ABSTRACT. The Electronic and Microelectronic Design Group of Universidad Politécnica de Madrid -GDEM- is in charge, between other issues, of improving the embedded system’s consumption. It is in this place and about this subject where the exposed project has taken shape and development. According to an article from de online magazine Revista de Electronica Embebida, an embedded system is “the one controlled by a microprocessor and, thanks to the programing that it includes, it carries out a specific function what it has been designed for, being integrated in it the most necessary elements for realizing the already said function”. The because of studying this subject, answers that each time there is more presence of the embedded system in our daily life. This is due to the tendency of providing “intelligence” to all what can make our lives easier. We can find this kind of systems in factories, offices, security systems, watchers, mobile phones, washing machines, ovens, hoovers and, definitely, in all kind of machines what we can think of. Despite its large vantages, there are still some inconveniences. Nowadays, the most important problem is the autonomy of the system itself when machines that have to be supplied by batteries –making easier the portability-. Therefore, this project is going after a save capacity of energy for the system as well as being able to take decisions in order to duplicate batteries’ autonomy. Smartphones are a clear example. They are a very successful product but the autonomy is just one day. This is not practical for users, at all, if they have to travel, to work or to do any activity that involves a huge use of the phone without a socket nearby. That is why the need of investigating a way to improve this situation. This project is working on this line, creating an automatic system that will generate the currents for verifying the acquisition system that, with the beagle board, will help taking decisions regarding the energy’s consumption. To carry out this system, we need different tools that we can find in the laboratory of the group previously mentioned, like power supply Agilent and the Beagle Board – as main working tools –. The main goal is the simulation of some signals that, after a conversion process, will represent de consumption of each of the parts in the embedded generic system. Therefore, the system will be a testing ground that simulate the consumption, once sent to the processor, to be processed and so the microprocessor system might take some decision.

ABSTRACT Today’s chemical, pharmaceutical and clinical industries generate pollutants that affect the soils and surface and ground waters of our country. Among these, phenol is a common organic pollutant that is extremely harmful to living organisms, even at low concentrations. Several protocols exist to minimize the effects of pollutants, but most are costly procedures or even generate other pollutants. The adsorption of hazardous materials onto clays is perhaps the most used, efficient and cost-saving method available. However, organic compounds such as phenol are difficult to adsorb and this has led to the development of materials known as organoclays, which are much better at remediating organic compounds. Organoclays are clays that have been modified using a surfactant. In turn, surfactants are organic molecules that confer a cationic rather than anionic charge to the clay surface, improving it’s capacity to adsorb phenol. For this doctorate project, kaolin was selected as an adsorbent material for the removal of phenol given its easy sourcing and relatively low cost. The materials investigated were kaolin sand, a directly extracted material, and washed kaolin, which is the byproduct of the kaolin sand washing process. The main difference between the materials is their quartz content, which is much higher in the kaolin sands. To generate a product from kaolin or kaolin sand capable of retaining organic pollutants such as phenol, both materials were subjected to several heat, chemical and/or mechanical treatments to give rise to compounds with a greater reactive surface area. To this end the two starting materials underwent heating at 750ºC for 3 h, grinding to the point of amorphization and/or activation with HCl 6M or NaOH 5M for 3 h at 90ºC. These treatments gave rise to 18 processed samples, which were characterized in terms of their morphological, mineralogical, and physical-chemical properties. The behaviour of these new materials was examined in terms of their pH, cation exchange capacity (CEC), water adsorption capacity (WCU and WCC), particle size distribution (PSD), specific surface area (SBET), and their X-ray diffraction (XRD), Fourier transform infrared spectroscopy (FTIR), thermal (DTG, DTA) and scanning and transmission electron microscopy (SEM and TEM) properties. The changes conferred by the different treatments were also examined in terms of Al and Si losses. Results for the materials derived from kaolin sands and washed kaolin were similar, with differences attributable to the kaolinite contents of the samples. The treatments heat and grinding produced amorphous materials, which when subjected to acid or alkali activation gave rise to Si and Al losses. This in turn led to a change in physico- chemical, mineralogical and morphological properties. Some samples showed a highly increased surface area and CEC. Further, among the changes produced, alkali treatment led to the generation of zeolites in different proportions depending on the sample. To produce the organoclays, the 18 samples were surfacted with hexadecyltrimethylammonium (HDTMA) 20 mM for 24 h at 60ºC. This surfactant concentration is higher than the CEC of each sample. The amount of HDTMA adsorbed onto the surface of each sample determined by FTIR varied according to treatment. A principle components analysis (PCA) was performed to examine correlations between sample Si/Al ratios and surfactant adsorption capacity. In addition, to explore HDTMA adsorption by the samples, DTG and DTA data were fitted to Freundllich and Langmuir models. The mechanisms of surfactant attachment to the sample surface were also addressed. The amount of phenol adsorbed by the resultant organoclays was determined when exposed to different phenol concentrations: 50, 500, 1000, 2000, and 2500 mg/l for 24 h. The quantity of adsorbed pollutant was estimated by gas chromatography and the data fitted to the models of Freundllich and Langmuir. Results indicate that the phenol adsorption capacity of the surfacted samples is dependent on the sample’s characteristics. In general, the washed kaolin samples showed a greater phenol adsorption capacity than the kaolon sands and alkali activation improved this capacity in the two types of sample. In conclusion, the treatments used gave rise to adsorbent materials with varying properties. Kaolin sands showed high quartz levels and their use in some industries is not recommended due to the costs involved in their washing and purification. The applications suggested by the data obtained for some of the kaolin sand samples indicate the added value of this industrial by-product. Nowadays, soil pollution with oil in Mexico is a very common issue due mainly to both oil spill and oil activities. For example, mainly in the southeast area of Mexico, polluted soil contains high concentrations of hydrocarbons, up to 450,000 mg/kg. For these reasons, enviromental authorities have the concern in developing economically feasible and efficient technology that allow the elimination of these type of contaminants. The sanitation in sites can be achieved through several procedures such as physical, chemical and biological methods (or a combination among them). The choice of a method depends on the nature and physical state of the contaminant, the concentration, type of soil, physical space available, time consumption and financial resources. Before any technological application, a diagnostic of the polluted soil is necessary in order to know the type, concentration and distribution of contaminants as well as the soil volume, climatic conditions and physical features of the place (access routes and services, among others). In this thesis, surfactants has been proposed as a technique to increase the mobility of hydrophobic-organic contaminants (HOCs), e.g. total hydrocarbons of petroleum, polychlorinated biphenyls, benzene, toluene, xylenes, explosives, chlorophenols, pesticides, among others, and, hence, to facilitate degradation. Since surfactants reduce the water surface tension, they are molecules comprised of hydrophilic polar groups and long-hydrophobic carbon chains. Surfactant’s polar groups form hydrogen bonding with water molecules while carbon chains, i.e. hydrocarbon chains, have hydrophobic interactios. In aqueous solutions, surfactants form self-organised spherical structures called micelles. The solubilisation of contaminants is carried out only when the micellar phase is formed. This is obtained when the surfactant concentration is higher than the crítical micelle concentration (CMC), i.e. above the concentration where the surfactant monomer begins to self-aggregate. The diesel efficiency desorption by surfactants depends on their nature, the dose use, the contaminant hydrophobicity, the surfactant-soil interaction and the contact time with surfactant soil. However, the best desorption is not always related with the best either mobility or solubility efficiency since high concentration of surfactant can inhibit mobilisation. According to information of the Federal Bureau of Environmental Protection (PROFEPA), up today, there is not any restauration of diesel-polluted sites using the washing-soil technique. Due to the above, there exist the necessity of employing the waching-soil technique ex situ. More specifically, a sample soil from the oil-refinery of “18 de marzo” in Mexico city was extracted and a surfactant solution with deionised water was put in contact with the diesel contaminated soil by means of cylindrical waching columns in order to remove the contaminant. The surfactants employed in this work were sodium lauryl sulfate, sodium lauryl ether sulfate and Glucopon AV-100 at different concentrations of 0.5 to 4 [g/L], obtaining a efficiency of 80 % with this last surfactant. The washing of diesel-polluted soil using surfactants is a technology which requires a deeper study of some variables such as the type of surfactant, concentration, washing time, difusión phenomena, desorption, thermodynamic properties, among others. These parameters determine the succes or failure of the employed technique.

ABSTRACT The present doctoral thesis, “Thermal utilisation of waste coal for electricity generation by deployment of efficient combustion and gasification technologies with minimum environmental impact”, develops an innovative waste-to-energy concept of waste coals produced during coal mining and washing. The energy system is at a dilemma, we are witnessing a shift in the energy paradigm and specifically in the field of electricity generation. A change in the generation and electrical consumption is foreseen. An increased health consciousness is forcing the containment and elimination of pollutants that are generated by the use of fossil fuels in the way that is being done. Increasing concern about climate change and to contain the rise of global temperature by 2°C by the end of this century, is promoting the development and final implementation of technology to control and reduce the CO2 emission. Electricity generation in a sustainable manner is becoming an obligation. This concept materialised in generating electricity while protecting the environment and deployment of natural resources at a competitive cost, considering the development of society and people´s benefit. Currently, coal is the main source of energy employ to generate electricity, and its use represents the most cost competitive form of energy to increase the standard of living of any group or society. Moreover, coal will keep playing a key role in the global electricity generation mix, maintaining a significant presence and being extracting in large amounts. However, coal production implies the production of waste, termed waste coal or culm in Pennsylvania anthracite extraction, produced during coal mining and coal washing activities. During the last decades, the potential use of waste coal has been studied, and currently, in a limited amount, waste coal is used in roads construction, embankments and fillings, and to produce some construction materials. This doctoral thesis evaluates the waste to energy of waste coals and assesses its potential use to generate electricity, implementing available technology to minimise the environment impact. Additionally, it pursues the significant advantage that presents sulphur content in waste coal to produce sulphuric acid (H2SO4) as a byproduct of the waste-to-energy process, a chemical compound highly demanded by the fertiliser industry and many applications in other markets. It analyses the characteristics of waste coal, and assesses the significant parameters and reference values for its use as fuel, being its fuel use for electricity generation very possible. While mining coal is taking place in Spain since the 1700s, it has been evaluated for a more recent period the waste coal available in Spain and the existing legislation that affects its application and deploy to generate electricity in the country. For the evaluation period has been calculated that may be available more than 68 million tons of waste coal that can be waste-toenergy. The potential available technology to deploy waste coal as fuel has been evaluated and assessed. After considering this, the doctoral thesis proposes four innovative alternatives of facility configuration, three of them based on a combustion process and one in a gasification process. After evaluating the four configurations for its technological, innovative and economic interest, the conceptual analysis of one of alternatives, based on a combustion process, takes place. The proposed alternative facility developed has a capacity of 65 MW, using as fuel a mixture of coal and waste coal 80/20 by weight. The facility comprises technology to remove 99.8% SO2 present in the flue gas and more than 99% of the particles. The facility includes a unit capable of producing 18.5 t/h of H2SO4, and another capture facility, removing 60% of CO2 present in the flue gas stream, producing 48 tCO2/h. The net capacity of the power station is 49.7 MW. The facility unitary cost of investment is 3,685 €/kW.

Superoxide-mediated clastogenesis is characteristic for various chronic inflammatory diseases with autoimmune reactions and probably plays a role in radiation-induced clastogenesis and in the congenital breakage syndromes. It is consistently prevented by exogenous superoxide dismutase (SOD), but not by heat-inactivated SOD, indicating that the anticlastogenic effect is related to the catalytic function of the enzyme. Increased superoxide production by activated monocytes/macrophages is followed by release of more long-lived metabolites, so-called clastogenic factors, which contain lipid peroxidation products, unusual nucleotides of inosine, and cytokines such as tumor necrosis factor α. Since these components are not only clastogenic, but can stimulate further superoxide production by monocytes and neutrophils, the genotoxic effects are self-sustaining. It is shown here that anticlastogenic effects of exogenous SOD are preserved despite extensive washing of the cells and removal of all extracellular SOD. Using flow cytometry and confocal laser microscopy, rapid adherence of the fluorescently labeled enzyme to the cell surface could be observed with slow uptake into the cell during the following hours. The degree of labeling was concentration and time dependent. It was most important for monocytes, compared with lymphocytes, neutrophils, and fibroblasts. The cytochrome c assay showed significantly diminished O2− production by monocytes, pretreated with SOD and washed thereafter. The preferential and rapid binding of SOD to monocytes may be of importance not only for the superoxide-mediated genotoxic effects, described above, but also from a therapeutic standpoint. It can explain the observation that beneficial effects of injected SOD lasted for weeks and months despite rapid clearance of the enzyme from the blood stream according to pharmacodynamic studies.

Δ9-Desaturase is a key enzyme in the synthesis of desaturated fatty acyl-CoAs. Desaturase is an integral membrane protein induced in the endoplasmic reticulum by dietary manipulations and then rapidly degraded. The proteolytic machinery that specifically degrades desaturase and other short-lived proteins in the endoplasmic reticulum has not been identified. As the first step in identifying cellular factors involved in the degradation of desaturase, liver subcellular fractions of rats that had undergone induction of this enzyme were examined. In livers from induced animals, desaturase was present in the microsomal, nuclear (P-1), and subcellular fractions (P-2). Incubation of desaturase containing fractions at physiological pH and temperature led to the complete disappearance of the enzyme. Washing microsomes with a buffer containing high salt decreased desaturase degradation activity. N-terminal sequence analysis of desaturase freshly isolated from the P-1 fraction without incubation indicated the absence of three residues from the N terminus, but the mobility of this desaturase preparation on SDS-PAGE was identical to the microsomal desaturase, which contains a masked N terminus under similar purification procedures. Addition of concentrated cytosol or the high-salt wash fraction did not enhance the desaturase degradation in the washed microsomes. Extensive degradation of desaturase in the high-salt washed microsomes could be restored by supplementation of the membranes with the lipid and protein components essential for the reconstituted desaturase catalytic activity. Lysosomotrophic agents leupeptin and pepstatin A were ineffective in inhibiting desaturase degradation. The calpain inhibitor, N-acetyl-leucyl-leucyl-methional, or the proteosome inhibitor, Streptomyces metabolite, lactacystin, did not inhibit the degradation of desaturase in the microsomal or the P-1 and P-2 fractions. These results show that the selective degradation of desaturase is likely to be independent of the lysosomal and the proteosome systems. The reconstitution of complete degradation of desaturase in the high-salt–washed microsomes by the components essential for its catalytic activity reflects that the degradation of this enzyme may depend on a specific orientation of desaturase and intramembranous interactions between desaturase and the responsible protease.

The extent of in vitro formation of the borate-dimeric-rhamnogalacturonan II (RG-II) complex was stimulated by Ca2+. The complex formed in the presence of Ca2+ was more stable than that without Ca2+. A naturally occurring boron (B)-RG-II complex isolated from radish (Raphanus sativus L. cv Aokubi-daikon) root contained equimolar amounts of Ca2+ and B. Removal of the Ca2+ by trans-1,2-diaminocyclohexane-N,N,N′,N′-tetraacetic acid induced cleavage of the complex into monomeric RG-II. These data suggest that Ca2+ is a normal component of the B-RG-II complex. Washing the crude cell walls of radish roots with a 1.5% (w/v) sodium dodecyl sulfate solution, pH 6.5, released 98% of the tissue Ca2+ but only 13% of the B and 22% of the pectic polysaccharides. The remaining Ca2+ was associated with RG-II. Extraction of the sodium dodecyl sulfate-washed cell walls with 50 mm trans-1,2-diaminocyclohexane-N,N,N′,N′-tetraacetic acid, pH 6.5, removed the remaining Ca2+, 78% of B, and 49% of pectic polysaccharides. These results suggest that not only Ca2+ but also borate and Ca2+ cross-linking in the RG-II region retain so-called chelator-soluble pectic polysaccharides in cell walls.

Cultured cells of rose (Rosa damascena) treated with an elicitor derived from Phytophthora spp. and suspension-cultured cells of French bean (Phaseolus vulgaris) treated with an elicitor derived from the cell walls of Colletotrichum lindemuthianum both produced H2O2. It has been hypothesized that in rose cells H2O2 is produced by a plasma membrane NAD(P)H oxidase (superoxide synthase), whereas in bean cells H2O2 is derived directly from cell wall peroxidases following extracellular alkalinization and the appearance of a reductant. In the rose/Phytophthora spp. system treated with N,N-diethyldithiocarbamate, superoxide was detected by a N,N′-dimethyl-9,9′-biacridium dinitrate-dependent chemiluminescence; in contrast, in the bean/C. lindemuthianum system, no superoxide was detected, with or without N,N-diethyldithiocarbamate. When rose cells were washed free of medium (containing cell wall peroxidase) and then treated with Phytophthora spp. elicitor, they accumulated a higher maximum concentration of H2O2 than when treated without the washing procedure. In contrast, a washing treatment reduced the H2O2 accumulated by French bean cells treated with C. lindemuthianum elicitor. Rose cells produced reductant capable of stimulating horseradish (Armoracia lapathifolia) peroxidase to form H2O2 but did not have a peroxidase capable of forming H2O2 in the presence of reductant. Rose and French bean cells thus appear to be responding by different mechanisms to generate the oxidative burst.

Amino acid analysis of internal sequences of purified NADH-hexacyanoferrate(III) oxidoreductase (NFORase), obtained from highly purified plasma membranes (PM) of spinach (Spinacia oleracea L.) leaves, showed 90 to 100% homology to internal amino acid sequences of monodehydroascorbate (MDA) reductases (EC 1.6.5.4) from three different plant species. Specificity, kinetics, inhibitor sensitivity, and cross-reactivity with anti-MDA reductase antibodies were all consistent with this identification. The right-side-out PM vesicles were subjected to consecutive salt washing and detergent (polyoxyethylene 20 dodecylether and 3-[(3-cholamido-propyl)-dimethylammonio]-1-propane sulfonate [CHAPS]) treatments, and the fractions were analyzed for NFORase and MDA reductase activities. Similar results were obtained when the 300 mm sucrose in the homogenization buffer and in all steps of the salt-washing and detergent treatments had been replaced by 150 mm KCl to mimic the conditions in the cytoplasm. We conclude that (a) MDA reductase is strongly associated with the inner (cytoplasmic) surface of the PM under in vivo conditions and requires washing with 1.0 m KCl or CHAPS treatment for removal, (b) the PM-bound MDA reductase activity is responsible for the majority of PM NFORase activity, and (c) there is another redox enzyme(s) in the spinach leaf PM that cannot be released from the PM by salt-washing and/or CHAPS treatment. The PM-associated MDA reductase may have a role in reduction of ascorbate in both the cytosol and the apoplast.

Pancreatic proteases in the duodenum inhibit the release of cholecystokinin (CCK) and thus exert feedback control of pancreatic exocrine secretion. Exclusion of proteases from the duodenum either by the diversion of bile-pancreatic juice or by the addition of protease inhibitors stimulates exocrine pancreatic secretion. The mechanism by which pancreatic proteases in the duodenum regulate CCK secretion is unknown. In this study, we isolated a trypsin-sensitive peptide that is secreted intraduodenally, releases CCK, and stimulates pancreatic enzyme secretion in rats. This peptide was found to be identical to the porcine diazepam binding inhibitor by peptide sequencing and mass spectrometry analysis. Intraduodenal infusion of 200 ng of synthetic porcine diazepam binding inhibitor1-86 in rats significantly stimulated pancreatic amylase output. Infusion of the CCK antagonist MK-329 completely blocked the diazepam binding inhibitor-stimulated amylase secretion. Similarly, diazepam binding inhibitor33-52 [corrected] also stimulated CCK release and pancreatic secretion in a dose-dependent manner although it was 100 times less potent than the whole peptide. Using a perfusion system containing isolated mucosal cells from the proximal intestine of rats, porcine diazepam binding inhibitor 10(-12) M) dose dependently stimulated CCK secretion. In separate studies, it was demonstrated that luminal secretion of the diazepam binding inhibitor immunoreactivity (7.5 X 10(11) M) could be detected in rat's intestinal washing following the diversion of bile-pancreatic juice. The secretion of this peptide was inhibited by atropine. In conclusion, we have isolated and characterized a CCK-releasing peptide that has a sequence identical to the porcine diazepam binding inhibitor from pig intestinal mucosa and that stimulates CCK release when administered intraduodenally in rat. This peptide may mediate feedback regulation of pancreatic enzyme secretion.

Recent evidence indicates that polyunsaturated long-chain fatty acids (PUFAs) prevent lethal ischemia-induced cardiac arrhythmias in animals and probably in humans. To increase understanding of the mechanism(s) of this phenomenon, the effects of PUFAs on Na+ currents were assessed by the whole-cell patch-clamp technique in cultured neonatal rat ventricular myocytes. Extracellular application of the free 5,8,11,14,17-eicosapentaenoic acid (EPA) produced a concentration-dependent suppression of ventricular, voltage-activated Na+ currents (INa). After cardiac myocytes were treated with 5 or 10 microM EPA, the peak INa (elicited by a single-step voltage change with pulses from -80 to -30 mV) was decreased by 51% +/- 8% (P < 0.01; n = 10) and 64% +/- 5% (P < 0.001; n = 21), respectively, within 2 min. Likewise, the same concentrations of 4,7,10,16,19-docosahexaenoic acid produced the same inhibition of INa. By contrast, 5 and 10 microM arachidonic acid (AA) caused less inhibition of INa, but both n - 6 and n - 3 PUFAs inhibited INa significantly. A monounsaturated fatty acid and a saturated fatty acid did not. After washing out EPA, INa returned to the control level. Raising the concentration of EPA to 40 microM completely blocked INa. The IC50 of EPA was 4.8 microM. The inhibition of this Na+ channel was found to be dose and time, but not use dependent. Also, the EPA-induced inhibition of INa was voltage dependent, since 10 microM EPA produced 83% +/- 7% and 29% +/- 5% inhibition of INa elicited by pulses from -80 to -30 mV and from -150 to -30 mV, respectively, in single-step voltage changes. A concentration of 10 microM EPA shifted the steady-state inactivation curve of INa by -19 +/- 3 mV (n = 7; P < 0.01). These effects of PUFAs on INa may be important for their antiarrhythmic effect in vivo.

Because previous studies showed that polyunsaturated fatty acids can reduce the contraction rate of spontaneously beating heart cells and have antiarrhythmic effects, we examined the effects of the fatty acids on the electrophysiology of the cardiac cycle in isolated neonatal rat cardiac myocytes. Exposure of cardiomyocytes to 10 microM eicosapentaenoic acid for 2-5 min markedly increased the strength of the depolarizing current required to elicit an action potential (from 18.0 +/- 2.4 pA to 26.8 +/- 2.7 pA, P < 0.01) and the cycle length of excitability (from 525 ms to 1225 ms, delta = 700 +/- 212, P < 0.05). These changes were due to an increase in the threshold for action potential (from -52 mV to -43 mV, delta = 9 +/- 3, P < 0.05) and a more negative resting membrane potential (from -52 mV to -57 mV, delta = 5 +/- 1, P < 0.05). There was a progressive prolongation of intervals between spontaneous action potentials and a slowed rate of phase 4 depolarization. Other polyunsaturated fatty acids--including docosahexaenoic acid, linolenic acid, linoleic acid, arachidonic acid, and its nonmetabolizable analog eicosatetraynoic acid, but neither the monounsaturated oleic acid nor the saturated stearic acid--had similar effects. The effects of the fatty acids could be reversed by washing with fatty acid-free bovine serum albumin. These results show that free polyunsaturated fatty acids can reduce membrane electrical excitability of heart cells and provide an electrophysiological basis for the antiarrhythmic effects of these fatty acids.
