O crambe é uma cultura promissora para produção de biodiesel, principalmente pelo alto conteúdo de óleo de suas sementes. No entanto, não há metodologias estabelecidas para avaliar a qualidade fisiológica das sementes desta espécie e lotes não podem ser comparados, especialmente por testes de vigor, como o de envelhecimento acelerado. O objetivo da presente pesquisa foi avaliar o efeito da alta temperatura e do período de exposição durante o teste de envelhecimento acelerado na qualidade fisiológica e atividade enzimática de sementes de crambe. Dois lotes de sementes de crambe, cultivar Brilhante, foram analisados por meio dos testes de teor de água, massa de 1.000 sementes, germinação, primeira contagem, condutividade elétrica, atividade enzimática (peroxidase e superóxido dismutase) e comprimento de plântulas. As avaliações foram conduzidas antes e após o envelhecimento acelerado, que foram testadas diferentes temperaturas (38, 40 e 42ºC) e períodos de exposição (24, 48 e 72h). O delineamento experimental foi o inteiramente casualizado com quatro repetições. Os dados foram submetidos à análise de variância e as médias foram comparadas pelo teste de Tukey (p < 0.05). O teste de Dunnet (p < 0.05) foi utilizado para comparar os valores da testemunha (antes do envelhecimento acelerado) com cada valor médio individualmente. O teste de correlação linear simples também foi aplicado. Conclui-se que a interação temperatura x período de exposição afeta a qualidade fisiológica das sementes e a atividade enzimática e que as melhores condições durante o teste de envelhecimento acelerado são dependentes do genótipo.

Os solos derivados de rochas serpentinizadas ou serpentinitos constituem um grupo especial de solos em toda superfície terrestre. de caráter ultramáfico, ou seja, rochas com mais de 70 % de minerais máficos (ferromagnesianos), os serpentinitos apresentam uma mineralogia pobre em sílica e escassa em Al, sendo, no entanto, muito enriquecida em Mg. São poucos os estudos sobre a morfologia, mineralogia, gênese e classificação dos solos desenvolvidos de tais rochas. em ambiente tropical úmido no sudoeste de Minas Gerais, na zona do greenstone belt do Morro de Ferro, em superfícies geomórficas jovens, três perfis de solos representativos dessa paisagem sobre rochas serpentinizadas foram caracterizados por meio de descrições macro e micromorfológicas, análises granulométricas, químicas e por mineralogia de raios X das frações argila e silte. Complementarmente, para acompanhamento da alteração geoquímica dos horizontes do solo, foram feitas microanálises das seções delgadas por EDRX. Os solos foram classificados como Chernossolo Háplico Férrico típico, Cambissolo Háplico eutroférrico léptico e Neossolo Regolítico eutrófico típico e, embora situados num clima que favorece o rápido intemperismo, do ponto de vista morfológico e mineralógico, mostraram-se similares aos solos derivados de rochas serpentinizadas das regiões subtropicais e temperada. No processo de formação de solo, a evolução da trama segue a seguinte seqüência: alteração da rocha ® trama frâgmica ® trama porfírica com cavidades ® trama porfírica aberta por coalescência de cavidades. O processo de argiluviação é evidente e se dá em dois estádios distintos: argiluviação primária, que ocorre nas fendas e cavidades que se formam por alteração de rocha, e argiluviação secundária, verificada na porosidade mais aberta e evoluída da coalescência das cavidades. Os solos apresentam mineralogia pouco comum para solos tropicais, com presença de minerais primários de fácil decomposição até mesmo na fração argila, com destaque para o talco, clorita trioctaedral e ocorrência limitada de tremolita, sendo esta última abundante na fração silte. Óxidos de Fe, caulinita e os interestratificados de clorita-esmectita e de clorita-vermiculita completam a assembléia mineralógica. A tendência de evolução é para B textural ou para B nítico com mineralogia 1:1 e alto conteúdo de óxidos de Fe. Nas fases iniciais de alteração, os alteromorfos já apresentam composição química similar aos agregados do solo, com forte perda de Mg, Ca e Si e acúmulo relativo de Al e Fe. Nas três situações estudadas, ocorreu um rejuvenescimento superficial por erosão diferencial, que acumulou material grosseiro e removeu os finos, contribuindo para o incremento da relação textural.

Estudou-se a evolução geoquímica e mineralógica em três perfis distintos de alteração de rochas serpentinizadas que ocorrem nas imediações dos municípios de Alpinópolis e Fortaleza de Minas, no sudoeste do Estado de Minas Gerais, sob regimes de umidade e de temperatura údico e térmico, respectivamente. Nas condições atuais, o grau de evolução química e mineralógica é moderado em relação ao desenvolvido sobre outros tipos de rochas básicas e ultrabásicas da mesma área, caracterizando-se por uma importante perda de Na e Mg e, em menor proporção, de Ca e Si. O Al (localmente também o Fe) é o elemento menos móvel dos sistemas. O K é escasso no material de origem e nas zonas de alteração, e ocorre enriquecimento desse elemento nos horizontes superficiais por aporte externo. Os minerais primários mais facilmente intemperizáveis, como o talco, a tremolita e a clorita trioctaédrica, são abundantes ainda na fração argila desses solos tropicais com composição mineralógica pouco comum, mas são todos termodinamicamente instáveis. do ponto de vista geoquímico, o processo de alteração atual pode ser definido como uma bissialitização, que pode coincidir com ferruginização, com formação de minerais trioctaédricos secundários por transformação direta de estrutura e também por neoformação, todos coexistindo com os minerais primários residuais. No entanto, as fases de maior evolução, em volumes com drenagem mais eficiente, tendem à monossialitização, com formação de caulinitas de diferentes graus de cristalinidade. A assembléia mineralógica existente evidencia a metaestabilidade e o caráter incipiente do sistema pedogenético.

The free form of the iron ion is one of the strongest oxidizing agents in the cellular environment. The effect of iron at different concentrations (0, 1, 5, 10, 50, and 100 µM Fe3+) on the normal human red blood cell (RBC) antioxidant system was evaluated in vitro by measuring total (GSH) and oxidized (GSSG) glutathione levels, and superoxide dismutase (SOD), catalase, glutathione peroxidase (GSH-Px) and reductase (GSH-Rd) activities. Membrane lipid peroxidation was assessed by measuring thiobarbituric acid reactive substance (TBARS). The RBC were incubated with colloidal iron hydroxide and phosphate-buffered saline, pH 7.45, at 37oC, for 60 min. For each assay, the results for the control group were: a) GSH = 3.52 ± 0.27 µM/g Hb; b) GSSG = 0.17 ± 0.03 µM/g Hb; c) GSH-Px = 19.60 ± 1.96 IU/g Hb; d) GSH-Rd = 3.13 ± 0.17 IU/g Hb; e) catalase = 394.9 ± 22.8 IU/g Hb; f) SOD = 5981 ± 375 IU/g Hb. The addition of 1 to 100 µM Fe3+ had no effect on the parameters analyzed. No change in TBARS levels was detected at any of the iron concentrations studied. Oxidative stress, measured by GSH kinetics over time, occurs when the RBC are incubated with colloidal iron hydroxide at concentrations higher than 10 µM of Fe3+. Overall, these results show that the intact human RBC is prone to oxidative stress when exposed to Fe3+ and that the RBC has a potent antioxidant system that can minimize the potential damage caused by acute exposure to a colloidal iron hydroxide in vitro.

Com o objetivo de avaliar a influência da relação P:Mg na fertilidade do solo, no estado nutricional e na produção de matéria seca da alfafa, foi realizado um experimento em vasos com Latossolo Vermelho Amarelo distrófico, segundo delineamento inteiramente casualizado, em esquema de parcelas subdivididas, com três repetições. Os tratamentos foram constituídos de combinações de P (doses: 0, 100, 200 e 400 mg kg-1, fonte: superfosfato triplo) e Mg (doses: 0, 100 e 200 mg kg-1, fonte: cloreto de magnésio), nas proporções de 0, 0,5, 1, 2 e 4. No período experimental, foram realizados três cortes, com intervalo de 30 dias (subparcelas). Os resultados demonstraram que o incremento da relação P:Mg e das doses de P e de Mg aumenta a produção de matéria seca. O teor de P e as combinações de P:Mg no tecido vegetal apresentaram estreita relação com a proporção desses nutrientes no solo. A relação 2:1 acarretou maior teor de clorofila e N na matéria seca, enquanto altas quantidades de P no solo diminuíram a absorção de K.

5-Aminolevulinic acid (ALA), a heme precursor that accumulates in acute intermittent porphyria patients and lead-exposed individuals, has previously been shown to autoxidize with generation of reactive oxygen species and to cause in vitro oxidative damage to rat liver mitochondria. We now demonstrate that chronically ALA-treated rats (40 mg/kg body wt every 2 days for 15 days) exhibit decreased mitochondrial enzymatic activities (superoxide dismutase, citrate synthase) in liver and soleus (type I, red) and gastrocnemius (type IIb, white) muscle fibers. Previous adaptation of rats to endurance exercise, indicated by augmented (cytosolic) CuZn-superoxide dismutase (SOD) and (mitochondrial) Mn-SOD activities in several organs, does not protect the animals against liver and soleus mitochondrial damage promoted by intraperitoneal injections of ALA. This is suggested by loss of citrate synthase and Mn-SOD activities and elevation of serum lactate levels, concomitant to decreased glycogen content in soleus and the red portion of gastrocnemius (type IIa) fibers of both sedentary and swimming-trained ALA-treated rats. In parallel, the type IIb gastrocnemius fibers, which are known to obtain energy mainly by glycolysis, do not undergo these biochemical changes. Consistently, ALA-treated rats under swimming training reach fatigue significantly earlier than the control group. These results indicate that ALA may be an important prooxidant in vivo.

At the present time, much attention is being paid to antioxidant substances because many pathological conditions are associated with oxidative stress. The purpose of the present study was to discover the potency of saponin (2-phenyl-benzopyrane), a soybean flavonoid, with respect to its hypoglycaemic and hypolipidaemic action, and the association of these effects with oxidative stress. Male Wistar rats were divided into two groups (n = 6): control group and saponin-treated group (60 mg/kg) during 30 days. Saponin had no effects on glucose tolerance. Although no changes had been observed in low-density lipoprotein-cholesterol, saponin-treated animals had increased low-density lipoprotein-cholesterol/triacylglycerol ratio and decreased triacylglycerol, very low-density lipoprotein-cholesterol and total/high-density lipoprotein-cholesterol ratio than the control group. Saponin-treated rats showed lower lipid hydroperoxide than control rats, indicating decreased potential to atherosclerosis. No alterations were observed in antioxidant enzymes, superoxide dismutase and glutathione peroxidase, while lipid hydroperoxide were decreased in saponin-treated rats. In conclusion, the beneficial effects of saponin on serum lipids were related to a direct saponin antioxidant activity.

BACKGROUND. The present report was carried out to determine whether alcohol intake could induce prostate lesions.METHODS. We tested male rats for 300 days. Animals were divided into three groups: controls received only tap water as liquid diet; the chronic alcohol intake group received only ethanol solution in semivoluntary research; and the withdrawal group received the same treatment as chronic alcohol intake until 240 days, after which they reverted to drinking water.RESULTS. Chronic alcohol intake increased lipoperoxide concentrations and acid phosphatase activities. Cu-Zn superoxide dismutase (SOD) was decreased at 60 days, but approached controls values at 300 days following treatment. The serum increased alkaline phosphatase, and alanine transaminase activities reflected the chronic toxic effect of ethanol.CONCLUSIONS. Since SOD activity was unable to scavenge superoxide radical and lipoperoxide formation, we can conclude that superoxide is an important intermediate in prostate damage of chronic alcohol intake. Superoxide radical (O-2(-)) is a free radical that may be involved in various toxic processes. Cu-Zn superoxide dismutase catalyzes the dismutation of the superoxide free radical and protects cells from oxidative damage. A rat bioassay validated for the identification of the toxic effects of azomethine H revealed increased serum activities of amylase, alanine transaminase, and alkaline phosphatase. The lipoperoxide and bilirubin concentrations were also increased in animals that received azomethine H (1 g/kg) from ascorbic or hydrochloric acid solutions. Azomethine H increased Cu-Zn superoxide dismutase activity. This elevation of Cu-Zn superoxide dismutase activity was highest on the 7th day and was at levels comparable with those of control rats from day 60 onwards. Superoxide is an important intermediate in the action and toxicity of azomethine H.

Long-term propranolol treatment reduces arterial blood pressure in hypertensive individuals mainly by reducing peripheral vascular resistance, but mechanisms underlying their vasodilatory effect remain poorly investigated. This study aimed to investigate whether long-term propranolol administration ameliorates the impairment of relaxing responses of aorta and mesenteric artery from rats made hypertensive by chronic nitric oxide (NO) deficiency, and underlying mechanisms mediating this phenomenon. Male Wistar rats were treated with N-omega-Nitro-L-arginine methyl ester (L-NAME; 20 mg/rat/day) for four weeks. DL-Propranolol (30 mg/rat/day) was given concomitantly to L-NAME in the drinking water. Treatment with L-NAME markedly increased blood pressure, an effect largely attenuated by DL-propranolol. In phenylephrine-precontracted aortic rings, the reduction of relaxing responses for acetylcholine (0.001-10 mu M) in L-NAME group was not modified by DL-propranolol, whereas in mesenteric rings the impairment of acetylcholine-induced relaxation by L-NAME was significantly attenuated by DL-propranolol. In mesenteric rings precontracted with KCl (80 MM), DL-propranolol failed to attenuate the impairment of acetylcholine-induced relaxation by L-NAME. The contractile responses to extracellular CaCl2 (1-10 mM) were increased in L-NAME group, and co-treatment with DL-propranolol reduced this response in both preparations in most Ca2+ concentrations used. The NO2/NO3 plasma levels and superoxide dismutase (SOD) activity were reduced in L-NAME-treated rats, both of which were significantly prevented by DL-propranolol. In conclusion, propranolol-induced amplification of the relaxation to acetylcholine in mesenteric arteries from L-NAME-treated rats is sensitive to depolarization. Additional mechanisms involving blockade of Ca2+ entry in the vascular smooth muscle and increase in NO bioavailability contributes to beneficial effects of long-term propranolol treatment. The contamination of water by metal compounds is a worldwide environmental problem. Concentrations of metals are widely related to biochemical values which are used in disease diagnosis due to environmental toxicity. The acute combined effects of cadmium and nickel on biochemical parameters were determined and compared with those of Cd2+ or Ni2+ alone in rats. Male adult rats were given drinking solutions of CdCl2 [Cd(II) cation, 100 mg/liter] or NiSO4 [Ni(II) cation, 100 mg/liter]. For the combined treatment, the animals (Ni+Cd) received both Ni(II)) cation (100 mg/liter) and Cd(II) cation (100 mg/liter). Nickel treatment induced increased alanine transaminase (ALT) activity and hepatotoxicity, but not renal injury. In contrast, cadmium exposure produced hepatic, renal and myocardial damage, characterized by increased creatinine, total and direct bilirubin concentrations and increased ALT and lactate dehydrogenase (LDH) activities. The combined effect Ni-Cd is less toxic than cadmium alone, suggesting antagonism between these toxicants. The toxicity of nickel and cadmium, alone and in combination, decreased Cu-Zn superoxide dismutase (SOD) activity and increased lipoperoxide formation. (C) 1998 Elsevier B.V. Ltd. Water contaminants have a high potential risk for the health of populations and for this reason their toxic effects urgently should be established. The present study was carried out to determine whether an environmentally realistic intake of water contaminants can induce tissue lesions, and to clarify the contribution of superoxide radical (O-2(.-)) formation to this effect. Male Wistar rats were given drinking water from the Tiett River (group A) and from the Capivara River (group B). The increased creatinine, glucose, alanine transaminase and amylase levels in serum reflected the toxic effects of river-water contaminants to renal, pancreatic and hepatic tissues of rats. As changes in lipoperoxide were observed in rats after river-water intake while superoxide dismutase activities decreased in these animals, it is assumed that the superoxide anion elicits lipoperoxide formation and induces tissue damage. There is evidence that oxygen tension reflects water pollution, since river-water with a-low oxygen tension induced more elevated toxicity in rat tissues. (C) 1999 Elsevier B.V. Ltd. The ability of high dietary carbohydrate to induce acute pancreatitis was investigated in groups of 16,2 1-day and 15-month old rats fed different carbohydrate diets for 30 days. Significantly increased levels of serum amylase (2-fold), phospholipids (50%),phosphorus (2-fold), and lipoperoxides (8-fold) were observed in 15-month old rats fed a high-carbohydrate diet, compared to rats fed a diet with normal carbohydrate levels, indicating peroxidation of membrane lipids which caused final cell death and pancreatic lesion. Serum Cu-Zn superoxide dismutase activity was not altered. Daily administration of bovine Cu-Zn superoxide dismutase conjugated with polyediylene glycol prevented the serum level alterations and pancreatic lesions, indicating that the superoxide radical has a role in dietary carbohydrate-induced acute pancreatitis. No biochemical changes were observed in rats in which treatment was initiated on the 21st day of life indicating that this is an age-related lesion.

The toxic effects of chronic ethanol ingestion were evaluated in male adult rats for 300 days. The animals were divided into three groups: the controls received only tap water as liquid diet; the chronic ethanol ingestion group received only ethanol solution (30%) in semivoluntary research; and the withdrawal group received the same treatment as chronic ethanol-treated rats until 240 days, after which they reverted to drinking water. Chronic ethanol ingestion induced increased lipoperoxide levels and acid phosphatase activities in seminal vesicles. Cu-Zn superoxide dismutase (SOD) decreased from its basal level 70.8 +/- 3.5 to 50.4 +/- 1.6 U/mg protein at 60 days of chronic ethanol ingestion. As changes in GSH-PX activity were observed in rats after chronic ethanol ingestion, while SOD activities were decreased in these animals, it is assumed that superoxide anion elicits lipoperoxide formation and induces cell damage before being converted to hydrogen peroxide by SOD. Ethanol withdrawal induced increased SOD activity and reduced seminar vesicle damage, indicating that the toxic effects were reversible, since increased SOD activity was adequate to scavenge superoxide radical formation. Superoxide radical is an important intermediate in the toxicity of chronic ethanol ingestion. Copyright (C) 1996 Elsevier B.V. Ltd
