The Drosophila compound eye has provided a genetic approach to understanding the specification of cell fates during differentiation. The eye is made up of some 750 repeated units or ommatidia, arranged in a lattice. The cellular composition of each ommatidium is identical. The arrangement of the lattice and the specification of cell fates in each ommatidium are thought to occur in development through cellular interactions with the local environment. Many mutations have been studied that disrupt the proper patterning and cell fating in the eye. The eyes absent (eya) mutation, the subject of this thesis, was chosen because of its eyeless phenotype. In eya mutants, eye progenitor cells undergo programmed cell death before the onset of patterning has occurred. The molecular genetic analysis of the gene is presented.
The eye arises from the larval eye-antennal imaginal disc. During the third larval instar, a wave of differentiation progresses across the disc, marked by a furrow. Anterior to the furrow, proliferating cells are found in apparent disarray. Posterior to the furrow, clusters of differentiating cells can be discerned, that correspond to the ommatidia of the adult eye. Analysis of an allelic series of eya mutants in comparison to wild type revealed the presence of a selection point: a wave of programmed cell death that normally precedes the furrow. In eya mutants, an excessive number of eye progenitor cells die at this selection point, suggesting the eya gene influences the distribution of cells between fates of death and differentiation.
In addition to its role in the eye, the eya gene has an embryonic function. The eye function is autonomous to the eye progenitor cells. Molecular maps of the eye and embryonic phenotypes are different. Therefore, the function of eya in the eye can be treated independently of the embryonic function. Cloning of the gene reveals two cDNA's that are identical except for the use of an alternatively-spliced 5' exon. The predicted protein products differ only at the N-termini. Sequence analysis shows these two proteins to be the first of their kind to be isolated. Trangenic studies using the two cDNA's show that either gene product is able to rescue the eye phenotype of eya mutants.
The eya gene exhibits interallelic complementation. This interaction is an example of an "allelic position effect": an interaction that depends on the relative position in the genome of the two alleles, which is thought to be mediated by chromosomal pairing. The interaction at eya is essentially identical to a phenomenon known as transvection, which is an allelic position effect that is sensitive to certain kinds of chromosomal rearrangements. A current model for the mechanism of transvection is the trans action of gene regulatory regions. The eya locus is particularly well suited for the study of transvection because the mutant phenotypes can be quantified by scoring the size of the eye.
The molecular genetic analysis of eya provides a system for uncovering mechanisms underlying differentiation, developmentally regulated programmed cell death, and gene regulation.

This paper conducts an exposition on the field identification of Clariid catfishes Heterobranchus, Clarias and their hybrid as an important tool in fish breeding and genetics. The paper explained the classification and aquacultural importance of Clariid catfishes. Parameters necessary for fish identification were highlighted. The identification of Heterobranchus, Clarias, their hybrid and sexual differences were also identified. The paper is of the position that the identification of Heterobranchus, Clarias and their hybrid is important in their genetic conservation and in achieving success in breeding and genetic studies

Dosidicus gigas is a large pelagic cephalopod of the eastern Pacific that has recently undergone an unexpected, significant range expansion up the coast of North America. The impact that such a range expansion is expected to have on local fisheries and marine ecosystems has motivated a thorough study of this top predator, a squid whose lifestyle has been quite mysterious until recently. Unfortunately, Dosidicus spends daylight hours at depths prohibitive to making observations without significant artificial interference. Observations of this squid‟s natural behaviors have thus far been considerably limited by the bright illumination and loud noises of remotely-operated-vehicles, or else the presence of humans from boats or with SCUBA. However, recent technological innovations have allowed for observations to take place in the absence of humans, or significant human intrusion, through the use of animal-borne devices such as National Geographic‟s CRITTERCAM. Utilizing the advanced video recording and data logging technology of this device, this study seeks to characterize unknown components of Dosidicus gigas behavior at depth. Data from two successful CRITTERCAM deployments reveal an assortment of new observations concerning Dosidicus lifestyle. Tri-axial accelerometers enable a confident description of Dosidicus orientation during ascents, descents, and depth maintenance behavior - previously not possible with simple depth tags. Video documentation of intraspecific interactions between Dosidicus permits the identification of ten chromatic components, a previously undescribed basal chromatic behavior, and multiple distinct body postures. And finally, based on visualizations of spermatophore release by D. gigas and repetitive behavior patterns between squid pairs, this thesis proposes the existence of a new mating behavior in Dosidicus. This study intends to provide the first glimpse into the natural behavior of Dosidicus, establishing the groundwork for a comprehensive ethogram to be supported with data from future CRITTERCAM deployments. Cataloguing these behaviors will be useful in accounting for Dosidicus‟ current range expansion in the northeast Pacific, as well as to inform public interest in the impacts this expansion will have on local fisheries and marine ecosystems.

This thesis deals with the oligochaete taxa (Annelida, Clitellata) from several karst units in the Cantabrian region, northern Iberian Peninsula. Groundwater oligochaetes are still poorly known fauna and the area seems to be a promising hotspot for groundwater taxa. Metodology is based on both morphological and molecular analyses. More than 7,000 specimens were collected from five karst units and >60 taxa were identified. Stygobiont oligochaete fauna in the northern Iberian Peninsula is diverse and mostly endemic (range areas <300 km). Three new stygogiont oligochaete species are described: Gianius navarroi Rodriguez & Achurra, 2010, Isochaetides gianii Rodriguez & Achurra, 2010 and Troglodrilus jugeti Achurra et al., 2012; and another four new taxa will be described in the near future. Taxonomic remarks on Lophochaeta ignota Stolc, 1886 and Troglodrilus galarzai (Giani & Rodriguez, 1988) are provided. The controversial separation of L. ignota and Heterochaeta costata Claparède, 1863 from Tubifex Lamarck, 1816 is corroborated by mitochondrial molecular data. Following the DNA barcoding method, individuals of the stygoxene species Stylodrilus heringianus Claparède, 1862 from different geographic areas are shown to represent a single metapopulation. The first phylogenetic analysis of the subfamily Tubificinae based on molecular data is attemped, which although incompletely resolved, evidences for the first time a close relationship between a stygobiont oligochaete (Troglodrilus Juget et al. 2006) and an estuarine especies (Heterochaeta costata Claparéde, 1863). A marine ancestor is hypothesised for Troglodrilus. Finally, Ereñozar karst unit (Biscay) is suggested to be a hotspot for groundwater oligochaetes (11 stygobiont taxa, of which 4 are endemic to the karst unit) and several biodiversity indices (Species richness, Rarity, Vulnerability and Complementarity) are shown to be useful tools for conservation management of groundwater habitats in that karst area.

Coherent ecological networks (EN) composed of core areas linked by ecological corridors are being developed worldwide with the goal of promoting landscape connectivity and biodiversity conservation. However, empirical assessment of the performance of EN designs is critical to evaluate the utility of these networks to mitigate effects of habitat loss and fragmentation. Landscape genetics provides a particularly valuable framework to address the question of functional connectivity by providing a direct means to investigate the effects of landscape structure on gene flow. The goals of this study are (1) to evaluate the landscape features that drive gene flow of an EN target species (European pine marten), and (2) evaluate the optimality of a regional EN design in providing connectivity for this species within the Basque Country (North Spain). Using partial Mantel tests in a reciprocal causal modeling framework we competed 59 alternative models, including isolation by distance and the regional EN. Our analysis indicated that the regional EN was among the most supported resistance models for the pine marten, but was not the best supported model. Gene flow of pine marten in northern Spain is facilitated by natural vegetation, and is resisted by anthropogenic landcover types and roads. Our results suggest that the regional EN design being implemented in the Basque Country will effectively facilitate gene flow of forest dwelling species at regional scale.

Background: Contrary to what is generally thought schizophrenia is a very common mental health issue. For this, several animal models are used to assess the illness in order to develop a definitive. The most widely spread paradigm is the use of pharmacological models. Aim: The aim of this review is to display which are the most used insults for the assessment of social behaviour related negative symptoms in animal models as well as to ascertain which is the most adequate regime. Design: Literature review. Methods: PubMed database was used for this article by the search of the indexed “schizophrenia”, “animal models”, “social behaviour” and “negative symptoms” descriptors. With the exception of a single article due to it value this review is based on articles from 10 years onwards. Besides, only clinical trials and reviews written in English or Spanish and that had laboratory rodents as target population were accepted. Results: The studies assessed agree that pharmacological models (specially those regarding the NMDA receptor antagonists) are a valuable means for the experimental investigation of negative symptoms in schizophrenia with the necessity to emphasise that only some negative symptoms (anhedonia and social interaction, mainly) can be experimentally assessed. Conclusions: There is not enough evidence regarding the fours aspects of this review. PCP, Ketamine or MK-801 in sub-acute dosage regimes are currently the most indicated insults to mimic schizophrenic symptoms in rodents, although further research in needed, albeit other substances are valuable as well. The FOB-3, anew type fiber optic biosensor, is designed to rapidly detect a variety of biological agents or analytes with better stability, sensitivity and specificity. In order to detect Y. Pestis, a sandwich immunoassay was developed by using the purified antibody against antigen FI immobilized on polystyrene probes as the capture antibody and the monoclonal antibody-Cy5 conjugate as the detector. After a series of optimization for the stability, sensitivity and specificity of the FOB-3, 50-1000 ng/ml of antigen FI and 6 x 10(1)-6 x 10(7) CFU/ml Y. pestis could be detected constantly in about 20 min, and Y pestis could be detected specifically from Y. pseudotuberculosis, Y. enterocolitica, B. anthracis and E. coli. Then, 39 blind samples, including 27 tissues of mice infected with Y pestis and 12 tissues of healthy mice as negative control, were detected with the FOB-3. 92.6% infected tissues were identified from the tissues of healthy mice and the tissues containing more than 100 CFU/ml bacteria could be detected by the biosensor. The results demonstrated the feasibility of the FOB-3 as an effective method to detect Y. pestis rapidly and directly from the infected animal specimens with the advantage of portability, simple-operation as well as high sensitivity and specificity. Panulirus argus (Latreille, 1804) é uma das principais espécies de lagosta no Atlântico, sendo um dos maiores recursos pesqueiros do Atlântico Ocidental, onde apresenta um alto valor comercial. A forte explotação da espécie resulta em uma grande pressão sobre suas populações. Recentemente, foi descoberto que sob o binômio P. argus estão contidas duas espécies crípticas que ocorrem em alopatria, uma na região do Caribe e outra na costa brasileira. Esta tese tem como objetivo estudar como se estruturam geneticamente as populações dessas duas espécies, com o propósito de fornecer mais informações para a determinação de estoques e um correto manejo das espécies, e analisar os processos históricos evolutivos que moldaram suas histórias demográficas. Para tal, foram estudados dois marcadores mitocondriais (região controle e o gene da Citocromo Oxidase I) e loci de microssatélites de indivíduos de 7 regiões do Caribe (Florida, Bahamas, Turks e Caicos, Porto Rico, Cuba, Colômbia e Venezuela) e 11 estados do Brasil (Pará, Maranhão, Piauí, Ceará, Rio Grande do Norte, Pernambuco, Alagoas, Bahia, Espírito Santo, Rio de Janeiro e São Paulo). Dentro de cada espécie foram observadas duas linhagens mitocondriais diferentes, que co-ocorriam, de maneira homogênea, ao longo de suas distribuições. Hipotetiso que essas linhagens foram formadas a partir de um evento de vicariância com contato secundário ou como consequência de um efeito gargalo seguido de expansão. As duas linhagens são evidentes nas sequências da região controle mitocondrial, mas no gene da COI foram evidentes apenas em P. cf. argus do Caribe. As linhagens do Brasil se separaram há aproximadamente 233 - 288 mil anos e cada uma sofreu expansão em tempos diferentes, a primeira se expandiu há 100 mil anos e a segunda linhagem há 50 mil anos. As linhagens do Caribe se separaram cerca de 1 milhão de anos atrás e possuem o mesmo tempo de expansão, 50 mil anos. Os microssatélites não revelaram subdivisão populacional para nenhuma das duas espécies, porém os marcadores, juntos, sugeriram um fluxo gênico diferenciado entre localidades expostas a diferentes correntes marítimas. Considerando que essas lagostas são intensamente explotadas, é importante ser cuidadoso no momento de definir estoques pesqueiros. Para a espécie do Brasil, dois estoques pesqueiros foram sugeridos, o primeiro do Pará à Bahia e o segundo do sul da Bahia a São Paulo. Para a espécie do Caribe, foi mantida e reforçada a hipótese de quatro estoques sugerida pela FAO (Norte, Sul, Centro-Norte e Centro-Sul).

Objective: Although dobutamine is widely used in neonatal clinical practice, the evidence for its use in this specific population is not clear. We conducted a systematic review of the use of dobutamine in juvenile animals to determine whether the evidence from juvenile animal experiments with dobutamine supported the design of clinical trials in neonatal/ paediatric population. Methods: Studies were identified by searching MEDLINE (1946-2012) and EMBASE (1974-2012). Articles retrieved were independently reviewed by three authors and only those concerning efficacy and safety of the drug in juvenile animals were included. Only original articles published in English and Spanish were included. Results: Following our literature search, 265 articles were retrieved and 24 studies were included in the review: 17 focused on neonatal models and 7 on young animal models. Although the aims and design of these studies, as well as the doses and ages analysed, were quite heterogeneous, the majority of authors agree that dobutamine infusion improves cardiac output in a dose dependent manner. Moreover, the cardiovascular effects of dobutamine are influenced by postnatal age, as well as by the dose used and the duration of the therapy. There is inadequate information about the effects of dobutamine on cerebral perfusion to draw conclusions. Conclusion: There is enough preclinical evidence to ensure that dobutamine improves cardiac output, however to better understand its effects in peripheral organs, such as the brain, more specific and well designed studies are required to provide additional data to support the design of clinical trials in a paediatric population.
