Geographically isolated wetlands, those entirely surrounded by uplands, provide numerous ecological functions, some of which are dependent on the degree to which they are hydrologically connected to nearby waters. There is a growing need for field-validated, landscape-scale approaches for classifying wetlands based on their expected degree of connectivity with stream networks. During the 2015 water year, flow duration was recorded in non-perennial streams (n = 23) connecting forested wetlands and nearby perennial streams on the Delmarva Peninsula (Maryland, USA). Field and GIS-derived landscape metrics (indicators of catchment, wetland, non-perennial stream, and soil characteristics) were assessed as predictors of wetland-stream connectivity (duration, seasonal onset and offset dates). Connection duration was most strongly correlated with non-perennial stream geomorphology and wetland characteristics. A final GIS-based stepwise regression model (adj-R2 = 0.74, p < 0.0001) described wetland-stream connection duration as a function of catchment area, wetland area and number, and soil available water storage.

Este estudo objetivou estimar a quantidade de sedimento que foi carreada da microbacia do Ribeirão Água Fria (Palmas, TO) entre fevereiro de 1998 e janeiro de 1999. Almejou-se ainda investigar as relações entre a produção de sedimento e alguns fatores antrópicos e ambientais. O método de Colby foi a técnica empregada no estudo. A produção específica de sedimento e o coeficiente de remoção de sedimentos foram parâmetros também investigados neste trabalho. Foi estimada uma quantidade de 138.619 toneladas de sedimento produzido e a produção específica de sedimentos foi estimada como sendo 827 t km-2 ano-1, enquanto que o coeficiente de remoção de sedimentos foi 6,2%. A fração suspensa foi a predominante durante quase todo o período de estudo.

There has been considerable interest in developing shape-changing soft materials for potential applications in drug delivery, microfluidics and biosensing. These shape- changing materials are inspired by the morphological changes exhibited by plants in nature, such as the Venus flytrap. One specific class of shape-change is that from a flat sheet to a folded structure (e.g., a tube). Such “self-folding” materials are usually composed of polymer hydrogels, and these typically fold in response to external stimuli such as pH and temperature. In order to develop these hydrogels for the previously described applications, it is necessary to expand the range of triggers. The focus of this dissertation is the advancement of shape-changing polymer hydrogels that are sensitive to uncommon cues such as specific biomolecules (enzymes), the substrates for such enzymes, or specific multivalent cations. First, we describe a hybrid gel that responds to the presence of low concentrations of a class of enzymes known as matrix metalloproteinases (MMPs). The hybrid gel was created by utilizing photolithographic techniques to combine two or more gels with distinct chemical composition into the same material. Certain portions of the hybrid gel are composed of a biopolymer derivative with crosslinkable groups. The hybrid gel is flat in water; however, in the presence of MMPs, the regions containing the biopolymer are degraded and the flat sheet folds to form a 3D structure. We demonstrate that hydrogels with different patterns can transform into different 3D structures such as tubes, helices and pancakes. Furthermore, this shape change can be made to occur at physiological concentrations of enzymes. Next, we report a gel with two layers that undergoes a shape change in the presence of glucose. The enzyme glucose oxidase (GOx) is immobilized in one of the layers. GOx catalyzes the conversion of glucose to gluconic acid. The production of gluconic acid decreases the local pH. The decrease in local pH causes one of the layers to swell. As a result, the flat sheet folds to form a tube. The tube unfolds to form a flat sheet when it is transferred to a solution with no glucose present. Therefore, this biomolecule- triggered shape transformation is reversible, meaning the glucose sensing gel is reusable. Furthermore, this shape change only occurs in the presence of glucose and it does not occur in the presence of other small sugars such as fructose. In our final study, we report the shape change of a gel with two layers in the presence of multivalent ions such as Ca2+ and Sr2+. The gel consists of a passive layer and an active layer. The passive layer is composed of dimethylyacrylamide (DMAA), which does not interact with multivalent ions. The active layer consists of DMAA and the biopolymer alginate. In the presence of Ca2+ ions, the alginate chains crosslink and the active layer shrinks. As a result, the gel converts from a flat sheet to a folded tube. What is particularly unusual is the direction of folding. In most cases, when flat rectangular gels fold, they do so about their short-side. However, our gels typically fold about their long-side. We hypothesize that non-homogeneous swelling determines the folding axis.

Environmental samples were collected at three surface water sites between 5/21/2011 and 11/21/2014 along the Upper Boulder River near Boulder Montana. The sites were located at Bernice (within the mountain block), near the High Ore drainage (near the mountain block/basin transition), and at the USGS Gauging Station near Boulder, Montana (within the basin). The parameters measured in the field were SC, temperature, and alkalinity with occasional pH measurements. We collected samples for anions, cations, and stable isotopes in the catchment. We identified endmembers by sampling snow and groundwater and determined from available data an approximate endmember for rain, snow, and groundwater. We used temporal and spatial variations of water chemistry and isotopes to generate an endmember mixing model. Groundwater was found to always be an important contributor to river flow and could increase by nearly an order of magnitude during large snowmelt events. This resulted in groundwater comprising ~20% of total river flow during snowmelt at all sites. At peak snowmelt we observed that near surface water contributions to the river were from a mixture of rain and snow. Soil water, though not sampled, was hypothesized to be an important part of the hydrologic story. If so, the endmember contributions determined in this study may be different. Groundwater may have the highest variation depending on water chemistry of shallow soil water.

ABSTRACT: This study aimed to estimate the probability of climatological water deficit in an experimental watershed in the Cerrado biome, located in the central plateau of Brazil. For that, it was used a time series of 31 years (1982?2012). The probable climatological water deficit was calculated by the difference between rainfall and probable reference evapotranspiration, on a decennial scale. The reference evapotranspiration (ET0) was estimated by the standard FAO-56 Penman-Monteith method. To estimate water deficit, it was used gamma distribution, time series of rainfall and reference evapotranspiration. The adherence of the estimated probabilities to the observed data was verified by the Kolmogorov-Smirnov nonparametric test, with significance level (a-0.05), which presented a good adjustment to the distribution models. It was observed a climatological water deficit, in greater or lesser intensity, between the annual decennials 2 and 32.

Graphene and carbon nanotube nanocomposite (GCN) was synthesised and applied in gene transfection of pIRES plasmid conjugated with green fluorescent protein (GFP) in NIH-3T3 and NG97 cell lines. The tips of the multi-walled carbon nanotubes (MWCNTs) were exfoliated by oxygen plasma etching, which is also known to attach oxygen content groups on the MWCNT surfaces, changing their hydrophobicity. The nanocomposite was characterised by high resolution scanning electron microscopy; energy-dispersive X-ray, Fourier transform infrared and Raman spectroscopies, as well as zeta potential and particle size analyses using dynamic light scattering. BET adsorption isotherms showed the GCN to have an effective surface area of 38.5m(2)/g. The GCN and pIRES plasmid conjugated with the GFP gene, forming π-stacking when dispersed in water by magnetic stirring, resulting in a helical wrap. The measured zeta potential confirmed that the plasmid was connected to the nanocomposite. The NIH-3T3 and NG97 cell lines could phagocytize this wrap. The gene transfection was characterised by fluorescent protein produced in the cells and pictured by fluorescent microscopy. Before application, we studied GCN cell viability in NIH-3T3 and NG97 line cells using both MTT and Neutral Red uptake assays. Our results suggest that GCN has moderate stability behaviour as colloid solution and has great potential as a gene carrier agent in non-viral based therapy, with low cytotoxicity and good transfection efficiency.
