The antioxidant effects of beta-carotene, oil-soluble (bixin) and water-soluble (norbixin) annatto preparations and mixtures of these carotenoids with virgin olive oil polar extract were assessed in bulk olive oil and oil-in-water emulsions stored at 60degreesC. Norbixin was the only carotenoid that inhibited the oxidative deterioration of lipids in both systems. Though bixin and beta-carotene did not retard autoxidation, their mixtures with the polar extract from virgin olive oil enhanced the antioxidant effect of the olive oil extract. Norbixin (2 mM) was of similar activity to delta-tocopherol (0.1 mM) in stored oil. The combination of norbixin with ascorbic acid or ascorbyl palmitate in oil showed a reduction in formation of volatile oxidation products but not in peroxide value, compared with the analogous sample lacking norbixin. In olive oil-in-water emulsions, norbixin (2 mM) reduced hydroperoxide formation to a similar extent as delta-tocopherol (0.1 mM), which in turn was a better antioxidant than alpha-tocopherol. A synergistic effect between norbixin and ascorbic acid or ascorbyl palmitate was observed in the emulsion systems. (C) 2003 Elsevier Ltd. All rights reserved.

Foods are complex biological materials, and the lipids within the food are susceptible to lipid oxidation, which is retarded by antioxidants. The precise structure and composition of the food may affect the antioxidant activity quite strongly in some cases. Solubility of the antioxidant in the phases present is one of the main parameters that affects the variation in antioxidant activity with phase composition of food. Polar antioxidants are more effective in oils, whereas non-polar antioxidants are more effective in oil-in-water emulsions. Antioxidant activity has been reported in a range of different media, including oils, emulsions, liposomes, microemulsions, fish and meat muscles, and the antioxidant activity may vary from one medium to another. Synergy between antioxidants may also vary from one medium to another. Interactions with metals and with proteins affect antioxidant activity and these interactions are also dependent on the phases present.

A poor representation of cloud structure in a general circulation model (GCM) is widely recognised as a potential source of error in the radiation budget. Here, we develop a new way of representing both horizontal and vertical cloud structure in a radiation scheme. This combines the ‘Tripleclouds’ parametrization, which introduces inhomogeneity by using two cloudy regions in each layer as opposed to one, each with different water content values, with ‘exponential-random’ overlap, in which clouds in adjacent layers are not overlapped maximally, but according to a vertical decorrelation scale. This paper, Part I of two, aims to parametrize the two effects such that they can be used in a GCM. To achieve this, we first review a number of studies for a globally applicable value of fractional standard deviation of water content for use in Tripleclouds. We obtain a value of 0.75 ± 0.18 from a variety of different types of observations, with no apparent dependence on cloud type or gridbox size. Then, through a second short review, we create a parametrization of decorrelation scale for use in exponential-random overlap, which varies the scale linearly with latitude from 2.9 km at the Equator to 0.4 km at the poles. When applied to radar data, both components are found to have radiative impacts capable of offsetting biases caused by cloud misrepresentation. Part II of this paper implements Tripleclouds and exponential-random overlap into a radiation code and examines both their individual and combined impacts on the global radiation budget using re-analysis data.

Tissue culture in the oil palm business is generally concerned with the multiplication (clonal production) of dura, pisifera and tenera palms. These are all normal diploids (2n=2x=36). Sumatra Bioscience has pioneered haploid tissue culture of oil palm (n=x=18). Haploid oil palm is the first step in producing doubled haploid palms which in turn provide parental lines for F1 hybrid production. Chromosome doubling is known to occur during embryogenesis in other haploid cultures, e.g. barley anther culture. Haploid tissue cultures in oil palm were therefore set up to investigate and exploit spontaneous chromosome doubling during embryogenesis. Flow cytometry of embryogenic tissue showed the presence of both haploid (n) and doubled haploid (2n) cells indicating spontaneous doubling. Completely doubled haploid ramets were regenerated suggesting that doubling occurred during the first mitoses of embryogenesis. This is the first report of doubled haploid production in oil palm via haploid tissue culture. The method provides a means of producing a range of doubled haploids in oil palm from the 1,000 plus haploids available at Sumatra Bioscience, in addition the method also produced doubled haploid (and haploid) clones. 1.

Microencapsulation of drugs into preformed polymers is commonly achieved through solvent evaporation techniques or spray drying. We compared these encapsulation methods in terms of controlled drug release properties of the prepared microparticles and investigated the underlying mechanisms responsible for the “burst release” effect. Using two different pH-responsive polymers with a dissolution threshold of pH 6 (Eudragit L100 and AQOAT AS-MG), hydrocortisone, a model hydrophobic drug, was incorporated into microparticles below and above its solubility within the polymer matrix. Although, spray drying is an attractive approach due to rapid particle production and relatively low solvent waste, the oil-in-oil microencapsulation method is superior in terms of controlled drug release properties from the microparticles. Slow solvent evaporation during the oil-in-oil emulsification process allows adequate time for drug and polymer redistribution in the microparticles and reduces uncontrolled drug burst release. Electron microscopy showed that this slower manufacturing procedure generated non-porous particles whereas thermal analysis and X-ray diffractometry showed that drug loading above the solubility limit of the drug in the polymer generated excess crystalline drug on the surface of the particles. Raman spectral mapping illustrated that drug was homogeneously distributed as a solid solution in the particles when loaded below saturation in the polymer with consequently minimal burst release.

Neuroprostheses interfaced with transected peripheral nerves are technological routes to control robotic limbs as well as convey sensory feedback to patients suffering from traumatic neural injuries or degenerative diseases. To maximize the wealth of data obtained in recordings, interfacing devices are required to have intrafascicular resolution and provide high signal-to-noise ratio (SNR) recordings. In this paper, we focus on a possible building block of a three-dimensional regenerative implant: a polydimethylsiloxane (PDMS) microchannel electrode capable of highly sensitive recordings in vivo. The PDMS 'micro-cuff' consists of a 3.5 mm long (100 µm × 70 µm cross section) microfluidic channel equipped with five evaporated Ti/Au/Ti electrodes of sub-100 nm thickness. Individual electrodes have average impedance of 640 ± 30 kΩ with a phase angle of −58 ± 1 degrees at 1 kHz and survive demanding mechanical handling such as twisting and bending. In proof-of-principle acute implantation experiments in rats, surgically teased afferent nerve strands from the L5 dorsal root were threaded through the microchannel. Tactile stimulation of the skin was reliably monitored with the three inner electrodes in the device, simultaneously recording signal amplitudes of up to 50 µV under saline immersion. The overall SNR was approximately 4. A small but consistent time lag between the signals arriving at the three electrodes was observed and yields a fibre conduction velocity of 30 m s−1. The fidelity of the recordings was verified by placing the same nerve strand in oil and recording activity with hook electrodes. Our results show that PDMS microchannel electrodes open a promising technological path to 3D regenerative interfaces.

Introduction Facing the challenging treatment of neurodegenerative diseases as well as complex craniofacial injuries such as those common after cancer therapy, the field of regenerative medicine increasingly relies on stem cell transplantation strategies. Here, neural crest-derived stem cells (NCSCs) offer many promising applications, although scale up of clinical-grade processes prior to potential transplantations is currently limiting. In this study, we aimed to establish a clinical-grade, cost-reducing cultivation system for NCSCs isolated from the adult human nose using cGMP-grade Afc-FEP bags. Methods We cultivated human neural crest-derived stem cells from inferior turbinate (ITSCs) in a cell culture bag system using Afc-FEP bags in human blood plasma-supplemented medium. Investigations of viability, proliferation and expression profile of bag-cultured ITSCs were followed by DNA-content and telomerase activity determination. Cultivated ITSCs were introduced to directed in vitro differentiation assays to assess their potential for mesodermal and ectodermal differentiation. Mesodermal differentiation was determined using an enzyme activity assay (alkaline phosphatase, ALP), respective stainings (Alizarin Red S, Von Kossa and Oil Red O), and RT-PCR, while immunocytochemistry and synaptic vesicle recycling were applied to assay neuroectodermal differentiation of ITSCs. Results When cultivated within Afc-FEP bags, ITSCs grew three-dimensionally in a human blood plasma-derived matrix, thereby showing unchanged morphology, proliferation capability, viability and expression profile in comparison to three dimensionally-cultured ITSCs growing in standard cell culture plastics. Genetic stability of bag-cultured ITSCs was further accompanied by unchanged telomerase activity. Importantly, ITSCs retained their potential to differentiate into mesodermal cell types, particularly including ALP-active, Alizarin Red S-, and Von Kossa-positive osteogenic cell types, as well as adipocytes positive in Oil Red O assays. Bag culture further did not affect the potential of ITSCs to undergo differentiation into neuroectodermal cell types coexpressing β-III-tubulin and MAP2 and exhibiting the capability for synaptic vesicle recycling. Conclusions Here, we report for the first time the successful cultivation of human NCSCs within cGMP-grade Afc-FEP bags using a human blood plasma-supplemented medium. Our findings particularly demonstrate the unchanged differentiation capability and genetic stability of the cultivated NCSCs, suggesting the great potential of this culture system for future medical applications in the field of regenerative medicine.

Avian intestinal spirochaetosis (AIS) caused by Brachyspira spp., and notably Brachyspira pilosicoli, is common in layer flocks and reportedly of increasing incidence in broilers and broiler breeders. Disease manifests as diar- rhoea, increased feed consumption, reduced growth rates and occasional mortality in broilers and these signs are shown in layers also associated with a delayed onset of lay, reduced egg weights, faecal staining of eggshells and non-productive ovaries. Treatment with Denagard® Tiamulin has been used to protect against B. pilosicoli colonisation, persistence and clinical presentation of AIS in commercial layers, but to date there has been no de- finitive study validating efficacy. Here, we used a poultry model of B. pilosicoli infection of layers to compare the impact of three doses of Denagard® Tiamulin. Four groups of thirty 17 week old commercial pre-lay birds were all challenged with B. pilosicoli strain B2904 with three oral doses two days apart. All birds were colonised within 2 days after the final oral challenge and mild onset of clinical signs were observed thereafter. A fifth group that was unchallenged and untreated was also included for comparison as healthy birds. Five days after the final oral Brachypira challenge three groups were given Denagard® Tiamulin in drinking water made up following the manufacturer's recommendations with doses verified as 58.7 ppm, 113 ppm and 225 ppm. Weight gain body condition and the level of diarrhoea of birds infected with B. pilosicoli were improved and shedding of the organism reduced significantly (p = 0.001) following treatment with Denagard® Tiamulin irrespective of dose given. The level and duration of colonisation of organs of birds infected with B. pilosicoli was also reduced. Confirming previous findings we showed that the ileum, caeca, colon, and both liver and spleen were colonised and here we demonstrated that treatment with Denagard® Tiamulin resulted in significant reduction in the numbers of Brachyspira found in each of these sites and dramatic reduction in faecal shedding (p b 0.001) to ap- proaching zero as assessed by culture of cloacal swabs. Although the number of eggs produced per bird and the level of eggshell staining appeared unaffected, egg weights of treated birds were greater than those of untreated birds for a period of approximately two weeks following treatment. These data conclusively demonstrate the ef- fectiveness of Denagard® Tiamulin in reducing B. pilosicoli infection in laying hens.

In a numerical, isopycnal, ocean model the mixing is investigated with the environment of two idealized Agulhas rings, one that splits, and one that remains coherent. The evolution of a passive tracer , initially contained within the rings, shows that tracer leakage is associated with the for mation of filaments in the early stage of ring evolution. These filaments reach down to the ther mocline. In the deepest layers leakage occurs on a larger scale. Self-advection of the rings is ver y irregular , and it is not possible to compute a Lagrangian boundar y i n order to estimate the transport of leakage from the rings. T o describe the processes that gover n tracer leakage, in a coordinate frame moving with the ring a kinematic separatrix is defined in the streamfunction field for the nondivergent flow . Initially , filaments arise because of the elongation of the ring, which is mainly gover ned by an m 5 2 instability that is collaborating with differential rotation. Because of beta, the symmetr y i s destroyed related to the separatrix associated with a stagnation point in the flow . The filament upstream of the stagnation point grows much faster and is associated with the bulk of tracer leakage. Mixing is enhanced by time dependence of the separatrix. As a result, there are no large differences between the leakage from a coherent ring, where the m 5 2 instability equilibrates, and from a splitting ring, where the m 5 2 instability keeps growing, which confir ms that the amount of leakage is mainly gover ned by the ring’ s initial defor mation combined with unsteady self-advection of the ring and not by the splitting of the ring. The decay of tracer content in the ther mocline shows that in the first months up to 40% of the ring water can be mixed with the environment. In deeper layers the decay of tracer content may reach up to 90%.

Until the beginning of the 1900:th century the paper making process was handicraft. The paper machines that have been developed since then are as impressing in size as fascinating when it comes to the technique. The process has always been improved to give a better paper for lower costs, with as short manufacture time as possible. Stratified forming has been a reality since 1830. From the beginning it was applied to board and paperboard. Now it is also the most common method for making tissue and one paper mill in Austria use it for fine paper. Stratified forming can be categorized as separate forming or simultaneous forming. Separate forming means using several headboxes and sometimes even several wires. Simultaneous forming means using only one stratified headbox. This method has many advantages over separate forming, inter alia improved economy, quality, wood exchange and higher runability of the paper machine. Several experiments have been done with simultaneous forming of different fibres by placing a fibre with high bulk in the middle layer and a smooth fibre on the surface layers. The results has shown that by using this method both the bending stiffness and surface properties are improved, or a lower grammage paper is obtained with maintaining quality. Simultaneous forming can also be used to stratify fillers in layers where they are most efficient. In that way both paper quality and economy is improved. Simultaneous forming is also used for fractionated pulp. Fractionation means separating springwood fibre from summerwood fibre, and placing them in separate layers. Research results of fractionated pulp show about the same result as simultaneous forming of different fibres.

Durante a análise sísmica de estruturas complexas, o modelo matemático empregado deveria incluir não só as distribuicões irregulares de massas e de rigidezes senão também à natureza tridimensional da ecitação sísmica. Na prática, o elevado número de graus de liberdade involucrado limita este tipo de análise à disponibilidade de grandes computadoras. Este trabalho apresenta um procedimento simplificado, para avaliar a amplificação do movimento sísmico em camadas de solos. Sua aplicação permitiria estabelecer critérios a partir dos quais avalia-se a necessidade de utilizar modelos de interação solo-estrutura mais complexos que os utilizados habitualmente. O procedimento proposto possui as seguientes características : A- Movimento rígido da rocha definido em termos de três componentes ortagonais. Direção de propagação vertical. B- A ecuação constitutiva do solo inclui as características de não linearidade, plasticidade, dependência da história da carga, dissipação de energia e variação de volume. C- O perfil de solos é dicretizado mediante um sistema de massas concentradas. Utiliza-se uma formulação incremental das equações de movimento com integração directa no domínio do tempo. As propriedades pseudo-elásticas do solo são avaliadas em cada intervalo de integração, em função do estado de tensões resultante da acção simultânea das três componentes da excitação. O correcto funcionamento do procedimento proposto é verificado mediante análises unidimensionais (excitação horizontal) incluindo estudos comparativos com as soluções apresentadas por diversos autores. Similarmente apresentam-se análises tridimensionais (acção simultânea das três componentes da excitação considerando registros sísmicos reais. Analisa-se a influência que possui a dimensão da análise (uma análise tridimensional frente a três análises unidimensionais) na resposta de camadas de solos submetidos a diferentes níveis de exçitação; isto é, a limitação do Princípio de Superposisão de Efeitos.

Trata-se nesta dissertação do emprego da reciclagem a frio de revestimentos asfálticos em usina com a finalidade de aproveitamento como camada de revestimento em vias de tráfego leve e muito leve. São revistos os conceitos de dimensionamento de pavimentos e da dosagem de misturas asfálticas. Dá-se especial atenção ao valor do revestimento asfáltico recuperado e a relevância que a reciclagem tem sobre a redução do descarte de resíduos sólidos no meio ambiente. Identifica-se a importância do emprego da reciclagem sobre o comportamento da sociedade. A metodologia adotada tem por base uma revisão bibliográfica dos registros de trabalhos de reciclagem a frio realizados no Brasil e no exterior. Faz-se um relato dos procedimentos de caracterização dos materiais envolvidos e dos cuidados a serem observados para obter-se um produto final de qualidade. Estão aqui descritos os processos industriais de usinagem da mistura reciclada em plantas industriais fixas. É estabelecida uma proposta de definição da viabilidade econômica de emprego da reciclagem a frio em comparação com o revestimento asfáltico tradicional. Ao final são registrados temas a serem estudados visando tornar a reciclagem a frio de revestimentos asfálticos em usinas de áreas urbanas mais conhecida e considerada como alternativa para a melhoria da qualidade de vida nas cidades.

Este trabalho teve como objetivo incluir flexibilidades gerenciais (tais como técnicas de injeção de gás e água) na avaliação de reservatórios. Concluimos que esta técnicas podem aumentar o valor dos reservatórios em até 25% segundo a teoria de opções reais. A principal vantagem da metodologia de teoria de opções face a tradicional técnica de fluxo de caixa descontado é levar em conta as questões operacionais da indústria do petróleo. Utilizamos dois modelos clássicos para a precificação de reservatórios de petróleo, e aplicamos uma análise de sensibilidade para determinarmos quais fatores são mais relevantes no seu valor econômico. Como era de se esperar em ambos os modelos, o tempo de concessão, bem como a taxa de convenience e/ou dividend yield foram os fatores mais importantes.

A PETROBRAS, conhecedora das mudanças ocorridas no ambiente de negócios durante a década de 90, esforça-se em aproveitar as oportunidades surgidas ou aquelas que ainda estão por aparecer no setor petroleiro. Tal esforço traduziu-se na formulação do Plano Estratégico do Sistema PETROBRAS 199212001. Este plano define objetivos e estratégias decenais. Inserida no Plano Estratégico, está a questão da automação da companhia, considerada de suma importância para o seu desempenho no setor. Dentre os 14 Projetos Estratégicos do Plano temos o Projeto Estratégico de Automação (PEA), e para a implementação de seus objetivos é fundamental o desempenho dos funcionários neste Projeto. O desempenho do quadro funcional irá influenciar decisivamente o caminho a ser percorrido pela empresa rumo à modernidade, em um ambiente de negócios altamente renovado pela tecnologia de base microeletrônica. Uma das variáveis , que permitirá ou não a efetividade da PETROBRAS está relacionada à sua Cultura Técnica, ou seja, ao conjuntod,e conhecimentos que os agentes técnicos da Companhia .,.~ I-r '\ .. utilizam no processo de tomada de decisão e que depende dos paradigmas tecnológicos internacionais, das normas internas da empresa e das características societais brasileiras. Investigar criticamente a situação do PEA, por meio da identificação da Cultura Técnica predominante, é o foco do trabalho ora proposto para apreciação. Este não é um tema que diz respeito somente aos engenheiros e técnicos diretamente envolvidos com o PEA, mas a todos aqueles que fazem a empresa, como também a todos os estudiosos do tema.
