The purpose of this study was to evaluate the effects of the platelet-rich plasma (PRP) when used in combination with autogenous bone graft and bioabsorbable membrane (Resolut® ) in the treatment of Class III furcation defects in dogs. Material and method: Class III furcation defects (5 mm in height and in depth) were surgically created in the mandibular third premolars of five mongrel dogs. After nine weeks, the lesions were treated with scaling and root planning and each defect received one of the following treatments: autogenous bone graft + membrane (group C) or PRP + autogenous bone graft + membrane (group T). After a healing period of 90 days, the animals were sacrificed. Routine histological processing and staining with hematoxilyn and eosin and Masson trichrome were performed and a histomorphometric analysis determined the effect of the treatments on periodontal tissue regereneration. Data were analyzed by Hotelling’s T-squared (p < 0.05). Result: No statistically significant difference between C and T groups was observed by the histomorphometric analysis of the furcation area. Both treatment groups demonstrated similar regenerative results with the furcation defects partially filled and periodontal regeneration limited to the experimental notches of the lesions. (p > 0.05). Conclusion: According to the present results, PRP does not enhance the periodontal regeneration in class III furcation defects treated with autogenous bone graft and bioabsorbable membrane.

This work examined the histological effects, on the rat palatal mucosa, of a denture base acrylic resin, submitted or not to a post-polymerization heat-treatment. Methods: Fifteen adult female Wistar rats, with sixty days old, weighting 150 g – 250 g were divided in G1: animals being maintained under the same conditions as the experimental groups following described, but without the use acrylic palatal plates (control group); G2: use of heat-polymerized acrylic resin palatal plates made of Lucitone 550; G3: use of palatal plates identical to G2, but subjected to a post-polymerization treatment in a water bath at 55°C for 60 min. The plates covered all the palate and were fixed in the molar region with light-cured resin, thus being kept there for 14 days. After the sacrifice, the palate was removed, fixed in formaldehyde 10% and decalcified with EDTA. Sections were stained using haematoxylin and eosin. Images in duplicate were made from the central region of the cuts, to measure the thickness (μm) of the keratin layers (TKC), epithelium total (TET) and connective tissue (TCC). Statistical analyses were carried out by one-way ANOVA and Tukey post-tests (α=0.05). Results: According to the results there was significant difference in the thickness of keratin between G2 and G3, with G1 having the intermediate value and similar to the other groups. BackgroundThe pathogenesis of melasma and the role of keratinocytes in disease development and maintenance are not completely understood. Dermal abnormalities, the expression of inflammatory mediators, growth factors, epithelial expression of melanocortin and sexual hormones receptors suggest that not only melanocytes, but entire epidermal melanin unit is involved in melasma physiopathology.ObjectivesTo compare nuclear morphological features and chromatin texture between basal keratinocytes in facial melasma and adjacent normal skin.MethodsWe took facial skin biopsies (2mm melasma and adjacent normal skin) from women processed for haematoxylin and eosin. Thirty non-overlapping basal keratinocyte nuclei were segmented and descriptors of area, highest diameter, perimeter, circularity, pixel intensity, profilometric index (Ra) and fractal dimension were extracted using ImageJ software.ResultsBasal keratinocyte nuclei from facial melasma epidermis displayed larger size, irregular shape, hyperpigmentation and chromatin heterogeneity by fractal dimension than perilesional skin.ConclusionBasal keratinocytes from facial melasma display changes in nuclear form and chromatin texture, suggesting that the phenotype differences between melasma and adjacent facial skin can result from complete epidermal melanin unit alterations, not just hypertrophic melanocytes.

This study aimed to evaluate possible histopathological liver of tilapia fed diets containing fish silage and level protein. Sample of 180 fed with tilapia fingerlings fed with diets containing three protein levels (20, 24 and 28% CP), and Proportions residue fermented silage of tilapia (1/4 and 1/2) of were analyzed during 75 days. The tissue fragments were fixed in Bouin and included in Histosec (R). After that, between 2 to 5 mu m slices were done in a rotation microtome. The methods used for tissue analysis were hematoxilin/eosin and PAS. The histological slices were examined under light microscope (Olympus BX-50). The disarray in the morphology of the liver of fish fed biological silage was influenced by high protein levels, and increased proportions of 1/2 of animal protein diets. It was observed that the variation of hepatocytes is directly related to the type of diet for fish. In fish fed diets containing 28% CP, the liver showed disruption of the structure cordon of hepatocytes, necrosis and shifting points of the core to the periphery. Elevated levels of biological fish silage cause deleterious changes in the liver. The level of protein required to maintain the health of the associated development fish is 24% crude protein.

In renovascular hypertensive rats, low doses of angiotensin converting enzyme (ACE) inhibitors have been found to prevent myocardial hypertrophy independent of blood pressure level. This finding would suggest humoral rather than mechanical control of myocyte growth. The aim of this study was to examine the effect of nonantihypertensive doses of ACE inhibitor on myocardial hypertrophy and necrosis in hypertensive rats. Renovascular hypertension (RHT) was induced in four-week-old Wistar rats. Twenty-eight animals were treated for four weeks with three doses of ramipril (0.01, 0.1 or 1. 0 mg/kg/day, which are unable to lower blood pressure. Fourteen animals were not treated (RHT group). A sham operated, age/sex-matched group was used as control (n = 10). Myocardial histology was analysed in 3 microm thick sections of the ventricle stained with either haematoxylin-eosin, reticulin silver stain or Masson's trichrome. There was a significant correlation between systolic blood pressure and left ventricular to body weight ratio in both sets of animals: untreated plus controls and ramipril-treated rats. ACE inhibition prevented myocyte and perivascular necrosis and fibrosis in a dose-dependent manner. We conclude that myocardial hypertrophy in rats with renovascular hypertension is directly related to arterial pressure, and that this relationship is not affected by nonantihypertensive doses of ACE inhibitor. Myocardial necrosis/fibrosis and coronary artery damage induced by angiotensin II are prevented by ACE inhibitor in a dose-dependent manner, despite the presence of arterial hypertension.

The clinical behavior of Concise and Prisma Shield sealants for pit and fissure was analyzed by clinical/photographic evaluation. A hematoxylin-based staining solution was applied to the occlusal surface 7 days, 18 months, 36 months, and 11 years after the occlusal sealing, thus allowing the sealant material on the surface to be checked. At each analysis time, each occlusal surface was photographed, and the photographs corresponding to each time were submitted to clinical/photographic evaluation. Although the sealant materials showed marginal alterations over time, they did not require reapplication because the region composed of the pit and fissure remained covered.

Canine visceral leishmaniasis (CVL) is an anthropozoonosis characterized by a clinically chronic progressive disease. Non lymphoid organs are also affected, especially the kidneys. Dogs with leishmaniasis usually die with renal failure despite treatment. Haematoxylin-eosin (HE) staining in kidney tissue sections has low sensitivity for parasite identification. Immunohistochemistry (IHC) and polymerase chain reaction (PCR) are efficient methods for Leishmania sp. antigen and DNA detection in cases of low parasite burden. The present study aims to identify renal lesions of CVL and correlate them with microscopic findings determined by histochemistry, IHC and PCR. Both IHC and PCR provided similar positivity for amastigote identification, 3/20 animals (15%), thus increasing detection of the parasite in renal tissues when compared with histopathologic examination. The lesion most commonly observed with visceral leishmaniasis-positive canine kidney tissue was membranoproliferative glomerulonephritis, followed by interstitial nephritis without correlation to the number of amastigotes.

This study evaluated the use of hipoosmotic swelling test (HOST) with deionized water (0 mOsmol), as a method of post thaw ram semen evaluation and correlate their findings with different techniques of semen evaluation. Therefore, twenty semen samples of 20 different adult rams were assessed as for kinetic sperm parameters through computerized system (IVOS 12, Hamiton Thorn Biosciences, Beverly, MA, EUA) and subjective analysis. The sperm membranes viability was carried out by the association of fluorescent probes (propidium iodide, JC-1 and FITC-PSA). The structural integrity of the plasma membrane was also studied through supravital test with eosin and the functional integrity of membrane evaluated by doing the hipoosmotic swelling test with deionized water (0 mOsmol), in the following proportions: One part of semen for 10 (HOST 10), 50 (HOST 50) and 100 (HOST 100) parts of water. After semen dilution in the different proportions it was fixed in formalin-buffered saline and analyzed with regard to percentage of HOST reactive sperm (bent/coiled). The percentage of reaction obtained for HOST 10 (33,1%); HOST 50 (32,8%) and HOST 100 (31,8%) did not differ significantly. HOST 10 presented positive correlation with the plasma membrane integrity by the EOS (r = 0,80; p < 0,05). Positive correlations between HOST 50 and HOST 100 with sperm subpopulation with membrane integrity by fluorescence were observed (r = 0,83 and r = 0,85; p < 0,01). The findings suggest that the HOST with deionized water can provide additional information for post thawing ram sperm viability evaluation.

Objective. - The aim of this study was to identify the effects of strength training on plasma parameters, body composition and the liver of ovariectomized rats. Methods. - Wistar sedentary (SHAM), ovariectomized (OVX), and ovariectomized trained rats (strength training [OVX-EXE]) of 85% of one maximal repetition (1 RM), three times per week, for 10 weeks, were used on this study. We monitored the body weight and visceral (uterine, mesenteric and retroperitoneal) and subcutaneous adiposity, total cholesterol, triglycerides, HDL, blood glucose and liver morphology to identify the presence of macrovesicular steotosis (haematoxylin and eosin staining). Results. - We observed that strength training changed body weight (SHAM 293.0 +/- 14.5 g; OVX 342.6 +/- 10.8 g; OVX-EXE 317.7 +/- 11.9 g, P < 0.05), visceral and subcutaneous adiposity, glucose (SHAM 111.2 +/- 10.0 mg/dL; OVX 147.4 +/- 18.8 mg/dL; OVX-EXE 118.5 +/- 2.2 mg/dL, P < 0.05), increased HDL (SHAM 82.7 +/- 1.4 mg/dL; OVX 64.6 +/- 2.8 mg/dL; OVX-EXE 91.4 +/- 2.6 mg/dL, P < 0.05) and reduced macrovesicular steatosis in liver tissue. Conclusions. - Considering the data obtained in this research, we emphasise the use of strength exercise training as a therapeutic means to combat or control the metabolic disturbances associated with menopause, including adiposity, and adverse changes in blood glucose, blood HDL and macrovesicular steatosis. O quati (Nasua nasua) é um animal que pertence à Família Procyonidae. Foram utilizados três animais ortotanasiados, de ambos os sexos, provenientes do Criatório Científico de Animais Silvestres, Centro Universitário Fundação de Ensino Octávio Bastos (Cecrimpas, Unifeob) autorizado pelo IBAMA (Proc.02027.003731/04-76). Para a análise macroscópica, as línguas foram retiradas, analisadas e foto-documentadas. Para análise microscópica, as línguas foram processadas rotineiramente pela técnica de microscopia eletrônica de varredura e inclusão em Paraplast; pela técnica de microscopia de luz os fragmentos foram cortados em micrótomo, com espessura média de 5mm e corados em HE e Picrosírius com fundo de hematoxilina. Os resultados macroscópicos e microscópicos mostram que a língua do quati apresenta papilas filiformes, fungiformes, valadas e cônicas sendo estas distribuídas nas regiões rostralis, medialis e caudalis. Histologicamente, a língua do quati é revestida por um epitélio pavimentoso estratificado queratinizado apresentando camada basal, espinhosa, granulosa e córnea com fibras de músculos estriados esqueléticos longitudinais e transversais e diversas glândulas. De acordo com os resultados pode-se concluir que a língua do quati possui características macroscópicas e microscópicas semelhantes aos canídeos, tendo como diferença o número de papilas valadas e o grau de queratinização.
