Females of different species might exert female mate choice for different reasons, one of them the aim of avoiding inbreeding. In this study I examine the implication of inbreeding avoidance as a mechanism driving female mate choice in Verreaux’s sifaka lemurs (Propithecus verreauxi). In fact, in this species females are dominant and appear to be able to choose certain males to mate with, while observations indicate that rank, body size, canine size and proportions of fights won are not factors influencing female mate choice. So I hypothesized that females mate choice is driven by inbreeding avoidance in Verreaux’s sifaka lemurs. Tissue and fecal samples were collected in the Kirindy Mitea National Park in western Madagascar as a source of DNA. Parentage was assigned for a sample of the population and relatedness coefficients between dams and sires were estimated and compared to those of between random female and male pairs, dams and other candidate sires within the population and within the groups were the offspring were conceived. I found that there were no significant differences in none of the comparisons which means that Verreaux’s sifaka females do not mate more with males that are more distantly related to them. I concluded that inbreeding avoidance does not appear to be the main force driving female mate choice in Verreaux’s sifaka lemurs and I addressed explanations for these findings. With this study I contribute to our knowledge of female mate choice in lemurs.

Nas Américas, a leishmaniose visceral (LV) experimenta um processo de urbanização e o cão doméstico é considerado o principal reservatório da doença neste cenário, embora seu papel no ciclo de transmissão não esteja totalmente explicado. Este estudo teve como objetivo investigar, por meio da análise de dados espaciais e imagens de sensoriamento remoto, a relação de fatores ambientais com a ocorrência de infecção canina por Leishmania chagasi e sua correlação espacial com a doença humana na cidade de Teresina (Piauí - Brasil), onde foi relatada a primeira epidemia urbana de LV no Brasil. Os resultados são apresentados na forma de dois manuscritos, nos quais são utilizados dados georreferenciados obtidos por meio de um inquérito sorológico canino realizado durante o ano de 2011, em diferentes bairros com transmissão moderada ou intensa. No primeiro, a regressão logística multinível foi utilizada para correlacionar a prevalência da infecção canina com variáveis ambientais de quadrículas de 900m2 (30mx30m) onde os domicílios estavam localizados, ajustando para as características individuais dos cães (sexo, idade e raça) e da residência. Participaram desta análise 717 cães distribuídos em 494 domicílios e 396 quadrículas. Um percentual >16,5% da área da quadrícula coberta por pavimentação clara (ruas de terra ou asfalto antigo) foi a única variável ambiental associada com a infecção canina por L. chagasi (Odds ratio [OR] = 2,00, intervalo de 95% de confiança [IC95%]: 1,22 - 3,26). Estas áreas provavelmente correspondem àquelas mais pobres e com pior infraestrutura urbana, sugerindo a ocorrência de um padrão de transmissão intra-urbano similar aos padrões rurais e peri-urbanos da LV. No segundo manuscrito, a partir da análise hierárquica do vizinho mais próximo foi verificada a presença de sete clusters de maior concentração de cães soropositivos em relação aos soro negativos em áreas menos urbanizadas e com vegetação pouco densa. Participaram desta análise 322 cães distribuídos em cinco bairros. A relação espacial entre os caninos soropositivos e os casos humanos foi investigada através do método da distância média entre os pontos e analisada por meio do teste t. Foi encontrada uma maior proximidade de casos humanos em relação a cães soropositivos quando comparada à distância em relação aos soro negativos, sugerindo a existência de uma relação espacial entre a LV humana e a soropositividade canina. Os resultados contribuem para uma maior compreensão sobre a dinâmica da doença em meio urbano além de fornecer informações úteis para a prevenção e controle da LV em seres humanos.

As más oclusões de Classe I com biprotrusão dentária são caracterizadas pela projeção dos incisivos superiores e inferiores e podem ser corrigidas com extrações de primeiros pré-molares, permitindo reposicionar os incisivos, usando como ancoragem os dentes posteriores. Esse trabalho teve como objetivo comparar, por meio de superposição de modelos tridimensionais, a movimentação de molares e de incisivos com duas técnicas para fechamento de espaços. Foram selecionados, tomografados (T0) e distribuídos, aleatoriamente em dois grupos, seis pacientes com indicação de exodontia de quatro primeiros pré-molares. No grupo 1 (n=3) foi feito o fechamento dos espaços das extrações em uma única etapa (fechamento em massa) e no grupo 2 (n=3), em duas etapas (distalização de caninos seguida da retração de incisivos). Após o completo fechamento dos espaços foi solicitada nova tomografia (T1). Os modelos tridimensionais em T0 e T1 para os dois grupos foram superpostos para descrever e quantificar a movimentação ocorrida nos primeiros molares. A retração de incisivos foi avaliada de maneira bidimensional. Os molares superiores e inferiores do grupo 1 sofreram movimentação mesial de translação, enquanto os do grupo 2, apresentaram deslocamento de coroa maior do que de ápice radicular, demonstrando movimento de inclinação mesial de coroa. Ao se comparar o movimento mesial dos molares entre os dois grupos, observou-se que no grupo 1 houve maior movimentação mesial da raiz mésio-vestibular dos molares superiores (p=0,009). No entanto, em ambos os grupos os molares superiores giraram sobre a raiz palatina. No arco inferior verificou-se maior movimento mesial de coroa para o grupo 2 (p=0,015). A análise da movimentação dos incisivos apresentou variações semelhantes para as medidas lineares e angulares dos incisivos superiores nos dois grupos. Já, para as medidas angulares dos incisivos inferiores, observou-se que o grupo 1 apresentou menor controle de torque. O tempo de tratamento de todos os pacientes da amostra foi superior a 30 meses, exceto um paciente que apresentou tempo de tratamento inferior a 24 meses. Pôde-se concluir que a perda de ancoragem ocorreu por movimento de translação no fechamento de espaços em massa, e por inclinação mesial no fechamento de espaços em duas etapas, para ambos os arcos. Parece haver maior movimentação mesial radicular nos molares superiores no fechamento em massa e maior movimento mesial de coroa dos molares inferiores para o fechamento em duas etapas. Sugere-se que não há diferença na retração dos incisivos superiores e maior retroinclinação dos incisivos inferiores no fechamento de espaços em duas etapas.

Domestic cats and dogs are important companion animals and model animals in biomedical research. The cat has a highly conserved karyotype, closely resembling the ancestral karyotype of mammals, while the dog has one of the most extensively rearranged mammalian karyotypes investigated so far. We have constructed the first detailed comparative chromosome map of the domestic dog and cat by reciprocal chromosome painting. Dog paints specific for the 38 autosomes and the X chromosomes delineated 68 conserved chromosomal segments in the cat, while reverse painting of cat probes onto red fox and dog chromosomes revealed 65 conserved segments. Most conserved segments on cat chromosomes also show a high degree of conservation in G-banding patterns compared with their canine counterparts. At least 47 chromosomal fissions (breaks), 25 fusions and one inversion are needed to convert the cat karyotype to that of the dog, confirming that extensive chromosome rearrangements differentiate the karyotypes of the cat and dog. Comparative analysis of the distribution patterns of conserved segments defined by dog paints on cat and human chromosomes has refined the human/cat comparative genome map and, most importantly, has revealed 15 cryptic inversions in seven large chromosomal regions of conserved synteny between humans and cats.

Forty chromosome-specific paint probes of the domestic dog (Canis familiaris, 2n = 78) were used to delineate conserved segments on metaphase chromosomes of the American mink (Mustela vison, 2n = 30) by fluorescence in situ hybridisation. Half of the 38 canine autosomal probes each painted one pair of homologous segments in a diploid mink metaphase, whereas the other 19 dog probes each painted from two to five pairs of discrete segments. In total, 38 canine autosomal paints highlighted 71 pairs of conserved segments in the mink. These painting results allow us to establish a complete comparative chromosome map between the American mink and domestic dog. This map demonstrates that extensive chromosome rearrangements differentiate the karyotypes of the dog and American mink. The 38 dog autosomes could be reconstructed from the 14 autosomes of the American mink through at least 47 fissions, 25 chromosome fusions, and six inversions. Furthermore, comparison of the current dog/mink map with the published human/dog map discloses 23 cryptic intrachromosomal rearrangements in 10 regions of conserved synteny in the human and American mink genomes and thus further refined the human/mink comparative genome map. Sexual dimorphism in the dentition and skeleton of the four extant species of snub-nosed langurs, Rhinopithecus (R.) bieti, R. (R.) brelichi, R. (R.) roxellana and R. (Presbytiscus) avunculus, was studied. The species shared a similar general pattern of sexual dimorphism, but were found to differ in respects that appear to reflect the influence of disparate socioecological and environmental factors. All the species showed marked canine dimorphism, but the very high degree of canine dimorphism in R. bieti appeared to be due to the intensity of intermale competition for mates during a temporally restricted breeding season, and possibly also to the intensity of competition between males for other resources during other times of the year. Sexual dimorphism in the postcranial skeleton of Rhinopithecus species was also most pronounced in R. bieti and may be related to the relatively higher frequency of terrestrial locomotion in males of the species. The masticatory apparatus for two endemic species of golden monkey in China, Rhinopithecus bieti and Rhinopithecus roxellana, were compared with those of macaques, Macaca and leaf monkeys, Presbytis. Multivariate analyses demonstrated that the two golden monkey species are distinct. Interspecies allometric analyses revealed that golden monkeys differ in their masticatory apparatus from both macaques and leaf monkeys. The prominent symphysial fusion, corpus, and sagittal condylar dimension of R. roxellana may produce efficient biting force on the incisal and posterior canine teeth, with the heavy reaction force barn on the temporomandibular joint. However, the well-developed bizygamatic width and mandibular height in R. bieti suggest that posterior canine function is similarly prominent in R. roxellana, while incisal function is not. Dental variation in the Chinese golden monkey (Rhinopithecus roxellana) is here evaluated by univariate, bivariate, and multivariate analyses. Allometric analyses indicate that canines and P3s are positively, but other dimensions negatively scaled to mandible and maxilla, and to body size. With the exception of the mesiodistal dimensions of I-1 and M-3, and the buccolingual dimension of Pq, mandibular dental variables show similar scaling relative to body size. Analysis of residuals shows that males have significantly larger canine, P-3 and buccolingual dimensions of the postcanine teeth (M-2 and M-3) than females. A significant difference in shape between the sexes is found in the buccolingual dimension of the upper teeth, but not in the mandible. Unlike the situation in some other species, Female golden monkeys do nor exhibit relatively larger postcanine teeth than males, in fact, the reverse is true, especially for M(2)s and M(3)s. The fact that most of the dental variables show low negative allometry to body size might be related a cold environment that has led to the development of larger body size with I-educed energy loss. When the raw data are examined by Discriminant Function Analysis the sexes are clearly distinguishable.

The current study investigates the feasibility of using a biodegradable polymeric stent in common bile duct (CBD) repair and reconstruction. Here, poly(l-lactide-co-glycolide) (PLGA, molar ratio LA/GA = 80/20) was processed into a circular tube- and dumbbell-shaped specimens to determine the in vitro degradation behavior in bile. The morphology, weight loss, and molecular weight changes were then investigated in conjunction with evaluations of the mechanical properties of the specimen. Circular tube-shaped PLGA stents with X-ray opacity were subsequently used in common bile duct exploration (CBDE) and primary suturing in canine models. Next, X-ray images of CBD stents in vivo were compared and levels of serum liver enzymes and a histological analysis were conducted after stent transplantation. The results showed that the PLGA stents exhibited the required biomedical properties and spontaneously disappeared from CBDs in 4-5 weeks. The degradation period and function match the requirements in repair and reconstruction of CBDs to support the duct, guide bile drainage, and reduce T-tube-related complications.

Perfusion experiments on an isolated, canine lateral saphenous vein segment preparation have shown that noradrenaline causes potent, flow dependent effects, at a threshold concentration comparable to that of plasma noradrenaline, when it stimulates the segment by diffusion from its microcirculation (vasa vasorum). The effects caused are opposite to those neuronal noradrenaline causes in vivo and that, in the light of the principle that all information is transmitted in patterns that need contrast to be detected – star patterns need darkness, sound patterns, quietness – has generated the hypothesis that plasma noradrenaline provides the obligatory contrast tissues need to detect and respond to the regulatory information encrypted in the diffusion pattern of neuronal noradrenaline. Based on the implications of that hypothesis, the controlled variable of the peripheral noradrenergic system is believed to be the maintenance of a set point balance between the contrasting effects of plasma and neuronal noradrenaline on a tissue. The hypothalamic sympathetic centres are believed to monitor that balance through the level of afferent sympathetic traffic they receive from a tissue and to correct any deviation it detects in the balance by adjusting the level of efferent sympathetic input it projects to the tissue. The failure of the centres to maintain the correct balance, for reasons intrinsic or extrinsic to themselves, is believed to be responsible for degenerative and genetic disorders. When the failure causes the balance to be polarised in favour of the effect of plasma noradrenaline that is believed to cause inflammatory diseases like dilator cardiac failure, renal hypertension, varicose veins and aneurysms; when it causes it to be polarised in favour of the effect of neuronal noradrenaline that is believed to cause genetic diseases like hypertrophic cardiopathy, pulmonary hypertension and stenoses and when, in pregnancy, a factor causes the polarity to favour plasma noradrenaline in all the maternal tissues except the uterus and conceptus, where it favours neuronal noradrenaline, that is believed to cause preeclampsia.

We have previously shown that intracardiac acoustic radiation force impulse (ARFI) imaging visualizes tissue stiffness changes caused by radiofrequency ablation (RFA). The objectives of this in vivo study were to (1) quantify measured ARFI-induced displacements in RFA lesion and unablated myocardium and (2) calculate the lesion contrast (C) and contrast-to-noise ratio (CNR) in two-dimensional ARFI and conventional intracardiac echo images. In eight canine subjects, an ARFI imaging-electroanatomical mapping system was used to map right atrial ablation lesion sites and guide the acquisition of ARFI images at these sites before and after ablation. Readers of the ARFI images identified lesion sites with high sensitivity (90.2%) and specificity (94.3%) and the average measured ARFI-induced displacements were higher at unablated sites (11.23 ± 1.71 µm) than at ablated sites (6.06 ± 0.94 µm). The average lesion C (0.29 ± 0.33) and CNR (1.83 ± 1.75) were significantly higher for ARFI images than for spatially registered conventional B-mode images (C = -0.03 ± 0.28, CNR = 0.74 ± 0.68).

© 2015, Springer-Verlag Berlin Heidelberg.The emotional-reactivity hypothesis proposes that problem-solving abilities can be constrained by temperament, within and across species. One way to test this hypothesis is with the predictions of the Yerkes–Dodson law. The law posits that arousal level, a component of temperament, affects problem solving in an inverted U-shaped relationship: Optimal performance is reached at intermediate levels of arousal and impeded by high and low levels. Thus, a powerful test of the emotional-reactivity hypothesis is to compare cognitive performance in dog populations that have been bred and trained based in part on their arousal levels. We therefore compared a group of pet dogs to a group of assistance dogs bred and trained for low arousal (N = 106) on a task of inhibitory control involving a detour response. Consistent with the Yerkes–Dodson law, assistance dogs, which began the test with lower levels of baseline arousal, showed improvements when arousal was artificially increased. In contrast, pet dogs, which began the test with higher levels of baseline arousal, were negatively affected when their arousal was increased. Furthermore, the dogs’ baseline levels of arousal, as measured in their rate of tail wagging, differed by population in the expected directions. Low-arousal assistance dogs showed the most inhibition in a detour task when humans eagerly encouraged them, while more highly aroused pet dogs performed worst on the same task with strong encouragement. Our findings support the hypothesis that selection on temperament can have important implications for cognitive performance.

We investigated the efficacy of liver-directed gene therapy using lentiviral vectors in a large animal model of hemophilia B and evaluated the risk of insertional mutagenesis in tumor-prone mouse models. We showed that gene therapy using lentiviral vectors targeting the expression of a canine factor IX transgene in hepatocytes was well tolerated and provided a stable long-term production of coagulation factor IX in dogs with hemophilia B. By exploiting three different mouse models designed to amplify the consequences of insertional mutagenesis, we showed that no genotoxicity was detected with these lentiviral vectors. Our findings suggest that lentiviral vectors may be an attractive candidate for gene therapy targeted to the liver and may be potentially useful for the treatment of hemophilia.
