Foam nest production in Hoplosternum littorale is described in detail. This fish produces foam, while swimming belly-up, by pumping water over the gills and using their pelvic fins to mix water and mucus. The bubbles resulting from this process are added to plant debris carried by the male, which possesses a specialized pectoral fin spine to perform such activity. Promoter hypermethylation of CDKN2A (p16INK4A protein) is the main mechanism of gene inactivation. However, its association with Helicobacter pylori infection is a controversial issue. Therefore, we examined a series of gastric adenocarcinomas to assess the association between p16INK4A inactivation and H. pylori genotype (vacA, cagA, cagE, virB11 and flaA) according to the location and histological subtype of the tumors. p16INK4A expression and CDKN2A promoter methylation were found in 77 gastric adenocarcinoma samples by immunohistochemistry and methylation-specific PCR, respectively. Helicobacter pylori infection and genotype were determined by PCR. A strong negative correlation between immunostaining and CDKN2A promoter region methylation was found. In diffuse subtype tumors, the inactivation of p16INK4A by promoter methylation was unique in noncardia tumors (p = 0.022). In addition, H. pylori-bearing flaA was associated with non-methylation tumors (p = 0.008) and H. pylori strain bearing cagA or vacAs1m1 genes but without flaA was associated with methylated tumors (p = 0.022 and 0.003, respectively). Inactivation of p16INK4A in intestinal and diffuse subtypes showed distinct carcinogenic pathways, depending on the tumor location. Moreover, the process of methylation of the CDKN2A promoter seems to depend on the H. pylori genotype. The present data suggest that there is a differential influence and relevance of H. pylori genotype in gastric cancer development.

Objective: We aimed to evaluate the inactivation of COX-2, HMLH1 and CDKN2A by promoter methylation and its relationship with the infection by different Helicobacter pylori strains in gastric cancer. Methods: DNA extracted from 76 H. pylori-positive gastric tumor samples was available for promoter methylation identification by methylation-specific PCR and H. pylori subtyping by PCR. Immunohistochemistry was used to determine COX-2, p16(INK4A) and HMLH1 expression. Results: A strong negative correlation was found between the expression of these markers and the presence of promoter methylation in their genes. Among cardia tumors, negativity of p16(INK4A) was a significant finding. on the other hand, in noncardia tumors, the histological subtypes had different gene expression patterns. In the intestinal subtype, a significant finding was HMLH1 inactivation by methylation, while in the diffuse subtype, CDKN2A inactivation by methylation was the significant finding. Tumors with methylated COX-2 and HMLH1 genes were associated with H. pylori vac A s1 (p = 0.025 and 0.047, respectively), and the nonmethylated tumors were associated with the presence of the gene flaA. Conclusions: These data suggest that the inactivation of these genes by methylation occurs by distinct pathways according to the histological subtype and tumor location and depends on the H. pylori genotype. The T-2 and the generalized variance vertical bar S vertical bar charts are used for monitoring the mean vector and the covariance matrix of multivariate processes. In this article, we propose for bivariate processes the use of the T-2 and the VMAX charts. The points plotted on the VMAX chart correspond to the maximum of the sample variances of the two quality characteristics. The reason to consider the VMAX statistic instead of the generalized variance vertical bar S vertical bar is the user's familiarity with the computation of simple sample variances; we can't say the same with regard to the computation of the generalized variance vertical bar S vertical bar.

During the growth of Kluyveromyces marxianus var. marxianus ATCC 10022 on lactose, peaks of glucose, but not β-galactosidase activity, were detected in culture medium. Harvested and washed whole cells produced glucose and galactose from lactose, or ortho-nitro-phenol from the chromogenic substrate ortho-nitro-phenyl-β-D-galactopyranoside (ONPG), indicating that β-galactosidase is physically associated with cells. ONPG hydrolysis by whole cells presented a monophasic kinetics (Km 36.6 mM) in lactose exponential growth phase cells, but a biphasic kinetics (Km 0.2 and 36.6 mM) in stationary growth phase cells. Permeabilization with digitonin or disruption of cells from both growth phases led to monosite ONPG hydrolysis (Km 2.2 to 2.5 mM), indicating that β-galactosidase is not located in the periplasm. In addition, the energy inhibitors fluoride or arsenate, as well as the uncouplercarbonyl cyanide m-chlorophenylhydrazone (CCCP) prevented ONPG hydrolysis by whole cells. These findings indicate that energy coupled transmembrane transport is the rate-limiting step for intracellular ONPG cleavage. The taxonomic and physiologic implications of the exclusive intracellular location of β-galactosidase of K. marxianus var. marxianus ATCC 10022 are discussed. © 1996 Kluwer Academic Publishers.

Agonistic encounters and facultative parental care in Hyla faber were observed in two localities in southeastern Brazil. Maximum male density was 0.9 and 3.3 males/m2 in Campinas and Ribeirão Branco, respectively. Aggression was escalated and the highly variable aggressive calls were specific to each phase of the encounter. The last, more aggressive phases rarely occurred in Campinas; in Ribeirão Branco they occurred frequently. Male parental care (egg attendance) was common in Ribeirão Branco while it was never observed in Campinas. Egg attendance lasted one to two nights and was observed only during high male density. The main benefit of egg attendance seemed to be avoiding nest intrusion by other males (sunken eggs and/or embryos invariably die). Males may build additional nests during egg attendance, but attending males did not attract females (they did not call).

The breeding biology of the only Scarlet Ibis Eudocimus ruber colony in southeastern Brazil was studied during the 1996-97 breeding season. The ibises began to visit their colony site by mid-September. Nest building and egg laying took place in early November and was synchronous, making the first nesting pulse. Mean clutch size in this pulse was 2.45 eggs/nest, and 0.67 young/nest reached age three weeks, when they were able to walk about the nest tree and environs. Predation was the main cause of nest failures (74% of all losses), followed by nest collapses (19%). A second nesting pulse, also synchronous, started in late December, when the young from the first nests were already able to wander about the colony and make short flights. Mean clutch size of this pulse was 2.05 eggs/nest and productivity was 0.34 young/nest. Nest collapses during storms accounted for 58% of the losses, and predation for a further 27%. A third pulse, with only a few nests, started when the second pulse young were in their third week, but no nest was successful. The incubation time was 21-24 days, and the young were able to fly well when 40 days old, deserting the colony by age 75 days. Nesting early in the breeding season yielded greater success. Nests were built close to each other (a sphere with a 1.8 m radius and centered on an average nest would include the four nearest neighbors) and there was always more than one nest per tree. Most nests were built on the upper third of the nest-tree and had some cover from overhanging branches. There was a trend for the ibises building their nests in even closer proximity during the second pulse, perhaps as a strategy to lessen individual predation risks. A pair of Planalto Woodcreepers (Dendrocolaptes platyrostris) fed young in a nest cavity 6 m up in a wooded botanic garden of an old eucalyptus grove near São Paulo, Brazil, during October 1982. The pair often visited together at first, one (likely male) raising its head feathers as noted in other species of the genus. Feedings were less frequent at midday. Songs of the southeastern moist forest D. p. platyrostris were strikingly different from inland dry forest D. p. intermedius, despite intermediate specimens where the two rather distinctly colored forms come together. There are several poorly studied forms in interior dry forests, which need much more conservation effort.
