A study was conducted, in association with the Sapelo Island and North Carolina National Estuarine Research Reserves (NERRs), to evaluate the impacts of coastal development on sentinel habitats (e.g., tidal creek ecosystems), including potential impacts to human health and well-being. Uplands associated with southeastern tidal creeks and the salt marshes they drain are popular locations for building homes, resorts, and recreational facilities because of the high quality of life and mild climate associated with these environments. Tidal creeks form part of the estuarine ecosystem characterized by high biological productivity, great ecological value, complex environmental gradients, and numerous interconnected processes. This research combined a watershed-level study integrating ecological, public health and human dimension attributes with watershed-level land use data. The approach used for this research was based upon a comparative watershed and ecosystem approach that sampled tidal creek networks draining developed watersheds (e.g., suburban, urban, and industrial) as well as undeveloped sites. The primary objective of this work was to clearly define the relationships between coastal development with its concomitant land use changes and non-point source pollution loading and the ecological and human health and well-being status of tidal creek ecosystems. Nineteen tidal creek systems, located along the southeastern United States coast from southern North Carolina to southern Georgia, were sampled during summer (June-August), 2005 and 2006. Within each system, creeks were divided into two primary segments based upon tidal zoning: intertidal (i.e., shallow, narrow headwater sections) and subtidal (i.e., deeper and wider sections), and watersheds were delineated for each segment. In total, we report findings on 24 intertidal and 19 subtidal creeks. Indicators sampled throughout each creek included water quality (e.g., dissolved oxygen concentration, salinity, nutrients, chlorophyll-a levels), sediment quality (e.g., characteristics, contaminants levels including emerging contaminants), pathogen and viral indicators, and abundance and genetic responses of biological resources (e.g., macrobenthic and nektonic communities, shellfish tissue contaminants, oyster microarray responses). For many indicators, the intertidally-dominated or headwater portions of tidal creeks were found to respond differently than the subtidally-dominated or larger and deeper portions of tidal creeks. Study results indicate that the integrity and productivity of headwater tidal creeks were impaired by land use changes and associated non-point source pollution, suggesting these habitats are valuable early warning sentinels of ensuing ecological impacts and potential public health threats. For these headwater creeks, this research has assisted the validation of a previously developed conceptual model for the southeastern US region. This conceptual model identified adverse changes that generally occurred in the physical and chemical environment (e.g., water quality indicators such as indicator bacteria for sewage pollution or sediment chemical contamination) when impervious cover levels in the watershed reach 10-20%. Ecological characteristics responded and were generally impaired when impervious cover levels exceed 20-30%. Estimates of impervious cover levels defining where human uses are impaired are currently being determined, but it appears that shellfish bed closures and the flooding vulnerability of headwater regions become a concern when impervious cover values exceed 10-30%. This information can be used to forecast the impacts of changing land use patterns on tidal creek environmental quality as well as associated human health and well-being. In addition, this study applied tools and technologies that are adaptable, transferable, and repeatable among the high quality NERRS sites as comparable reference entities to other nearby developed coastal watersheds. The findings herein will be of value in addressing local, regional and national needs for understanding multiple stressor (anthropogenic and human impacts) effects upon estuarine ecosystems and response trends in ecosystem condition with changing coastal impacts (i.e., development, climate change). ENGLISH: A study of the temporal and spatial distribution of larval tunas and the concomitant oceanic conditions was made in cooperation with the Direccion General de Pesca e Industrias Conexas of Mexico. Field work consisted of eight hydrographic cruises made from October 1966 through August 1967 near the entrance of the Gulf of California. From January through April, surface currents were southerly at velocities up to 20 cm/sec; currents in June were variable in direction and mostly less than 10 cm/sec; by August the surface current was northerly at 10-15 cm/sec. Surface winds were usually secondary to the distribution of mass as an influence on the surface circulation. Currents at 100 m were generally similar in direction to those at the surface, but the water moved more slowly. Between the surface and 100 m, southbound currents crossed the entrance of the Gulf at velocities of 5-10 cm/sec during January and April, forming frontal boundaries with the California Current water, which often occurred south of the entrance. From April to August, the median concentration of surface chlorophyll a increased from 0.65 to 0.97 mg/m3, while the median productivity increased from 5.6 mgC/m3/day in April to 17.8 mgC/m3/day in June before returning to 2.6 mgC/m3/day in August. Primary productivity was closely correlated with the concentration of surface chlorophyll a. Productivity was generally higher in the vicinity of the Gulf than that found for water in the open Pacific. Productivity was highest near Islas Las Tres Marias and second highest near Cabo San Lucas, both locations of local upwelling. The standing crop of phytoplankton was shown to be subjected to progressively heavier grazing pressure in the spring and summer by zooplankton. SPANISH: Un estudio de la distribución temporal y espacial de las larvas de atún y de las condiciones oceánicas concomitantes fue realizado en cooperación con la Dirección General de Pesca e Industrias Conexas de México. El trabajo experimental consistió en ocho cruceros hidrográficos realizados desde octubre 1966 hasta agosto 1967, cerca a la entrada del Golfo de California. De enero a abril, las corrientes superficiales fueron meridionales alcanzando velocidades hasta de 20 cm/seg; las corrientes en junio fueron variables en dirección y la mayoría con una velocidad de menos de 10 cm/seg; en agosto la corriente superficial fue septentrional a 10-15 cm/seg, Los vientos superficiales fueron por lo común secundarios a la dístríbucíón de la masa, como una influencia de la circulación superficial. Las corrientes a 100 m fueron generalmente similares en dirección a las de la superficie, pero el agua se movió más lentamente. Entre la superficie y los 100 m, las corrientes que se dirigen hacia el sur cruzaron la entrada del Golfo a velocidades de 5-10 cm/seg durante enero y abril formando límites frontales con el agua de la Corriente de California, que apareció a menudo al sur de la entrada. De abril a agosto, la concentración media de la clorofila a superficial aumentó de 0.65 a 0.97 mg/m3, mientras que la productividad mediana aumentó de 5.6 mgC/m3/día en abril hasta 17.8 mgC/m3/día en junio antes de regresar a 2.6 mgC/m3/día en agosto. La productividad primaria se correlacionó estrechamente con la concentración de clorofila a superficial. La productividad fue generalmente más alta en la vecindad del Golfo que aquella encontrada en el agua de alta mar del Pacífico. La productividad fue más alta cerca a las Islas Tres Marías, y el segundo máximo fue cerca al Cabo San Lucas, ambas localidades de afloramiento local. Limnological studies in Lake Victoria (Kenyan portion) have been sporadic. Water quality and nutrient dynamics studies are being undertaken in fifteen sampling sites that have been divided into four ecological zones namely: Nyanza Gulf, Rusinga Channel, open waters inshore and open waters. The ongoing study will show how the physical and chemical paramenters affect fish distribution and abundance.

Production of mince from Tilapia using a combination of physical and chemical methods was found to improve the storage life of the mince in the deep freezer. Though the chemical composition of the mince was slightly affected, the mince was microbiologically stable throughout the five weeks frozen storage. Fish cakes prepared traditionally from tilapia minces were more acceptable than oven dried cakes. Production of fish cakes form tilapia will improve utilization of this species in areas where small size tilapia are regarded as fish of low economic value

This thesis describes the use of multiply-substituted stable isotopologues of carbonate minerals and methane gas to better understand how these environmentally significant minerals and gases form and are modified throughout their geological histories. Stable isotopes have a long tradition in earth science as a tool for providing quantitative constraints on how molecules, in or on the earth, formed in both the present and past. Nearly all studies, until recently, have only measured the bulk concentrations of stable isotopes in a phase or species. However, the abundance of various isotopologues within a phase, for example the concentration of isotopologues with multiple rare isotopes (multiply substituted or 'clumped' isotopologues) also carries potentially useful information. Specifically, the abundances of clumped isotopologues in an equilibrated system are a function of temperature and thus knowledge of their abundances can be used to calculate a sample’s formation temperature. In this thesis, measurements of clumped isotopologues are made on both carbonate-bearing minerals and methane gas in order to better constrain the environmental and geological histories of various samples.

Clumped-isotope-based measurements of ancient carbonate-bearing minerals, including apatites, have opened up paleotemperature reconstructions to a variety of systems and time periods. However, a critical issue when using clumped-isotope based measurements to reconstruct ancient mineral formation temperatures is whether the samples being measured have faithfully recorded their original internal isotopic distributions. These original distributions can be altered, for example, by diffusion of atoms in the mineral lattice or through diagenetic reactions. Understanding these processes quantitatively is critical for the use of clumped isotopes to reconstruct past temperatures, quantify diagenesis, and calculate time-temperature burial histories of carbonate minerals. In order to help orient this part of the thesis, Chapter 2 provides a broad overview and history of clumped-isotope based measurements in carbonate minerals.

In Chapter 3, the effects of elevated temperatures on a sample’s clumped-isotope composition are probed in both natural and experimental apatites (which contain structural carbonate groups) and calcites. A quantitative model is created that is calibrated by the experiments and consistent with the natural samples. The model allows for calculations of the change in a sample’s clumped isotope abundances as a function of any time-temperature history.

In Chapter 4, the effects of diagenesis on the stable isotopic compositions of apatites are explored on samples from a variety of sedimentary phosphorite deposits. Clumped isotope temperatures and bulk isotopic measurements from carbonate and phosphate groups are compared for all samples. These results demonstrate that samples have experienced isotopic exchange of oxygen atoms in both the carbonate and phosphate groups. A kinetic model is developed that allows for the calculation of the amount of diagenesis each sample has experienced and yields insight into the physical and chemical processes of diagenesis.

The thesis then switches gear and turns its attention to clumped isotope measurements of methane. Methane is critical greenhouse gas, energy resource, and microbial metabolic product and substrate. Despite its importance both environmentally and economically, much about methane’s formational mechanisms and the relative sources of methane to various environments remains poorly constrained. In order to add new constraints to our understanding of the formation of methane in nature, I describe the development and application of methane clumped isotope measurements to environmental deposits of methane. To help orient the reader, a brief overview of the formation of methane in both high and low temperature settings is given in Chapter 5.

In Chapter 6, a method for the measurement of methane clumped isotopologues via mass spectrometry is described. This chapter demonstrates that the measurement is precise and accurate. Additionally, the measurement is calibrated experimentally such that measurements of methane clumped isotope abundances can be converted into equivalent formational temperatures. This study represents the first time that methane clumped isotope abundances have been measured at useful precisions.

In Chapter 7, the methane clumped isotope method is applied to natural samples from a variety of settings. These settings include thermogenic gases formed and reservoired in shales, migrated thermogenic gases, biogenic gases, mixed biogenic and thermogenic gas deposits, and experimentally generated gases. In all cases, calculated clumped isotope temperatures make geological sense as formation temperatures or mixtures of high and low temperature gases. Based on these observations, we propose that the clumped isotope temperature of an unmixed gas represents its formation temperature — this was neither an obvious nor expected result and has important implications for how methane forms in nature. Additionally, these results demonstrate that methane-clumped isotope compositions provided valuable additional constraints to studying natural methane deposits.

This new project is multidisciplinary, with physical and chemical palaeolimnological aspects mainly the responsibility of Swiss and Russian scientists, and the biological limnology and palaeolimnology components mainly undertaken by the British and Russian groups. The overall project aim is to improve palaeoclimate reconstructions using sedimentary diatoms by promoting better understanding of diatom ecology and sediment-forming processes. The initial work plan is divided into four main parts: To understand diatom phytoplankton ecology more fully, to assess taphonomic changes associated with the transformation of phytoplankton diatom communities into sediment assemblages, to demonstrate sediment core integrity and representativity and to calibrate modern diatom assemblages against contemporary climate records. The preliminary results from the interrelated studies of phytoplankton, sediment traps and sediment cores used in GEOPASS-NERC, demonstrate the complexity of links between the living and fossil systems. Furthermore, the nature of recent sedimentation in Lake Baikal is spatially variable and incompletely known. This poses a major challenge to palaeolimnological interpretation. Turbidite deposits and differential preservation of microfossils, combined with inadequate knowledge of the modern ecology of endemic diatoms, all conspire to obfuscate the sedimentary record of environmental change.

Trata-se de um estudo quantitativo, descritivo, visando uma análise das condições de trabalho em unidades intensivas de um hospital universitário no município do Rio de Janeiro. Foi definido como objeto de estudo a percepção dos trabalhadores de enfermagem sobre os riscos ocupacionais e os problemas de saúde inerentes às condições de trabalho em unidades intensivas e como problema de pesquisa: quais os riscos ocupacionais e problemas de saúde relacionados às condições de trabalho, percebidos pelos trabalhadores de enfermagem, em unidades intensivas de um hospital universitário? O objetivo geral foi estudar nas unidades intensivas os riscos ocupacionais e problemas de saúde da equipe de enfermagem e sua relação com condições de trabalho, a partir da percepção dos mesmos. Os objetivos específicos traçados foram: identificar as características pessoais e profissionais dos trabalhadores de enfermagem de unidades intensivas; descrever os fatores de risco do ambiente de trabalho percebidos pelos trabalhadores de enfermagem; levantar os problemas de saúde percebidos pelos trabalhadores e sua relação com o trabalho; analisar a associação entre os problemas de saúde percebidos pelos trabalhadores de enfermagem e as condições do trabalho em unidades intensivas. Participaram da pesquisa 125 profissionais de enfermagem de quatro unidades intensivas do Hospital Universitário (HU) entre Maio e Julho de 2009. A predominância foi de profissionais do sexo feminino, com idade acima dos 40 anos, com mais de um vínculo empregatício e trabalhando no HU há mais de 10 anos. Os riscos ocupacionais mais percebidos pelos trabalhadores foram os ergonômicos, seguido dos biológicos, de acidentes, físicos e químicos. Os problemas de saúde mais frequentes foram varizes, problemas oculares, lombalgias, estresse e depressão, transtornos do sono, lesões de coluna vertebral, dores de cabeça, mudanças no humor, dores musculares crônicas e hipertensão arterial. Pela associação entre riscos ocupacionais e problemas de saúde, conclui-se que os trabalhadores expostos a fatores de riscos ergonômicos e físicos têm maior probabilidade de adquirir problemas de saúde osteoarticulares e circulatórios (varizes). Diante dos dados desta pesquisa faz-se necessário aprofundamento da investigação sobre os fatores de riscos encontrados e possíveis medidas para minimizá-los, mediante novos estudos. Como recomendações destacam-se a criação de um espaço de discussão entre os gerentes e trabalhadores para a elaboração de um programa que vise a promoção e proteção da saúde do trabalhador de enfermagem de unidades intensivas; implementação de medidas de controle específicas para cada tipo de risco evidenciado e a criação de um Comitê de Ergonomia para operacionalizar a implementação das melhorias no HU, a fim de consolidar as transformações esperadas.

To study and qualify the factors influencing interactions between various trophic levels in natural hard-water streams, a recirculating artificial stream channel was constructed. This structure has enabled patterns of population change of stream fauna to be observed under partially controlled physical and chemical conditions. Initial colonization of the substratum by invertebrates and subsequent succession was studied along with depth distribution and growth and production studies of invertebrates.
