Objective: To explore the non-pharmacological correlates of the perceived effectiveness of antidepressants (ADs), thereby enhancing understanding of the mechanisms involved in recovery from depression while taking ADs. Method: An online survey was completed by 1781 New Zealand adults who had taken ADs in the previous 5 years. Results: All 18 psychosocial variables measured were associated with depression reduction, and 16 with improved quality of life (QoL). Logistic regression models revealed that the quality of the relationship with the prescriber was related to both depression reduction and improved QoL. In addition, depression reduction was related to younger age, higher income, being fully informed about ADs by the prescriber, fewer social causal beliefs for depression and not having lost a loved one in the 2 months prior to prescription. Furthermore, both outcome measures were positively related to belief in ‘chemical’ rather than ‘placebo’ effects. Conclusion: There are multiple non-pharmacological processes involved in recovery while taking ADs. Enhancing them, for example focusing on the prescriber–patient relationship and giving more information, may enhance recovery rates, with or without ADs.

No livro IV das Geórgicas, afamado poema didáctico de Virgílio, é descrita a vida das abelhas, apontadas como modelo de organização social e destacando-se a sua dedicação ao bem comum. O tópico tornou-se num dos paradigmas literários da Antiguidade, tendo sido repetidamente retomado, por exemplo nas obras de poetas didácticos modernos, quer europeus, quer do Novo Mundo. Referimo-nos especificamente ao poema Praedium rusticum (Toulose, 1730), do escritor inaciano francês Jacques Vanière, que aborda a sociedade das abelhas (Apes) no livro XIV, e a Rusticatio mexicana (Modena, 1781; Bolonha, 1782), do jesuíta guatemalteco Rafael Landívar, cujo livro VI descreve a sociedade dos castores (Fibri), procurando emular a obra Vanière. Na sociedade colectiva de Vanière não existe riqueza privada e cada abelha se dedica à tarefa que prefere. Em vez da ganância e do ódio prevalecem a generosidade e o amor. Na res publica dos castores, por seu lado, tal como a apresenta Landívar, existe divisão das tarefas a desempenhar e um forte sentido de comunidade. Os castores são felizes no seu trabalho, altamente inteligentes, nobres, amantes da paz e igualmente libertos de sentimentos como o ódio, a vingança e outras preocupações mais profundas. Esta comunicação pretende avaliar o modo como são apresentadas nestes textos concepções políticas e sociais que, embora procurem adaptar e integrar os ideais do Iluminismo, seguem na esteira do modelo literário virgiliano.

Conspiracy theories can be treated as both rational narratives of the world as well as outcomes of underlying maladaptive traits. Here, we examined associations between belief in conspiracy theories and individual differences in personality disorders. An Internet-based sample (N=259) completed measures of belief in conspiracy theories and the 25 facets of the Personality Inventory for DSM-5 (PID-5). Preliminary analyses showed no significant differences in belief in conspiracy theories across participant sex, ethnicity, and education. Regression analyses showed that the PID-5 facets of Unusual Beliefs and Experiences and, to a lesser extent, Suspiciousness, significantly predicted belief in conspiracy theories. These findings highlight a role for maladaptive personality traits in understanding belief in conspiracy theories, but require further investigation.

This paper is a contribution for the assessment and comparison of magnet properties based on magnetic field characteristics particularly concerning the magnetic induction uniformity in the air gaps. For this aim, a solver was developed and implemented to determine the magnetic field of a magnetic core to be used in Fast Field Cycling (FFC) Nuclear Magnetic Resonance (NMR) relaxometry. The electromagnetic field computation is based on a 2D finite-element method (FEM) using both the scalar and the vector potential formulation. Results for the magnetic field lines and the magnetic induction vector in the air gap are presented. The target magnetic induction is 0.2 T, which is a typical requirement of the FFC NMR technique, which can be achieved with a magnetic core based on permanent magnets or coils. In addition, this application requires high magnetic induction uniformity. To achieve this goal, a solution including superconducting pieces is analyzed. Results are compared with a different FEM program.

STRIPPING is a software application developed for the automatic design of a randomly packing column where the transfer of volatile organic compounds (VOCs) from water to air can be performed and to simulate it’s behaviour in a steady-state. This software completely purges any need of experimental work for the selection of diameter of the column, and allows a choice, a priori, of the most convenient hydraulic regime for this type of operation. It also allows the operator to choose the model used for the calculation of some parameters, namely between the Eckert/Robbins model and the Billet model for estimating the pressure drop of the gaseous phase, and between the Billet and Onda/Djebbar’s models for the mass transfer. Illustrations of the graphical interface offered are presented.

O tolueno tem uma vasta aplicação industrial e apresenta alguns riscos para a saúde humana. Legislação comunitária, recentemente publicada, exige que se cumpra a médio prazo padrões mínimos de qualidade apontando como limite máximo de concentração de tolueno na água destinada a consumo humano o valor de 1 ppm. O processo de arrastamento por ar em colunas com enchimento desordenado é uma das técnicas de remoção disponíveis, embora ainda pouco implementada no nosso país. Neste trabalho simulou-se, laboratorial e numericamente, a remoção do tolueno da água através desta tecnologia sob condições operatórias pré-estabelecidas. A simulação à escala laboratorial foi executada numa instalação piloto constituída por uma coluna de vidro acrílico com 0.137metros de diâmetro interno preenchida desordenadamente com anéis Raflux de polietileno com 15milímetros de calibre numa altura de 1.10 metros. A experimentação efectuada abrangeu ensaios realizados quer em regime permanente quer em regime transitório. As simulações numéricas do comportamento de uma coluna com idênticas características, em estado estacionário e em estado transiente, foram efectuadas com “software” específico concebido para MatLab. Este software permite a descrição do regime hidrodinâmico e da transferência de massa deste processo de acordo com os modelos de Robbins [Robbins, 1991] e Onda [Onda, 1968], respectivamente, inseridos num modelo global fenomenológico traduzido num sistema de equações às derivadas parciais de 1ª ordem, do tipo hiperbólico, com uma variável espacial axial e uma variável temporal. A resolução deste modelo é efectuada através de algoritmos e estratégias específicas nomeadamente a implementação numérica dos métodos explícito de Euler para resolução do sistema de ED0s que traduz o estado estacionário primitivo e de diferenças finitas com esquema explícito associado a manipulação de matrizes esparsas para resolução do sistema de EDPs que traduz o estado dinâmico. A concordância entre os resultados obtidos na teoria e na prática, pese embora as dificuldades encontradas no controlo analítico das amostras, permite concluir que o modelo proposto possui um rigor fenomenológico suficiente e que o modelo numérico associado possibilita um elevado grau de exactidão e de fiabilidade. A aplicação desenvolvida pode constituir uma importante ferramenta de apoio à análise dinâmica desta operação unitária à escala industrial, reduzindo o esforço experimental e os respectivos custos associados. Constitui igualmente a base para uma implementação de um potencial sistema de controlo por actuação antecipada com base num modelo que reveste a forma de um sistema de parâmetros distribuídos.

OBJECTIVE: Tuberculosis (TB) is highly prevalent among HIV-infected people, including those receiving combination antiretroviral therapy (cART), necessitating a well tolerated and efficacious TB vaccine for these populations. We evaluated the safety and immunogenicity of the candidate TB vaccine M72/AS01 in adults with well controlled HIV infection on cART. DESIGN: A randomized, observer-blind, controlled trial (NCT00707967). METHODS: HIV-infected adults on cART in Switzerland were randomized 3 : 1 : 1 to receive two doses, 1 month apart, of M72/AS01, AS01 or 0.9% physiological saline (N = 22, N = 8 and N = 7, respectively) and were followed up to 6 months postdose 2 (D210). Individuals with CD4⁺ cell counts below 200 cells/μl were excluded. Adverse events (AEs) including HIV-specific and laboratory safety parameters were recorded. Cell-mediated (ICS) and humoral (ELISA) responses were evaluated before vaccination, 1 month after each dose (D30, D60) and D210. RESULTS: Thirty-seven individuals [interquartile range (IQR) CD4⁺ cell counts at screening: 438-872 cells/μl; undetectable HIV-1 viremia] were enrolled; 73% of individuals reported previous BCG vaccination, 97.3% tested negative for the QuantiFERON-TB assay. For M72/AS01 recipients, no vaccine-related serious AEs or cART-regimen adjustments were recorded, and there were no clinically relevant effects on laboratory safety parameters, HIV-1 viral loads or CD4⁺ cell counts. M72/AS01 was immunogenic, inducing persistent and polyfunctional M72-specific CD4⁺ T-cell responses [medians 0.70% (IQR 0.37-1.07) at D60] and 0.42% (0.24-0.61) at D210, predominantly CD40L⁺IL-2⁺TNF-α⁺, CD40L⁺IL-2⁺ and CD40L⁺IL-2⁺TNF-α⁺IFN-γ⁺]. All M72/AS01 vaccines were seropositive for anti-M72 IgG after second vaccination until study end. CONCLUSION: M72/AS01 was clinically well tolerated and immunogenic in this population, supporting further clinical evaluation in HIV-infected individuals in TB-endemic settings.

Circulating levels of adiponectin, a hormone produced predominantly by adipocytes, are highly heritable and are inversely associated with type 2 diabetes mellitus (T2D) and other metabolic traits. We conducted a meta-analysis of genome-wide association studies in 39,883 individuals of European ancestry to identify genes associated with metabolic disease. We identified 8 novel loci associated with adiponectin levels and confirmed 2 previously reported loci (P = 4.5×10(-8)-1.2×10(-43)). Using a novel method to combine data across ethnicities (N = 4,232 African Americans, N = 1,776 Asians, and N = 29,347 Europeans), we identified two additional novel loci. Expression analyses of 436 human adipocyte samples revealed that mRNA levels of 18 genes at candidate regions were associated with adiponectin concentrations after accounting for multiple testing (p<3×10(-4)). We next developed a multi-SNP genotypic risk score to test the association of adiponectin decreasing risk alleles on metabolic traits and diseases using consortia-level meta-analytic data. This risk score was associated with increased risk of T2D (p = 4.3×10(-3), n = 22,044), increased triglycerides (p = 2.6×10(-14), n = 93,440), increased waist-to-hip ratio (p = 1.8×10(-5), n = 77,167), increased glucose two hours post oral glucose tolerance testing (p = 4.4×10(-3), n = 15,234), increased fasting insulin (p = 0.015, n = 48,238), but with lower in HDL-cholesterol concentrations (p = 4.5×10(-13), n = 96,748) and decreased BMI (p = 1.4×10(-4), n = 121,335). These findings identify novel genetic determinants of adiponectin levels, which, taken together, influence risk of T2D and markers of insulin resistance.

OBJECTIVES: The goal of this study was to determine whether subclinical thyroid dysfunction was associated with incident heart failure (HF) and echocardiogram abnormalities. BACKGROUND: Subclinical hypothyroidism and hyperthyroidism have been associated with cardiac dysfunction. However, long-term data on the risk of HF are limited. METHODS: We studied 3,044 adults>or=65 years of age who initially were free of HF in the Cardiovascular Health Study. We compared adjudicated HF events over a mean 12-year follow-up and changes in cardiac function over the course of 5 years among euthyroid participants, those with subclinical hypothyroidism (subdivided by thyroid-stimulating hormone [TSH] levels: 4.5 to 9.9, >or=10.0 mU/l), and those with subclinical hyperthyroidism. RESULTS: Over the course of 12 years, 736 participants developed HF events. Participants with TSH>or=10.0 mU/l had a greater incidence of HF compared with euthyroid participants (41.7 vs. 22.9 per 1,000 person years, p=0.01; adjusted hazard ratio: 1.88; 95% confidence interval: 1.05 to 3.34). Baseline peak E velocity, which is an echocardiographic measurement of diastolic function associated with incident HF in the CHS cohort, was greater in those patients with TSH>or=10.0 mU/l compared with euthyroid participants (0.80 m/s vs. 0.72 m/s, p=0.002). Over the course of 5 years, left ventricular mass increased among those with TSH>or=10.0 mU/l, but other echocardiographic measurements were unchanged. Those patients with TSH 4.5 to 9.9 mU/l or with subclinical hyperthyroidism had no increase in risk of HF. CONCLUSIONS: Compared with euthyroid older adults, those adults with TSH>or=10.0 mU/l have a moderately increased risk of HF and alterations in cardiac function but not older adults with TSH<10.0 mU/l. Clinical trials should assess whether the risk of HF might be ameliorated by thyroxine replacement in individuals with TSH>or=10.0 mU/l.
