Common bean (Phaseolus vulgaris L.) is a leguminous in high demand for human nutrition and a very important agricultural product. Production of common bean is constrained by environmental stresses such as drought. Although conventional plant selection has been used to increase production yield and stress tolerance, drought tolerance selection based on phenotype is complicated by associated physiological, anatomical, cellular, biochemical, and molecular changes. These changes are modulated by differential gene expression. A common method to identify genes associated with phenotypes of interest is the characterization of Single Nucleotide Polymorphims (SNPs) to link them to specific functions. In this work, we selected two drought-tolerant parental lines from Mesoamerica, Pinto Villa, and Pinto Saltillo. The parental lines were used to generate a population of 282 families (F3:5) and characterized by 169 SNPs. We associated the segregation of the molecular markers in our population with phenotypes including flowering time, physiological maturity, reproductive period, plant, seed and total biomass, reuse index, seed yield, weight of 100 seeds, and harvest index in three cultivation cycles. We observed 83 SNPs with significant association (p < 0.0003 after Bonferroni correction) with our quantified phenotypes. Phenotypes most associated were days to flowering and seed biomass with 58 and 44 associated SNPs, respectively. Thirty-seven out of the 83 SNPs were annotated to a gene with a potential function related to drought tolerance or relevant molecular/biochemical functions. Some SNPs such as SNP28 and SNP128 are related to starch biosynthesis, a common osmotic protector; and SNP18 is related to proline biosynthesis, another well-known osmotic protector.

Purpose: To investigate the anti-hyperprolactinemic activity of Prunella vulgaris L. extract (PVE) in vivo and in vitro. Methods: Rats were given intraperitoneal (i. p.) metoclopramide (MCP, 150 mg/kg daily) for 10 days to prepare hyperprolactinemia (hyperPRL) model. Bromocriptine was used as positive control drug. High (5.6 g/kg), medium (2.8 g/kg) and low (1.4 g/kg) doses of PVE were administered to hyperPRL rats. The effect of PVE on serum prolactin (PRL), estradiol (E2), progesterone (PGN), follicle stimulating hormone (FSH) and luteinizing hormone (LH) levels were investigated in the rats. MMQ cells derived from rat pituitary adenoma cells and GH3 cells from rat pituitary lactotropictumoral cells were used for in vitro experiments. The effect of PVE on PRL secretion were studied in MMQ cells and GH3 cells respectively. Results: Compared with the control group (446.21 ± 32.43 pg/mL), high (219.23 ± 10.62 pg/mL) and medium (245.47 ± 13.52 pg/mL) reduced PRL level of hyperPRL rats significantly (p 0.05). In MMQ cells, treatment with 5 mg/mL PVE or 10 mg/mL PVE) significantly suppressed PRL secretion and synthesis at 24h compared with controls (p < 0.01). Consistent with D2- action, PVE did not affect PRL in rat pituitary lactotropic tumor-derived GH3 cells that lack the D2 receptor expression, compared with controls. Conclusion: PVE showed anti-hyperPRL activity and can potentially be used for the treatment of hyperprolactinemi, but further studies are required to ascertain this

The stomach contents of Diplodus vulgaris and Spondyliosoma cantharus were: analysed using three simple methods (numeric, gravimetric and frequency of occurrence) and a composite index (I.R.I - Index of Relative Importance). To compare the species, the Schoener index was used. The diet of D. vulgaris consisted mainly of ophiuroids, polychaetes, amphipods and echinoids, while polychaetes, amphipods and hydrozoans dominated in the case of S. cantharus. There were some size-related differences in S. cantharus feeding. Diet overlap was relatively slight, with significant differences in feeding between the two species, notably in terms of greater consumption of echinoderms by D. vulgaris and hydrozoans by S. cantharus. As is the case for the majority of sea breams, D. vulgaris and S. cantharus are characterised by a diverse diet in terms of prey reflecting available prey items in their environment.

Acoustic telemetry and standard tag-recapture were used to determine the home range and residency of juveniles and sub-adults of Diplodus sargus and Diplodus vulgaris in the Ria Formosa (Portugal) coastal lagoon. Maximum time between recaptures for the standard tag-recapture method was 128 days for D. sargus and 30 days for D. vulgaris. The majority of the fish were recaptured in the vicinity of the tagging location. Fish tagged with acoustic transmitters had a maximum period of time between first and last detections of 62 days for D. sargus and 260 days for D. vulgaris. Minimum convex polygons areas ranged between 148 024 m(2) and 525 930 m(2) for D. sargus and between 23 786 m(2) and 42134 m(2) for D. vulgaris. Both species presented a high residency index between first and last detections. Two D. sargus tagged with acoustic tags were recaptured by fishermen outside the coastal lagoon at distances of 12 km and 90 km from the tagging position, providing evidence that this species leaves the Ria Formosa during the winter time for the adjacent coastal waters. The results of this study reinforce the importance of Ria Formosa as a nursery for D. sargus and D. vulgaris in the south coast of Portugal. (C) 2009 Elsevier Ltd. All rights reserved.

The common two-banded sea bream (Diplodus vulgaris) is an important fish in the marine ecosystems of the NW Atlantic and Mediterranean. In southern Portugal it is a major fishery resource being targeted mainly by the artisanal fleets. Although there is some knowledge of the age, growth and reproductive biology of the species, information about its population structure is scarce and somewhat limited to the Mediterranean Sea. In this study the otolith elemental signatures of 90 specimens of D. vulgaris of the same age group (2+ years) and cohort collected from the important fishery regions of SW Portugal (Sesimbra, Sagres and Faro) have been analysed by inductively coupled plasma mass spectrometry (ICP-MS). Two different methodologies have been applied: solution based analysis of the whole otoliths; representative of the entire life-history prior to capture, and laser ablation analysis of otolith cores; representative of the larval and early post-settlement phase. Whole otolith comparisons utilised Sr/Ca, Ba/Ca, Mn/Ca, Li/Ca and Ni/Ca to demonstrate regional population structure. Classification accuracy rates from linear discriminant function analyses (LDFA) of whole otolith chemistry data were high for each region; Faro - 93%, Sagres - 90% and Sesimbra - 80%. Comparison of the otolith core chemistry utilised Sr/Ca, Ba/Ca, Mn/Ca and Mg/Ca and Zn/Ca. LDFA for the otolith core chemistry also achieved accurate classification for samples from Sesimbra (73%), but there was high overlap of otolith chemistry between samples from Faro and Sagres (47 and 43% classification accuracy respectively). The whole otolith results suggest that D. vulgaris are resident in the regional fishing areas during the juvenile phase. Both the core and whole otolith chemistry data supported separation of the Sesimbra fishery region from the more southern and closely associated Faro and Sagres regions for management purposes. However, while the whole otolith data indicated that the populations at Faro and Sagres likely remained separated in the juvenile stage, the otolith core chemistry data was inconclusive as to whether recruitment to these two areas was derived, or not, from different spawning areas.

The study of the reproduction of Diplodus vulgaris (Geoff.) as part of a base-line study of the fishery resources of the southwest coast of Portugal, was based on the analysis of the spawning season, gonad maturation, size at maturity, fecundity and hermaphroditism. The spawning season is relatively long, from December to March, with peaks in January and February. No significant differences were found either in the sex ratio (M:F = 1.01) over the year or by size. The size at first maturity (L-50) for all sexes and undetermined individuals combined was 18.33 cm total length (TL), with no significant differences between males and females. The estimated L-50 is considerably greater than the minimum legal size in Portugal of 15.0 cm. Mean absolute fecundity (F-a) was 131 127 oocytes, ranging from 31 523 to 250 608. The relationship between absolute fecundity and total length (TL) (F-a = 25 398 TL-484 426) and somatic weight (SW) (F-a = 878.8SW-71 416) was of the linear type. The mean number of oocytes per gram of somatic weight was 526, ranging from 194 to 887. The reproductive strategy of this species is characterized by a rudimentary hermaphroditism with possible protandry, as evidenced by the existence of individuals in sexual transition.

The feeding habits of two major species of sole, the common sole Solea vulgaris Quensel, 1806 and the Senegalese sole Solea senegalensis Kaup, 1858 were studied in the lower estuary of the Guadiana River (Algarve, southern Portugal). An evaluation of the number, weight, and feeding coefficient of prey types showed that S. vulgaris feed on a limited variety of prey (only Polychaeta and Tanaidacea) and present low-intensity feeding activity, with small differences in diet between seasons. S. senegalensis also have a low-diversity diet (with only one more taxa, Amphipoda), but exhibit more intense feeding activity which varies seasonally, although with little seasonal variation in the relative importance of the main preys. The diet composition of these two species suggests feeding specialization.

The super early genotypes (SEG) of dry bean (Phaseolus vulgaris L.) have a shorter life cycle (65-75 days) when compared with the season length of traditional cultivars (90-100 days). Timing of nitrogen top-dressing fertilization could be different because of this reduction in length of the SEG life cycle. This study aimed at characterizing, by using growth analysis and vegetation index, super early genotypes of dry bean development as affected by timing of nitrogen application. Field experiments were conducted in the 2014 and 2015 growing seasons in central Brazil with a randomized block experimental design with split plots scheme and four replicates. The plots comprised the dry bean genotypes (Colibri ? check cultivar, CNFC 15873, CNFC 15874, and CNFC 15875), and subplots comprised applications of N at different timings: 90 kg of N at sowing, 90 kg N at top-dressing; 45 kg of N at sowing plus 45 kg at top-dressing, with urea as the source of N. We also used a control treatment without N application. The CNFC 15874 super early genotype of dry bean had the higher grain yield (2776 kg ha-1) and differed from the CNFC 15873 genotype (2492 kg ha-1). Nitrogen fertilization allowed higher grain yield (2619 kg ha-1, when applied N at sowing, 2605 kg ha-1, when applied N at sowing and at top-dressing, and 2680 kg ha-1, when applied N at top-dressing) than the control, 2360 kg ha-1 (no N fertilization). The time of N fertilization in super early genotype of dry bean did not affect grain yield.

Octopus vulgaris, il polpo comune, è un cefalopode in grado di esprimere specifici comportamenti o body pattern, costituiti da componenti locomotorie, cromatiche, posturali e tessiturali. A seconda di come si combinano tra di loro tali componenti, i comportamenti generati possono assumere diversi significati, spaziando da situazioni di riposo e mimetismo a quelle di interazioni sociali e conflittualità intra-inter specifica. I polpi sono influenzati a livello visivo e tattile dal loro ambiente, pertanto l’arricchimento ambientale può influenzare il comportamento di questo invertebrato. Maggiore è la varietà e complessità dell’habitat maggiore sarà il repertorio comportamentale manifestato. Diversi studi hanno dimostrato l’importanza e l’efficacia dell’arricchimento ambientale sul benessere delle specie ittiche, ma ancora poche sono le conoscenze su come rispondano gli invertebrati. Nel presente lavoro, 12 esemplari sono stati suddivisi in due vasche distinte con differenti condizioni ambientali: un ambiente arricchito (ENR) e un ambiente spoglio (POR). La prova, durata 30 giorni, ha raccolto osservazioni comportamentali a livello quantitativo dei polpi, sottoposti nei due diversi ambienti. I risultati hanno dimostrato come l’ambiente influenzi significativamente il comportamento dei polpi, in quanto gli esemplari in ambiente spoglio manifestavano un numero inferiore di body pattern, riconducibili a situazioni di stress, aggressività e scarsa interazione sociale; inoltre erano spesso impauriti, permanevano per lungo tempo all’interno della tana ed erano inappetenti. Al contrario, i polpi in ambiente arricchito hanno espresso un vasto repertorio comportamentale, interagendo molto con l’habitat circostante e conspecifici e dimostrando una elevata capacità di variare rapidamente body pattern in relazione ai vari stimoli. Inoltre, l’accrescimento è stato maggiore nei polpi in ambiente arricchito.

The Fourier transform-infrared (FT-IR) signature of dry samples of DNA and DNA-polypeptide complexes, as studied by IR microspectroscopy using a diamond attenuated total reflection (ATR) objective, has revealed important discriminatory characteristics relative to the PO2(-) vibrational stretchings. However, DNA IR marks that provide information on the sample's richness in hydrogen bonds have not been resolved in the spectral profiles obtained with this objective. Here we investigated the performance of an all reflecting objective (ARO) for analysis of the FT-IR signal of hydrogen bonds in DNA samples differing in base richness types (salmon testis vs calf thymus). The results obtained using the ARO indicate prominent band peaks at the spectral region representative of the vibration of nitrogenous base hydrogen bonds and of NH and NH2 groups. The band areas at this spectral region differ in agreement with the DNA base richness type when using the ARO. A peak assigned to adenine was more evident in the AT-rich salmon DNA using either the ARO or the ATR objective. It is concluded that, for the discrimination of DNA IR hydrogen bond vibrations associated with varying base type proportions, the use of an ARO is recommended.

Kocuria rosea belongs to genus Kocuria (Micrococcaceae family, suborder Micrococcineae, order Actinomycetales) that includes about 11 species of bacteria. Usually, Kocuria sp are commensal organisms that colonize oropharynx, skin and mucous membrane; Kocuria sp infections have been described in the last decade commonly affecting immunocompromised patients, using intravenous catheter or peritoneal dialysis. These patients had mainly bacteremia/recurrent sepsis. We hereby describe the case of a 10-year-old girl, immunocompetent, who had endocarditis/sepsis by K. rosea which was identified in five different blood cultures by Vitek 2 ID-GPC card (BioMérieux, France). Negative HIV serology, blood count within normal range of leukocytes/neutrophils and lymphocytes, normal fractions of the complement, normal level of immunoglobulins for the age; lymphocyte immunophenotyping was also within the expected values. Thymus image was normal at chest MRI. No catheters were required. Identification of K. rosea was essential to this case, allowing the differentiation of coagulase-negative staphylococci and use of an effective antibiotic treatment. Careful laboratory analysis of Gram-positive blood-born infections may reveal more cases of Kocuria sp infections in immunocompetent patients, which may collaborate for a better understanding, prevention and early treatment of these infections in pediatrics.

Objetivou-se avaliar os efeitos da aplicação de calcário e boro nos atributos químicos de um Latossolo, estado nutricional e produtividade da cultura do feijoeiro. As doses de calcário empregadas foram crescentes, considerando-se a saturação por bases igual a 70%, correspondendo às doses: zero; 0,6; 1,2 (v = 70%); 1,8 e 2,4 t ha-1. O calcário utilizado apresentava poder relativo de neutralização total (PRNT), poder de neutralização (PN) e reatividade (RE) de 131; 137 e 95%, respectivamente. As doses de boro estudadas foram iguais a 0 (zero); 0,6; 1,2; 1,8; e 2,4 mg dm-3 de B, sendo a referência para definição das mesmas a de 1,2 mg dm-3 de B (ou 1,2 kg ha-1 de B). Utilizou-se como fonte o ácido bórico (H3BO3) p.a. (17% B). Na época do florescimento foi avaliado o estado nutricional das plantas e, aos 90 dias foram avaliadas a fertilidade do solo e a produção de grãos. No solo houve melhoria dos atributos pH, SB, V e na concentração de Ca e Mg, e do B com o emprego da adubação boratada. A interação da calagem com a adubação boratada promoveu maior acúmulo de Ca, Mg e B nas plantas. Os dados apresentados demonstram que o feijoeiro foi responsivo à calagem e à adubação boratada, tendo atingido bons índices de produção com a utilização de 1,8 kg ha-1 de B com doses crescentes de calcário.
