Con el objetivo de evitar los altos costes que supone un servicio LVoD hemos implementado una solución basada en el paradigma P2P. De esta forma se ha diseñado un sistema de servidores virtuales en las que son ubicados los diferentes peer dependiendo que parte de un vídeo determinado solicitan al servidor principal.

In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

Pseudomonas fluorescens EPS62e was selected during a screening procedure for its high efficacy in controlling infections by Erwinia amylovora, the causal agent of fire blight disease, on different plant materials. In field trials carried out in pear trees during bloom, EPS62e colonized flowers until the carrying capacity, providing a moderate efficacy of fire-blight control. The putative mechanisms of EPS62e antagonism against E. amylovora were studied. EPS62e did not produce antimicrobial compounds described in P. fluorescens species and only developed antagonism in King’s B medium, where it produced siderophores. Interaction experiments in culture plate wells including a membrane filter, which physically separated the cultures, confirmed that inhibition of E. amylovora requires cell-to-cell contact. The spectrum of nutrient assimilation indicated that EPS62e used significantly more or different carbon sources than the pathogen. The maximum growth rate and affinity for nutrients in immature fruit extract were higher in EPS62e than in E. amylovora, but the cell yield was similar. The fitness of EPS62e and E. amylovora was studied upon inoculation in immature pear fruit wounds and hypanthia of intact flowers under controlled-environment conditions. When inoculated separately, EPS62e grew faster in flowers, whereas E. amylovora grew faster in fruit wounds because of its rapid spread to adjacent tissues. However, in preventive inoculations of EPS62e, subsequent growth of EPS101 was significantly inhibited. It is concluded that cell-to-cell interference as well as differences in growth potential and the spectrum and efficiency of nutrient use are mechanisms of antagonism of EPS62e against E. amylovora

The presence of the antimicrobial peptide (AMP) biosynthetic genes srfAA (surfactin), bacA (bacylisin), fenD (fengycin), bmyB (bacyllomicin), spaS (subtilin), and ituC (iturin) was examined in 184 isolates of Bacillus spp. obtained from plant environments (aerial, rhizosphere, soil) in the Mediterranean land area of Spain. Most strains had between two and four AMP genes whereas strains with five genes were seldom detected and none of the strains had six genes. The most frequent AMP gene markers were srfAA, bacA, bmyB, and fenD, and the most frequent genotypes srfAA-bacA-bmyB and srfAAbacA-bmyB-fenD. The dominance of these particular genes in Bacillus strains associated with plants reinforces the competitive role of surfactin, bacyllomicin, fengycin, and bacilysin in the fitness of strains in natural environments. The use of these AMP gene markers may assist in the selection of putative biological control agents of plant pathogens

Four methods were tested to assess the fire-blight disease response on grafted pear plants. The leaves of the plants were inoculated with Erwinia amylovora suspensions by pricking with clamps, cutting with scissors, local infiltration, and painting a bacterial suspension onto the leaves with a paintbrush. The effects of the inoculation methods were studied in dose-time-response experiments carried out in climate chambers under quarantine conditions. A modified Gompertz model was used to analyze the disease-time relatiobbnships and provided information on the rate of infection progression (rg) and time delay to the start of symptoms (t0). The disease-pathogen-dose relationships were analyzed according to a hyperbolic saturation model in which the median effective dose (ED50) of the pathogen and maximum disease level (ymax) were determined. Localized infiltration into the leaf mesophile resulted in the early (short t0) but slow (low rg) development of infection whereas in leaves pricked with clamps disease symptoms developed late (long t0) but rapidly (high rg). Paintbrush inoculation of the plants resulted in an incubation period of medium length, a moderate rate of infection progression, and low ymax values. A cultivation-independent approach based on polymerase chain reaction (PCR)-amplified partial small subunit rRNA genes was used to characterize bacterial populations in the surface soil of a commercial pear orchard consisting of different pear cultivars during two consecutive growing seasons. Pyrus communis L. cvs Blanquilla, Conference, and Williams are among the most widely cultivated cultivars in Europe and account for the majority of pear production in Northeastern Spain. To assess the heterogeneity of the community structure in response to environmental variables and tree phenology, bacterial populations were examined using PCR-denaturing gradient gel electrophoresis (DGGE) followed by cluster analysis of the 16S ribosomal DNA profiles by means of the unweighted pair group method with arithmetic means. Similarity analysis of the band patterns failed to identify characteristic fingerprints associated with the pear cultivars. Both environmentally and biologically based principal-component analyses showed that the microbial communities changed significantly throughout the year depending on temperature and, to a lesser extent, on tree phenology and rainfall. Prominent DGGE bands were excised and sequenced to gain insight into the identities of the predominant bacterial populations. Most DGGE band sequences were related to bacterial phyla, such as Bacteroidetes, Cyanobacteria, Acidobacteria, Proteobacteria, Nitrospirae, and Gemmatimonadetes, previously associated with typical agronomic crop environments

El objetivo de nuestro estudio fue determinar la eficacia y seguridad del uso de mallas sintéticas profilácticas en laparotomías urgentes para la prevención de hernia incisional y evisceración. Planteamos un estudio retrospectivo de datos informatizados de 266 pacientes sometidos a laparotomía media urgente durante dos años. Se analizaon las complicaciones postoperatorias y la incidencia de hernia incisional según el tipo de cierre de pared. Podemos concluir que el uso de malla profiláctica sintética en laparotomía urgente es factible para la prevención de hernias incisionales sin añadir de forma significativa morbilidad al procedimiento incluso en terrenos contaminados y/o infectados.

Este proyecto ha sido desarrollado en el Institut d'Estudis Territorials, y se plantea como una solución visual que permita mapificar variables estadísticas de la Región Metropolitana de Barcelona, mediante una aplicación Web dinámica con la que el usuario pueda consultar y representar datos en el territorio a través de una cartografía de soporte.El Institut d’Estudis Territorials, a petición del Departament de Política Territorial i Obres Públiques, es el encargado de elaborar el Pla Territorial Metropolità de Barcelona. Esta aplicación complementa a los estudios socioeconómicos que se están llevando a cabo, y pretende ser una herramienta de gestión que permita caracterizar el territorio, hacer diagnósticos y evaluar las diferentes propuestas presentadas para dicho plan.En una etapa preliminar se desarrolló una aplicación Excel, pero pronto se manifestaron las numerosas carencias que esta sufría, tales como la dificultad para acceder a ella de manera concurrente, no ofrecer garantías en lo que a medidas de seguridad se refiere y no poder tratar los datos de manera centralizada. El archivo Excel debía ser distribuido entre los diferentes usuarios por lo que cualquier cambio en los datos implicaba una replicación del archivo y en consecuencia una nueva distribución. Resultó obvia la necesidad de una única base de datos centralizada, y un acceso distribuido a ésta. Se planteó entonces una aplicación Web como solución, la cual incluiría herramientas capaces de gestionar información gráfica (cartografía) y alfa-numérica (datos) de manera conjunta. Conociendo las facilidades que los Sistemas de Información Geográfica (SIG) ofrecen para el manejo de este tipo de datos y su interoperabilidad con Internet, se optó por construir un servicio de mapas con el que el usuario, de manera intuitiva, pudiese construir distintos mapas en función de la variable territorial que desease.Así mismo se plantean mejoras futuras, como la predicción a corto plazo de la evolución de diferentes variables, siempre ofreciendo los mapas como componente principal de la interfaz de usuario.

Seu isolamento serviu para fortalecer sua singular cultura crioula, porem a cllsta de contactos frequentes com influencias e inforrna90es externas. Cabo Verde e muito pequeno. Sua popula9ao de 300.000 habitantes, que mal chegaria para tornar uma cidade auto-suficiente num complexo industrial, esta dividida entre nove ilhas. A capital, Praia, conta somente com 23.000 habitantes, e e, essencialmente, urna pequena e simpatica cidade onde a maior parte dos vercursos pode ser feita a pe e onde encontros casuais entre funcionarios do governo sao provavelmente tao irnportantes quanto os formais meios de comunica9ao pelo telefone ou por escrito. Cabo Verde e tambem pequeno ern termos de area territorial, embora ocupe urna area relativamente grande de ocea~o. Sua superficie total e pouco mais de 4.000 km2 , porem suas nove ilhas principais estao espalhadas por uma POr9aO grande, quadrangular, do Atlantico medindo aproximadamente 240 qUilometros de urn lado. A superficie total (nao incluindo partes da plataforma continental que se estende para alem do litoral externo das ilhas) e comparavel as republicas de Togo ou Sri Lanka, ou cerca de metade da Guatemala.
