Inorganic phosphorus (Pi) usually controls the P availability in tropical soils, but the contribution of organic P (Po) should not be neglected, mainly in systems with low P input or management systems that promote organic matter accumulation. The aims of this study were to evaluate the changes in the Po fractions over time in soil fertilized and not fertilized with cattle manure and to correlate Po forms with available P extracted by anion exchange resin. The experiment was carried out under field conditions, in a sandy-clay loam Haplustox. The experimental design was a 2 × 9 randomized complete block factorial design, in which the first factor was manure application (20 t ha-1) or absence, and the second the soil sampling times (3, 7, 14, 21, 28, 49, 70, 91, and 112 days) after manure incorporation. Labile, moderately labile and non-labile Po fractions were determined in the soil material of each sampling. Manure fertilization increased the Po levels in the moderately labile and non-labile fractions and the total organic P, but did not affect the Po fraction proportions in relation to total organic P. On average, 5.1 % of total Po was in the labile, 44.4 % in the moderately labile and 50.5 % in the non-labile fractions. Available P (resin P) was more affected by the manure soluble Pi rather than by the labile Po forms. The labile and non-labile Po fractions varied randomly with no defined trend in relation to the samplings; for this reason, the data did not fit any mathematical model.

Bacterial reporters are live, genetically engineered cells with promising application in bioanalytics. They contain genetic circuitry to produce a cellular sensing element, which detects the target compound and relays the detection to specific synthesis of so-called reporter proteins (the presence or activity of which is easy to quantify). Bioassays with bacterial reporters are a useful complement to chemical analytics because they measure biological responses rather than total chemical concentrations. Simple bacterial reporter assays may also replace more costly chemical methods as a first line sample analysis technique. Recent promising developments integrate bacterial reporter cells with microsystems to produce bacterial biosensors. This lecture presents an in-depth treatment of the synthetic biological design principles of bacterial reporters, the engineering of which started as simple recombinant DNA puzzles, but has now become a more rational approach of choosing and combining sensing, controlling and reporting DNA 'parts'. Several examples of existing bacterial reporter designs and their genetic circuitry will be illustrated. Besides the design principles, the lecture also focuses on the application principles of bacterial reporter assays. A variety of assay formats will be illustrated, and principles of quantification will be dealt with. In addition to this discussion, substantial reference material is supplied in various Annexes.

This station was required to have air temperature, relative humidity, wind speed and direction, and pavement temperature sensors of similar quality to the traditional RWIS sensors, have an integrated solar powered battery system, and be trailer-mounted for ease of transport. The station was tested by the Iowa DOT for basic reliability and sensor performance for a month and a half in Ames, Iowa before being moved to near Osceola, Iowa in early February 2010 for further field testing and evaluation. DOT field maintenance staff was able to successfully set up the station with minimal instruction and found the station to be relatively intuitive in its installation. Air temperature, wind speed, and wind direction observations from the station were compared to a nearby RWIS station and had good agreement. Pavement temperature readings were compared to sites within 40 miles of the station, and the readings correlated. The station has had good reliability.
