Rising CO2 levels in the oceans are predicted to have serious consequences for many marine taxa. Recent studies suggest that non-genetic parental effects may reduce the impact of high CO2 on the growth, survival and routine metabolic rate of marine fishes, but whether the parental environment mitigates behavioural and sensory impairment associated with high CO2 remains unknown. Here, we tested the acute effects of elevated CO2 on the escape responses of juvenile fish and whether such effects were altered by exposure of parents to increased CO2 (transgenerational acclimation). Elevated CO2 negatively affected the reactivity and locomotor performance of juvenile fish, but parental exposure to high CO2 reduced the effects in some traits, indicating the potential for acclimation of behavioural impairment across generations. However, acclimation was not complete in some traits, and absent in others, suggesting that transgenerational acclimation does not completely compensate the effects of high CO2 on escape responses.

Coral reefs are globally threatened by climate change-related ocean warming and ocean acidification (OA). To date, slow-response mechanisms such as genetic adaptation have been considered the major determinant of coral reef persistence, with little consideration of rapid-response acclimatization mechanisms. These rapid mechanisms such as parental effects that can contribute to trans-generational acclimatization (e.g. epigenetics) have, however, been identified as important contributors to offspring response in other systems. We present the first evidence of parental effects in a cross-generational exposure to temperature and OA in reef-building corals. Here, we exposed adults to high (28.9°C, 805 µatm PCO2) or ambient (26.5°C, 417 µatm PCO2) temperature and OA treatments during the larval brooding period. Exposure to high treatment negatively affected adult performance, but their larvae exhibited size differences and metabolic acclimation when subsequently re-exposed, unlike larvae from parents exposed to ambient conditions. Understanding the innate capacity corals possess to respond to current and future climatic conditions is essential to reef protection and maintenance. Our results identify that parental effects may have an important role through (1) ameliorating the effects of stress through preconditioning and adaptive plasticity, and/or (2) amplifying the negative parental response through latent effects on future life stages. Whether the consequences of parental effects and the potential for trans-generational acclimatization are beneficial or maladaptive, our work identifies a critical need to expand currently proposed climate change outcomes for corals to further assess rapid response mechanisms that include non-genetic inheritance through parental contributions and classical epigenetic mechanisms.

This paper presents an algorithm for generating scale-free networks with adjustable clustering coefficient. The algorithm is based on a random walk procedure combined with a triangle generation scheme which takes into account genetic factors; this way, preferential attachment and clustering control are implemented using only local information. Simulations are presented which support the validity of the scheme, characterizing its tuning capabilities.

As advanced Cloud services are becoming mainstream, the contribution of data centers in the overall power consumption of modern cities is growing dramatically. The average consumption of a single data center is equivalent to the energy consumption of 25.000 households. Modeling the power consumption for these infrastructures is crucial to anticipate the effects of aggressive optimization policies, but accurate and fast power modeling is a complex challenge for high-end servers not yet satisfied by analytical approaches. This work proposes an automatic method, based on Multi-Objective Particle Swarm Optimization, for the identification of power models of enterprise servers in Cloud data centers. Our approach, as opposed to previous procedures, does not only consider the workload consolidation for deriving the power model, but also incorporates other non traditional factors like the static power consumption and its dependence with temperature. Our experimental results shows that we reach slightly better models than classical approaches, but simul- taneously simplifying the power model structure and thus the numbers of sensors needed, which is very promising for a short-term energy prediction. This work, validated with real Cloud applications, broadens the possibilities to derive efficient energy saving techniques for Cloud facilities.

El presente trabajo consistió en el desarrollo de una intervención nutricional a largo plazo llevada a cabo con jugadores profesionales de baloncesto, en función al cumplimiento de las recomendaciones nutricionales, con los siguientes dos objetivos: 1) valorar los cambios que dicha intervención produce sobre las prácticas nutricionales diarias de estos deportistas y 2) conocer la influencia de las modificaciones nutricionales producidas sobre la tasa de percepción del esfuerzo por sesión (RPE-Sesión) y la fatiga, a lo largo de una temporada competitiva, tanto para entrenamientos como partidos oficiales. Los objetivos del estudio se fundamentan en: 1) la numerosa evidencia científica que muestra la inadecuación de los hábitos nutricionales de los jugadores de baloncesto y otros deportistas respecto a las recomendaciones nutricionales; 2) el hecho ampliamente reconocido en la literatura especializada de que una ingesta nutricional óptima permite maximizar el rendimiento deportivo (a nivel físico y cognitivo), promoviendo una rápida recuperación y disminuyendo el riesgo de enfermedades y lesiones deportivas. No obstante, pocos estudios han llevado a cabo una intervención nutricional a largo plazo para mejorar los hábitos alimentarios de los deportistas y ninguno de ellos fue realizado con jugadores de baloncesto; 3) la elevada correlación entre la percepción del esfuerzo (RPE) y variables fisiológicas relacionadas al desarrollo de un ejercicio (por ej.: frecuencia cardíaca, consumo máximo de oxígeno o lactato sanguíneo) y los múltiples estudios que muestran la atenuación de la RPE durante la realización del ejercicio mediante una ingesta puntual de nutrientes, (especialmente de hidratos de carbono) aunque ninguno fue desarrollado en baloncesto; 4) el estudio incipiente de la relación entre la ingesta nutricional y la RPE-Sesión, siendo éste un método validado en baloncesto y otros deportes de equipo como indicador de la carga de trabajo interna, el rendimiento deportivo y la intensidad del ejercicio realizado; 5) el hecho de que la fatiga constituye uno de los principales factores influyentes en la percepción del esfuerzo y puede ser retrasada y/o atenuada mediante la ingesta de carbohidratos, pudiendo disminuir consecuentemente la RPE-Sesión y la carga interna del esfuerzo físico, potenciando el rendimiento deportivo y las adaptaciones inducidas por el entrenamiento; 6) la reducida evidencia acerca del comportamiento de la RPE-Sesión ante la modificación de la ingesta de nutrientes, encontrándose sólo un estudio llevado a cabo en baloncesto y 7) la ausencia de investigaciones acerca de la influencia que puede tener la mejora del patrón nutricional de los jugadores sobre la RPE-Sesión y la fatiga, desconociéndose si la adecuación de los hábitos nutricionales conduce a una disminución de estas variables en el largo plazo para todos los entrenamientos y partidos oficiales a nivel profesional. Por todo esto, este trabajo comienza con una introducción que presenta el marco teórico de la importancia y función de la nutrición en el deporte, así como de las recomendaciones nutricionales actuales a nivel general y para baloncesto. Además, se describen las intervenciones nutricionales llevadas a cabo previamente con otros deportistas y las consecuentes modificaciones sobre el patrón alimentario, coincidiendo este aspecto con el primer objetivo del presente estudio. Posteriormente, se analiza la RPE, la RPE-Sesión y la fatiga, focalizando el estudio en la relación de dichas variables con la carga de trabajo físico, la intensidad del entrenamiento, el rendimiento deportivo y la recuperación post ejercicio. Finalmente, se combinan todos los aspectos mencionados: ingesta nutricional, RPE percepción del esfuerzo y fatiga, con el fin de conocer la situación actual del estudio de la relación entre dichas variables, conformando la base del segundo objetivo de este estudio. Seguidamente, se exponen y fundamentan los objetivos antes mencionados, para dar lugar después a la explicación de la metodología utilizada en el presente estudio. Ésta consistió en un diseño de estudios de caso, aplicándose una intervención nutricional personalizada a tres jugadores de baloncesto profesional (cada jugador = un estudio de caso; n = 1), con el objetivo de adecuar su ingesta nutricional en el largo plazo a las recomendaciones nutricionales. A su vez, se analizó la respuesta individual de cada uno de los casos a dicha intervención para los dos objetivos del estudio. Para ello, cada jugador completó un registro diario de alimentos (7 días; pesada de alimentos) antes, durante y al final de la intervención. Además, los sujetos registraron diariamente a lo largo del estudio la RPE-Sesión y la fatiga en entrenamientos físicos y de balón y en partidos oficiales de liga, controlándose además en forma cuantitativa otras variables influyentes como el estado de ánimo y el sueño. El análisis de los datos consistió en el cálculo de los estadísticos descriptivos para todas las variables, la comparación de la ingesta en los diferentes momentos evaluados con las recomendaciones nutricionales y una comparación de medias no paramétrica entre el período pre intervención y durante la intervención con el test de Wilcoxon (medidas repetidas) para todas las variables. Finalmente, se relacionaron los cambios obtenidos en la ingesta nutricional con la percepción del esfuerzo y la fatiga y la posible influencia del estado de ánimo y el sueño, a través de un estudio correlacional (Tau_b de Kendall). Posteriormente, se presentan los resultados obtenidos y la discusión de los mismos, haciendo referencia a la evidencia científica relacionada que se encuentra publicada hasta el momento, la cual facilitó el análisis de la relación entre RPE-Sesión, fatiga y nutrición a lo largo de una temporada. Los principales hallazgos y su correspondiente análisis, por lo tanto, pueden resumirse en los siguientes: 1) los tres jugadores de baloncesto profesional presentaron inicialmente hábitos nutricionales inadecuados, haciendo evidente la necesidad de un nutricionista deportivo dentro del cuerpo técnico de los equipos profesionales; 2) las principales deficiencias correspondieron a un déficit pronunciado de energía e hidratos de carbono, que fueron reducidas con la intervención nutricional; 3) la ingesta excesiva de grasa total, ácidos grasos saturados, etanol y proteínas que se halló en alguno/s de los casos, también se adecuó a las recomendaciones después de la intervención; 4) la media obtenida durante un período de la temporada para la RPE-Sesión y la fatiga de entrenamientos, podría ser disminuida en un jugador individual mediante el incremento de su ingesta de carbohidratos a largo plazo, siempre que no existan alteraciones psico-emocionales relevantes; 5) el comportamiento de la RPE-Sesión de partidos oficiales no parece estar influido por los factores nutricionales modificados en este estudio, dependiendo más de la variación de elementos externos no controlables, intrínsecos a los partidos de baloncesto profesional. Ante estos resultados, se pudo observar que las diferentes características de los jugadores y las distintas respuestas obtenidas después de la intervención, reforzaron la importancia de utilizar un diseño de estudio de casos para el análisis de los deportistas de élite y, asimismo, de realizar un asesoramiento nutricional personalizado. Del mismo modo, la percepción del esfuerzo y la fatiga de cada jugador evolucionaron de manera diferente después de la intervención nutricional, lo cual podría depender de las diferentes características de los sujetos, a nivel físico, psico-social, emocional y contextual. Por ello, se propone que el control riguroso de las variables cualitativas que parecen influir sobre la RPE y la fatiga a largo plazo, facilitaría la comprensión de los datos y la determinación de factores desconocidos que influyen sobre estas variables. Finalmente, al ser la RPE-Sesión un indicador directo de la carga interna del entrenamiento, es decir, del estrés psico-fisiológico experimentado por el deportista, la posible atenuación de esta variable mediante la adecuación de los hábitos nutricionales, permitiría aplicar las cargas externas de entrenamiento planificadas, con menor estrés interno y mejor recuperación entre sesiones, disminuyendo también la sensación de fatiga, a pesar del avance de la temporada. ABSTRACT This study consisted in a long-term nutritional intervention carried out with professional basketball players according to nutritional recommendations, with the following two main objectives: 1) to evaluate the changes produced by the intervention on daily nutritional practices of these athletes and 2) to determine the influence of long term nutritional intake modifications on the rate of perceived exertion per session (Session-RPE) and fatigue, throughout a competitive season for training as well as competition games. These objectives are based on: 1) much scientific evidence that shows an inadequacy of the nutritional habits of basketball players and other athletes regarding nutritional recommendations; 2) the fact widely recognized in the scientific literature that an optimal nutrition allows to achieve the maximum performance of an athlete (both physically and cognitively), promoting fast recovery and decreasing risks of sports injuries and illnesses. However, only few studies carried out a long term nutritional intervention to improve nutritional practices of athletes and it could not be found any research with basketball players; 3) the high correlation between the rate of perceived exertion (RPE) and physiological variables related to the performance of physical exercise (e.g.: heart rate, maximum consumption of oxygen or blood lactate) and multiple studies showing the attenuation of RPE during exercise due to the intake of certain nutrients (especially carbohydrates), while none of them was developed in basketball; 4) correlation between nutritional intake and Session-RPE has been recently studied for the first time. Session-RPE method has been validated in basketball players and other team sports as an indicator of internal workload, sports performance and exercise intensity; 5) fatigue is considered one of the main influential factor on RPE and sport performance. It has also been observed that carbohydrates intake may delay or mitigate the onset of fatigue and, thus, decrease the perceived exertion and the internal training load, which could improve sports performance and training-induced adaptations; 6) there are few studies evaluating the influence of nutrient intake on Session-RPE and only one of them has been carried out with basketball players. Moreover, it has not been analyzed the possible effects of the adequacy of players’ nutritional habits through a nutritional intervention on Session-RPE and fatigue, variables that could be decreased for all training session and competition games because of an improvement of daily nutritional intake. Therefore, this work begins with an introduction that provides the conceptual framework of this research focused on the key role of nutrition in sport, as well as on the current nutritional recommendations for athletes and specifically for basketball players. In addition, previous nutritional interventions carried out with other athletes are described, as well as consequential modifications on their food pattern, coinciding with the first objective of the present study. Subsequently, RPE, Session-RPE and fatigue are analyzed, with focus on their correlation with physical workload, training intensity, sports performance and recovery. Finally, all the aforementioned aspects (nutritional intake, RPE and fatigue) were combined in order to know the current status of the relation between each other, this being the base for the second objective of this study. Subsequently, the objectives mentioned above are explained, continuing with the explanation of the methodology used in the study. The methodology consisted of a case-study design, carrying out a long term nutritional intervention with three professional basketball players (each player = one case study; n = 1), in order to adapt their nutritional intake to nutritional recommendations. At the same time, the individual response of each player to the intervention was analyzed for the two main objectives of the study. Each player completed a food diary (7 days; weighing food) in three moments: before, during and at the end of the intervention. In addition, the Session-RPE and fatigue were daily recorded throughout the study for all trainings (training with ball and resistance training) and competition games. At the same time, other potentially influential variables such as mood state and sleeping were daily controlled throughout the study. Data analysis consisted in descriptive statistics calculation for all the variables of the study, the comparison between nutritional intake (evaluated at different times) and nutritional recommendations and a non-parametric mean comparison between pre intervention and during intervention periods was made by Wilcoxon test (repeated measurements) for all variables too. Finally, the changes in nutritional intake, mood state and sleeping were correlated with the perceived exertion and fatigue through correctional study (Tau_b de Kendall). After the methodology, the study results and the associated discussion are presented. The discussion is based on the current scientific evidence that contributes to understand the relation between Session-RPE, fatigue and nutrition throughout the competitive season. The main findings and results analysis can be summarized as follows: 1) the three professional basketball players initially had inadequate nutritional habits and this clearly shows the need of a sports nutritionist in the coaching staff of professional teams; (2) the major deficiencies of the three players’ diet corresponded to a pronounced deficit of energy intake and carbohydrates consumption which were reduced with nutritional intervention; (3) the excessive intake of total fat, saturated fatty acids, ethanol and protein found in some cases were also adapted to the recommendations after the intervention; (4) Session-RPE mean and fatigue of a certain period of the competition season, could be decreased in an individual player by increasing his carbohydrates intake in the long term, if there are no relevant psycho-emotional disorders; (5) the behavior of the Session-RPE in competition games does not seem to be influenced by the nutritional factors modified in this study. They seem to depend much more on the variation of external non-controllable factors associated with the professional basketball games. Given these results, the different characteristics of each player and the diverse responses observed after the intervention in each individual for all the variables, reinforced the importance of the use of a case study design for research with elite athletes as well as personalized nutritional counselling. In the same way, the different responses obtained for RPE and fatigue in the long term for each player due to modification of nutritional habits, show that there is a dependence of such variables on the physical, psychosocial, emotional and contextual characteristics of each player. Therefore it is proposed that the rigorous control of the qualitative variables that seem to influence the RPE and fatigue in the long term, may facilitate the understanding of data and the determination of unknown factors that could influence these variables. Finally, because Session-RPE is a direct indicator of the internal load of training (psycho-physiological stress experienced by the athlete), the possible attenuation of Session-RPE through the improvement in nutritional habits, would allow to apply the planned external loads of training with less internal stress and better recovery between sessions, with a decrease in fatigue, despite of the advance of the season.

Studies of mouse models of human cancer have established the existence of multiple tumor modifiers that influence parameters of cancer susceptibility such as tumor multiplicity, tumor size, or the probability of malignant progression. We have carried out an analysis of skin tumor susceptibility in interspecific Mus musculus/Mus spretus hybrid mice and have identified another seven loci showing either significant (six loci) or suggestive (one locus) linkage to tumor susceptibility or resistance. A specific search was carried out for skin tumor modifier loci associated with time of survival after development of a malignant tumor. A combination of resistance alleles at three markers [D6Mit15 (Skts12), D7Mit12 (Skts2), and D17Mit7 (Skts10)], all of which are close to or the same as loci associated with carcinoma incidence and/or papilloma multiplicity, is significantly associated with increased survival of mice with carcinomas, whereas the reverse combination of susceptibility alleles is significantly linked to early mortality caused by rapid carcinoma growth (χ2 = 25.22; P = 5.1 × 10−8). These data indicate that host genetic factors may be used to predict carcinoma growth rate and/or survival of individual backcross mice exposed to the same carcinogenic stimulus and suggest that mouse models may provide an approach to the identification of genetic modifiers of cancer survival in humans.

Hereditary hemochromatosis (HH) is a common disorder of iron metabolism caused by mutation in HFE, a gene encoding an MHC class I-like protein. Clinical studies demonstrate that the severity of iron loading is highly variable among individuals with identical HFE genotypes. To determine whether genetic factors other than Hfe genotype influence the severity of iron loading in the murine model of HH, we bred the disrupted murine Hfe allele onto three different genetically defined mouse strains (AKR, C57BL/6, and C3H), which differ in basal iron status and sensitivity to dietary iron loading. Serum transferrin saturations (percent saturation of serum transferrin with iron), hepatic and splenic iron concentrations, and hepatocellular iron distribution patterns were compared for wild-type (Hfe +/+), heterozygote (Hfe +/−), and knockout (Hfe −/−) mice from each strain. Although the Hfe −/− mice from all three strains demonstrated increased transferrin saturations and liver iron concentrations compared with Hfe +/+ mice, strain differences in severity of iron accumulation were striking. Targeted disruption of the Hfe gene led to hepatic iron levels in Hfe −/− AKR mice that were 2.5 or 3.6 times higher than those of Hfe −/− C3H or Hfe −/− C57BL/6 mice, respectively. The Hfe −/− mice also demonstrated strain-dependent differences in transferrin saturation, with the highest values in AKR mice and the lowest values in C3H mice. These observations demonstrate that heritable factors markedly influence iron homeostasis in response to Hfe disruption. Analysis of mice from crosses between C57BL/6 and AKR mice should allow the mapping and subsequent identification of genes modifying the severity of iron loading in this murine model of HH.

The placenta contains several types of feto-maternal interfaces where zygote-derived cells interact with maternal cells or maternal blood for the promotion of fetal growth and viability. The genetic factors regulating the interactions between different cell types within feto-maternal interfaces and the relative contributions of the maternal and zygotic genomes are poorly understood. Genomic imprinting, the epigenetic process responsible for parental origin-dependent functional differences between homologous chromosomes, has been proposed to contribute to these events. Previous studies showed that mouse conceptuses with an absence of imprinted differences between the two copies of chromosome 12 (upon paternal inheritance of both copies) die late in gestation and have a variety of defects, including placentomegaly. Here we examined the role of chromosome 12 imprinting in these placentae in more detail. We show that the spatial interactions between different cell types within feto-maternal interfaces are defective and identify abnormal behaviors in both zygote-derived and maternal cells that are attributed to the genome of the zygote but not the mother. These include compromised invasion of the maternal decidualized endometrium and the central maternal artery situated within it by zygote-derived trophoblast, abnormalities in the wall of the central maternal artery, and defects within the zygote-derived cellular layer of the labyrinth, which is in direct contact with maternal blood. These findings demonstrate multiple roles for chromosome 12 imprinting in the placenta that have not previously been associated with imprinting effects. They provide insights into the function of imprinting in placental development and have evolutionary and clinical implications.

External (environmental) factors affecting the speciation of birds are better known than the internal (genetic) factors. The opposite is true for several groups of invertebrates, Drosophila being the outstanding example. Ideas about the genetics of speciation in general trace back to Dobzhansky who worked with Drosophila. These ideas are an insufficient guide for reconstructing speciation in birds for two main reasons. First, speciation in birds proceeds with the evolution of behavioral barriers to interbreeding; postmating isolation usually evolves much later, perhaps after gene exchange has all but ceased. As a consequence of the slow evolution of postmating isolating factors the scope for reinforcement of premating isolation is small, whereas the opportunity for introgressive hybridization to influence the evolution of diverging species is large. Second, premating isolation may arise from nongenetic, cultural causes; isolation may be affected partly by song, a trait that is culturally inherited through an imprinting-like process in many, but not all, groups of birds. Thus the genetic basis to the origin of bird species is to be sought in the inheritance of adult traits that are subject to natural and sexual selection. Some of the factors involved in premating isolation (plumage, morphology, and behavior) are under single-gene control, most are under polygenic control. The genetic basis of the origin of postmating isolating factors affecting the early development of embryos (viability) and reproductive physiology (sterility) is almost completely unknown. Bird speciation is facilitated by small population size, involves few genetic changes, and occurs relatively rapidly.

Male infertility, affecting as many as 10% of the adult population, is an extremely prevalent disorder. In most cases, the cause of the condition is unknown, and genetic factors that might affect male fertility, other than some sequences on the Y chromosome, have not been identified. We report here that male mice heterozygous for a targeted mutation of the apolipoprotein B (apo B) gene exhibit severely compromised fertility. Sperm from these mice failed to fertilize eggs both in vivo and in vitro. However, these sperm were able to fertilize eggs once the zona pellucida was removed but displayed persistent abnormal binding to the egg after fertilization. In vitro fertilization-related and other experiments revealed reduced sperm motility, survival time, and sperm count also contributed to the infertility phenotype. Recognition of the infertility phenotype led to the identification of apo B mRNA in the testes and epididymides of normal mice, and these transcripts were substantially reduced in the affected animals. Moreover, when the genomic sequence encoding human apo B was introduced into these animals, normal fertility was restored. These findings suggest that this genetic locus may have an important impact on male fertility and identify a previously unrecognized function for apo B.

Hormonal and genetic factors strongly influence the susceptibility of inbred mice to hepatocarcinogenesis. Female C57BR/cdJ (BR) mice are extremely susceptible to liver tumor induction relative to other strains because they are genetically insensitive to the inhibition of hepatocarcinogenesis by ovarian hormones. To determine the genetic basis for the sensitivity of BR mice relative to resistant C57BL/6J (B6) mice, we treated 12-day-old B6BRF1 x B6 and B6BRF1 x B6BRF1 (F2) animals with N,N-diethylnitrosamine (0.1 micromol/g of body weight) and enumerated liver tumors at 32 weeks of age in males and at 50 weeks in females. Genomic DNA samples from backcross and F2 mice were analyzed for 70 informative simple sequence length polymorphism markers. Genetic markers on chromosome 17 (D17Mit21) and chromosome 1 (D1Mit33) cosegregated with high tumor multiplicity in both sexes. Together, these loci [designated Hcf1 and Hcf2 (Hepatocarcinogenesis in females), respectively] account for virtually all of the difference in sensitivity between BR and B6 mice. The Hcf1 locus accounts for a majority of the higher susceptibility of BR mice of both sexes. Backcross female mice heterozygous at both loci (33 +/- 23 tumors per mouse) and at Hcf1 only (17 +/- 18) were 15- and 8-fold more sensitive, respectively, than mice homozygous for the B6 alleles at Hcf1 and Hcf2 (2.2 +/- 3.9). In backcross male mice, the double heterozygotes (35 +/- 22) and Hcf1 heterozygotes (28 +/- 12) were 5.4- and 4.3-fold more sensitive than mice homozygous for B6 alleles at both loci (6.5 +/- 5.4).

The prevalence of cholesterol gallstones differs among inbred strains of mice fed a diet containing 15% (wt/wt) dairy fat, 1% (wt/wt) cholesterol, and 0.5% (wt/wt) cholic acid. Strains C57L, SWR, and A were notable for a high prevalence of cholelithiasis; strains C57BL/6, C3H, and SJL had an intermediate prevalence; and strains SM, AKR, and DBA/2 exhibited no cholelithiasis after consuming the diet for 18 weeks. Genetic analysis of the difference in gallstone prevalence rates between strains AKR and C57L was carried out by using the AKXL recombinant inbred strain set and (AKR x C57L)F1 x AKR backcross mice. Susceptibility to gallstone formation was found to be a dominant trait determined by at least two genes. A major gene, named Lith1, mapped to mouse chromosome 2. When examined after 6 weeks on the lithogenic diet, the activity of hepatic 3-hydroxy-3-methylglutaryl-CoA reductase (EC 1.1.1.88) was downregulated as expected in the gallstone-resistant strains, AKR and SJL, but this enzyme failed to downregulate in C57L and SWR, the gallstone-susceptible strains. This suggests that regulation of the rate-limiting enzyme in cholesterol biosynthesis may be pivotal in determining the occurrence and severity of cholesterol hypersecretion and hence lithogenicity of gallbladder bile. These studies indicate that genetic factors are critical in determining gallstone formation and that the genetic resources of the mouse model may permit these factors to be identified.

INTRODUÇÃO: A infecção por HIV-1 é um grave problema de saúde pública causando elevada taxa de morbidade e mortalidade. Entretanto, alguns indivíduos são considerados resistentes à infecção por HIV-1, mesmo após repetidas exposições ao vírus. Vários fatores imunológicos e genéticos podem estar associados a resistência à infecção, como ativação de componentes da imunidade inata e também devido ao baixo perfil de ativação das células T. É possível que nos indivíduos expostos e não infectados por HIV-1 (ENI) ocorra uma importante atuação das células T secretoras de IL-17 e IL-22, e também as células T reguladoras, pois são necessárias para a manutenção e homeostase das mucosas associadas ao intestino (GALT). OBJETIVO: Avaliar o fenótipo e a função de células TCD4+ e TCD8+ em casais sorodiscordante ao HIV-1, compostos por indivíduos ENI e os parceiros infectados por HIV-1. MÉTODOS: Os casais sorodiscordantes ao HIV-1, consistiam de 23 indivíduos expostos não-infectados (ENI), 14 mulheres e 9 homens, com mediana de 41 anos e 21 parceiros infectados por HIV-1 (HIV), 20 homens e 1 mulher com mediana de 41 anos. Os controles saudáveis foram 24 indivíduos (14 mulheres e 10 homens) com mediana de 37 anos. Os casais sorodiscordantes foram compostos por 16 heterossexuais e 7 homossexuais, com tempo de relacionamento de 13 anos. As frequências de células Th17, Th22 e Tc22, as células T polifuncionais foram analisadas em células mononucleares (CMNs) do sangue periférico, estimulados com peptídeos da região Gag do HIV-1 e da enterotoxina B do Staphylococcus aureus (SEB), a frequência de células T reguladoras, o perfil fenotípico de exaustão/diferenciação e a expressão da integrina alfa4?7 e CCR9 em células T, foram realizados por citometria de fluxo. RESULTADOS: No grupo HIV, as células T CD4+ e CD8+ do sangue periférico mostrou maior frequência de CD95 e PD-1 e baixa expressão de CD127 comparado ao grupo ENI e controle. A frequência de células Th17 em CMNs aumentou nos grupos ENI e HIV-1 na condição sem estímulo, contudo, após estímulo com os peptídeos da região p24 da Gag do HIV-1 induziu resposta somente no grupo HIV-1. O grupo ENI mostrou resposta antígeno-especifica somente para IL-22. Além disto, avaliando as células Tc22 e Th22, foi verificado aumento da resposta aos peptídeos da Gag e também ao SEB, nos grupos HIV e ENI. A presença de células T polifuncionais antígeno-especificas, secretoras de 5-4 citocinas, foi detectada apenas em células T CD38+ no grupo HIV, enquanto os indivíduos ENI mostraram resposta polifuncional por células T CD38- somente ao estímulo policlonal por SEB. Uma diminuição do número absoluto de células T reguladoras (CD4+CD25+CD127low/-Foxp3+) foi detectada no grupo HIV comparado ao ENI e controle, com maior expressão de moléculas HLA-DR e CD95. Além disto, foi detectado diminuição na frequência de células TCD8+ ?4?7+ no grupo ENI e de células TCD4+ alfa4beta7+ nos grupos ENI e HIV. Houve uma correlação positiva entre as células Tc22 e Th22 com as células TCD8+ e TCD4+ que expressam alfa4beta7, no grupo ENI e HIV-1. CONCLUSÃO: Os indivíduos ENI são capazes de desenvolver resposta antígeno-específicas relacionadas com a IL-22, que possui importante função na imunidade de mucosas. Além disto, mostram presença de células T polifuncionais com baixo perfil de ativação a estímulo policlonal. Os dados evidenciam que os indivíduos ENI, mostram indução de células Tc22, aumento de expressão de moléculas de migração para o intestino e equilíbrio entre as células efetoras e Treg, que em conjunto, devem exercer importante papel para a resistência à infecção por HIV-1

A literatura acadêmica sobre o comportamento do investidor financeiro é bastante escassa. A pesquisa sobre o processo de decisão, em geral, aborda tradeoffs na aquisição de produtos e pouco se discute o processo de decisão de investimento. Esta tese pretende contribuir para a redução deste gap ao discutir fatores determinantes para a tomada de decisão do investidor pessoal em produtos financeiros. A decisão de investimento é complexa, envolve, entre outros, o tradeoff entre renunciar o consumo presente pela possibilidade de maior bem estar no futuro. Adicionalmente, em muitas situações, existe possibilidade real de perda dos recursos financeiros investidos. Para investigar os percursos desta decisão foram realizadas entrevistas em profundidade com executivos ligados ao setor de fundos de investimento e ao de distribuição de produtos de investimento dos maiores bancos brasileiros atuantes no segmento de varejo. Os conhecimentos recolhidos e a revisão de literatura efetuada subsidiaram a elaboração do questionário de pesquisa aplicado em plataforma web junto a potenciais investidores. Os atributos rentabilidade, possibilidade de perda (proxy de risco), liquidez, taxa de administração e recomendação do gerente foram identificados como os mais relevantes para a decisão do investidor. Para construção dos estímulos e decomposição da utilidade da decisão foi utilizada a técnica conjoint based choice (CBC) que simula uma decisão real. Os resultados apontaram ser a recomendação do gerente o atributo mais importante para a formação da preferência por uma alternativa de investimento, resultado que, por si só, indica que fatores não racionais exercem influência na decisão. Estudou-se, então, o impacto da aversão ao risco e do estilo cognitivo do investidor. Os resultados denotam que os mais avessos e os mais intuitivos são mais suscetíveis à recomendação do gerente, mas que seus efeitos são independentes entre si. As evidências sugerem que os mais intuitivos utilizam o gerente para alcançar conforto cognitivo na decisão e que os mais avessos para mitigar a sensação de risco associada ao produto. Uma análise de cluster indicou ser possível segmentar a amostra em dois grupos, um mais propenso à recomendação do gerente e outro aos atributos do produto. A recomendação do gerente mostrou ser o atributo mais forte na distinção dos grupos. Os resultados indicam que uma segmentação de mercado baseada na propensão à recomendação do gerente pode ser efetiva para direcionar a construção de uma estratégia de relacionamento que busque incrementar os resultados de longo prazo.

More than ever alternative energy solutions are being discussed including potential legislative action. With the use of fossil fuels being the most abundant and most controversial sources of energy, new solutions must be found. This paper looks at the economic and technical feasibility of different types of alternative energy and cogeneration applications within a steel mill. This paper examines alternative energy systems. The systems examined include Solar systems, thermal electric materials, and an Organic Rankine Cycle cogeneration project. None of the projects has limiting technical feasibility issues however each of the projects face economically limiting factors. Taking into account tangible and non-tangible factors the solar system and Organic Rankine Cycle cogeneration project are recommended for further study and potential installation.
