The radial growth of samples of thalli of Parmelia glabratula ssp. fuliginosa were measured in situ on a south-facing and a northwest-facing rock surface each month from August 1973 to July 1974. In the periods August to October 1973 and March to July 1974 the radial growth of thalli in the northwest population was greater than in the south population. In the period November 1973 to February 1974 the radial growth of thalli in the south population was greater than in the northwest population. A physiological basis for the differences in seasonal growth in the two populations was suggested. The mean annual radial growth rate (in units of mm/year) was not significantly different in the two populations. However, the variability in radial growth rate between thalli was signifiacntly larger in the northwest than in the south population. These results may be explained by genetic difference between the populations and environmental differences between the rock surfaces.

The objective of the present thesis was to use the manipulation of oocytes enclosed in preantral follicles (MOEPF) as a tool for the female gametes rescue and optimization, from wild species of Caatinga biome. The thesis was divided into 4 experiments. At first experiment, it was performed the estimative and description of the agouti (Dasyprocta leporina) preantral follicles (PF) histologic and ultrastructural features, in which it was estimated 4419.8 ± 532.26 and 5397.52 ± 574.91 follicles for the right and left ovary, respectively, and the majority (86,63%) belonged to the primordial follicles category (P<0.05). Most of the population consists of morphologically normal follicles (70.78%), presenting a large and central nuclei and uniform cytoplasm. At ultrastructural evaluation it was verified the presence of a great number of round mitochondrias associated to lipid droplets. In the second experiment, it was performed the estimative and description of yellow-toothed cavies (Galea spixii) PF characteristics, also, the evaluation of the effect of solid surface vitrification (SSV) on the in situ PF morphology. The total of 416.0 ± 342.8 PF was estimated for the ovary pair and the presence of a large quantity of primary follicles (P<0.05) was evidenced. Most of the PF was morphologically normal (94.6%), in which the oocyte nuclei presented condensed granules of heterochromatin. Round or elongated shaped mitochondria constituted the most abundant organelles. In regard of the SSV, the protocol using the dimethylsulfoxide (DMSO) 3M possibility the preservation of 69.5% of morphologically normal PF, which was evidenced by the light and transmission electronic microscopy. At third experiment, the evaluation of the SSV procedure on the morphology and viability in situ PF form collared peccaries (Pecari tajacu) was performed. No differences were observed among treatments, in which the use of DMSO, ethylene glycol (EG) and dimethylformamide (DMF) as cryoprotectants, regardless its concentration, promoted the morphology preservation of much than 70% of PF. Concerning the PF viability, the DMSO and EG promoted the best preservation. The fourth experiment aimed to evaluate the effect of α MEM+ or TCM199 associated or not to 50 ng of FSHr on the morphology, activation and growth of collared peccaries PF, in vitro cultured (IVC) during 1 or 7 days and the effect on the extracellular matrix (ECM). After 7 days of IVC only the use of TCM199/FSH maintained the proportion of intact PF, similar to day 1(63.2%), however, no differences were observed among treatments (P>0.05). Also, an improvement of the proportion of intact growing PF was verified (P>0.05). By the Ag-NOR analysis it was observed that only the treatment using TCM199/FSH promoted the maintenance of cell proliferation similar to day 1 (P>0.05). The picrosirius red stain revealed that ECM remained intact in all treatments (P>0.05). Thus, as the general conclusion, the use of MOEPF in the refereed species allowed the knowledge of aspects related to its reproductive morphology and physiology, enabling the germplasm conservation, with the possibility of germplasm bank formation, as the elucidation of mechanisms related to the PF survive and in vitro development.

The umbu tree (Spondias tuberosa Arruda) is a fruit native to the northeast of Brazil with great economic, social and ecological importance for the northeastern semi-arid region. Despite its role, the umbu tree has suffered negative pressure thanks to cluttered extractivism and to negative selection of its fruits, which as the deforestation and the dormancy of seeds contribute to the decrease of its production year after year, making necessary studies that contribute to the improvement of this specie and its conservation. Given the risks to the conservation of the specie and its usefulness to the population, the association between plant biotechnology, for being a tool that can be used to increase its production. and the perception of gathering communities, by valuing the point of view and the knowledge of the population, can facilitate its conservation. This work aimed to develop methods of propagation for umbu tree as well as contribute to its conservation by using biotechnology, with specific objectives to contribute to the conservation of this species; determine concentrations of BAP and ANA in the formation of buds; testing the efficiency of different substrates and concentrations of gibberellic acid on germination in vitro and ex vitro, as well as capture the perception of families in communities that engage in the gathering of umbu. To study the germination, the seeds were inoculated in different substrates (vermiculite, vermiculite + clay, clay, clay + manure and manure + vermiculite) and in different concentrations of gibberellic acid (0 mg, 250 g and 500 mg). For the formation of buds BAP to 0.1 mg-1 was associated with different concentrations of ANA (0.2; 0.4; 0.8mg.L-1). The study of perception was conducted by applying semi-structured questionnaire with Malhada Vermelha community. The experiments resulted in vermiculite and concentration of 500 mg gibberellic acid as the best for germination. The association of 0.1 mg.L-1 of BAP to 0.2 mg.L-1 of ANA provided better formation of buds. As to the application of questionnaires, they revealed that the population understands the decreased amount of umbu plants and umbu fruit in the region, as well as shows concern for its conservation.

Worldwide, families Carangidae and Rachycentridae represent one of the groups most important commercial fish, used for food, and great potential for marine aquaculture. However, the genetic bases that can underpin the future cultivation of these species, cytogenetic between these aspects are very weak. The chromosomal patterns have provided basic data for the exploration of biotechnological processes aimed at handling chromosomal genetic improvement, such as induction of polyploidy, androgenesis and ginogenesis, as well as obtaining monosex stocks and interspecific hybridizations. This paper presents a comprehensive cytogenetic survey in 10 species, seven of the family Carangidae and the monotypic family Rachycentridae. Classical cytogenetic analysis and in situ mapping of multigene sequences were employed, and additionally for the genus Selene and morphotypes of Caranx lugubris, comparisons were made using geometric morphometrics. In general, conservative species exhibit a marked chromosome number (2n=48). Although present in large part, different karyotypic form, retain many characteristics typical of chromosomal Order Perciformes, the high number of elements monobrachyal, Ag-NORs/18S rDNA sites and heterochromatin simply reduced, preferably centromeric. The main mechanisms involved in karyotypic diversification are the pericentric inversions, with secondary action of centric fusions. In addition to physical mapping and chromosome detail for the species are presented and discussed patterns of intra-and interspecific diversity, cytotaxonomic markers. A escassez de parâmetros técnicos e científicos e o caráter subjetivo que em regra preside à escolha de determinado material, produto ou técnica suscitaram o interesse pelo presente tema onde se procurou aumentar as ferramentas de análise e de apoio à decisão em intervenções de conservação do património azulejar. Procurou compreender-se o azulejo a nível histórico, estético, técnico e material, estudando-o do ponto de vista físico, químico e mineralógico. Comparando o azulejo com as réplicas produzidas para intervenções de conservação e restauro estudadas, concluiu-se que as suas composições, química e mineralógica, são diferentes, embora não se verifiquem tantas dissemelhanças ao nível das propriedades físicas. As linhas orientadoras da conservação e restauro de azulejo foram lançadas por Santos Simões a partir dos anos 40 do séc. XX e, desde então, foi-se desenvolvendo um pouco à margem das restantes áreas de especialidade, mesmo ao nível do ensino. Os resultados do inquérito internacional junto dos profissionais que trabalham em conservação e restauro de azulejo in situ corroboram de certo modo este facto e, simultaneamente, permitem uma apreensão da situação atual. Constatou-se uma quase total ausência de documentação sobre intervenções realizadas, mas a análise de casos de estudo e de relatórios permitiu uma primeira sistematização da informação disponível segundo os materiais correntemente utilizados. Procedeu-se a uma identificação e tipificação de patologias decorrentes das intervenções procurando entender qual o papel que tiveram os materiais utilizados e as técnicas de intervenção no desempenho das intervenções. Os processos de reintegração volumétrica e cromática mereceram uma maior atenção, procedendo-se à simulação de todo o sistema, em contexto laboratorial, com base nos materiais correntemente utilizados. Os resultados revelam comportamento diferenciado dos materiais de origem orgânica e inorgânica, evidenciando estes últimos uma maior compatibilidade com o azulejo, mas um pior desempenho a médio/longo prazo. O reconhecimento das soluções mais ou menos compatíveis permitiu delinear uma metodologia de avaliação da compatibilidade, da eficácia e do desempenho dos materiais e das técnicas utilizadas, contribuindo também para a definição de algumas recomendações e de critérios de atuação; Abstract The lack of information found, when selecting a material, product or technique, to apply on a specific conservation treatment contributed to the interest on this topic. The historical, aesthetic and technical characteristics of the azulejo was researched by studying their physical, chemical and mineralogical properties. Comparing original azulejos with the replicas produced for conservation treatments studied, it was concluded that the chemical and mineralogical composition is different, although there are less dissimilarities in terms of physical properties. The guidelines for conservation and restoration were launched by Santos Simões in the 40s and since then, it has been developing a bit on the sidelines of the remaining areas of expertise, even at an educational level. The results of an international survey, near professionals working in conservation and restoration of azulejo in situ, corroborate this fact and simultaneously allow an insight of the current situation. It was found an almost complete lack of technical documentation about previous conservation interventions, but the analysis of existent case studies and intervention reports permitted a systematization of the available information, also considering the materials commonly used. The identification and characterization of damages resulting from interventions, was performed trying to establish a correlation with the materials used and intervention techniques. The attention focused on the study of the infills and chromatic reintegration and a simulation of the entire system was developed, in a laboratory context, on the basis of commonly used materials. The results reveal a distinct behavior between the organic and inorganic materials, showing the latter greater compatibility with the azulejos but a worse medium/long-term performance. The recognition of more or less compatible solutions allowed to outline a methodology to evaluate the compatibility, efficiency and performance of materials and techniques used, also contributing to define some recommendations and action criteria.

Leguminosae com cerca de 650 gêneros e 19.000 espécies compreende a terceira maior família de angiospermas, superada apenas por Orchidaceae (20.000 spp.) e Asteraceae (24.000 spp.). Para o Brasil, são citados 222 gêneros e 2.803 espécies distribuídas nos diversos domínios fitogeográficos, e para região Nordeste estão registradas 1.085 espécies em 168 gêneros. No Ceará, são reportadas 340 espécies em 105 gêneros, dos quais Mimosa L. (26 spp.) é o mais diversificado. No bioma Caatinga, é a principal família de fanerógamas com 83 gêneros e 354 espécies, muitas das quais com diversas potencialidades de uso. Em virtude da importância deste táxon no contexto do semiárido, objetivou-se com este trabalho catalogar as Leguminosae ocorrentes em uma área de Caatinga no distrito de Taperuaba, Sobral, Ceará. O estudo foi realizado na Unidade de Conservação Refúgio de Vida Silvestre Pedra da Andorinha, localizada a 70 km da sede municipal e possuindo aproximadamente 600 ha. A área é composta por Caatinga em duas fitofisionomias: uma terrícola e outra rupícola. As expedições de coletas foram realizadas entre março/2015 e março/2016. Os espécimes foram identificados com o auxílio de literatura e sites especializados. O material encontra-se depositado no acervo do Herbário Prof. Francisco José de Abreu Matos (HUVA). Foram catalogadas 15 espécies em oito gêneros: Mimosa (4 spp.), Chamaecrista Moench (3 spp.), Poincianella Britton & Rose (2 spp.). Bauhinia L., Centrosema (DC.) Benth., Desmodium Desv., Senna Mill., Stylosanthes Sw. e Zornia J.F. Gmel. apresentaram uma espécie cada. Dentre as espécies coletadas, duas apresentam propriedades medicinais: B. forficata (Vogel) Fortunato & Wunderlin e M. tenuiflora (Willd.) Poir. Centrosema pascuorum Mart. ex Benth, Mimosa pudica (Humb. & Bonpl. ex Willd.) DC., Stylosathes humilis Kunth e Zornia cearensis Huber são tratadas como invasoras de cultivos. Entre as espécies com alto potencial forrageiro, estão Chamaecrista linearis (H.S. Irwin & Barneby) Afr. Fern. & E.P. Nunes e Desmodium tortuosum (Sw.) DC. Por outro lado, Senna obtusifolia (L.) H.S. Irwin & Barneby, é tóxica aos bovinos. Alguns estudos ainda sugerem que os extratos das raízes de Poincianella bracteosa (Tul.) L.P. Queiroz possuem atividade biológica sobre larvas de Aedes aegypti. Diante dos resultados, verificou-se que Leguminosae é a família mais representativa da área de estudo e que 11 espécies apresentaram algum uso ou potencialidade econômica.

As relações entre populações humanas e Unidades de Conservação no bioma Mata Atlântica são, muitas vezes, marcadas por conflitos que envolvem as diferentes percepções que as mesmas possuem sobre essas áreas. Entender como se manifestam tais relações constitui-se num desafio para a preservação da biodiversidade in situ nos seus ecossistemas remanescentes. No âmbito do Sistema de Unidades de Conservação brasileiro a realização desse trabalho em uma Área de Proteção Ambiental (APA) justifica-se por esta categoria ser menos restritiva quanto ao uso da terra, o que possibilita uma maior ação do uso social sobre fragmentos florestais. A APA escolhida para o estudo de caso deve-se ao seu papel ecológico de conexão espacial no contexto da figura do Mosaico de UCs do Central Fluminense - RJ. Este trabalho objetiva entender como as práticas sociais mediadas pelas percepções ambientais dessas populações, estabelecidas em diferentes paisagens geográficas (rural, urbana e rururbana), matrizes de fragmentos florestais remanescentes, podem interferir no processo de sucessão ecológica dos mesmos. A partir de entrevistas de populações em quatro matrizes diferentes. Apreendem-se padrões de percepções de diferentes atores e suas possíveis resultantes para cenários de conflitos sociais nessa mediação de preservação da sucessão ecológica dos fragmentos florestais.

This research studied the effect of low density polyethylene packaging and storage temperature on the preservation of fresh-cut (minimally processed) cabbage. The cabbages, previously cooled to a temperature of 10 ºC, were selected, washed, cut in four parts (with the central stalk removed), sanitized, cut in strips, rinsed, put in the centrifuge, weighed and stored in plastic packaging of low density polyethylene (70 µm), and then stored in cold chambers at temperatures of 1 and 10 ºC for 20 days. The following aspects were evaluated: carbon dioxide, oxygen and ethylene in the internal atmosphere of the package as well as, pH, titratable acidity, total soluble solids, vitamin C, loss of fresh mass and the total soluble solids/acidity in the fresh-cut cabbage ratio. The experimental design was entirely casual, with three repetitions. The analysis parameters, except for the vitamin C, loss of fresh mass and ethylene, presented significant variation between the temperatures and days of storage. The cabbage stored at a temperature of 1 ºC presented a shelf life of around 15 days, significantly higher than that stored at 10 ºC. At this temperature, on the 8th day of storage, the product was completely decayed, unfit for commercialization or consumption.

Postharvest losses vary among the different vegetable products. However, among fruits and vegetables the losses generally range from 30% to 50%. Thus, this paper aimed the application of 1-methylcycloprene (1-MCP) and fast cooling with forced air (PC) on peaches, in order to estimate their effects in the ripening process of this fruit. Physiological analyses were performed, such as loss of fresh mass, firmness, pH, titratable acidity, soluble solids, ratio and CO2 production, as well as sensorial analyses such as color, texture and flavor. The experiment was divided in two phases. In the first one, concentrations of 30, 60, and 90 nL/L 1-MCP, applied at 0 ºC and 20 ºC, were tested. The fruits treated without 1-MCP were denominated control for both temperatures studied. The second phase was composed by the following treatments: cold storage (CS) or control, cooling with forced air (CFA), cooling with forced air followed by 1-MCP application (CFA + 1-MCP) and 1-MCP application (1-MCP). Among these, the CFA + 1-MCP treatment provided more firmness of the fruits in comparison to the control fruits. The respiratory rate of peaches under CFA and CFA + 1-MCP treatments decreased in comparison to the control fruit respiratory rates.

Several native herbaceous and subshrub species native to the Cerrado in Brazil are geophytes, that is, they survive the unfavorable dry season and low temperatures, that sometimes coincide with fire, with only the underground system intact. Vernonia oxylepis is one of these species and the aim of this study was to describe the morpho-anatomy of the tuberous root and bud formation on this structure. The main axis of this root is perpendicular to the soil surface, and from which aerial shoots arise periodically throughout the life cycle. On the upper portion of the root, self-grafting of the shoots occurs. The root stores lipids and fructans, exhibits contraction and produces reparatory buds; adventitious buds arise from proliferated pericycle. These characteristics may be related to adaptation of this species to conditions in the Cerrado.

Nectandra Rol. ex Rottb. has 14 species in Paraná: Nectandra angustifolia (Schrader) Nees & Mart., N. cissiflora Nees, N. cuspidata Nees & Mart., N. grandiflora Nees & Mart., N. hihua (Ruiz & Pav.) Rohwer, N. lanceolata Nees & Mart., N. leucantha Nees & Mart., N. megapotamica (Sprengel) Mez, N. membranacea (Sw.) Griseb., N. nitidula Nees & Mart., N. oppositifolia Nees & Mart., N. paranaensis Coe-Teixeira, N. puberula (Schott) Nees and N. reticulata (Ruiz & Pav.) Mez. We present an identification key and species descriptions, as well as illustrations and data on phenology and geographic distribution. N. hihua is cited for the first time in Paraná State.

The durability and postharvest quality of cut flowers are fundamental attributes in value along the production chain and in consumer satisfaction. The objective of this study was to evaluate the effect of chemical inhibitors of ethylene action on maintaining the postharvest quality of chrysanthemum stems (Chrysanthemum morifolium Ramat cv. Dragon). The experiment tested maintenance solutions with silver thiosulfate (STS) under five levels (distilled water, a 0.2 mM STS, the STS 0.2 mM + sucrose at 50 g L-1, STS at 0.4 mM; STS at 0.4 mM + sucrose at 50 g L-1), and date of sampling, for three levels (0, 3, 6 days). Three replications with two flower stems in each treatment were used in the experiment. Physical assessments were made: color, fresh mass and relative water content; chemical evaluations: reducing sugars and pigments, and qualitative assessments: turgidity, flower color, and number of buds, open flowers and partially open flowers. Treatment with 0.2 mM STS resulted in better maintenance of fresh mass of stems. The concentration of pigments and reducing sugar was higher in those treatments in which sucrose was associated. The color and relative water content were favored in treatments STS 0.2 mM and 0.4 mM. The concentration of 0.2 mM STS obtained the best results, prolonging the vase life the stems. The quality of these stems was higher, with the best assessments of water content, color and turgidity.
