Usually, competitive stress, work life or just the training load in athletes can cause a phenomenon called overtraining syndrome. It is described as imbalance between stimulus and recovery in which decreases performance of athletes without being apparently immersed in a frame of injury or disease. That prolonged fatigue may affect the ability to adapt and thus severely reduce performance. This situation can be prolonged in time despite the existence of weekly or monthly periods of recovery. We want to see if the parameters of HRV, RESTQ-Sport questionnaire and strength values vary significantly in front 2 types of training (an intensive two daily sessions, large volume of work and little rest between the sessions; and another with 4 weekly workouts, with longer recovery time and less volume of work), in national judo players (n=14) to study the relationship between VFC and the imbalance of stress-recovery. METHODOLOGY: We performed a longitudinal and experimental study of HRV in 14 subjects, judo players at the national, randomly divided into 2 groups. One of these was conducted intensive training (high volume and low recovery) and another who made a smoother workout (less volume and less recovery) during a period of 4 weeks. CONCLUTIONS: The correct interpretation of the fluctuation parameters of HRV during the training judo players, may be a good indicator of stress levels, the recovery workload and therefore the observation of VFC is becomes an economic and noninvasive method of monitoring training in judo.

O objetivo deste trabalho foi padronizar uma técnica de reação em cadeia da polimerase (PCR), para detecção de Clostridium chauvoei, em tecidos fixados em formol e incluídos em parafina de cobaias (Cavia porcellus) experimentalmente infectadas com esse microrganismo. Os animais foram sacrificados, e amostras do músculo da área de inoculação (MAI), fígado, miocárdio e baço foram disponibilizadas para a técnica de PCR. O clostrídio foi detectado em todas as secções do MAI, fígado e miocárdio, mas não foi observado em secções do baço. Reações cruzadas não foram observadas a partir de secções do MAI dos animais com inoculação de outras espécies de clostrídios, bem como nenhuma amplificação foi observada a partir de secções do MAI dos animais controle. Esses resultados mostram que a técnica de PCR desenvolvida neste estudo, pode ser usada para detecção de Clostridium chauvoei em tecidos fixados em formol e incluídos em parafina.

In patients suffering from sickle cell disease (SCD), bone is a preferred site of infection. We report the case of a five-year-and-eight-month-old black African boy with homozygous-SS disease who developed a cranial epidural abscess. This intracranial infectious complication originated from a Salmonella enteritidis osteitis of the frontal bone. Antibiotic treatment alone did not control the disease, so surgery was necessary to remove the necrotic bone and to evacuate the epidural pus. The numerous factors interfering with normal healing of a septic focus in sickle cell anemia, particularly in this previously undescribed intracranial complication, emphasize the need for a primary and early surgical treatment in similar situations.

We uncover the global organization of clustering in real complex networks. To this end, we ask whether triangles in real networks organize as in maximally random graphs with given degree and clustering distributions, or as in maximally ordered graph models where triangles are forced into modules. The answer comes by way of exploring m-core landscapes, where the m-core is defined, akin to the k-core, as the maximal subgraph with edges participating in at least m triangles. This property defines a set of nested subgraphs that, contrarily to k-cores, is able to distinguish between hierarchical and modular architectures. We find that the clustering organization in real networks is neither completely random nor ordered although, surprisingly, it is more random than modular. This supports the idea that the structure of real networks may in fact be the outcome of self-organized processes based on local optimization rules, in contrast to global optimization principles.

Background: In recent years, microRNA (miRNA) pathways have emerged as a crucial system for the regulation of tumorogenesis. miR-SNPs are a novel class of single nucleotide polymorphisms that can affect miRNA pathways. Design and Methods: We analyzed eight miR-SNPs by allelic discrimination in 141 patients with Hodgkin lymphoma and correlated the results with treatment-related toxicity, response, disease-free survival (DFS) and overall survival (OS). Results: The KRT81 (rs3660) GG genotype was associated with an increased risk of neurological toxicity (P=0.016), while patients with XPO5 (rs11077) AA or CC genotypes had a higher rate of bleomycin-associated pulmonary toxicity (P=0.048). Both miR-SNPs emerged as independent factors in the multivariate analysis. The XPO5 AA and CC genotypes were also associated with a lower response rate (P=0.036). XPO5 (P=0.039) and TRBP (rs784567) (P=0.022) genotypes emerged as prognostic markers for DFS, and XPO5 was also associated with OS (P=0.033). In the multivariate analysis, only XPO5 emerged as an independent prognostic factor for DFS (HR: 2.622; 95%CI 1.039-6.620; P=0.041). Given the influence of XPO5 and TRBP as individual markers, we then investigated the combined effect of these miR-SNPs. Patients with both the XPO5 AA/CC and TRBP TT/TC genotypes had the shortest DFS (P=0.008) and OS (P=0.008). Conclusion: miR-SNPs can add useful prognostic information on treatment-related toxicity and clinical outcome in Hodgkin lymphoma and can be used to identify patients likely to be chemoresistant or to relapse.

Esteve (1836) inventarià la documentació antiga, estructurada en seccions per Quintana. Solà (1854), seguit per Rancés, classificà per negociats i matèries i millorà la descripció dels lligalls. Vallès confeccionà un Registre-inventari que descriu cronològicament per negociats i matèries tots els fons. També inventarià col•leccions i fons afegits, que la Mancomunitat destinà a l’IEC, la Biblioteca de Catalunya i els Museus d’Art. Rapid advection of extremely warm and dry air is studied during two events in the Mediterranean Basin. On 27 August 2010 a rapid advection of extremely warm and dry air affected the northeast Iberian Peninsula during a few hours. At the Barcelona city center, the temperature reached 39.3 ° C, which is the maximum temperature value recorded during 230 yr of daily data series. On 23 March 2008 a rapid increase of temperature and drop of relative humidity were recorded for a few hours in Heraklion (Crete). During the morning on that day, the recorded temperature reached 34 °C for several hours on the northern coastline of this island.According to the World Meteorological Organization none of these events can be classified as a heat wave, which requires at least two days of abnormally high temperatures; neither are they a heat burst as defined by the American Meteorological Society, where abnormal temperatures take place during a few minutes. For this reason, we suggest naming this type of event flash heat. By using data from automatic weather stations in the Barcelona and Heraklion area and WRF mesoscale numerical simulations, these events are analyzed. Additionally, the primary risks and possible impacts on several fields are presented.

The objective of this work was to assess the potential of interspecific hybridization of Vitis labruscana and Muscadinia rotundifolia by using artificial cross-pollinations. Microsatellite markers were used to confirm interspecific hybridizations and the identity of the parental genotypes. In crosses in which M. rotundifolia was used as the female parent, no true hybrids were obtained. In the reciprocal crosses, 114 seedlings were identified as true V. labruscana x M. rotundifolia hybrids. Self pollination occurred in direct and in reciprocal crosses. The crossings between 'Bordo' x 'Carlos', 'Magnolia', 'Regale' and' Roanoke', and between' Isabel' x 'Bountiful', 'Carlos', 'Magnolia', 'Regale' and 'Roanoke' were confirmed. The 15 markers evaluated showed that two M. rotundifolia parental genotypes had the same fingerprint profile, indicating a like lyplanting error. The success of hybridization depends mainly on the species and on the cultivar used as the female parent. Microsatellite markers are efficient to confirm the paternity of interspecific F1 hybrids and to determine the correct identity of M. rotundifolia cultivars.

Der Aufsatz gilt den ,,Texten hinter den Texten" der aus dem elsässischen Produktionszusammenhang um Diebold Lauber im 15. Jahrhundert überlieferten Handschriften. Dass das rekonstruktive Potential dieses im deutschsprachigen Mittelalter einmaligen Corpus einer so grossen Gruppe von Handschriften eines einzelnen Skriptoriums bisher nahezu ungenutzt blieb, wird als Resultat einer unbemerkten perspektivischen Verschränkung zweier Wissenschaftsgruppen beschrieben: Wo die Texte der Handschriften überhaupt in den Blick gerieten, geschah dies entweder aus editorisch-kritischem Erkenntnisinteresse oder, den Impulsen jüngerer kunstgeschichtlicher Lauber-Forschung folgend, auf der Suche nach bewusstem, gar gestaltendem Umgang mit dem Text durch das Lauber-Team. Beiden Zugriffen liegen diametral entgegengesetzte Begriffe von textueller ,,Qualität" zugrunde, die schon auf terminologischer Ebene wiederholt zu Missverständnissen geführt haben. Was die systematische, vergleichende Untersuchung der Texte Lauberscher Handschriften - hier anhand von Mehrfachüberlieferungen - sowohl für die Erschliessung von Informationen über den elsässischen Betrieb als auch für den Census der älteren volkssprachigen Überlieferung im deutschen Südwesten zu leisten vermag, zeigen die Beispielanalysen der Lauber-Texte von ,Flore und Blanscheflur' (H, B) und ,Parzival' (m, n, o). In beiden Fällen lassen sprachliche Untersuchungen und paläographische Fehleranalysen der erhaltenen Textzeugen eine Vorlage mit frühem, aus kritischer Perspektive bemerkenswert gutem - von den Lauber-Redaktoren nach diesen Kriterien gewähltem? - Text erkennen. Im Fall von ,Flore und Blanscheflur' stammte mit hoher Wahrscheinlichkeit die Vorlagenhandschrift selber aus dem 13. oder spätestens frühen 14. Jahrhundert; den ,Parzival'-Abschriften dagegen lagen zwei verschiedene zeitgenössische Handschriften zugrunde. Erhellt wird auch der praktische Umgang mit Vorlagenhandschriften im Herstellungsprozess, der bisher im Dunkeln lag: Der Aufsatz belegt, dass in beiden untersuchten Beispielen Handschriften fremder Provenienz als Vorlagenexemplare zur wiederholten Benutzung aufbereitet und dauerhaft verwahrt wurden. Dabei müssen die laubertypischen, gliedernden Zwischenüberschriften auf einem separaten Anweisungsblatt bzw. Faszikel notiert gewesen sein. Ihre vorgesehene Positionierung wurde dem Schreiber vermutlich durch Randmarkierungen im Vorlagenexemplar angezeigt. Besondere Umstände galten für die Vorlagen der drei erhaltenen ,Parzival'-Handschriften: Lauber benutzte offenbar zunächst eine den Lauber-Produkten im Typ ähnliche Bilderhandschrift, die er, da sie beschädigt war, später durch ein anderes Exemplar ersetzte. Dabei handelte es sich wieder um eine Bilderhandschrift gleichen Typs, die nun allerdings nach werkstattüblichem Verfahren über ein separates Anweisungsblatt so aufbereitet wurde, dass sich die Zahl der Illustrationen und Zwischenüberschriften erhöhte. Als wahrscheinlichste Quelle dieser zweimal verfügbaren, textlich zusammengehörenden bebilderten Handschriften ist die sogenannte ,Werkstatt von 1418' auszumachen. Auch andere Hinweise deuten darauf, dass ab etwa den 1440er Jahren eine grössere Gruppe von Handschriften aus dieser älteren Produktion, deren Schwerpunkt die höfische Literatur des 13. Jahrhunderts bildete, in den Bestand Laubers geriet und dessen Programm erweiterte. Die ab dieser Phase in der jüngeren Forschung (Saurma-Jeltsch) beobachtete, scheinbar marktorientiert gezielte Neugestaltung des Programms erweist sich so möglicherweise nur als Folge einer plötzlich verfügbaren neuen Vorlagengruppe.

Este trabalho objetivou testar diferentes doses de Paclobutrazol (PBZ), ethephon e nitrato de cálcio na indução do florescimento e na produção da mangueira (Mangifera indica L.) cv. Tommy Atkins, localizada no pomar didático da ESAM em Mossoró-RN, no ano de 1999/2000. O delineamento experimental utilizado foi em blocos casualizados, no esquema fatorial 2x2x3, assim distribuídos: T1 1000 mg.L-1 de PBZ+ 2% de nitrato de cálcio; T2 1000 mg.L-1 de PBZ + 2% de nitrato de cálcio + 1,0 mL.L-1 de ethephon; T3 1000 mg.L-1 de PBZ + 2% de nitrato de cálcio + 3,0 mL.L-1 de ethephon; T4 1000 mg. L-1 de PBZ + 3% de nitrato de cálcio; T5 1000 mg.L-1 de PBZ + 3% de nitrato de cálcio + 1,0 mL.L-1 de ethephon; T6 1000 mg.L-1 de PBZ + 3% de nitrato de cálcio + 3,0 mL.L-1 de ethephon ; T7 1500 mg.L-1 de PBZ + 2% de nitrato de cálcio; T8 1500 mg.L-1 de PBZ + 2% de nitrato de cálcio + 1,0 mL.L-1 de ethephon; T9 1500 mg.L-1 de PBZ + 2% de nitrato de cálcio + 3,0 mL.L-1 de ethephon; T10 1500 mg.L-1 de PBZ+ 3% de nitrato de cálcio; T11 1500 mg.L-1 de PBZ + 3% de nitrato de cálcio + 1,0 mL.L-1 de ethephon.L; T12 1500 mg.L-1 de PBZ + 3% de nitrato de cálcio + 3,0 mL.L-1 de ethephon, com 4 repetições. A mangueira teve um maior florescimento (81,75%) com 2% de nitrato de cálcio e 1500 mg. L-1 de PBZ, o número de frutos por planta teve seu maior valor (86 frutos) com 3% de nitrato de cálcio e 1500 mg L-1 de PBZ e o peso do fruto foi maior (425,5g) na dosagem de 3mL.L-1 de ethephon. Os produtos aplicados não diferenciaram entre si em relação à produção.

Currently, MVA virus vectors carrying HIV-1 genes are being developed as HIV-1/AIDS prophylactic/therapeutic vaccines. Nevertheless, little is known about the impact of these vectors on human dendritic cells (DC) and their capacity to present HIV-1 antigens to human HIV-specific T cells. This study aimed to characterize the interaction of MVA and MVA expressing the HIV-1 genes Env-Gag-Pol-Nef of clade B (referred to as MVA-B) in human monocyte-derived dendritic cells (MDDC) and the subsequent processes of HIV-1 antigen presentation and activation of memory HIV-1-specific T lymphocytes. For these purposes, we performed ex vivo assays with MDDC and autologous lymphocytes from asymptomatic HIV-infected patients. Infection of MDDC with MVA-B or MVA, at the optimal dose of 0.3 PFU/MDDC, induced by itself a moderate degree of maturation of MDDC, involving secretion of cytokines and chemokines (IL1-ra, IL-7, TNF-α, IL-6, IL-12, IL-15, IL-8, MCP-1, MIP-1α, MIP-1β, RANTES, IP-10, MIG, and IFN-α). MDDC infected with MVA or MVA-B and following a period of 48 h or 72 h of maturation were able to migrate toward CCL19 or CCL21 chemokine gradients. MVA-B infection induced apoptosis of the infected cells and the resulting apoptotic bodies were engulfed by the uninfected MDDC, which cross-presented HIV-1 antigens to autologous CD8+ T lymphocytes. MVA-B-infected MDDC co-cultured with autologous T lymphocytes induced a highly functional HIV-specific CD8+ T cell response including proliferation, secretion of IFN-γ, IL-2, TNF-α, MIP-1β, MIP-1α, RANTES and IL-6, and strong cytotoxic activity against autologous HIV-1-infected CD4+ T lymphocytes. These results evidence the adjuvant role of the vector itself (MVA) and support the clinical development of prophylactic and therapeutic anti-HIV vaccines based on MVA-B.

Adjuvant chemotherapy decisions in breast cancer are increasingly based on the pathologist's assessment of tumor proliferation. The Swiss Working Group of Gyneco- and Breast Pathologists has surveyed inter- and intraobserver consistency of Ki-67-based proliferative fraction in breast carcinomas. METHODS: Five pathologists evaluated MIB-1-labeling index (LI) in ten breast carcinomas (G1, G2, G3) by counting and eyeballing. In the same way, 15 pathologists all over Switzerland then assessed MIB-1-LI on three G2 carcinomas, in self-selected or pre-defined areas of the tumors, comparing centrally immunostained slides with slides immunostained in the different laboratoires. To study intra-observer variability, the same tumors were re-examined 4 months later. RESULTS: The Kappa values for the first series of ten carcinomas of various degrees of differentiation showed good to very good agreement for MIB-1-LI (Kappa 0.56-0.72). However, we found very high inter-observer variabilities (Kappa 0.04-0.14) in the read-outs of the G2 carcinomas. It was not possible to explain the inconsistencies exclusively by any of the following factors: (i) pathologists' divergent definitions of what counts as a positive nucleus (ii) the mode of assessment (counting vs. eyeballing), (iii) immunostaining technique, and (iv) the selection of the tumor area in which to count. Despite intensive confrontation of all participating pathologists with the problem, inter-observer agreement did not improve when the same slides were re-examined 4 months later (Kappa 0.01-0.04) and intra-observer agreement was likewise poor (Kappa 0.00-0.35). CONCLUSION: Assessment of mid-range Ki-67-LI suffers from high inter- and intra-observer variability. Oncologists should be aware of this caveat when using Ki-67-LI as a basis for treatment decisions in moderately differentiated breast carcinomas.

Pint lincRNA connects the p53 pathway with epigenetic silencing by the Polycomb repressive complex 2

BACKGROUND: The p53 transcription factor is located at the core of a complex wiring of signaling pathways that are critical for the preservation of cellular homeostasis. Only recently it has become clear that p53 regulates the expression of several long intergenic noncoding RNAs (lincRNAs). However, relatively little is known about the role that lincRNAs play in this pathway. RESULTS: Here we characterize a lincRNA named Pint (p53 induced noncoding transcript). We show that Pint is a ubiquitously expressed lincRNA that is finely regulated by p53. In mouse cells, Pint promotes cell proliferation and survival by regulating the expression of genes of the TGF-β, MAPK and p53 pathways. Pint is a nuclear lincRNA that directly interacts with the Polycomb repressive complex 2 (PRC2), and is required for PRC2 targeting of specific genes for H3K27 tri-methylation and repression. Furthermore, Pint functional activity is highly dependent on PRC2 expression. We have also identified Pint human ortholog (PINT), which presents suggestive analogies with the murine lincRNA. PINT is similarly regulated by p53, and its expression significantly correlates with the same cellular pathways as the mouse ortholog, including the p53 pathway. Interestingly, PINT is downregulated in colon primary tumors, while its overexpression inhibits the proliferation of tumor cells, suggesting a possible role as tumor suppressor. CONCLUSIONS: Our results reveal a p53 autoregulatory negative mechanism where a lincRNA connects p53 activation with epigenetic silencing by PRC2. Additionally, we show analogies and differences between the murine and human orthologs, identifying a novel tumor suppressor candidate lincRNA.
