Current knowledge of the bioavailability of lycopene in humans is limited due to the inability to distinguish newly administered lycopene from the body reserves of lycopene. A quantitative method to assess the absorption and relative bioavailability of newly absorbed synthetic or natural lycopene was developed using two deuterated lycopene sources, in conjunction with an advanced LC/APCI-MS (liquid chromatography/atmospheric pressure chemical ionization-mass spectrometry) to analyze newly absorbed lycopene in blood samples of study subjects. Two subjects (1 male and 1 female) consumed hydroponically grown tomatoes containing deuterium-enriched lycopene (8084 g wet weight tomato containing 16.3 and 17.4 mu mol lycopene, respectively) and two subjects (1 male, and 1 female) consumed 11 mu mol synthetic H-2(10) lycopene in 6 g of corn oil. Tomatoes were steamed and pureed. The doses were given together with a liquid formulated drink with 25% energy from fat. Our results showed that up to 34 days after taking an oral 2 1110 lycopene dose (synthetic or from tomato) with a liquid formula drink, the area under the curve of the average serum percent enrichment response of synthetic lycopene reached 33.9 (+/- 1.7) nmol-day/mu mol lycopene in the dose, whereas that of lycopene from the tomato dose was 11.8 (+/- 0.3) nmol-day/mu mol lycopene in the dose. Our study provides evidence that the absorption of physiological levels of lycopene in intrinsically labeled tomatoes can be studied in humans. From these preliminary investigations, we find that the bioavailability of synthetic lycopene in oil appears to be about three times higher than that of lycopene from steamed and pureed tomatoes. The objective of the present research was to study the degradation and the persistence of phenthoate (Elsan 50 CE) residues in tomato fruits. The experiment was set in a tomato field located in Mombuca, State of São Paulo, Brazil, after 45 days from seedlings transplantation. Dosages of 600 (A), 800 (B), 1200 (C) and 1600 g a.i./ha (D) were sprayed at the beginning of the ripening period of fruits. Samples of 15 fruits each were collected 1, 4, 10, 20 and 40 days after products spraying. The utilized procedure consisted of extraction with acetone and purification by partition with dichloromethane. The purified extract was concentrated and injected into a gas chromatograph equipped with alkaly flame ionization detector (AFID). The limits of detection were of 0,01 ppm for this product. The recovery percentage from fortified samples varied from 60 to 81%. The degradation and the persistence half-lives were 1 to 2 and 3 to 6 days for phenthoate, respectively.

An automated system with a C-18 bonded silica gel packed minicolumn is proposed for spectrophotometric detection of arsenic using flow-injection hydride generation following sorbent extraction preconcentration. Complexes formed between arsenic(III) and ammonium diethyl dithiophosphate (ADDP) are retained on a C-18 sorbent. The eluted As-DDP complexes are merged with a 1.5% (w/v) NaBH4 and the resulting solution is thereafter injected into the hydride generator/gas-liquid separator. The arsine generated is carried out by a stream of N-2 and trapped in an alkaline iodine solution in which the analyte is determined by the arsenomolybdenum blue method. With preconcentration time of 120 s, calibration in the 5.00-50.0 mu g As l(-1) range and sampling rate of about 20 samples h(-1) are achieved, corresponding to 36 mg ADDP plus 36 mg ammonium heptamolybdate plus 7 mg hydrazine sulfate plus 0.7 mg stannous chloride and about 7 mi sample consumed per determination. The detection limit is 0.06 mu g l(-1) and the relative standard deviation (n = 12) for a typical 17.0 mu g As l(-1) sample is ca. 6%. The accuracy was checked for arsenic determination in plant materials from the NIST (1572 citrus leaves; 1573 tomato leaves) and the results were in agreement with the certified values at 95% confidence level. Good recoveries (94-104%) of spiked tap waters, sugars and synthetic mixtures of trivalent and pentavalent arsenic were also found. (C) 1999 Elsevier B.V. B.V. All rights reserved.

The objective of the present research was to determine the allelopathic potential of Ipomoea fistulosa plants on germination and growth of seedlings. In laboratory, lettuce and tomato seeds were placed to germinate in the presence of water or Ipomoea fistulosa leaf extract, in the concentrations of 5, 10 e 15% (p/v). In greenhouse, seeds were placed to germinate in trays containing Plantmax (R) substrate and Plantmax (R) substrate + Ipomoea fistulosa dry leaf residue, at 3% (w/w). The aqueous extract inhibited germination, raised the mean germination time of the seeds and reduced the radicle and hypocotyl growth in all used concentrations. In greenhouse, the plant height, leaf number, and dry mass accumulation also were reduced in the presence of the leaf residue in the substrate. The results suggest the presence of allelochemicals in Ipomoea fistulosa and demonstrate the highest sensitivity of lettuce that tomato to these compounds.

The castor bean plant is a tropical species and is subject to various diseases, which cause great losses. Among these diseases, the gray mold (Botryotinia ricini) is one of the most important. The fungus spore production was evaluated in the media of cultures BDA; Oats-Agar; Mazeina-Agar; Rice-Agar; castor-bean crushed leaves-agar (FM), FM-CaCO3; V8 juice to 5% (V8-5%); V8-10%; V8-20% and tomato juice at 5% (TJ-5%); TJ-10% and TJ-20%. The production of spore in different media of cultures was evaluated at 8(th) days of incubation. The data were analyzed using the comparison of means, through the test Tukey a 5% probability, and the data processed for (X + 1) (0.5). All means of crops tested were able to produce conidia, but the best results was obtained with the culture medium V8-20% (5.7 x 10(6) conidia/mL) and BDA (3.5 x 10(6) conidia/mL).

Meloidogyne mayaguensis is the plant-parasitic nematode responsible for a great impact on guava production in Brazil, since it has been the cause for eradication of thousand of hectares of guava plantations in the Northeastern region. In the present study, this species was detected in soybean fields of Ituverava municipality, São Paulo state, and in different vegetable crops (lettuce, cucumber, pepper and cherry tomato) in Chapada dos Guimaraes municipality, Mato Grosso state, causing root galls and other symptoms. The species was identified on the basis of the perineal pattern of females, and on the morphology and morphometry of anterior region of males. Isozyme phenotype for esterase was used for confirmation. This constitutes the first report on the occurrence of M. mayaguensis on lettuce, cucumber, pepper and cherry tomato cultures in Mato Grosso state and the first one in soybean in São Paulo state. It was found that morphological features of male anterior region and female perineal pattern are enough for the safe distinction between M. mayaguensis and M. incognita.

The objective of this research was to evaluate the residue from cassava, known as manipueira, as an alternative for tomato (Lycopersicon esculentum) fertilization. This experiment was carried out in UNESP with the cultivar Rio Grande. The experimental design was a completely randomized blocks in a 3 × 3 factorial arrangement with five blocks. Treatments constituted the following: 1) witness; 2) 54 m3/ha of manipueira; 3) 108 m3/ha of manipueira; 4) half recommended mineral fertilization; 5) half recommended mineral fertilization + 54 m3/ha of manipueira; 6) half recommended mineral fertilization + 108 m3/ha of manipueira; 7) recommended mineral fertilization; 8) recommended mineral fertilization + 54 m3/ha of manipueira and 9) recommended mineral fertilization + 108 m3/ha of manipueira. Tomato yield, number of fruits per plant, fruit diameter and lenght, total soluble solids (TSS), total titratable acidity (TTA) and the ratio TSS/TTA were evaluated. The results obtained indicated that manipueira contributed significantly to yield and quality of pos-harvested tomato fruits.

To determine lycopene uptake and tissue distribution in ferrets (Mustela putorius furo) and F344 rats, we supplemented orally 4.6 mg/(kg body wt-d) lycopene in a tomato oleoresin-com oil mixture (experimental groups). After 9 wk of supplementation, the animals were killed and blood and organs were collected. Plasma and tissue carotenoids were extracted and measured using HPLC. Mean concentrations of lycopene (nmol/kg wet tissue) in saponified tissues of ferrets were as follows: liver 933, intestine 73, prostate 12.7 and stomach 9.3. Levels of lycopene (nmol/kg wet tissue) in saponified tissue of rats were as follows: liver 14213, intestine 3125, stomach 78.6, prostate 24 and testis 3.9. When these organs were extracted without saponification, the lycopene levels were lower, except for rat testis. All-translycopene was the predominant isomer found in tomato oleoresin and in the majority of rat tissues, whereas cislycopenes were predominant in rat prostate and plasma. This pattern was reversed in ferrets. The results show the following: 1) lycopene from tomato oleoresin is absorbed and stored primarily in the liver of both animals; 2) saponification generally improves the extraction of lycopene from most tissues of both animals; 3) cis-lycopene and all- translycopene are the predominant isomers in ferret and rat tissues, respectively; and 4) rats absorb lycopene more effectively than ferrets.

A new strategy for minimization of Cu2+ and Pb2+ interferences on the spectrophotometric determination of Cd2+ by the Malachite green (MG)-iodide reaction using electrolytic deposition of interfering species and solid phase extraction of Cd2+ in flow system is proposed. The electrolytic cell comprises two coiled Pt electrodes concentrically assembled. When the sample solution is electrolyzed in a mixed solution containing 5% (v/v) HNO3, 0.1% (v/v) H2SO4 and 0.5 M NaCl, Cu2+ is deposited as Cu on the cathode, Pb2+ is deposited as PbO2 on the anode while Cd2+ is kept in solution. After electrolysis, the remaining solution passes through an AG1-X8 resin (chloride form) packed minicolumn in which Cd2+ is extracted as CdCl4/2-. Electrolyte compositions, flow rates, timing, applied current, and electrolysis time was investigated. With 60 s electrolysis time, 0.25 A applied current, Pb2+ and Cu2+ levels up to 50 and 250 mg 1-1, respectively, can be tolerated without interference. For 90 s resin loading time, a linear relationship between absorbance and analyte concentration in the 5.00-50.0 μg Cd 1-1 range (r2 = 0.9996) is obtained. A throughput of 20 samples per h is achieved, corresponding to about 0.7 mg MG and 500 mg KI and 5 ml sample consumed per determination. The detection limit is 0.23 μg Cd 1-1. The accuracy was checked for cadmium determination in standard reference materials, vegetables and tap water. Results were in agreement with certified values of standard reference materials and with those obtained by graphite furnace atomic absorption spectrometry at 95% confidence level. The R.S.D. for plant digests and water containing 13.0 μg Cd 1-1 was 3.85% (n = 12). The recoveries of analyte spikes added to the water and vegetable samples ranged from 94 to 104%. The aim of the present work was to determine the oviposition preference of the silverleaf whitefly on tomato (Lycopersicon esculentum). Preferred plant age and leaflet position on the plant were studied and the number of adults per plant necessary for selecting hosts was determined. Bruna VFN hybrid plants at different ages (30-, 45- and 60-days old) were offered to adult whiteflies in free choice test and 30-day old plants in no-choice test. Leaves were collected from upper and lower part of the plant; the leaflets were divided into three categories: basis (those near the steam of plant), middle and apex (terminal leaflet). The 30-day old plants were more preferred for oviposition in free choice test. In the upper part of the plant, whitefly adults preferred to oviposit on the leaflets at the apex and the middle of the leaves. However, in the lower part of the plant, the basal leaflets were preferred in relation to middle and apex. The densities of 100 or 150 adults per tomato plant were considered adequate for genotypes screening, using no-choice test.

The aim of present work was to study, through means and variances of generations, the genetic control of fruit-set percentage in tomatoes, in a biparental cross between line Jab-95 (resistant to fruit-set) and 'Caribe' cultivar (susceptible to fruit-set) after being subjected to high temperatures. F1, F2, and backcross generations for both parents (P 1, P2) were produced in a greenhouse at FCAVUNESP, Jaboticabal-SP, Brazil. A randomized complete block experimental design with four replications was used to evaluate the different generations. Estimates of genetic parameters were obtained according to Mather and Jinks (1982). An additive-dominant model was fitted to explain the variation observed. The mean degree of dominance (0.54), indicated incomplete dominance in the direction of increasing fruit-set percentage at high temperatures. The genetic control of fruit-set percentage, at high temperatures, for tomato was found to be oligogenic or polygenic (3.54 genes). The heritability in a limited sense was 83.91%, suggesting that the selection of individuals based on the characteristic evaluated can be efficient.

The construction of ecological life tables is a useful tool to understand the population dynamics of insects in the field. In this work, ecological life tables were developed for Phyllocnistis citrella (Stainton) in an orchard of Citrus sinensis situated in Santa Rita do Passa Quatro, Sao Paulo State, during 2002. The occurrence of generation overlay in the field was avoided by obtaining the eggs from infestation on young citrus plants (Citrus limonia) kept under controlled conditions. The samples were taken every other day and the number of live, dead, predated and parasitized individuals were recorded. The occurrence of arthropod predators was determined through visual search on the plants of the orchard. The key factors of mortality acting on P.citrella populations were: pupae parasitism caused by Ageniaspis citricola (Logvinovskaya), action of uncertain factors on prepupae and predation of 3rd-instar larvae. The most abundant arthropod predators belonged to the families Formicidae, Chrysopidae, Coccinellidae, Reduviidae e Araneae. In two of five generations of P. citrella, approximately 4% of A. citricola pupae were hyperparasitized by Galeopsomyia fausta (LaSalle) (Hymenoptera: Eulophidae).

The aim of this paper was to apply a multiresidue method using Supercritical Fluid Extraction (SFE) and capillary gas chromatography with electron capture and mass spectrometry detections in the analysis of the levels of pesticide residues in fruits and vegetables. Single laboratory validation of both solid-liquid and supercritical fluid extraction methods was carried out for 32 compounds selected from four pesticide classes (organochlorine, organonitrogen, organophosphorus and pyretroid) in blank and fortified samples of fresh lettuce, potato, apple and tomato. Recoveries for the majority of pesticides from fortified samples at fortification level of 0.04-0.10 mg kg -1 ranged 74-96% for both methods and confirmation of pesticide identity was performed by gas-chromatography-mass spectrometry in a selected-ion monitoring mode. Both methods showed good limits of detection (less 0.01 mg kg-1, depending on the pesticide and matrix) and the SFE method minimized environmental concerns, time, and laboratory work. ©2005 Sociedade Brasileira de Química.
