Bacterial type III secretion systems deliver protein virulence factors to host cells. Here we characterize the interaction between HrpB2, a small protein secreted by the Xanthomonas citri subsp. citri type III secretion system, and the cytosolic domain of the inner membrane protein HrcU, a paralog of the flagellar protein FlhB. We show that a recombinant fragment corresponding to the C-terminal cytosolic domain of HrcU produced in E. coli suffers cleavage within a conserved Asn264-Pro265-Thr266-His267 (NPTH) sequence. A recombinant HrcU cytosolic domain with N264A, P265A, T266A mutations at the cleavage site (HrcU(AAAH)) was not cleaved and interacted with HrpB2. Furthermore, a polypeptide corresponding to the sequence following the NPTH cleavage site also interacted with HrpB2 indicating that the site for interaction is located after the NPTH site. Non-polar deletion mutants of the hrcU and hrpB2 genes resulted in a total loss of pathogenicity in susceptible citrus plants and disease symptoms could be recovered by expression of HrpB2 and HrcU from extrachromossomal plasmids. Complementation of the Delta hrcU mutant with HrcU(AAAH) produced canker lesions similar to those observed when complemented with wild-type HrcU. HrpB2 secretion however, was significantly reduced in the Delta hrcU mutant complemented with HrcU(AAAH), suggesting that an intact and cleavable NPTH site in HrcU is necessary for total functionally of T3SS in X. citri subsp. citri. Complementation of the Delta hrpB2 X. citri subsp. citri strain with a series of hrpB2 gene mutants revealed that the highly conserved HrpB2 C-terminus is essential for T3SS-dependent development of citrus canker symptoms in planta.

Advanced titanosaurian sauropods, such as nemegtosaurids and saltasaurids, were diverse and one of the most important groups of herbivores in the terrestrial biotas of the Late Cretaceous. However, little is known about their rise and diversification prior to the Late Cretaceous. Furthermore, the evolution of their highly-modified skull anatomy has been largely hindered by the scarcity of well-preserved cranial remains. A new sauropod dinosaur from the Early Cretaceous of Brazil represents the earliest advanced titanosaurian known to date, demonstrating that the initial diversification of advanced titanosaurians was well under way at least 30 million years before their known radiation in the latest Cretaceous. The new taxon also preserves the most complete skull among titanosaurians, further revealing that their low and elongated diplodocid-like skull morphology appeared much earlier than previously thought.

Background to the debate: The tobacco control community is divided on whether or not to inform the public that using oral, smokeless tobacco (Swedish snus) is less hazardous to health than smoking tobacco. Proponents of 'harm reduction' point to the Swedish experience. Snus seems to be widely used as an alternative to cigarettes in Sweden, say these proponents, contributing to the low overall prevalence of smoking and smoking-related disease. Harm reduction proponents thus argue that the health community should actively inform inveterate cigarette smokers of the benefits of switching to snus. However, critics of harm reduction say that snus has its own risks, that no form of tobacco should ever be promoted, and that Sweden’s experience is likely to be specific to that culture and not transferable to other settings. Critics also remain deeply suspicious that the tobacco industry will use snus marketing as a 'gateway' to promote cigarettes. In the interests of promoting debate, the authors (who are collaborators on a research project on the future of tobacco control) have agreed to outline the strongest arguments for and against promoting Swedish snus as a form of harm reduction.

A doença mental continua imbuída de mitos, preconceitos e estereótipos, apesar da crescente aposta na investigação e na melhoria de tratamento nesta área da saúde. Como consequência, as pessoas com doença mental são discriminadas e estigmatizadas quer pelo público geral e pelos meios de comunicação, quer pelas próprias famílias e pelos profissionais de saúde mental que lhes prestam cuidados. Uma vez que os profissionais de saúde mental estabelecem uma ponte entre a doença e a saúde, espera-se que as suas atitudes e práticas contribuam para o recovery da pessoa com doença mental. No entanto, se os profissionais também apresentarem atitudes e crenças estigmatizantes face à doença mental, este processo reabilitativo pode ficar comprometido. Nesse sentido, e perante as lacunas de investigação nesta área, este trabalho tem como objectivo explorar e clarificar a presença ou ausência de atitudes estigmatizantes dos profissionais de saúde mental e, quando presentes, como se caracterizam. Para tal realizaramse 24 entrevistas de carácter qualitativo a profissionais de saúde mental que trabalham em três instituições na região do Porto, nomeadamente num serviço de psiquiatria de um hospital geral, num hospital especializado e em estruturas comunitárias. A análise do material discursivo recolhido junto de Assistentes Sociais, Enfermeiros, Médicos Psiquiatras, Psicólogos e Terapeutas Ocupacionais evidencia a presença de crenças e atitudes de carácter estigmatizante face à doença mental, independentemente da idade, formação ou local onde exercem funções, salvo escassos aspectos onde parece haver influência da idade e da profissão. Significa isto que é provável que as variações de atitudes dos profissionais sejam fundamentalmente consequência das suas características pessoais.

Traditionally, a country's electoral system requires the voter to vote at a specific day and place, which conflicts with the mobility usually seen in modern live styles. Thus, the widespread of Internet (mobile) broadband access can be seen as an opportunity to deal with this mobility problem, i.e. the adoption of an Internet voting system can make the live of voter's much more convenient; however, a widespread Internet voting systems adoption relies on the ability to develop trustworthy systems, i.e. systems that are verifiable and preserve the voter's privacy. Building such a system is still an open research problem. Our contribution is a new Internet voting system: EVIV, a highly sound End-to-end Verifiable Internet Voting system, which offers full voter's mobility and preserves the voter's privacy from the vote casting PC even if the voter votes from a public PC, such as a PC at a cybercafe or at a public library. Additionally, EVIV has private vote verification mechanisms, in which the voter just has to perform a simple match of two small strings (4-5 alphanumeric characters), that detect and protect against vote manipulations both at the insecure vote client platform and at the election server side. D. João de Magalhães e Avelar (1754-1833) formou aquela que, ao tempo, era a maior biblioteca privada portuguesa. Com cerca de 36000 volumes, foi elogiada por personalidades nacionais e estrangeiras, por aliar à quantidade de volumes inúmeros e valiosíssimos manuscritos. Formada ao longo dos séculos XVIII e XIX, durante mais de 30 anos, originou, em 1833, o primeiro núcleo da actual Biblioteca Pública Municipal do Porto. Numa época em que possuir livros era sinónimo de prestígio social mas num período em que quase não havia tradição de bibliotecas públicas no nosso país, contrariamente ao que acontecia noutras realidades, a livraria privada de Avelar formou, com outras, a Real Biblioteca Pública da Cidade do Porto. Em 1833, aquando do primeiro aniversário da entrada do exército liberal no Porto, por decreto, criou-se a biblioteca portuense. Estabelecida na casa que servia de Hospício dos Religiosos de Santo António do Val da Piedade, à praça da Cordoaria, tinha como objectivo satisfazer a utilidade pública, estando aberta todos os dias, excepto domingos e feriados. Propriedade da cidade do Porto, ficava sujeita à administração da Câmara que se obrigava à sua guarda, manutenção, conservação, bem como à constante aquisição de espólio. Como veremos, tratou-se de um processo conflituoso mas o Porto obtinha, definitivamente, a sua biblioteca pública.

The Iberian viticultural regions are convened according to the Denomination of Origin (DO) and present different climates, soils, topography and management practices. All these elements influence the vegetative growth of different varieties throughout the peninsula, and are tied to grape quality and wine type. In the current study, an integrated analysis of climate, soil, topography and vegetative growth was performed for the Iberian DO regions, using state-of-the-art datasets. For climatic assessment, a categorized index, accounting for phenological/thermal development, water availability and grape ripening conditions was computed. Soil textural classes were established to distinguish soil types. Elevation and aspect (orientation) were also taken into account, as the leading topographic elements. A spectral vegetation index was used to assess grapevine vegetative growth and an integrated analysis of all variables was performed. The results showed that the integrated climate-soil-topography influence on vine performance is evident. Most Iberian vineyards are grown in temperate dry climates with loamy soils, presenting low vegetative growth. Vineyards in temperate humid conditions tend to show higher vegetative growth. Conversely, in cooler/warmer climates, lower vigour vineyards prevail and other factors, such as soil type and precipitation acquire more important roles in driving vigour. Vines in prevailing loamy soils are grown over a wide climatic diversity, suggesting that precipitation is the primary factor influencing vigour. The present assessment of terroir characteristics allows direct comparison among wine regions and may have great value to viticulturists, particularly under a changing climate.

This paper analyses forest fires in the perspective of dynamical systems. Forest fires exhibit complex correlations in size, space and time, revealing features often present in complex systems, such as the absence of a characteristic length-scale, or the emergence of long range correlations and persistent memory. This study addresses a public domain forest fires catalogue, containing information of events for Portugal, during the period from 1980 up to 2012. The data is analysed in an annual basis, modelling the occurrences as sequences of Dirac impulses with amplitude proportional to the burnt area. First, we consider mutual information to correlate annual patterns. We use visualization trees, generated by hierarchical clustering algorithms, in order to compare and to extract relationships among the data. Second, we adopt the Multidimensional Scaling (MDS) visualization tool. MDS generates maps where each object corresponds to a point. Objects that are perceived to be similar to each other are placed on the map forming clusters. The results are analysed in order to extract relationships among the data and to identify forest fire patterns.
