Eight reproductive rams with no prior reproductive disease were distributed into three groups of infection with T. gondii: GI, 3 rams, 2.0 x 10(5) P strain oocysts; GII, 3 rams, 1.0 x 10(6) RH strain tachyzoites; GIII, 2 control rams. Clinical parameters were measured and serological evaluations (IIF) were performed. Presence of the parasite in the semen was investigated by PCR and bioassay techniques. The rams presented clinical alterations (hyperthermia and apathy) related to toxoplasmosis in both groups infected with Toxoplasma gondii. All the inoculated rams responded to antigenic stimulus, producing antibodies against T. gondii from postinoculation day 5 onwards. In ovine groups I and II, the greatest titers observed were 1 : 4096 and 1 : 8192, respectively. In semen samples collected from these two groups, the presence of T. gondii was detected by bioassay and PCR. This coccidian was isolated (bioassay and PCR) in tissue pools (testicles, epididymis, seminal vesicle, and prostrate) from two rams infected presenting oocysts and in one presenting tachyzoites.

This study aimed to verify the influence of adjuvants on the droplet spectrum of an air induction nozzle. The experiment used nine spray solutions, one including only water and eight containing adjuvants: Nimbus® (mineral oil), Óleo vegetal Nortox (vegetal oil), Li-700® (a mixture of lecithin and propionic acid), Agral® (nonyl phenoxy poly ethanol), In-Tec® (nonyl phenol ethoxylate), Antideriva (nonyl phenol ethoxylate), Silwet® L-77 Ag (copolymer polyester and silicon) and TA 35 (sodium lauryl ether sulfate). A flat fan air induction nozzle Hypro® Guardian Air 110 03 was used for the droplet spectrum evaluation. The study was conducted at the Laboratory for Particle Size Analysis (Lapar), at FCAV/UNESP, Jaboticabal/SP - Brazil. The determination of the droplet spectrum characteristics (Volume Median Diameter/VMD, percentage of droplets smaller than 100 micrometers and span) was carried out by a particle size analyzer by laser diffraction Mastersizer S (Malvern Instruments). For statistical analysis the mean values were compared using Confidence Interval at 95% (CI 95%). The results showed that for the Hypro® GA air induction nozzle the oil based adjuvants (Óleo Vegetal Nortox e Nimbus®) increased the VMD. The percentage of droplets smaller than 100 micrometers was lower for the Agral®, Antideriva, In-Tec® e TA 35, in comparison with the Óleo Vegetal Nortox and Li-700®. The span was higher for the oil based adjuvants (Óleo Vegetal Nortox e Nimbus®) and lower for the TA 35 (sodium lauryl ether sulfate), showing that the TA 35 adjuvant has a potential to improve the quality of the droplet spectrum of the Hypro® GA 11003 nozzle.

Homalometron elongatum is reexamined using heat-killed material that was not subjected to pressure during fixation from Gerres cinereus collected from San Juan Harbor, Puerto Rico, U.S.A. The new material is compared with some paratype specimens and differs by having a much less variable forebody length, and a median rather than submedian genital pore. Tegumental spines reportedly cover the anterior end of the body but we observed tegumental spines covering the entire body surface in both the paratype and new material. Homalometron lesliorum n. sp. is described from Eucinostomus currani from the Pacific coasts of Costa Rica and Nicaragua. The new species has three pairs of oral papillae surrounding the mouth and thus resembles three other congeners: H. elongatum, Homalometron carapevae, and Homalometron papilliferum. Homalometron lesliorum n. sp. is distinguished from the three species by having the anterior extent of the vitelline follicles at or above the base of the ventral sucker, compared with posterior to the ventral sucker at the level of the seminal vesicle (H. elongatum) or further posterior at the posterior margin of the ovary (H. carapevae and H. papilliferum). The four species are further differentiated from one another by sucker width ratio, tegumental spine size and distribution, egg size, host preference, and biogeography. Comparison of nuclear ribosomal DNA (3' end of 18S, internal transcribed spacer [ITS]1, ITS2, and 5' end of 28S) between H. elongatum and H. lesliorum n. sp. revealed one variable base (n = 162) at the 3' end of 18S, 12 variable bases (n = 476) at ITS1, 10 variable bases (n = 310) at ITS2, and 11 variable bases (n = 1,325) at the 5' end fragment of 28S. Nuclear ribosomal DNA from Homalometron pallidum and Homalometron armatum are included for further comparison with H. elongatum and H. lesliorum n. sp.

Contents The aim of this study was to determine the effect of temporary inhibition of meiosis using the cyclin-dependent kinase inhibitor butyrolactone I (BLI) on gene expression in bovine oocytes and cumulus cells. Immature bovine cumulusoocyte complexes (COCs) were assigned to groups: (i) Control COCs collected immediately after recovery from the ovary or (ii) after in vitro maturation (IVM) for 24 h, (iii) Inhibited COCs collected 24 h after incubation with 100 mu m BLI or (iv) after meiotic inhibition for 24 h followed by IVM for a further 22 h. For mRNA relative abundance analysis, pools of 10 denuded oocytes and respective cumulus cells were collected. Transcripts related to cell cycle regulation and oocyte competence were evaluated in oocytes and cumulus cells by quantitative real-time PCR (qPCR). Most of the examined transcripts were downregulated (p < 0.05) after IVM in control and inhibited oocytes (19 of 35). Nine transcripts remained stable (p > 0.05) after IVM in control oocytes; only INHBA did not show this pattern in inhibited oocytes. Seven genes were upregulated after IVM in control oocytes (p < 0.05), and only PLAT, RBP1 and INHBB were not upregulated in inhibited oocytes after IVM. In cumulus cells, six genes were upregulated (p < 0.05) after IVM and eight were downregulated (p < 0.05). Cells from inhibited oocytes showed the same pattern of expression regarding maturation profile, but were affected by the temporary meiosis inhibition of the oocyte when the same maturation stages were compared between inhibited and control groups. In conclusion, changes in transcript abundance in oocytes and cumulus cells during maturation in vitro were mostly mirrored after meiotic inhibition followed by maturation.
