Resumo:
Although genome sequencing of microbial pathogens has shed light on the evolution of virulence, the drivers of the gain and loss of genes and of pathogenicity islands (gene clusters), which contribute to the emergence of new disease outbreaks, are unclear. Recent experiments with the bean pathogen Pseudomonas syringae pv. phaseolicola illustrate how exposure to resistance mechanisms acts as the driving force for genome reorganization. Here we argue that the antimicrobial conditions generated by host defences can accelerate the generation of genome rearrangements that provide selective advantages to the invading microbe. Similar exposure to environmental stress outside the host could also drive the horizontal gene transfer that has led to the evolution of pathogenicity towards both animals and plants.

A major challenge in deciphering the development of left-right asymmetry in vertebrates is uncovering how subtle early differences between the left and right sides are translated into robust differences in gene expression. Recently, Raya and colleagues suggested that asymmetric localization of extracellular calcium ions at the node could differentially modulate Notch signalling, resulting in asymmetric expression of the signalling molecule Nodal on the left side of the node. In this article, I examine the implications of this finding and explore their relevance to the evolution of asymmetry in vertebrates and invertebrates.

The immense social and economic impact of bacterial pathogens, from drug-resistant infections in hospitals to the devastation of agricultural resources, has resulted in major investment to understand the causes and conse- quences of pathogen evolution. Recent genome se- quencing projects have provided insight into the evolution of bacterial genome structures; revealing the impact of mobile DNA on genome restructuring and pathogenicity. Sequencing of multiple genomes of relat- ed strains has enabled the delineation of pathogen evo- lution and facilitated the tracking of bacterial pathogens globally. Other recent theoretical and empirical studies have shown that pathogen evolution is significantly influenced by ecological factors, such as the distribution of hosts within the environment and the effects of co- infection. We suggest that the time is ripe for experi- mentalists to use genomics in conjunction with evolu- tionary ecology experiments to further understanding of how bacterial pathogens evolve.

The growing human population will require a significant increase in agricultural production. This challenge is made more difficult by the fact that changes in the climatic and environmental conditions under which crops are grown have resulted in the appearance of new diseases, whereas genetic changes within the pathogen have resulted in the loss of previously effective sources of resistance. To help meet this challenge, advanced genetic and statistical methods of analysis have been used to identify new resistance genes through global screens, and studies of plant-pathogen interactions have been undertaken to uncover the mechanisms by which disease resistance is achieved. The informed deployment of major, race-specific and partial, race-nonspecific resistance, either by conventional breeding or transgenic approaches, will enable the production of crop varieties with effective resistance without impacting on other agronomically important crop traits. Here, we review these recent advances and progress towards the ultimate goal of developing disease-resistant crops.

The past decade has seen significant increases in combustion-generated ambient particles, which contain a nanosized fraction (less than 100 nm), and even greater increases have occurred in engineered nanoparticles (NPs) propelled by the booming nanotechnology industry. Although inhalation of these particulates has become a public health concern, human health effects and mechanisms of action for NPs are not well understood. Focusing on the human airway smooth muscle cell, here we show that the cellular mechanical function is altered by particulate exposure in a manner that is dependent upon particle material, size and dose. We used Alamar Blue assay to measure cell viability and optical magnetic twisting cytometry to measure cell stiffness and agonist-induced contractility. The eight particle species fell into four categories, based on their respective effect on cell viability and on mechanical function. Cell viability was impaired and cell contractility was decreased by (i) zinc oxide (40-100 nm and less than 44 mu m) and copper(II) oxide (less than 50 nm); cell contractility was decreased by (ii) fluorescent polystyrene spheres (40 nm), increased by (iii) welding fumes and unchanged by (iv) diesel exhaust particles, titanium dioxide (25 nm) and copper(II) oxide (less than 5 mu m), although in none of these cases was cell viability impaired. Treatment with hydrogen peroxide up to 500 mu M did not alter viability or cell mechanics, suggesting that the particle effects are unlikely to be mediated by particle-generated reactive oxygen species. Our results highlight the susceptibility of cellular mechanical function to particulate exposures and suggest that direct exposure of the airway smooth muscle cells to particulates may initiate or aggravate respiratory diseases.

Stainless steels are well known to be prone to cold welding and material transfer in sliding contacts and therefore difficult to cold form unless certain precautions as discussed in this paper are taken. In the present study different combinations of tool steels/stainless steels/lubricants has been evaluated with respect to their galling resistance using pin-on-disc testing. The results show that a high galling resistance is favored by a high stainless steel sheet hardness and a blasted stainless steel sheet surface topography. The effect of type of lubricant was found to be more complex. For example, the chlorinated lubricants failed to prevent metal-to-metal contact on a brushed sheet surface but succeeded on a blasted sheet surface of the same stainless steel material. This is believed to be due to a protective tribofilm which is able to form on the blasted surface, but not on the brushed surface.

O maior objetivo deste trabalho foi estudar os efeitos da composição dos fluxos para arco submerso sobre algumas importantes características de um metal de solda ferrítico, como composição química, microestrutura, propriedades mecânicas e geometria do cordão. Para realizar tal pesquisa, vinte oito fluxos aglomerados foram elaborados de compostos de pureza comercial e utilizados em soldagens ao aorco submerso, mantendo constante todas as demais condições de soldagem. Houve uma notável influência da composição química e do índice de basicidade dos fluxos sobre os níveis de oxig~enio, silício e manganês do metal de solda. Um modelo termodinâmico, baseado no conceito de potencial de oxigênio, foi proposto para explicar as reações gás-meta-escória ocorrendo durante a soldagem. Este modelo mostrou-se usável para as reações entre silício, oxigênio e carbono, permitindo um melhor entendimento das mesmas. A composição química do metal de solda pôde ser relacionada a sua microestrutura, através de metalografia quantitativa. Foi observado que oxigênio, silício, manganês e titânio têm grande influência sobre ela. A fase mais tenaz encontrada foi a ferrita acicular. A respeito da geometria do cordão, a sílica mostrou o efeito mais forte, com os outros compostos influenciando somente o acabamento do cordão soldado.

A intervenção vocacional aparece,inevitavelmente, associada ao trabalho do psicólogo escolar, constituindo uma referência importante da sua actividade profissional. Ainda que esta forma de intervenção se realize, diversas vezes, em grupo deve contemplar a individualidade e as características de grupos específicos. Com a realização deste trabalho pretendemos apresentar o programa de orientação vocacional “Para lá do 9º ano!” e reflectir sobre a sua aplicação, no 3º período do ano lectivo, junto de um grupo de alunos a frequentar o 9º ano de escolaridade numa Escola EB2,3. Este grupo é constituído por 14 alunos, que num estudo anterior, realizado quando frequentavam o 6º ano de escolaridade, foram sinalizados como alunos mais capazes, tomando critérios de avaliação ao nível da criatividade (TPCT), da cognição (BPR-5/6), da inteligência geral (TIG-1) e do rendimento académico (notas escolares). Seguimos uma abordagem mais qualitativa ao analisarmos as suas expectativas e crenças face ao desenvolvimento escolar e profissional, e ao caracterizarmos as suas preferências e o processo de tomado de decisão. A generalidade destes alunos pretende ingressar no Ensino Superior, optando por inscrever-se no Ensino Secundário em Cursos Científico-Humanísticos, mais precisamente em Ciências e Tecnologias. Terminamos, numa perspectiva de desenvolvimento de carreira, com algumas reflexões em relação ao percurso educativo destes alunos e da organização do sistema educativo.

A identificação dos alunos sobredotados e talentosos implica a ponderação e a adopção de critérios e procedimentos de avaliação que permitam caracterizar com validade os alunos sinalizados. Nesse sentido, a necessidade de contemplar várias fontes de informação pressupõe uma certa complexidade no processo de identificação, sendo que o recurso a uma identificação multi-referencial permite o alargamento do número de alunos que poderá beneficiar de medidas educativas. O principal objectivo desta comunicação é analisar e reflectir sobre os critérios utilizados para a sinalização e a avaliação de alunos com altas habilidades. Procuramos comparar os desempenhos ao nível do raciocínio cognitivo (BPR-5/6 e BPR-7/9) e académico (classificações escolares) de um grupo de alunos identificado com altas habilidades, quando frequentavam o 2º ciclo (5º e 6º ano) [conjugando critérios de avaliação ao nível da criatividade (TPCT), da cognição (BPR-5/6),da inteligência geral (TIG-1) e do rendimento académico (notas escolares)], com os seus desempenhos em anos lectivos posteriores, quando frequentam o 3º ciclo (8º e 9ºano). Além disso, consideramos nesta análise a percepção dos Directores de Turma ao nível da sinalização dos alunos (BISAST-NP e IDT). Genericamente, os resultados sugerem que nem todos os alunos identificados apresentam um elevado rendimento académico nos anos escolares posteriores e que os professores revelam alguma dificuldade na sinalização de alunos com altas habilidades, sendo os resultados dessa sinalização discrepantes face aos resultados nas provas psicológicas.

The failure of materials is always an unwelcome event for several reasons: human lives are put in danger, economic losses, and interference in the availability of products and services. Although the causes of failures and behaviour of materials can be known, the prevention of such a condition is difficult to be guaranteed. Among the failures, wear abrasion by the low voltage is the kind of failure that occurs in more equipment and parts industry. The Plants Sucroalcooleiras suffer significant losses because of such attrition, this fact that motivated their choice for the development of this work. For both, were considered failures in the swing hammers desfibradores stopped soon after the exchange provided in accordance with tonnage of cane processed, then were analyzed by the level of wear testing of rubber wheel defined by the standard ASTM G65-91.The failures were classified as to the origin of the cause and mechanism, moreover, were prepared with samples of welding procedures according to ASME code, sec. IX as well, using the technique of thermal spraying to analyze the performance of these materials produced in laboratories, and compares them with the solder used in the plant. It was observed that the bodies-of-proof prepared by the procedure described as welding, and the thermal spraying the results of losing weight have been minimized significantly compared to the preparations in the plant. This is because the use of techniques more appropriate and more controlled conditions of the parameters of welding. As for the thermal spraying, this technique has presented a satisfactory result, but requires the use of these coatings in the best condition for real affirmation of the results

The main objective is to analyze the abrasive wear resistance to the low stress of the elements that make up the organs of road machinery that are exposed directly to contact with abrasives. These samples were analyzed after these elements are coated superficially by the process of welding electrode coated with (SAER) and the manual process of coating type LVOF thermal spraying. As well, is to provide suggestions for a better recovery and return of these elements, which are reducing costs and avoiding downtime in the fronts of service. The samples were made from a substrate of carbon ABNT 1045 tempered steel, following the same specifications and composition of metals and alloys of constituents was followed the standard governing the dimensions of these samples and in accordance with the corresponding size. The results were evaluated by testing the hardness, abrasion resistance to wear by the low stress and the loss of volume involving the microstructure of coatings analyzed

This work a studied the high energy milling effect in microstructure and magnetic properties of the WC-10wt.%Co composite. The composite powders were prepared by mechanical mixed and milled at 2 hours, 100 hours, 200 hours and 300 hours in planetary milling. After this process the composite were compacted in stainless steel die with cylindrical county of 10 mm of diameter, at pressure 200 Mpa and sintered in a resistive furnace in argon atmosphere at 1400 oC for 5 min. The sintered composite were cutted, inlaid, sandpapered, and polished. The microestrutural parameters of the composite was analyzed by X-ray diffraction, scanning electronic microscopy, optical microscopy, hardness, magnetic propriety and Rietveld method analyze. The results shows, with milling time increase the particle size decrease, it possibility minor temperature of sintering. The increase of milling time caused allotropic transformation in cobalt phase and cold welding between particles. The cold welding caused the formation of the particle composite. The X-ray diffraction pattern of composite powders shows the WC peaks intensity decrease with the milling time increase. The X-ray diffraction pattern of the composite sintered samples shows the other phases. The magnetic measurements detected a significant increase in the coercitive field and a decrease in the saturation magnetization with milling time increase. The increase coercitive field it was also verified with decrease grain size with milling time increase. For the composite powders the increase coercitive field it was verified with particle size reduction and saturation magnetization variation is relate with the variation of free cobalt. The Rietveld method analyze shows at milling time increase the mean crystalline size of WC, and Co-cfc phases in composite sintered sample are higher than in composite powders. The mean crystallite size of Co-hc phase in composite powders is higher than in composite sintered sample. The mean lattice strains of WC, Co-hc and Co-cfc phases in composite powders are higher than in composite sintered samples. The cells parameters of the composite powder decrease at milling time increase this effect came from the particle size reduction at milling time increase. In sintered composite the cells parameters is constant with milling time increase

In this work, was studied the formation of a composite of the refractory metal niobium with copper, through the process of high-energy milling and liquid phase sintering. The HEM can be used to synthesize composite powders with high homogeneity and fine size particle distribution. It may also produce the solid solubility in immiscible systems such as Nb-Cu, or extend the solubility of systems with limited solubility. Therefore, in the immiscible system Cu-Nb, the high-energy milling was successfully used to obtain the composite powder particles. Initially, the formation of composite particles during the HEM and the effect of preparation technique on the microstructure of the material was evaluated. Four loads of Nb and Cu powders containing 20%wt Cu were synthesized by MAE in a planetary type ball mill under different periods of grinding. The influence of grinding time on the metal particles is evaluated during the process by the withdrawal of samples at intermediate times of milling. After compaction under different forces, the samples were sintered in a vacuum furnace. The liquid phase sintering of these samples prepared by HEM produced a homogeneous and fine grained. The composite particles forming the sintered samples are the addition of a hard phase (Nb) with a high melting point, and a ductile phase (Cu) with low melting point and high thermal and electrical conductivities. Based on these properties, the Nb-Cu system is a potential material for many applications, such as electrical contacts, welding electrodes, coils for generating high magnetic fields, heat sinks and microwave absorbers, which are coupled to electronic devices. The characterization techniques used in this study, were laser granulometry, used to evaluate the homogeneity and particle size, and the X-ray diffraction, in the phase identification and to analyze the crystalline structure of the powders during milling. The morphology and dispersion of the phases in the composite powder particles, as well the microstructures of the sintered samples, were observed by scanning electron microscopy (SEM). Subsequently, the sintered samples are evaluated for density and densification. And finally, they were characterized by techniques of measuring the electrical conductivity and microhardness, whose properties are analyzed as a function of the parameters for obtaining the composite
