In order to assess a new strategy of DNA vaccine for a more complete understanding of its action in immune response, it is important to determine the in vivo biodistribution fate and antigen expression. In previous studies, our group focused on the prophylactic and therapeutic use of a plasmid DNA encoding the Mycobacterium leprae 65-kDa heat shock protein (Hsp65) and achieved an efficient immune response induction as well as protection against virulent M. tuberculosis challenge. In the present study, we examined in vivo tissue distribution of naked DNA-Hsp65 vaccine, the Hsp65 message, genome integration and methylation status of plasmid DNA. The DNA-Hsp65 was detectable in several tissue types, indicating that DNA-Hsp65 disseminates widely throughout the body. The biodistribution was dose-dependent. In contrast, RT-PCR detected the Hsp65 message for at least 15 days in muscle or liver tissue from immunized mice. We also analyzed the methylation status and integration of the injected plasmid DNA into the host cellular genome. The bacterial methylation pattern persisted for at least 6 months, indicating that the plasmid DNA-Hsp65 does not replicate in mammalian tissue, and Southern blot analysis showed that plasmid DNA was not integrated. These results have important implications for the use of DNA-Hsp65 vaccine in a clinical setting and open new perspectives for DNA vaccines and new considerations about the inoculation site and delivery system. © 2006 Coelho-Castelo et al; licensee BioMed Central Ltd.

Neospora caninum, bovine viral diarrhea virus (BVDV) and bovine herpes virus 1 (BHV-1) are worldwide spread pathogens associated with reproductive problems in cattle. The present work aimed to observe the infection pattern of these three pathogens in two dairy herds with distinct reproductive managements from Triângulo Mineiro, Minas Gerais State, Brazil. The herds were not vaccinated against either N. caninum, BVDV or BHV-1. Blood samples were collected and analyzed for presence of specific antibodies, and N. caninum IgG avidity was measured in N. caninum positive samples. In herd 1, 34 out of 174 sampled cows (20%) had antibodies to N. caninum and the seropositivity of BVDV and BHV-1 were 62% and 86%, respectively. Of 69 sampled cows in herd 2. 7 (10%) had antibodies to N. caninum, and 49% and 39% were seropositive to BVDV and BHV-1, respectively. The IgG avidity profiles indicated that N. caninum had been present in both herds for some years and that herd 1 had an ongoing horizontal spread of the parasite. The results indicate that the studied reproductive pathogens were present in the herds and partly may have contributed to their reproductive problems.

The objective of this study was to investigate the spatial and temporal distribution of Libinia spinosa H. MILNE EDWARDS 1834 on unconsolidated sublittoral bottoms in two regions off the northern coast of the state of São Paulo, and to analyze the influence of environmental factors on the occurrence of this species and observe the recruitment pattern of its young. Crabs were collected monthly (July 2001 through June 2003) at depths of 5, 10, 15, 20, 25, 30 and 35 In, from a fishing boat equipped with two double-rig nets. Samples of water and sediment were collected for analysis of environmental factors. A total of 2112 spider crabs was obtained (701 juveniles and 1411 adults). The highest abundance was observed at depths of 20 and 25 m, in both regions. These localities were characterized by substrate composed of very fine sand and silt-clay. In regard to the temporal distribution, juveniles and adults predominated in the summer and winter months respectively. From these results, one can infer that the distribution of L. spinosa is related to environmental factors favorable for its life cycle; sediment type is the factor which most strongly determines its presence. © E. A multi-agent system with a percolation approach to simulate the driving pattern of Plug-In Electric Vehicle (PEV), especially suited to simulate the PEVs behavior on any distribution systems, is presented. This tool intends to complement information about the driving patterns database on systems where that kind of information is not available. So, this paper aims to provide a framework that is able to work with any kind of technology and load generated of PEVs. The service zone is divided into several sub-zones, each subzone is considered as an independent agent identified with corresponding load level, and their relationships with the neighboring zones are represented as network probabilities. A percolation approach is used to characterize the autonomy of the battery of the PVEs to move through the city. The methodology is tested with data from a mid-size city real distribution system. The result shows the sub-area where the battery of PEVs will need to be recharge and gives the planners of distribution systems the necessary input for a medium to long term network planning in a smart grid environment. © 2012 IEEE.

Triatoma lenti and Triatoma sherlocki are hemipterans that belong to the brasiliensis subcomplex. In triatomines, the constitutive heterochromatin pattern is species-specific and allows, in many cases, for the grouping of species. Thus, we cytogenetically analyzed T. sherlocki and T. lenti using C-banding, and we compared the results with previous ones obtained in other species of the brasiliensis subcomplex. Both species were found to have a male diploid chromosome number of 22 chromosomes (2n = 20A. +. XY) with heterochromatic blocks at one or both chromosomal ends of all autosomal pairs. During early meiotic prophase, they showed a large heteropycnotic chromocenter constituted by the association of both sex chromosomes plus two autosomal pairs and many heterochromatic blocks dispersed inside the nucleus. All of these cytogenetic characteristics are similar to those observed in other species of brasiliensis subcomplex, results which confirm the grouping of T. sherlocki and T. lenti within this subcomplex. However, we emphasize the importance of other approaches, such as molecular analysis, to confirm the placement of T. lenti within the brasiliensis subcomplex. © 2012 Elsevier B.V.

Objective. This study aimed to investigate the stress distribution in screwed implant-supported prostheses with different implant-abutment connections by using a photoelastic analysis. Materials and methods. Four photoelastic models were fabricated in PL-2 resin and divided according to the implant-abutment connection (external hexagon (EH) and Morse taper (MT) implants (3.75 × 11.5 mm)) and the number crowns (single and 3-unit piece). Models were positioned in a circular polariscope and 100-N axial and oblique (45) loading were applied in the occlusal surface of the crowns by using a universal testing machine. The stresses were photographically recorded and qualitatively analyzed using software (Adobe Photoshop). Results. Under axial loading, the MT implants exhibited a lower number of fringes for single-unit crowns than EH implants, whereas for a 3-unit piece the MT implants showed a higher number of fringes vs EH implants. The oblique loading increased the number of fringes for all groups. Conclusion. In conclusion, the MT implant-abutment connection reduced the amount of stress in single-unit crowns, for 3-unit piece crowns the amount of stress was lower using an external hexagon connection. The stress pattern was similar for all groups. Oblique loading promoted a higher stress concentration than axial loading. © Informa Healthcare.

A distribuição espacial e temporal da densidade e biomassa dos copépodos planctônicos Pseudodiaptomus richardi e P. acutus, ao longo de um gradiente de salinidade, foi estudada no Estuário do Rio Caeté (Norte do Brasil) durante os meses de junho e dezembro de 1998 (estação seca) e fevereiro e maio de 1999 (estação chuvosa). A biomassa dos copépodos foi estimada a partir de parâmetros da regressão baseada na relação entre o peso seco e o comprimento do corpo (prossoma) de organismos adultos. O Estuário do Rio Caeté caracterizou-se por uma grande variação espacial e sazonal na salinidade (0,8-37,2). A relação peso-comprimento para ambas as espécies de Pseudodiaptomus foi do tipo exponencial. Os valores de densidade e biomassa oscilaram entre 0,28-46,18 ind. m-3 e 0,0022-0,3507 mg DW. m-3 para P. richardi; e entre 0,01-17,02 ind. m-3 e 0,0005-0,7181 mg DW. m-3 para P. acutus. Os resultados revelaram que a contribuição de P. richardi para a produção secundária no Estuário do Rio Caeté é mais importante na zona liminética que em outras zonas onde foram dominantes os regimes eurihalino-polihalino. Contudo, para P. acutus não foi possível observar de forma clara um padrão de distribuição espacial e temporal para a área estudada.

Características ambientais e distâncias espaciais entre os locais foram utilizadas para explicar a distribuição das espécies no ambiente, através das predições da teoria Neutra (espaço) e teoria do Nicho (ambiente). Foram avaliados os efeitos de fatores geográficos e ambientais sobre a distribuição de larvas de Odonata ao longo da Bacia do Rio Suiá-Missu, no estado de Mato Grosso. Nós testamos a hipótese de que (1) o ambiente é o principal fator de estruturação da comunidade devido às suas exigências ecofisiológicas; e (2) o padrão, se presente, é mais expressivo para Zygoptera. As amostras foram feitas em 12 locais na Bacia do Rio Suiá-Missu, em três campanhas (2007/2008), com um total de 1.382 larvas de Odonata, composta por 10 famílias, 51 gêneros e 100 morfoespécies. Os Anisoptera foram mais abundantes que Zygoptera, que compreende 81% de todas as amostras. O ambiente afetou Zygoptera (R = 0,291; p = 0,007) e foi o principal fator de estruturação da assembléia. Assim, a teoria do nicho foi confirmada. A ausência deste efeito sobre Anisoptera pode ser devido às adaptações ecofisiológicos que lhes permitem ocupar diferentes habitats. Larvas de Zygoptera são indicadores de mudanças na estrutura do habitat. Os efeitos das variáveis ambientais sobre a ecologia das larvas enfatizam a forte relação entre esses organismos e integridade ambiental.

Species richness and geographical distribution of Cyclopoida freshwater copepods were analyzed along the La Plata River basin. Ninety-six samples were taken from 24 sampling sites, twelve sites for zooplankton in open waters and twelve sites for zooplankton within macrophyte stands, including reservoirs and lotic stretches. There were, on average, three species per sample in the plankton compared to five per sample in macrophytes. Six species were exclusive to the plankton, 10 to macrophyte stands, and 17 were common to both. Only one species was found in similar proportions in plankton and macrophytes, while five species were widely found in plankton, and thirteen in macrophytes. The distinction between species from open water zooplankton and macrophytes was supported by nonmetric multidimensional analysis. There was no distinct pattern of endemicity within the basin, and double sampling contributes to this result. This lack of sub-regional faunal differentiation is in accordance with other studies that have shown that cyclopoids generally have wide geographical distribution in the Neotropics and that some species there are cosmopolitan. This contrasts with other freshwater copepods such as Calanoida and some Harpacticoida. We conclude that sampling plankton and macrophytes together provided a more accurate estimate of the richness and geographical distribution of these organisms than sampling in either one of those zones alone.

The feeding behaviour and the swimming pattern of the reef manta (Manta alfredi) are described based on video recordings conducted in Sudan, Red Sea, Indian Ocean, during June 2001. Three circular swimming patterns are described: horizontal circle pattern, inclined circle pattern and double circle pattern, related to feedings on zooplankton distributed in the water column. The observation of circle-like swimming patterns for reef manta rays in shallow waters expands our knowledge about M. alfredi and suggests that the distribution and abundance of zooplankton influence the occurrence and group size of this species in the studied area, as reported from other localities for Manta species.
