power system signals is essential for the secure and reliable operation of power systems. The algorithm is readily applicable to such occasions where signal processing is affected by frequency deviation. Both mathematical proof and numerical simulation are given in this paper to illustrate robustness and effectiveness of the proposed algorithm. Background: Although early in life there is little discernible difference in bone mass between boys and girls, at puberty sex differences are observed. It is uncertain if these differences represent differences in bone mass or just differences in anthropometric dimensions. Aim: The study aimed to identify whether sex independently affects bone mineral content (BMC) accrual in growing boys and girls. Three sites are investigated: total body (TB), femoral neck (FN) and lumbar spine (LS). Subjects and methods: 85 boys and 67 girls were assessed annually for seven consecutive years. BMC was assessed by dual energy X-ray absorptiometry (DXA). Biological age was defined as years from age at peak height velocity (PHV). Data were analysed using a hierarchical (random effects) modelling approach. Results: When biological age, body size and body composition were controlled, boys had statistically significantly higher TB and FN BMC at all maturity levels (p < 0.05). No independent sex differences were found at the LS (p > 0.05). Conclusion: Although a statistical significant sex effect is observed, it is less than the error of the measurement, and thus sex difference are debatable. The epidemic that is osteoporosis has led to an increasing interest in bone mineral, and the factors that influence the levels of bone mineral, in recent years. While it is unrealistic to try and turn back the clock, a return to an increased level of physical activity may be an important consideration in terms of skeletal health. Peak bone mass is largely determined by heredity, but lifestyle and dietary patterns also influence the level of bone mineral accrued during the growing years. In this review, we summarize the evidence that vigorous weight-bearing physical activity and adequate calcium intake represent the best possibility for enhancing the attainment of an optimal level of bone mineral, within genetic limits.

Adolescents and adults with CF have lower bone mineral density (BMD) than normal, but its relationship with phenotype is not well understood. Point FEV1% predicted (FEV) and rate of change of FEV are biased estimates of disease severity, because progressively older subjects represent a selected survivor population, with females at greater risk of death than males. To investigate the relationship between BMD and phenotype we used an index (predicted age at death) derived from Bayesian estimates of slope and intercept of FEV, age at last measurement and survival status. Predictive equations for the index were derived from 97 subjects (78 survivors) from the RCH CF clinic, and applied to a group of 102 comparable subjects who had BMD measured, classified as having‘mild’ ()75th), ‘moderate’ (25– 75th), or ‘severe’ (-25th centile) phenotype. Total body (TB) and lumbar spine (LS) BMD z-scores (Z) were compared, adjustingfor gender effects, using 2-way ANOVA. Annual mean change in FEV segregated, as expected, according to phenotype, ‘severe’ (ns25), ‘moderate’ (ns51) and ‘mild’ (ns25) y3.01(y3.73 to y2.30)%, y0.85(y1.36 to y0.35)%, 2.70(1.92 to 3.46)%, respectively, with no gender difference. LS and TB BMDZ were different in each phenotype (P-s 0.002), LS BMDZ for ‘severe’, ‘moderate’ and ‘mild’ y1.63(CI: y2.07 to y 1.19), y0.86(CI: y1.17 to y0.55), y0.06(CI: y0.54 to 0.41). Males had lower LS BMDZ than females overall (y1.22 (CI: y1.54 to y0.91) vs. y0.48(CI: y 0.84 to y0.12) Ps0.002). In the ‘severe’ group, males had lower TB BMDZ and LS BMDZ (PF0.002). Low BMD is associated with ‘moderate’ and ‘severe’ phenotypes, with relative preservation in females in the ‘severe’ group. Female biology (reproductive fitness) might promote resistance to bone resorption at a critical level of BMD loss.

A cultura do cafeeiro sempre ocupou lugar de destaque na economia do país, dada sua importância na área econômica e social, buscando cada vez mais, um mercado diferenciado, com novas tecnologias para a melhoria da qualidade da bebida. A atividade de lavagem e despolpa de frutos do cafeeiro, necessária para a redução do custo de secagem e a melhoria da qualidade de bebida, é geradora de grandes volumes de resíduos sólidos e líquidos, ricos em material orgânico e inorgânico. A água residuária da atividade de beneficiamento do café (ARC) é gerada anualmente em grande volume no Espírito Santo, e aliado aos nutrientes existentes nesse efluente indica a sua viabilidade de reaproveitamento na fertirrigação de culturas agrícolas. O objetivo do presente trabalho foi avaliar o efeito de diferentes doses de água residuária de café no crescimento, na absorção e interação entre nutrientes e no estado nutricional do milho. Para tanto, foi conduzido um experimento em casa de vegetação utilizando-se o delineamento experimental inteiramente casualizado, onde foram aplicadas 7 doses de ARC, com 3 repetições, em unidades experimentais constituídas por vasos com 2 dm³ de solo. As doses foram equivalentes a 0,00, 15,17, 30,35, 45,52, 60,70, 75,87 e 91,05 litros de ARC por m² de solo. Realizou-se a semeadura de cinco sementes de milho híbrido BR 206 por vaso e cinco dias após a germinação das plantas foi feito o desbaste, mantendo-se três plantas por vaso. Aos trinta dias após a germinação determinou-se o diâmetro do caule (DC), área foliar (AF), matéria seca da parte aérea (MSPA), matéria seca do sistema radicular (MSR), relação parte aérea/raiz (MSPA/MSR), razão massa radicular (matéria seca raiz/matéria seca total) e razão área foliar (área foliar/matéria seca total). Na parte aérea das plantas foi determinado os teores dos macronutrientes (N, P, K, Ca, Mg e S). Os dados foram submetidos a análise de variância e as variáveis em função das doses de ARC submetidas a análise de regressão. Para as variáveis dependentes foi calculado o coeficiente de correlação linear de Pearson. A ARC serviu como fonte de nutrientes para as plantas de milho, aumentou o rendimento da maioria das variáveis de crescimento e os teores de N, K e S. Porém diminuiu o teor de Ca, Mg e P da parte aérea das plantas além de indicar que altas doses promovem desbalanceamento na relação entre nutrientes.

Elastin isolated from fresh bovine ligaments was dissolved in a mixture of 1,1,1,3,3,3-Hexafluoro-2-propanol and water and electrospun into fiber membranes under different processing conditions. Fiber mats of randomly and aligned fibers were obtained with fixed and rotating ground collectors and fibrils were composed by thin ribbons whose width depends on electrospinning conditions; fibrils with 721 nm up to 2.12 m width were achieved. After cross-linking with glutaraldehyde, -elastin can uptake as much as 1700 % of PBS solution and a slight increase on fiber thickness was observed. The glass transition temperature of electrospun fiber mats was found to occur at ~ 80 ºC. Moreover, -Elastin showed to be a perfect elastomeric material, and no mechanical hysteresis was found in cycle mechanical measurements. The elastic modulus obtained for oriented and random fibers mats in a PBS solution was 330 ± 10 kPa and 732 ± 165 kPa, respectively. Finally, the electrospinning and cross-linking process does not inhibit MC-3T3-E1 cell adhesion. Cell culture results showed good cell adhesion and proliferation in the cross-linked elastin fiber mats.

Bone weakening can occur due to the absence of load on the skeleton or even short periods of decreased physical activity. Therefore, musculoskeletal diseases that involve temporary immobilization by casts, inactivity or tension increases the risk of fractures. Physical activity is the most studied procedure both to prevent damage and to restore bone structure. The present study aimed at evaluating, by bone densitometry on rat femurs, the influence of hindlimb unloading and later running activity on treadmill or free movement. Sixty-four Wistar rats were used, aged 65 days with a mean corporal mass of 316.11g, randomly divided into eight experimental groups: group 1, the suspended control with seven animals under hindlimb unloading regimen for 28 days, then euthanized; groups 2 and 3, the trained suspended comprising of 7 and five animals, respectively, subjected to hindlimb unloading for 28 days, followed by treadmill exercise for 28 days (group 2) or 56 days (group 3), then euthanized; groups 4 and 5, designated free suspended, comprised of 7 animals each under hindlimb unloading regimen for 28 days followed by free activity in cages for 28 days (group 4) or 56 days (group 5), then euthanized; groups 6, 7 and 8, negative controls, each with 8 animals allowed to free activity in cages and euthanized at the ages of 93, 121 and 149 days, respectively. Bone mineral density (BMD) of the left femur was analyzed by bone densitometry. Unloading by tail-suspension decreased BMD while treadmill training and free activity in cages promoted its recovery in a similar way and over time.

A grande expansão das indústrias e do mercado consumidor tem provocado, nas últimas décadas, a geração de elevadas quantidades de resíduos, os quais têm constituído constante preocupação econômica e ambiental. Objetivou-se, neste trabalho, analisar a viabilidade econômica do uso de três doses de composto orgânico, oriundo da compostagem de resíduos da extração da celulose, em substituição à adubação mineral, na cultura do eucalipto, no município de Selvíria, MS. A metodologia de custos adotada foi a do custo operacional total e do custo total. Concluiu-se que, por causa do elevado custo de transporte e aplicação, a adubação orgânica mostrou-se mais onerosa em relação à mineral, sendo sua utilização viável, economicamente, apenas nas proximidades da indústria produtora.

Existem diversos meios nutritivos utilizados no cultivo in vitro de orquídeas, com diferentes composições e concentrações de sais, resultando em respostas distintas entre eles. O objetivo deste trabalho foi avaliar o crescimento in vitro de plântulas de orquídea (Cattleya walkeriana Gardner) sob diferentes doses de fertilizante NPK, adicionado ao meio de cultivo, como fonte de nutrientes. Foi utilizado o fertilizante Peters® na formulação NPK 10-30-20 + Mg + micronutrientes, nas doses de: 0,0; 1,0; 2,0; 3,0; 4,0; 5,0 e 10,0 g L-1 de meio, acrescido de água de coco (150 mL L-1), sacarose (20 g L-1), carvão ativado (2 g L-1) e pH corrigido para 5,6. Foram utilizados frascos de vidro de 320 mL de capacidade, contendo 35 mL de meio onde foram inoculadas dez plântulas com seis meses de idade, previamente germinadas em meio Knudson C. O experimento foi mantido em sala de crescimento a 27 ± 2 ºC, 16/8 h luz/escuro e irradiância de 48 µmol m-2 s-1. A produção máxima de matéria seca da parte aérea das plantas, aos seis meses de idade, foi estimada em 0,255 g/frasco com uso de 5,22 g L-1 do fertilizante adicionado ao meio de cultura. Considerando a possibilidade de salinidade elevada do meio, para algumas espécies de orquídea, estimou-se a dose de fertilizante (3,55 g L-1) necessária para obtenção de 90% da produção máxima. As raízes apresentaram drástica redução do crescimento (menor número e curtas) em condições de alta concentração de fertilizante.

A utilização de fungicida para o controle de doenças foliares na cultura do trigo é prática comum entre os triticultores. Contudo, devido ao impacto ambiental provocado pelo seu uso generalizado, buscam-se alternativas para controle dos fitopatógenos. Sendo assim, este trabalho teve como objetivo avaliar o efeito da nutrição mineral foliar sobre as doenças foliares do trigo, bem como seu efeito sobre o desenvolvimento da planta. O delineamento experimental foi em blocos casualizados com quatro repetições e quatro tratamentos: 1- testemunha; 2- adubação foliar aos 30 dias após a emergência (DAE) e aos 75 DAE; 3- fungicida aos 30 e 75 DAE; e 4- adubação foliar aos 30 DAE e adubação foliar mais fungicida aos 75 DAE. Foram realizadas nove avaliações da incidência e da severidade do oídio, cujos dados foram utilizados para o cálculo das áreas abaixo das curvas de progresso da doença; três avaliações da severidade da ferrugem e da mancha da gluma; e duas avaliações da mancha amarela. Foram determinadas a massa de matéria fresca e seca da parte aérea e do sistema radicular e a produtividade da cultura. Foi observada redução da incidência e da severidade de oídio em função da adubação foliar, apenas na primeira avaliação de incidência e nas segunda e quinta avaliações de severidade da doença. O efeito complementar dos nutrientes com o fungicida para redução das doenças foi observado apenas para a mancha amarela, com redução de aproximadamente 11,16 % da severidade. Os maiores valores de produtividade foram observados para os tratamentos apenas com fungicida e, com fungicida mais nutrientes. A adubação foliar proporcionou aumento da massa fresca de raízes e massa seca de raízes. Apesar disso, não houve aumento da massa fresca da parte aérea em função da adubação foliar.

Protein aggregation became a widely accepted marker of many polyQ disorders, including Machado-Joseph disease (MJD), and is often used as readout for disease progression and development of therapeutic strategies. The lack of good platforms to rapidly quantify protein aggregates in a wide range of disease animal models prompted us to generate a novel image processing application that automatically identifies and quantifies the aggregates in a standardized and operator-independent manner. We propose here a novel image processing tool to quantify the protein aggregates in a Caenorhabditis elegans (C. elegans) model of MJD. Confocal mi-croscopy images were obtained from animals of different genetic conditions. The image processing application was developed using MeVisLab as a platform to pro-cess, analyse and visualize the images obtained from those animals. All segmenta-tion algorithms were based on intensity pixel levels.The quantification of area or numbers of aggregates per total body area, as well as the number of aggregates per animal were shown to be reliable and reproducible measures of protein aggrega-tion in C. elegans. The results obtained were consistent with the levels of aggrega-tion observed in the images. In conclusion, this novel imaging processing applica-tion allows the non-biased, reliable and high throughput quantification of protein aggregates in a C. elegans model of MJD, which may contribute to a significant improvement on the prognosis of treatment effectiveness for this group of disor-ders
