Le cancer du col utérin (CCU) est dans plus de 99% des cas provoqué par une infection avec le virus du papillome humain (VPH), dont le potentiel oncogénique réside dans l'expression des proto-oncogènes viraux E6/E7. Le potentiel carcinogénique de ces protéines virales réside essentiellement dans leurs actions sur les produits des gènes suppresseurs de tumeur p53 et RB. Les produits de ces gènes, p53 et Rb, font parti des voies de signalisation de réponse aux dommages de l'ADN cellulaire (RDA) et leur perte entraine une perte de fonctionnalité qui mène à une instabilité génomique. À long terme et en présence de d'autres facteurs ceux-ci mèneront au développement d'un cancer. Les protéines E6 et E7 sont constitutivement exprimées dans les cellules du CCU ainsi que dans les cellules de tout autre cancer induit par le VPH et seulement dans ces dernières. La prise en charge des cas avancés de ces cancers se fait principalement par radiothérapie et chimiothérapie concomitante. La chimio-radiothérapie utilisée en traitement est efficace mais résulte en un taux élevé de morbidité et un nombre important de patientes récidiveront. Nous proposons que l'exploitation de l'expression spécifique d’E6 et d’E7 dans les cellules du CCU permette d’envisager une stratégie de létalité synthétique afin d'amplifier l'effet létal de l'irradiation sur les cellules CCU. Ceci permettrait potentiellement d'augmenter l'efficacité du traitement et de diminuer les récidives, ainsi que la morbidité liée au traitement. En s'appuyant sur cette hypothèse, notre objectif est d’identifier des composés dont l'action seule ou couplée à l'irradiation provoquerait préférentiellement la mort des cellules exprimant les protéines E6 et E7 du VPH. Les cellules testées comprennent des cellules isogéniques humaines issues de kératinocytes normaux que nous avons modifiées séquentiellement pour obtenir les modifications associées aux cellules CCU (hTERT, E6 et E7), ainsi que les lignées de cellules de CCU HeLa et CaSki .Nous avons procédé à la mise au point et à la validation du protocole de criblage et des méthodes d’évaluation de la sensibilisation, qui se définit comme une perte de viabilité, un arrêt ou ralentissement de la croissance, par détection d’ATP ainsi que par coloration d’ADN génomique au DRAQ5. Suite à un criblage ciblé impliquant des inhibiteurs connus de la voie de réparation des dommages à l’ADN, nous avons identifié l’inhibiteur de mdm2, Nutlin-3, comme étant un composé sensibilisant et radio-sensibilisant préférentiellement les cellules exprimant E6 et E7 du VPH. La Nutlin-3 a été testée sur des cellules HEKn-hTERT-E6-E7, des cellules CaSki et HeLa. L’effet de sensibilisation et de radio-sensibilisation a été confirmé dans ces trois lignées. Tel que suggéré par son action sur mdmd2, la Nutlin-3 permet la stabilisation de p53 dans les cellules HEKn-hTERT-E6-E7 et CaSki et sa réactivation dans les lignées cellulaires HeLa et CaSki. Malgré cette stabilisation de p53, de façon surprenante, l’effet de la Nutlin-3 sur la sensibilisation et la radio-sensibilisation des cellules HeLa et CaSki semble indépendant de p53, tel qu’observé en utilisant des cellules HeLa-GSE et CaSki-GSE dont le p53 est déficient. In vivo la Nutlin-3a montre dans un essai préliminaire l’inhibition de la croissance tumorale des xénogreffes HeLa chez des souris RAG2γc. Ce résultat reste à confirmer avec un essai impliquant un nombre d’échantillons plus grand. À plus long terme, nous comptons étudier l’implication de mdm2 dans l’effet de sensibilisant de la Nutlin-3 dans les cellules CCUs, ainsi que les autres cibles pouvant être impliquées dans la création de cet effet sensibilisant observé.

Effect of pyridoxine on growth, metabolism and cellular activity of freshwater prawn Macrobrachiuni rosenbergii was studied. Postlarvae (PL-10) of M. rosenbergii were fed with clam meat containing various concentrations of pyridoxine. After 30 days RNA and DNA of the abdominal tissues were estimated. Length, weight and RNA to DNA ratio increased significantly with increasing concentrations of pyridoxine. The effect of pyridoxine on the metabolic enzyme, malate dehydrogenase, was also studied. Vmax showed a significant decrease and the (Km) showed a significant increase in experimental groups compared to control.

The increasing interest in the interaction of light with electricity and electronically active materials made the materials and techniques for producing semitransparent electrically conducting films particularly attractive. Transparent conductors have found major applications in a number of electronic and optoelectronic devices including resistors, transparent heating elements, antistatic and electromagnetic shield coatings, transparent electrode for solar cells, antireflection coatings, heat reflecting mirrors in glass windows and many other. Tin doped indium oxide (indium tin oxide or ITO) is one of the most commonly used transparent conducting oxides. At present and likely well into the future this material offers best available performance in terms of conductivity and transmittivity combined with excellent environmental stability, reproducibility and good surface morphology. Although partial transparency, with a reduction in conductivity, can be obtained for very thin metallic films, high transparency and simultaneously high conductivity cannot be attained in intrinsic stoichiometric materials. The only way this can be achieved is by creating electron degeneracy in a wide bandgap (Eg > 3eV or more for visible radiation) material by controllably introducing non-stoichiometry and/or appropriate dopants. These conditions can be conveniently met for ITO as well as a number of other materials like Zinc oxide, Cadmium oxide etc. ITO shows interesting and technologically important combination of properties viz high luminous transmittance, high IR reflectance, good electrical conductivity, excellent substrate adherence and chemical inertness. ITO is a key part of solar cells, window coatings, energy efficient buildings, and flat panel displays. In solar cells, ITO can be the transparent, conducting top layer that lets light into the cell to shine the junction and lets electricity flow out. Improving the ITO layer can help improve the solar cell efficiency. A transparent ii conducting oxide is a material with high transparency in a derived part of the spectrum and high electrical conductivity. Beyond these key properties of transparent conducting oxides (TCOs), ITO has a number of other key characteristics. The structure of ITO can be amorphous, crystalline, or mixed, depending on the deposition temperature and atmosphere. The electro-optical properties are a function of the crystallinity of the material. In general, ITO deposited at room temperature is amorphous, and ITO deposited at higher temperatures is crystalline. Depositing at high temperatures is more expensive than at room temperature, and this method may not be compatible with the underlying devices. The main objective of this thesis work is to optimise the growth conditions of Indium tin oxide thin films at low processing temperatures. The films are prepared by radio frequency magnetron sputtering under various deposition conditions. The films are also deposited on to flexible substrates by employing bias sputtering technique. The films thus grown were characterised using different tools. A powder x-ray diffractometer was used to analyse the crystalline nature of the films. The energy dispersive x-ray analysis (EDX) and scanning electron microscopy (SEM) were used for evaluating the composition and morphology of the films. Optical properties were investigated using the UVVIS- NIR spectrophotometer by recording the transmission/absorption spectra. The electrical properties were studied using vander Pauw four probe technique. The plasma generated during the sputtering of the ITO target was analysed using Langmuir probe and optical emission spectral studies.

The radio frequency plasma generated during the sputtering of Indium Tin Oxide target using Argon was analyzed by Langmuir probe and optical-emission spectroscopy. The basic plasma parameters such as electron temperature and ion density were evaluated. These studies were carried out by varying the RF power from 20 to 50 W. A linear increase in ion density and an exponential decrease in electron temperature with rf power were observed. The measured plasma parameters were then correlated with the properties of ITO thin films deposited under similar plasma conditions.

In recent years, there is a visible trend for products/services which demand seamless integration of cellular networks, WLANs and WPANs. This is a strong indication for the inclusion of high speed short range wireless technology in future applications. In this context UWB radio has a significant role to play as an extension/complement to existing cellular/access technology. In the present work, we have investigated two major types of wide band planar antennas: Monopole and Slot. Four novel compact broadband antennas, suitable for poratble applications, are designed and characterized, namely 1. Elliptical monopole 2. Inverted cone monopole 3. Koch fractal slot 4. Wide band slot The performance of these designs have been studied using standard simulation tools used in industry/academia and they have been experimentally verified. Antenna design guidelines are also deduced by accounting the resonances in each structure. In addition to having compact sized, high efficiency and broad bandwidth antennas, one of the major criterion in the design of impulse-UWB systems have been the transmission of narrow band pulses with minimum distortion. The key challenge is not only to design a broad band antenna with constant and stable gain but to maintain a flat group delay or linear phase response in the frequency domain or excellent transient response in time domain. One of the major contributions of the thesis lies in the analysis of the frequency and time-domain response of the designed UWB antennas to confirm their suitability for portable pulsed-UWB systems. Techniques to avoid narrowband interference by engraving narrow slot resonators on the antenna is also proposed and their effect on a nano-second pulse have been investigated.

The present investigation revealed three types of circulating haemocytes in the haemolymph of F. indicus: hyalinocytes, small-granule haemocytes, and large-granule haemocytes. Intermediate stages indicate the maturing process of a single cell. The presence of enzymes such as peroxidase, phenoloxidase and acid phosphatase in the haemocytes, and the substantial production of oxygen radicals during phagocytosis show that the haemocytes are capable of mounting a fme cellular defense mechanism. The enzyme activities of the serum and the presence of agglutinins in the serum, which may act as opsonins, agglutinate foreign particles and augment phagocytosis, confirm the presence of a superior humoral immune system in F. indicus.Bacterial infection caused considerable variations in the cellular and humoral factors, such as the number of circulating cells and haemagglutinating activity, especially in the initial hours of infection. The total haemocyte count, haemagglutination titer and phenoloxidase enzyme showed significant reductions on bacterial presence and could be used as indicators of bacterial infection.The number of circulating cells showed drastic fluctuation on exposure to pollutants. Nuvan at low concentrations was able to produce changes in the haemolymph factors and in the tissue organization, which implies that the animal is under stress and is easily prone to infections. Exposure to nuvan resulted in significant variation in all of the cellular and humoral factors, especially, the total haemocyte count, percentage of small granule haemocytes, phagocytic activity and the haemagglutinating activity, which might be good indicators of pesticide pollution. Heavy metal exposure caused significant increase in total haemocyte count and reduction in phenoloxidase enzyme activity Even changes in the physio-chemical parameters, such as salinity caused fluctuations in the defense factors, indicating stress in this euryhaline species. The dietary incorporation of a commercial immunostimulant containing P-l,3 glucan resulted in stimulation of some of the humoral defense factors of F indicus, but was time dependent. The modulations, on exposure to various external factors, in the cellular and humoral factors, especially, total haemocyte count, phagocytic activity, haemagglutinating activity and the phenoloxidase and acid phosphatase enzymes suggest that these parameters could be used as indicators of the health status of F indicus, which assist in better monitoring and effective health management of this important cultured species.

Numerous low - pressure systems form in the Arabian Sea and Bay of Bengal. These low-pressure systems are highly useful in bringing the rainfall over the Indian sub continent. The developments of these systems are accompanied by the reduction in air temperature and an increase in atmospheric humidity. The radio refractivity, which is a function of the atmospheric pressure, temperature and humidity, also changes following the development of these systems. Variation of radio refractive index and its vertical gradient are analysed for many low pressure systems formed over the Arabian Sea and Bay of Bengal. It is found that the atmosphere becomes super refractive associated with the formation of these systems, caused by the increase in humidity and decrease in temperature. The maximum gradient is observed near the surface layers, especially in the lowest 1 km. Super refraction leads to increased radar detection range and extension of radio horizon

Multimodal imaging agents that combine magnetic and fluorescent imaging capabilities are desirable for the high spatial and temporal resolution. In the present work, we report the synthesis of multifunctional fluorescent ferrofluids using iron oxide as the magnetic core and rhodamine B as fluorochrome shell. The core–shell structure was designed in such a way that fluorescence quenching due to the inner magnetic core was minimized by an intermediate layer of silica. The intermediate passive layer of silica was realized by a novel method which involves the esterification reaction between the epoxy group of prehydrolysed 3-Glyidoxypropyltrimethoxysilane and the surfactant over iron oxide. The as-synthesized ferrofluids have a high saturation magnetization in the range of 62–65 emu/g and were found to emit light of wavelength 640 nm ( excitation = 446 nm). Time resolved life time decay analysis showed a bi-exponential decay pattern with an increase in the decay life time in the presence of intermediate silica layer. Cytotoxicity studies confirmed the cell viability of these materials. The in vitro MRI imaging illustrated a high contrast when these multimodal nano probes were employed and the R2 relaxivity of these ∗Author to whom correspondence should be addressed. The carrier transport mechanism of polyaniline (PA) thin films prepared by radio frequency plasma polymerization is described in this paper. The mechanism of electrical conduction and carrier mobility of PA thin films for different temperatures were examined using the aluminium–PA–aluminium (Al–PA–Al) structure. It is found that the mechanism of carrier transport in these thin films is space charge limited conduction. J –V studies on an asymmetric electrode configuration using indium tin oxide (ITO) as the base electrode and Al as the upper electrode (ITO–PA–Al structure) show a diode-like behaviour with a considerable rectification ratio

Der Janus Kinase / signal transducer and activator of transcription (JAK/STAT) Signal- transduktionsweg wird für viele Entwicklungsvorgänge benötigt und spielt eine zentrale Rolle bei der Hämatopoese und bei der Immunantwort. Obwohl der JAK/STAT-Signalweg in den vergangenen Jahren Gegenstand intensiver Forschung war, erschwert die Redundanz des Signalwegs bei Wirbeltieren genetische Untersuchungen zur Identifizierung derjenigen Mechanismen, die den JAK/STAT-Signalweg regulieren. Der JAK/STAT-Signaltransduktionsweg ist evolutionär konserviert und ebenfalls bei der Taufliege Drosophila melanogaster vorhanden. Im Gegensatz zu Wirbeltieren ist der Signaltransduktionsweg von Drosophila weniger redundant und beinhaltet folgende Hauptkomponenten: den Liganden Unpaired (Upd), den Transmembranrezeptor Domeless (Dome), die einzige JAK-Tyrosinkinase Hopscotch (hop), sowie den Transkriptionsfaktor STAT92E. In der vorliegenden Arbeit wird die Rolle des JAK/STAT-Signalwegs bei der zellulären Proliferation mithilfe der Modellsysteme der Flügel- und der Augen-Imaginalscheiben von Drosophila charakterisiert. "Loss-of-function"- und "Gain-of-function"-Experimente zur Verminderung beziehungs-weise Erhöhung der Signalaktivität zeigten, dass der JAK/STAT-Signalweg eine Rolle bei der zellulären Proliferation der Flügel-Imaginalscheiben spielte, ohne die Zellgröße oder Apoptose zu verändern. Bei der Flügelentwicklung während des zweiten und des frühen dritten Larvalstadiums war die Aktivität des JAK/STAT-Signalwegs sowohl notwendig für die zelluläre Proliferation als auch hinreichend, um Überproliferation anzutreiben. Allerdings änderte sich während der späten dritten Larvalstadien die JAK/STAT-Signalaktivität, sodass endogene STAT92E-Mengen einen anti-proliferativen Effekt im gleichen Gewebe aufwiesen. Weiterhin reichte die ektopische Aktivierung des JAK/STAT-Signalwegs zu diesem späten Entwicklungszeitpunkt aus, um die Mitose zu inhibieren und die Zellen in der Phase G2 des Zellzyklus zu arretieren. Diese Ergebnisse legen den Schluss nahe, dass der JAK/STAT-Signalweg sowohl pro-proliferativ in frühen Flügelscheiben als auch anti-proliferativ zu späten Stadien der Flügelscheiben-Entwicklung wirken kann. Dieser späte anti-proliferative Effekt wurde durch einen nicht-kanonischen Mechanismus der STAT92E-Aktivierung vermittelt, da späte hop defiziente Zellverbände im Vergleich zu Wildtyp-Zellen keine Veränderungen im Ausmaß der zellulären Proliferation aufwiesen. Ferner konnte gezeigt werden, dass eine während der Larvalstadien exprimierte dominant-negative und im N-Terminus deletierte Form von STAT92E (?NSTAT92E) nicht für den anti-proliferativen Effekt verantwortlich ist. Diese Tatsache ist ein weiteres Indiz dafür, dass das vollständige STAT92E den späten anti-proliferativen Effekt verursacht. Um Modulatoren für die von JAK/STAT vermittelte zelluläre Proliferation zu identifieren, wurde ein P-Element-basierter genetischer Interaktions-Screen in einem sensibilisierten genetischen Hintergrund durchgeführt. Insgesamt wurden dazu 2267 unabhängige P-Element-Insertionen auf ihre Wechselwirkung mit der JAK/STAT-Signalaktivität untersucht und 24 interagierende Loci identifiziert. Diese Kandidaten können in folgende Gruppen eingeordnet werden: Zellzyklusproteine, Transkriptionsfaktoren, DNA und RNA bindende Proteine, ein Mikro-RNA-Gen, Komponenten anderer Signaltransduktionswege und Zelladhäsionsproteine. In den meisten Fällen wurden mehrere Allele der interagierenden Kandidatengene getestet. 18 Kandidatengene mit übereinstimmend interagierenden Allelen wurden dann zur weiteren Analyse ausgewählt. Von diesen 18 Kandidaten-Loci wurden 7 mögliche JAK/STAT-Signalwegskomponenten und 6 neue Zielgene des Signalwegs gefunden. Zusammenfassend wurde das Verständnis um STAT92E verbessert. Dieses Protein hat die gleiche Funktion wie das STAT3-Protein der Wirbeltiere und treibt die zelluläre Proliferation voran. Analog zu STAT1 hat STAT92E aber auch einen anti-proliferativen Effekt. Ferner wurden 24 mögliche Modulatoren der JAK/STAT-Signalaktivität identifiziert. Die Charakterisierung dieser Wechselwirkungen eröffnet vielversprechende Wege zu dem Verständnis, wie JAK/STAT die zelluläre Proliferation reguliert und könnte bei der Entwicklung von neuartigen therapeutischen Targets zur Behandlung von Krebskrankheiten und Entwicklungsstörungen beitragen.

Software Defined Radio (SDR) hardware platforms use parallel architectures. Current concepts of developing applications (such as WLAN) for these platforms are complex, because developers describe an application with hardware-specifics that are relevant to parallelism such as mapping and scheduling. To reduce this complexity, we have developed a new programming approach for SDR applications, called Virtual Radio Engine (VRE). VRE defines a language for describing applications, and a tool chain that consists of a compiler kernel and other tools (such as a code generator) to generate executables. The thesis presents this concept, as well as describes the language and the compiler kernel that have been developed by the author. The language is hardware-independent, i.e., developers describe tasks and dependencies between them. The compiler kernel performs automatic parallelization, i.e., it is capable of transforming a hardware-independent program into a hardware-specific program by solving hardware-specifics, in particular mapping, scheduling and synchronizations. Thus, VRE simplifies programming tasks as developers do not solve hardware-specifics manually.

Circadiane Schrittmacher koordinieren die täglichen Rhythmen in Physiologie und Verhalten in lebenden Organismen. Die Madeira Schabe Rhyparobia maderae (Synonym: Leucophaea maderae) ist ein gut etabliertes Modell, um die neuronalen Mechanismen der circadianen Rhythmen bei Insekten zu studieren. Die akzessorische Medulla (AME) in den optischen Loben des Gehirns wurde als das circadiane Schrittmacherzentrum der Madeira Schabe identifiziert, das circadiane Rhythmen in der Laufaktivität steuert. Über die Neurotransmitter der Eingangswege in das circadiane System der Madeira Schabe ist noch nicht viel bekannt. Das Hauptziel dieser Arbeit war es, mögliche Eingangssignale in die innere Uhr der Madeira Schabe zu bestimmen. An primären Zellkulturen von AME-Neuronen wurden Calcium-Imaging Experimente durchgeführt, um die Neurotransmitter-abhängigen Veränderungen in der intrazellulären Calcium-Konzentration zu messen. Darüber hinaus wurde die Signalkaskade des Neuropeptids Pigment Dispersing Factor (PDF), dem wichtigsten Kopplungsfaktor in circadianen Schrittmachern von Insekten, in Calcium-Imaging und Förster-Resonanzenergietransfer (FRET) Experimenten untersucht. Acetylcholin (ACh) erhöht die intrazelluläre Calcium-Konzentration in der Mehrzahl der circadianen Schrittmacherneurone der Madeiraschabe. Applikation von GABA, Serotonin und Octopamin erhöhten oder reduzierten die intrazelluläre Calcium-Konzentration in den AME-Neuronen, während Histamin und Glutamat die intrazelluläre Calcium-Konzentration ausschließlich reduzierten. Pharmakologische Experimente zeigten, dass die AME-Neurone ACh über ionotrope nikotinische ACh-Rezeptoren detektierten, während GABA über ionotrope GABAA-Rezeptoren und metabotrope GABAB-Rezeptoren detektiert wurde. Diese Ergebnisse deuten darauf hin, dass die circadiane Aktivität der Schabe durch verschiedene Eingänge, einschließlich ACh, GABA, Glutamat, Histamin, Octopamin und Serotonin, moduliert wird. Bei den FRET Studien wurde ein Proteinkinase A (PKA)-basierter FRET Sensor zur Detektion von cyclischem AMP (cAMP) verwendet. Es wurde gezeigt, dass PDF über Adenylylcyclase-abhängige und -unabhängige Signalwege wirken kann. Zusätzlich wurden Laufrad-Assays durchgeführt, um Phasenverschiebungen im Rhythmus der circadianen Laufaktivität zu detektieren, nachdem der Neurotransmitter Histamin zu verschiedenen circadianen Zeiten injiziert wurde. Histamin-Injektionen durch die Komplexaugen der Schabe ergaben eine biphasische Phasenantwortkurve (phase response curve) mit Phasenverzögerungen in der Laufaktivität am späten subjektiven Tag und am Beginn der subjektiven Nacht und Phasenbeschleunigungen in der späten subjektiven Nacht. Schließlich wurde eine extrazelluläre Ableittechnik an lebenden Schaben etabliert, die gleichzeitige Langzeit-Ableitungen von der AME, des Komplexauges (Elektroretinogramm = ERG), und der Beinmuskulatur (Elektromyogramm = EMG) für mehrere Tage ermöglichte.
