The term water stress refers to the effects of low water availability on microbial growth and physiology. Water availability has been proposed as a major constraint for the use of microorganisms in contaminated sites with the purpose of bioremediation. Sphingomonas wittichii RW1 is a bacterium capable of degrading the xenobiotic compounds dibenzofuran and dibenzo-p-dioxin, and has potential to be used for targeted bioremediation. The aim of the current work was to identify genes implicated in water stress in RW1 by means of transposon mutagenesis and mutant growth experiments. Conditions of low water potential were mimicked by adding NaCl to the growth media. Three different mutant selection or separation method were tested which, however recovered different mutants. Recovered transposon mutants with poorer growth under salt-induced water stress carried insertions in genes involved in proline and glutamate biosynthesis, and further in a gene putatively involved in aromatic compound catabolism. Transposon mutants growing poorer on medium with lowered water potential also included ones that had insertions in genes involved in more general functions such as transcriptional regulation, elongation factor, cell division protein, RNA polymerase β or an aconitase.

The integrative and conjugative element ICEclc is a mobile genetic element in Pseudomonas knackmussii B13, and an experimental model for a widely distributed group of elements in Proteobacteria. ICEclc is transferred from specialized transfer competent cells, which arise at a frequency of 3-5% in a population at stationary phase. Very little is known about the different factors that control the transfer frequency of this ICE family. Here we report the discovery of a three-gene operon encoded by ICEclc, which exerts global control on transfer initiation. The operon consists of three consecutive regulatory genes, encoding a TetR-type repressor MfsR, a MarR-type regulator and a LysR-type activator TciR. We show that MfsR autoregulates expression of the operon, whereas TciR is a global activator of ICEclc gene expression, but no clear role was yet found for MarR. Deletion of mfsR increases expression of tciR and marR, causing the proportion of transfer competent cells to reach almost 100% and transfer frequencies to approach 1 per donor. mfsR deletion also caused a two orders of magnitude loss in population viability, individual cell growth arrest and loss of ICEclc. This indicates that autoregulation is an important feature maintaining ICE transfer but avoiding fitness loss. Bioinformatic analysis showed that the mfsR-marR-tciR operon is unique for ICEclc and a few highly related ICE, whereas tciR orthologues occur more widely in a large variety of suspected ICE among Proteobacteria.

Uusia keinoja kullan erottamiseksi malmista on etsitty viimeaikoina taloudellisista ja ympäristöllisistä syistä kautta maailman. Syanidointimenetelmä on hallinnut kullan talteenottoayli sata vuotta. Menetelmässä kulta liuotetaan laimeaan syanidiliuokseen, jostase otetaan talteen aktiivihiilen avulla. Syanidin käyttöä pyritään kuitenkin vähentämään sen myrkyllisyyden takia. Lisäksi nykyään louhitaan enenemässä määrin malmia, josta on hankala rikastaa kulta kustannustehokkaasti syanidia käyttäen. Kullan talteenottoa syanidi- ja kloridiliuoksesta on selvitetty kirjallisuuden avulla. Kullan kemiaan liuotuksen aikana on perehdytty ennen kullan talteenottoa aktiivihiilellä. Aktiivihiilen elinkaari kullan adsorbenttinaon käsitelty valmistuksesta hylkäämiseen mukaan lukien hiilen myrkyttyminen prosessissa ja regenerointi. Aktiivi-hiilen käyttäytyminen syanidi- ja kloridiliuoksessa on selvitetty erikseen. Kullan talteenottoa kuparipitoisista malmeista on käsitelty. Kullan talteenottoa kloridiliuoksesta aktiivihiiltä käyttäen on tutkittu kokeellisesti. Pääasialliset tutkimuskohteet ovat adsorption kinetiikka, kuparin vaikutus adsorptioon, aktiivihiilen vaikutus adsorptioonja adsorboituneiden metallien strippaus hiilestä selektiivisesti. Hapettavan stippauksen vaikutus kullan desorptioon hiilestä on tutkittu yksityiskohtaisesti. Kullan erotusmenetelmät kuparimalmista aktiivihiiltä käyttäen on selvitetty diplomityön tulosten pohjalta. Diplomityön keskeisten tulosten perusteella kulta ei välttämättä saostu aktiivihiilen pinnalle kloridiliuoksesta. Havainto varmistettiin ladattujen hiilipartikkelien pyyhkäisyelektronimikroskooppikuvista ja partikkeleille tehdyistä mikroanalyyseistä. Kullan pelkistyminen metalliseksi kullaksi aktiivihiilessä voitaneen välttää käyttämällä erittäin hapettavia olosuhteita. Aktiivihiili ilmeisesti hapettuu näissä olosuhteissa, mikä mahdollistaa kultakloridin adsorboitumisen hiileen.

Both isolates were examined by PFGE and SDS-PAGE, which led to the identification of a new insertion sequence, ISPa133. This element was shown to have distinct chromosomal locations in each of the original isolates that appeared to explain the differences in imipenem susceptibilty. In strain PA110514, ISPa133 is located 56 nucleotides upstream of the translational start codon, which has no effect on expression of the porin OprD. However, in strain PA116136 ISPa133 it is located in front of nucleotide 696 and, by interrupting the coding region, causes a loss of OprD expression, thus conferring imipenem resistance. In vitro experiments mimicking the natural conditions of selective pressure yielded imipenem-resistant strains in which ISPa133 similarly interrupted oprD. The Munc13 gene family encodes molecules located at the synaptic active zone that regulate the reliability of synapses to encode information over a wide range of frequencies in response to action potentials. In the CNS, proteins of the Munc13 family are critical in regulating neurotransmitter release and synaptic plasticity. Although Munc13-1 is essential for synaptic transmission, it is paradoxical that Munc13-2 and Munc13-3 are functionally dispensable at some synapses, although their loss in other synapses leads to increases in frequency-dependent facilitation. We addressed this issue at the calyx of Held synapse, a giant glutamatergic synapse that we found to express all these Munc13 isoforms. We studied their roles in the regulation of synaptic transmission and their impact on the reliability of information transfer. Through detailed electrophysiological analyses of Munc13-2, Munc13-3, and Munc13-2-3 knock-out and wild-type mice, we report that the combined loss of Munc13-2 and Munc13-3 led to an increase in the rate of calcium-dependent recovery and a change in kinetics of release of the readily releasable pool. Furthermore, viral-mediated overexpression of a dominant-negative form of Munc13-1 at the calyx demonstrated that these effects are Munc13-1 dependent. Quantitative immunohistochemistry using Munc13-fluorescent protein knock-in mice revealed that Munc13-1 is the most highly expressed Munc13 isoform at the calyx and the only one highly colocalized with Bassoon at the active zone. Based on these data, we conclude that Munc13-2 and Munc13-3 isoforms limit the ability of Munc13-1 to regulate calcium-dependent replenishment of readily releasable pool and slow pool to fast pool conversion in central synapses.

In this paper, a new two-dimensional shear deformable beam element based on the absolute nodal coordinate formulation is proposed. The nonlinear elastic forces of the beam element are obtained using a continuum mechanics approach without employing a local element coordinate system. In this study, linear polynomials are used to interpolate both the transverse and longitudinal components of the displacement. This is different from other absolute nodal-coordinate-based beam elements where cubic polynomials are used in the longitudinal direction. The accompanying defects of the phenomenon known as shear locking are avoided through the adoption of selective integration within the numerical integration method. The proposed element is verified using several numerical examples, and the results are compared to analytical solutions and the results for an existing shear deformable beam element. It is shown that by using the proposed element, accurate linear and nonlinear static deformations, as well as realistic dynamic behavior, can be achieved with a smaller computational effort than by using existing shear deformable two-dimensional beam elements.

The fiber recovery process is an essential part of the modern paper mill. It creates the basisfor mill's internal recirculation of the most important raw materials ¿ water and fiber. It is normally also a start point for further treatment of wastewater and if it works efficiently, it offers excellent basis to minimize effluents. This dissertation offers two different approaches to the subject. Firstly a novel save-all disc filter feeding system is developed and presented. This so-called precoat method is tested both in the laboratory and full-scale conditions. In laboratory scale it beats the traditional one clearly, when low freeness pulps are used as a sweetener stock. The full-scale application needs still some development work before it can be implemented to the paper mills. Secondly, the operationenvironment of save-all disc filter is studied mostly in laboratory conditions.The focus of this study is in cases, where low-freeness pulps are used as a sweetener stock of save-all filter. The effects of CSF-value, pressure drop, suspension consistency and retention chemicals to the quantity and quality of the filtrate was studied. Also the filtration resistance of the low freeness pulps was one studied.

El present treball proposa l’addició d’un 10% de sal al pinso d’engreix d’11 vedells de la raça Parda de Montaña, com a sistema per limitar-ne el consum sense haver de fer ús de mà d’obra complementària ni de sistemes mecanitzats que en controlin la quantitat de consum. Aquest % de sal, pretén aconseguir una reducció en la palatabilitat del pinso de consum dels animals, aconseguint així uns menors valors d’ingestió de concentrat per un major consum de farratge. Aquesta relació quantitativa té com a objectiu, aconseguir la introducció d’aquests animals en el marc de la producció ecològica, que estableix una ràtio de consum de farratge vs concentrat de 60 : 40. El grup d’11 vedells, prèviament esmentat, és comparat amb una altre grup, també d’11 vedells i de la mateixa raça i edat, que són alimentats amb el mateix fenc d’alfals, però amb pinso convencional i no amb el complementat amb sal
