Experiments were conducted over 5 years to understand the seasonal phenology of bare-rooted ?Festival? strawberry plants (Fragaria ?ananassa) growing at Nambour in southeastern Queensland, Australia. Yields ranged from 661 to 966 g/plant, and average seasonal fruit fresh weight ranged from 15 to 18 g. The growth of the leaves, crowns, roots, flowers and fruit over time followed a linear or sigmoid pattern. Maximum values of leaf, crown and root dry weight towards the end of the growing season about 190 days after planting were 30, 15 and 7 g/plant, respectively. The rates of leaf and crown growth were lower than those achieved in California under a Mediterranean climate. There were strong relationships between the allocation of dry matter to the leaves, crowns and roots and plant dry weight. Allocation to the leaves, and especially to the crowns and roots, declined as the plants grew. The number of fruit/plant increased initially over time with a decline later in the season. Average fruit fresh weight was generally higher early in the season and then declined as fruit production increased. There were strong relationships between the growth of the whole plant and the growth of the flowers and immature fruit, and leaf expansion, across the growing season and across the 5 different years. These results indicate that seasonal growth and potential productivity were strongly linked to the expansion of the leaves in this environment.

Campylobacter is an important food borne pathogen, mainly associated with poultry. A lack of through-chain quantitative Campylobacter data has been highlighted within quantitative risk assessments. The aim of this study was to quantitatively and qualitatively measure Campylobacter and Escherichia coli concentration on chicken carcasses through poultry slaughter. Chickens (n = 240) were sampled from each of four flocks along the processing chain, before scald, after scald, before chill, after chill, after packaging and from individual caeca. The overall prevalence of Campylobacter after packaging was 83% with a median concentration of 0.8 log10 CFU/mL. The processing points of scalding and chilling had significant mean reductions of both Campylobacter (1.8 and 2.9 log10 CFU/carcase) and E. coli (1.3 and 2.5 log10 CFU/carcase). The concentration of E. coli and Campylobacter was significantly correlated throughout processing indicating that E. coli may be a useful indicator organism for reductions in Campylobacter concentration. The carriage of species varied between flocks, with two flocks dominated by Campylobacter coli and two flocks dominated by Campylobacter jejuni. Current processing practices can lead to significant reductions in the concentration of Campylobacter on carcasses. Further understanding of the variable effect of processing on Campylobacter and the survival of specific genotypes may enable more targeted interventions to reduce the concentration of this poultry associated pathogen.

The influence of grazing management on total soil organic carbon (SOC) and soil total nitrogen (TN) in tropical grasslands is an issue of considerable ecological and economic interest. Here we have used linear mixed models to investigate the effect of grazing management on stocks of SOC and TN in the top 0.5 m of the soil profile. The study site was a long-term pasture utilization experiment, 26 years after the experiment was established for sheep grazing on native Mitchell grass (Astrebla spp.) pasture in northern Australia. The pasture utilization rates were between 0% (exclosure) and 80%, assessed visually. We found that a significant amount of TN had been lost from the top 0.1 m of the soil profile as a result of grazing, with 80% pasture utilization resulting in a loss of 84 kg ha−1 over the 26-year period. There was no significant effect of pasture utilization rate on TN when greater soil depths were considered. There was no significant effect of pasture utilization rate on stocks of SOC and soil particulate organic carbon (POC), or the C:N ratio at any depth; however, visual trends in the data suggested some agreement with the literature, whereby increased grazing pressure appeared to: (i) decrease SOC and POC stocks; and, (ii) increase the C:N ratio. Overall, the statistical power of the study was limited, and future research would benefit from a more comprehensive sampling scheme. Previous studies at the site have found that a pasture utilization rate of 30% is sustainable for grazing production on Mitchell grass; however, given our results, we conclude that N inputs (possibly through management of native N2-fixing pasture legumes) should be made for long-term maintenance of soil health, and pasture productivity, within this ecosystem.

In the yeast, mobilization of triacylglycerols (TAG) is facilitated by TGL3, TGL4 and TGL5 gene products. Interestingly, experiments using [32P] orthophosphate as a precursor for complex glycerophospholipids revealed that tgl mutants had a lower steady-state level of these membrane lipids. To understand a possible link between TAG lipolysis and phospholipid metabolism, we performed overexpression studies with Tgl3p and Tgl5p which clearly demonstrated that these two enzymes enhanced the level of phospholipids. Domains and motifs search analyses indicated that yeast TAG hydrolases posses a GXSXG lipase motif but also a HX4D acyltransferase motif. Purified Tgl3p and Tgl5p did not only exhibit TAG lipase activity but also catalyzed acyl-CoA dependent acylation of lyso-phosphatidylethanolamine and lyso-phosphatidic acid (LPA), respectively. Search for lipase/hydrolase homologues in the Arabidopsis thaliana genome led to the identification of At4g24160 which possess three motifs that are conserved across the plant species such as GXSXG motif, a HX4D motif and a probable lipid binding motif V(X)3HGF. Characterization of At4g24160 expressed in bacteria revealed that the presence of an acyl-CoA dependent LPA acyltransferase activity. In addition, the purified recombinant At4g24160 protein hydrolyzed both TAG and phosphatidylcholine. We hypothesize that the plant enzyme may be involved in membrane repair. In summary, our results indicate that these TAG lipases play a dual role and thereby contribute to both anabolic and catabolic processes in yeast and plants.

The application of variable-number tandem repeats (VNTR) genotyping of Mycobacterium avium subsp. paratuberculosis isolates to assist in investigating incidents of bovine Johne’s disease in a low-prevalence region of Australia is described in the current study. Isolates from a response to detection of bovine Johne’s disease in Queensland were compared with strains from national and international sources. The tandem application of mycobacterial interspersed repetitive unit (MIRU) and multilocus short sequence repeats (MLSSR) genotyping identified 2 strains, 1 that infected cattle on multiple properties with trace-forward histories from a common infected property, and 1 genotypically different strain recovered from a single property. The former strain showed an identical genotype to an isolate from India. Neither strain showed a genotypic link to regions of Australia with a higher prevalence of the disease. Genotyping has indicated incursions from 2 independent sources. This intelligence has informed investigations into potential routes of entry and the soundness of ongoing control measures, and supported strategy and policy decisions regarding management of Mycobacterium avium subsp. paratuberculosis incursions for Queensland.

Glucosinolates are a group of sulphur-containing glycosides found in the plant order Brassicales which includes the Brassica vegetables such as broccoli, cabbage and cauliflower. When brought into contact with the plant enzymes, myrosinases, the glucosinolates break down releasing glucose and other products which serve principally in plant defence against herbivores. The most important of the products from a human nutritional viewpoint, are the isothiocyanates. These potent inducers of detoxifying enzymes bestow the distinct anti-cancer properties on these plants. Unique among tropical fruits, papaya is known to contain an abundance of one particular glucosinolate, glucotropaeolin. Other compounds that play a pivotal role in the chemical defence system of many plants are the cyanogenic glycosides. Cyanogenic glycosides are activated by plant enzymes in the event of pest attack, releasing the deterrent: toxic hydrogen cyanide. Papaya, in addition to glucosinolates, also contains low levels of cyanogenic glycosides, an unusual occurrence because it was assumed that the two classes of metabolites were mutually exclusive. Studies measuring the levels of both in the edible parts of the papaya fruit and other utilised tissues are discussed and considered in the context of potential human health ramifications. Grasses, legumes, saltbushes and herbs were evaluated at 6 sites in southern inland Queensland to identify potential pasture and forage plants for use on marginal cropping soils. The region experiences summer heat waves and severe winter frosts. Emphasis was on perennial plants, and native species were included. Seedlings were transplanted into the unfertilized fields in either summer or autumn to suit the growing season of plants, and watered to ensure estab-lishment. Summer-growing grasses were the most successful group, while cool season-growing perennials mostly failed. Summer legumes were disappointing, with Stylosanthes scabra and Indigofera schimperi performing best. Some lines such as I. schimperi and the Eragrostis hybrid cv. Cochise were assessed as potential weeds owing to low animal acceptance. Native Rhynchosia minima grew well at some sites and deserves more study. Cenchrus ciliaris was always easy to establish and produced the highest yields. Persistence of some Digitaria and Bothriochloa species, Eragrostis curvula and Fingerhuthia africana at specific sites was encouraging, but potential weediness needs careful assessment. Standard species were identified to represent the main forage types, such as Austrostipa scabra for cool season-growing grasses, for incorporation into future trials with new genetic materials. The early field testing protocol used should be considered for use elsewhere, if unreliable rainfall poses a high risk of establishment failure from scarce seed.

Mikania micrantha Kunth (mikania vine) is a highly invasive tropical weed that was first discovered in Australia in 1997, and has been the target of a nationally cost-shared weed eradication program since 2003. Field crews have been effectively treating the weed with herbicide solutions containing 1 g a.i. L−1 of fluroxypyr. During the eradication program there have been limited opportunities to test alternative foliar herbicides or rates. A newly discovered infestation provided sufficient immature vines to compare the effectiveness of eight herbicide treatments.

Bellyache bush (Jatropha gossypifolia L. (Euphorbiaceae)) is a serious weed of dry tropical regions of northern Australia, with the potential to spread over much of the tropical savannah. It is well adapted to the harsh conditions of the dry tropics, defoliating during the dry season and rapidly producing new leaves with the onset of the wet season. In this study we examined the growth and biomass allocation of the three Queensland biotypes Queensland Green, Queensland Bronze and Queensland Purple) under three water regimes (water-stressed, weekly watering and constant water). Bellyache bush plants have a high capacity to adjust to water stress. The impact of water stress was consistent across the three biotypes. Water stressed plants produced significantly less biomass compared to plants with constant water, increased their biomass allocation to the roots and increased biomass allocation to leaf material. Queensland Purple plants allocated more resources to roots and less to shoots than Queensland Green (Queensland Bronze being intermediate). Queensland Green produced less root biomass than the other two biotypes.

In the sub-tropical grain region of Australia, cotton and grains systems are now dominated by flaxleaf fleabane (Conyza bonariensis (L.) Cronquist), feathertop Rhodes grass (Chloris virgata Sw.) and awnless barnyard grass (Echinochloa colona (L.) Link). While control of these weed species is best achieved when they are young, previous studies have shown a potential for reducing seed viability and minimising seed bank replenishment by applying herbicides when plants are reproductive. Pot trials were established over two growing seasons to examine the effects of 2,4-D, 2,4-D + picloram, glyphosate and glufosinate which had been successful on other species, along with paraquat and haloxyfop (grasses only). Herbicides were applied at ¾ field rates in an attempt not to kill the plants. Flaxleaf fleabane plants were sprayed at two growth stages (budding and flowering) and the grasses were sprayed at two stages (late tillering/booting and flowering). Spraying flaxleaf fleabane at flowering reduced seed viability to 0% (of untreated) in all treatments except glyphosate (51%) and 2,4-D + picloram (8%). Seed viability was not reduced with the first and second regrowths with the exception of 2,4-D + picloram where viability was reduced to 20%. When sprayed at budding only 2,4-D + picloram reduced seed viability in both trials. Spraying the grasses at late tillering/booting did not reduce viability except for glufosinate on awnless barnyard grass (50%). Applying herbicides at flowering resulted in 0% seed viability in awnless barnyard grass from glufosinate, paraquat and glyphosate and 0% viability in feathertop Rhodes grass for glufosinate. These herbicides were less effective on heads that emerged and flowered after spraying, only slightly reducing seed viability. These trials have shown that attempts to reduce seed viability have potential, however flaxleaf fleabane and feathertop Rhodes grass are able to regrow and will need on-going monitoring and control measures.
